Stand transpiration (E) estimated using the sap flux methods is affected by the azimuthal, radial, and tree-to-tree variations of sap flux. Although several studies have examined the relative importance of the three variations in estimating E, the seasonality of the three variations remains unknown. In the current study, we attempted to clarify whether the relative importance of these three variations could show seasonal changes. Using sap flux data measured in a subtropical cloud forest from August 2010 to July 2011, we calculated the differences resulting from omitting the three variations in estimating E. The effects of the three variations in estimating E showed seasonality. The azimuthal and tree-to-tree variations were more pronounced during winter, whereas the radial variation was more pronounced during summer. However, the effect of tree-to-tree variation was consistently much larger than the other two variations throughout the study period. The tree-to-tree variation is more important in estimating E monthly, seasonally and annually than both the azimuthal and radial variations, although all three variations have shown seasonality. In addition, the sensor allocation for summer would be acceptable for the practical estimation of E if aiming at the long time scale.  相似文献

Spontaneous and induced waxy phenotype, associated with endosperm containing little or no amylose, has been recognized in rice (Oryza sativa L.). Mutation of a dominant gene Wx into a recessive gene wx, which causes the inactivation or absence of granule bound starch synthase, is believed to be responsible for the change in endosperm starch leading to the waxy grain. In the present study, the nucleotide polymorphism in the Wx gene of rice genotype Tainung 67 (wild type) and its 35 NaN3-induced wx mutants were examined. Iodine staining confirmed that all the mutants had waxy grain trait. The G-to-T single base substitution analysis indicated that the wild type genotype Tainung 67 and its waxy mutants carried Wxb allele. Moreover, 23-bp duplication in exon 2 was detected in all the waxy mutants. Microsatellite polymorphism (CT)n was also detectable on the Wx gene of the tested genotypes and mutants, with at least 5 classes of (CT)n microsatellites identified at the Wx locus. Electrophoretic analyses also confirmed the observed nucleotide polymorphsim. Thus, nucleotide polymorphsim exist among NaN3-induced waxy mutants in rice. However, only the 23-bp duplication in exon 2 may be used as a molecular marker to characterize waxy grain trait in rice genotypes.  相似文献

Portulaca oleracea (common purslane) is a summer annual weed with wide geographic distribution and is problematic in many crops worldwide. Field experiments were conducted to determine the effects of different management practices on P. oleracea emergence in soyabean fields. Two tillage systems [conventional tillage (CT) and no‐till (NT)], three soyabean seeding rates (SR) (200 000, 300 000 and 400 000 seeds ha?1) and three imazethapyr doses (0, 50, and 100 g a.i. ha?1 applied pre‐emergence) were considered as experimental factors. Portulaca oleracea emergence was affected by management practices including tillage system, soyabean SR and imazethapyr dose. Conventional tillage required a thermal time (TT) of 195.95 and 221.30 d °C to reach 50% emergence in 2016 and 2017, respectively, while for NT, the respective TT requirements were 182.34 and 203.32 d °C. On increasing soyabean SR from 200 000 to 400 000 seeds ha?1, the TT requirements for 50% emergence (T50) of P. oleracea also increased. The T50 at the herbicide dose of 100 g a.i. ha?1 was 193.05 and 220.67 d °C in 2016 and 2017, respectively, while for the non‐herbicide treatment, the respective TT requirements were 165.98 and 202.94 d °C. From an integrated weed management perspective, a combination of CT with a SR of 400 000 seeds ha?1 and a 100 g a.i. ha?1 imazethapyr dose not only resulted in the lowest P. oleracea seedling density m?2 but also caused the longest delay in the time to reach the T50. Findings from our study may facilitate the development of effective P. oleracea management strategies.  相似文献

Solanum nigrum L. (SN) has been used in traditional folk medicine to treat different cancers. It is also used as a hepatoprotective and anti-inflammatory agent. In this study, we demonstrated that the extract of SN (SNE) induced a strong cytotoxic effect toward HepG2 cells but much less to Chang liver and WRL-68 cells. The mechanisms of the cytotoxic effect were concentration-dependent. High doses of SNE (2 and 5 mg/mL) induced apoptotic cell death in HepG2 cells, as evidenced by increases in the expressions of p-JNK and Bax, mitochodrial release of cytochrome c, and caspase activation. On the other hand, cells treated with low concentrations of SNE (50-1000 microg/mL) revealed morphological and ultrastructural changes of autophagocytic death under electron microscopic observation. Furthermore, these cells showed increased levels of autophagic vacuoles and LC3-I and LC3-II proteins, specific markers of autophagy. The levels of Bcl-2 and Akt that have been implicated in the down-regulation of autophagy were decreased upon SNE treatment. Taken together, these findings indicate that SNE induced cell death in hepatoma cells via two distinct antineoplastic activities of SNE, the ability to induce apoptosis and autophagocytosis, therefore suggesting that it may provide leverage to treat liver cancer.  相似文献

Adult Japanese eels, Anguilla japonica Temminck & Schlegel, (200–250 g, 45–55 cm) were immunized by intramuscular injection with goat IgG. After 5 weeks, eel immunoglobulin (Ig) was purified using affinity chromatography. The purified eel Ig was used to immunize rabbits to produce anti-eel Ig antibody. The highest antibody ELISA value in eels was reached 3 weeks after initial immunization with goat IgG, and then gradually decreased. The antibody could still be detected at 140 days post-immunization. The optimal temperature for antibody production was 30°C. Freund's complete adjuvant and secondary immunization both increased antibody production in eels.  相似文献
