ABSTRACT Ascocarps of Phaeosphaeria nodorum, which causes Stagonospora nodorum blotch (SNB) of wheat, have not been found by others in the eastern United States despite extensive searches. We sampled tissues from living wheat plants or wheat debris in Kinston, NC, each month except June from May to October 2003. Additional wheat samples were gathered in Kinston, Salisbury, and Plymouth, NC, in 2004 and 2005. For the 3 years, in all, 2,781 fruiting bodies were dissected from the wheat tissues and examined microscopically. Fruiting bodies were tallied as P. nodorum pycnidia or ascocarps, "unknown" (not containing spores, potentially P. nodorum or other fungi), or "other fungi." In the 2003 sample, asco-carps of P. nodorum were present each month after May at a frequency of 0.8 to 5.4%, and comprised a significantly higher percentage of fruiting bodies from wheat spikes than of those from lower stems and leaves. Ascocarps also were found at frequencies <10% in some wheat debris samples from 2004 and 2005. Analysis of the nucleotide sequences of internally transcribed spacer regions of 18 genetically distinct North Carolina isolates from 2003 suggested that all were P. nodorum, not the morphologically similar P. avenaria f. sp. triticea. Neither the 18 isolates from 2003 nor a set of 77 isolates derived from 2004 Kinston leaf samples gave reason to suspect a mating-type imbalance in the larger P. nodorum population (P >/= 0.4). We conclude that, in North Carolina, sexual reproduction plays a role in initiation of SNB epidemics and the creation of adaptively useful genetic variability, although its relative importance in structuring this population is uncertain. 相似文献

The variation obtained in storage fatty acids induced by the procedures of tissue culture and transformation with Agrobacterium tumefaciens was investigated and compared in rapeseed, Brassica napus, cv. Hanna. An increased variation in the fatty acid profiles was noted after tissue culture and transformation compared
with plants derived directly from seeds. In the second generation of rapeseed transformants, T2, the content of oleic acid ranged from 39–72%, 12–31% for linoleic acid and 7–16% for linolenic acid. This could be compared
with the oleic acid content in the T2 generation of tissue culture-derived plants which ranged between 47–76% and in seed-derived material where oleic acid ranged
between 55–69%.In the T3 generation the ranges in transgenic seeds were decreased but still larger than in the seed derived plants. The range in transgenic
plants was 49–64% for oleic acid, 20–28% for linoleic acid and 9–18% for linolenic acid. The most extreme individuals, both
highest and lowest in the common fatty acids, were found in the group of transformed plants independent of generation. The
total lipid content was also affected by the two treatments and seeds with the lowest and highest lipid content were both
found among the transformed plants. In conclusion, care should be taken to use proper controls when performing transformation
experiments in order to distinguish variation in the fatty acid profiles induced by the transformation procedure and tissue
culture treatments from the changes due to transgenic expression.

