Phytosiderophores (PS) of the mugineic acid family can be separated effectively by HPLC on resin‐based anion exchange columns. Using gradient elution with aqueous NaOH, separation of 2'‐deoxymugineic acid (DMA), mugineic acid (MA), 3‐hydroxymugineic acid (HMA), 3‐epi‐hyydroxymugineic acid (epi‐HMA), and nicotianamine was obtained within 16 min with a complete cycle time of 30 min. Fluorimetric detection was performed after post‐column derivatization using sodium hypochlorite (NaOCl) and orthophtaldialdehyde. External standardization revealed a linear range between 0.1–2.5 nmol of each compound applied to the column, with a detection limit of approximately 0.05 nmol. Only minimal sample pre‐treatment by addition of sodium hydroxide (NaOH) was required prior to HPLC‐injection of natural occurring samples such as root exudates, collected from the whole root system or from single apical root zones, xylem sap, and hot water extracts of root material, obtained from iron (Fe)‐deficient maize and barley plants. The method is discussed in comparison with cation exchange HPLC which is conventionally employed for the separation of phytosiderophores.  相似文献

Glutathione (GSH) and phenolics play an important role in plant defense against metal‐ion toxicity. The antioxidant activity and metal‐binding capacity of these compounds can account for the protective effects. In contrast to animal‐cell models, however, the possible interplay among these substances in stress defense of plants is poorly investigated. This study compares the influence of cadmium (Cd) on the profiles of both soluble phenolics and GSH in shoots of different Thlaspi and Noccaea species: two ecotypes of the nonhyperaccumulator T. arvense differing in Cd resistance (ecotype Aigues Vives, Cd‐sensitive, and ecotype Jena, Cd‐resistant) and two Cd‐tolerant Cd‐Zn hyperaccumulators N. praecox and N. caerulescens (formerly Thlaspi praecox and T. caerulescens). To reveal the possible influence of Cd‐induced sulfur (S) shortage on the stress response, plants receiving normal S concentrations (500 μM MgSO4) and plants treated with surplus S (500 μM MgSO4 + 500 μM K2SO4) were analyzed. Our working hypothesis was that species differences in tolerance to high tissue Cd concentrations should be reflected by differences in endogenous levels of GSH and phenolic compounds. The results reveal clear species‐dependent differences in both the constitutive patterns and the Cd‐ and S‐induced changes in shoot concentrations of GSH and phenolics. However, no simple relationship between these shoot concentrations and Cd accumulation and tolerance can be established.  相似文献

Purpose

Many Mediterranean drylands are characterized by strong erosion in headwater catchments, where connectivity processes play an important role in the redistribution of water and sediments. Sediment connectivity describes the ease with which sediment can move through a catchment. The spatial and temporal characterization of connectivity patterns in a catchment enables the estimation of sediment contribution and transfer paths. Apart from topography, vegetation cover is one of the main factors driving sediment connectivity. This is particularly true for the patchy vegetation cover typical of many dryland environments. Several connectivity measures have been developed in the last few years. At the same time, advances in remote sensing have enabled an improved catchment-wide estimation of ground cover at the subpixel level using hyperspectral imagery.

Materials and methods

The objective of this study was to assess the sediment connectivity for two adjacent subcatchments (~70 km2) of the Isábena River in the Spanish Pyrenees in contrasting seasons using a quantitative connectivity index based on fractional vegetation cover and topography data. The fractional cover of green vegetation, non-photosynthetic vegetation, bare soil and rock were derived by applying a multiple endmember spectral mixture analysis approach to the hyperspectral image data. Sediment connectivity was mapped using the index of connectivity, in which the effect of land cover on runoff and sediment fluxes is expressed by a spatially distributed weighting factor. In this study, the cover and management factor (C factor) of the Revised Universal Soil Loss Equation (RUSLE) was used as a weighting factor. Bi-temporal C factor maps were derived by linking the spatially explicit fractional ground cover and vegetation height obtained from the airborne data to the variables of the RUSLE subfactors.

Results and discussion

The resulting connectivity maps show that areas behave very differently with regard to connectivity, depending on the land cover and on the spatial distribution of vegetation abundances and topographic barriers. Most parts of the catchment show higher connectivity values in August as compared to April. The two subcatchments show a slightly different connectivity behaviour that reflects the different land cover proportions and their spatial configuration.

Conclusions

The connectivity estimation can support a better understanding of processes controlling the redistribution of water and sediments from the hillslopes to the channel network at a scale appropriate for land management. It allows hot spot areas of erosion to be identified and the effects of erosion control measures, as well as different land management scenarios, to be studied.  相似文献

We examined the effect of TiO2 nanoparticles (NPs) on the growth of maize and soybean plants and associated soil microbial communities. Plants were grown in a greenhouse, and low levels of undoped or nitrogen-doped TiO2 NPs were applied. Plant growth and nutrient content were determined, and effects of NPs on composition of soil microbial communities were examined using terminal restriction fragment length polymorphism analysis (TRFLP) of rDNA. We found no significant effects of TiO2 NPs on plant growth, nutrient content, or the composition of bacterial communities within the rhizosphere. However, arbuscular mycorrhizal fungal communities were affected by application of undoped and nitrogen-doped TiO2 NPs. This observation may be partially attributed to the small but significant TiO2 NP uptake levels in the root tissues of both plants. Our results suggest that even low concentrations of TiO2 NPs may influence some important groups of soil microbes, such as mycorrhizal fungi, but changes in the composition of microbial communities may not affect plant growth under conditions of adequate moisture and nutrients.  相似文献