Amplified fragment length polymorphism (AFLP) analysis was used to study genetic variation among 76 isolates of Verticillium. A dendrogram based on the AFLP data revealed three main groups. One group consisted of 35 European isolates derived from Brassica napus together with five Californian isolates taken from B. oleracea. This group displayed a high degree of genetic similarity and included three isolates earlier classified as Verticillium longisporum, indicating that all isolates in this group probably should be regarded as members of V. longisporum. V. dahliae isolates constituted the second group while the third group contained four V. albo-atrum isolates. In addition to these three groups, a cluster of six V. nigrescens isolates was observed. However, the genetic distances between the isolates of V. nigrescens were much higher than those between members in the other groups and the bootstrap value for the V. nigrescens cluster was subsequently low. Four isolates classified as V. tricorpus were highly diverse and did not cluster together. Analysis of molecular variance revealed that the isolates of V. longisporum were separated into four subgroups, based on geographic origin. Midazolam was administered at 0.3 mg kg−1 IV as pre-medication at least 1 minute prior to induction. Direct arterial blood pressure, arterial blood gases, and heart rate were obtained at baseline, before induction, after induction, and for every 5 minutes afterwards until the dog began to swallow and the trachea was extubated. The dogs were allowed to breathe room air with the endotracheal tube in place.ResultsThe systolic arterial pressure (SAP) was higher in the etomidate group compared with the propofol group after induction. The SAP and mean arterial pressure (MAP) were higher in the etomidate group compared with the propofol group at 5 minutes. The recovery quality and ataxia score were worse in the etomidate group compared with the propofol group. Time from extubation to sternal recumbency and sternal recumbency to standing was longer in the etomidate group compared with the propofol group. The heart rate, PaCO2, and HCO3 were higher in the propofol group compared with the etomidate group after induction. The PaO2 and SaO2 were lower in the propofol group compared with the etomidate group after induction. The SAP and MAP were lower in the propofol group at 5 minutes compared with baseline.Conclusion and clinical relevancePropofol caused a decrease in SAP and MAP which was not observed with etomidate. General anaesthesia was induced and maintained using isoflurane in oxygen with volume-controlled ventilation [tidal volume (VT) (10 mL kg?1), respiratory rate (8–12 breaths minute?1)] to maintain end-tidal carbon dioxide between 35 and 40 mmHg (4.66–5.32 kPa). Regional distribution of ventilation was assessed with EIT (plane of fifth to sixth intercostal space) at 30, 60 and 90 minutes after intubation. The resulting functional images were divided into four regions of interest (ROI, A–D) to determine: 1) the ratio of VT distribution between dependent to nondependent lung (D/ND) and 2) changes in distribution within the lungs in the latero-lateral direction. The centre of ventilation (COV) was calculated. The dimension of the chest in the latero-lateral (W) and dorso-ventral (H) direction was measured at the height of the withers and the W/H ratio was calculated. The influence of time on the D/ND ratio, COV and ROI A–D were analysed with anova followed by post hoc Bonferroni tests. Pearson correlation coefficient and the coefficient of determination (r2) were calculated to evaluate the relationship between the W/H ratio and D/ND ratio, and COV. Statistical significance was set at p < 0.05.Results
The median D/ND ratio (T30–T90) was 0.67 (0.40–1.01) and for COV 31.15% (11.20–45.20), confirming the expected uneven distribution of ventilation. The D/ND ratio had a moderate negative correlation with the W/H ratio (r = ?0.68, r2 = 0.46, p < 0.001), while the COV did not correlate with the W/H ratio (r = ?0.04).Conclusions and clinical relevance
Uneven ventilation in mechanically ventilated right laterally recumbent anaesthetized ponies occurs within 30 minutes and changed little over the following 60 minutes. 相似文献118.

A 15-year-old Haflinger gelding presented with a mass in the left nictitating membrane. Two biopsies and the excised nictitating membrane were taken at different time points as a result of reoccurrence of the mass and submitted for histopathologic evaluation. The horse was euthanized as a result of poor prognosis following the reoccurrence of the mass after surgical removal. Histologically, the mass consisted of dilated, thin-walled vascular clefts and channels, lined by flattened to cuboidal endothelial cells with moderate cellular pleomorphism. There was up to 1 mitotic figure per high power field. The channels were empty or contained few erythrocytes. In the collagen-rich stroma, few lymphocytes, focal follicular lymphoid aggregations, and marked lymphangiectasia were observed. Immunohistochemically, the neoplastic cells stained positive for vimentin and partially positive for factor VIII-related antigen. Ultrastructural analysis revealed discontinuous endothelial lining vascular channels that partially lacked a basal membrane. Based on the histopathologic, immunohistochemical, and ultrastructural features lymphangiosarcoma was diagnosed. 相似文献

Invasive, non-native species are a major threat to global biodiversity. Stoats were introduced from Britain to New Zealand in the 1880s and have since caused grave conservation problems. A histopathological and serological survey of disease and infection in stoats from New Zealand was undertaken to identify agents that might be used or modified to control this population. Of 60 stoats examined, 63% exhibited inflammation of the lung, mostly occurring as local or diffuse interstitial pneumonia, 30% showed signs of inflammatory liver disease and 14% were positive for antibodies reactive with feline calicivirus. In Britain only 11% of 44 stoats exhibited symptoms of pulmonary inflammatory disease, suggesting higher rates of infection or compromise of the pulmonary immune system among invasive stoats, possibly related to genetic founder effects or environmental variation.