The properties of a white wheat bread could be changed by adding normal or heat‐treated barley flour in small amounts (2 and 4%) to a white wheat bread recipe. Differences regarding gelatinization as well as retrogradation properties were found when analyzing the two flours in model systems. The heat‐treated flour was fully gelatinized due to prior time, temperature, and pressure treatment and could therefore absorb larger amounts of water than the other flours. In gelatinized model systems with 40% flour (dwb), the heat‐treated barley flour contained less retrograded amylopectin as compared with normal barley flour after storage for up to 14 days, whereas no differences were found with 20% flour (dwb). However, stored breads showed an increased retrogradation of amylopectin (as measured by differential scanning calorimetry [DSC]) when 2% pretreated barley flour was added as compared with addition of 2% normal barley flour. On the other hand, there were no significant differences at the 4% level. Addition of either of the barley flours resulted in less firm breads during storage as compared with the control breads. Increased water absorption in barley flour and thus increased water content in the breads or different water‐binding capacities of the flour blends could explain these results. The present study indicated that water had a stronger influence on bread firmness than the retrogradation of amylopectin. This conclusion was based on breads with pretreated barley flour being less firm than breads with normal barley flour, although the retrogradation, as determined by DSC, was higher.  相似文献

Background

The objective of this study was to investigate the association between average daily gain and the number of Lawsonia intracellularis bacteria in faeces of growing pigs with different levels of diarrhoea.

Methods

A longitudinal field study (n = 150 pigs) was performed in a Danish herd from day 29 to 47 post weaning. Every third day all pigs were weighed, subjected to a clinical examination and faecal samples were obtained. Faecal samples were subjected to dry matter determination and absolute quantification by PCR for L. intracellularis and porcine circovirus type 2 (PCV2). Association between average daily gain, faecal dry matter content, numbers of L. intracellularis bacteria and PCV2 genome copies in faeces was investigated in a multilevel mixed-effects linear model.

Results

Increasing numbers of L. intracellularis log10 bacteria/g faeces were significantly associated with decreasing average daily gain (P < 0.001). The association was decreasing with increasing faecal dry matter content (P < 0.01). The number of PCV2 log10 copies/g faeces was not significantly associated with average daily gain of the pigs (P > 0.5).

Conclusion

The results suggest a potential application of a PCR quantifying L. intracellularis in growing pigs. Faecal dry matter content must be taken into consideration in interpretation of such test results.  相似文献

Because of its role in human disease, there are increasing global requirements for reliable diagnostic and control methods for Trichinella in food animals to ensure meat safety and to facilitate trade. Consequently, there is a need for standardization of methods, programs, and best practices used in the control of Trichinella and trichinellosis. This review article describes the biology and epidemiology of Trichinella, and describes recommended test methods as well as modified and optimized procedures that are used in meat inspection programs. The use of ELISA for monitoring animals for infection in various porcine and equine pre- and post-slaughter programs, including farm or herd certification programs is also discussed. A brief review of the effectiveness of meat processing methods, such as freezing, cooking and preserving is provided. The importance of proper quality assurance and its application in all aspects of a Trichinella diagnostic system is emphasized. It includes the use of international quality standards, test validation and standardization, critical control points, laboratory accreditation, certification of analysts and proficiency testing. Also described, are the roles and locations of international and regional reference laboratories for trichinellosis where expert advice and support on research and diagnostics are available.  相似文献

Planting three rows of potatoes in a bed the width of two conventional rows offers an easily managed way to increase seed piece populations, with the potential of increasing tuber yield and enhancing tuber quality. A wide bed production system (3 rows of potatoes planted on a 1.9 m flat-topped raised bed) was compared to a conventional-ridged system (1 row of potatoes in sharply sloped ridges on 96 cm centers) in 1996 and 1997 on a Norfolk sandy loam soil and a Portsmouth fine sandy loam soil in eastern North Carolina. Potato plant stands, leaf area index at approximately 9 WAP, yield, and quality were measured. Soil temperature, soil moisture, and cone index, as a measure of soil penetration resistance, were also measured, wide beds were more moist than conventional ridges early in the season. Cone index was greater throughout the root profile in wide beds in two of three tests. The row on the west side of an individual wide bed was most similar to conventional ridges in daily soil temperature fluctuations between minimum and maximum temperatures, and had greater fluctuations than the middle and eastern rows of the wide bed. Total yield and yield of grade A potatoes were not significantly different between wide beds and conventional ridges at either site. At one site, yield of grade B potatoes was significantly less in the wide bed; among the three rows in the wide bed, the eastern row had significantly lower yield of grade B potatoes. Conventional ridges had a higher percent of green grade A potatoes than the wide beds in one of three trials. Under North Carolina conditions, changing production systems would be unadvisable for most growers because wide beds do not increase yield enough to justify spending the money for more seed and to change equipment.  相似文献
