Grain protein concentration (GPC) affects wheat nutritional value and several critical parameters for bread and pasta quality. A gene designated Gpc-B1, which is not functional in common and durum wheat cultivars, was recently identified in Triticum turgidum ssp. dicoccoides. The functional allele of Gpc-B1 improves nitrogen remobilization from the straw increasing GPC, but also shortens the grain filling period resulting in reduced grain weight in some genetic backgrounds. We developed isogenic lines for the Gpc-B1 introgression in six hexaploid and two tetraploid wheat genotypes to evaluate its effects on bread-making and pasta quality. In common wheat, the functional Gpc-B1 introgression was associated with significantly higher GPC, water absorption, mixing time and loaf volume, whereas in durum wheat, the introgression resulted in significant increases in GPC, wet gluten, mixing time, and spaghetti firmness, as well as a decrease in cooking loss. On the negative side, the functional Gpc-B1 introgression was associated in some varieties with a significant reduction in grain weight, test weight, and flour yield and significant increases in ash concentration. We present an overview of a research platform that provides essential germplasm, genotypic and phenotypic data and analytical tools for dissecting phenotype–genotype associations in rice. These resources include a diversity panel of 400 Oryza sativa and 100 Oryza rufipogon accessions that have been purified by single seed descent, a custom-designed Affymetrix array consisting of 44,100 SNPs, an Illumina GoldenGate assay consisting of 1,536 SNPs, and a suite of low-resolution 384-SNP assays for the Illumina BeadXpress Reader that are designed for applications in breeding, genetics and germplasm management. Our long-term goal is to empower basic research discoveries in rice by linking sequence diversity with physiological, morphological, and agronomic variation. This research platform will also help increase breeding efficiency by providing a database of diversity information that will enable researchers to identify useful DNA polymorphisms in genes and germplasm of interest and convert that information into cost-effective tools for applied plant improvement. 相似文献

The objective of this retrospective study was to evaluate the expression of receptor tyrosine kinases (RTKs) in canine adrenal tumors and correlate this expression with features of tumor aggressiveness and survival in dogs undergoing adrenalectomy.Forty-three canine adrenal tumors were evaluated for expression of c-kit, fms-like tyrosine kinase 3 (flt-3), platelet-derived growth factor receptor-β (PDGFR-β), and vascular endothelial growth factor receptor 2 (VEGFR2) using immunohistochemistry. Tumor RTK staining characteristics were compared to normal adrenals. Medical records were reviewed for data regarding patient outcome and tumor characteristics. Expression of c-kit, flt-3, PDGFR-β, and VEGFR2 was detected in 26.9%, 92.3%, 96.2%, and 61.5% of cortical tumors and 0%, 63.2%, 47.4%, and 15.8% of pheochromocytomas, respectively. Expression of RTKs was not significantly increased when compared to normal adrenals and did not correlate with survival after adrenalectomy. Receptor tyrosine kinases are not overexpressed in canine adrenal tumors compared to normal adrenal tissue. Therapeutic inhibition of these receptors may still represent an effective approach in cases where receptor activation is present. 相似文献

Holm oak is the hardwood most used for reforestation in Mediterranean Spain, which makes the development of stock quality
standards in order to improve establishment success, a priority. However, its nursery culture is characterized by a wide range
of practices resulting in stock heterogeneity and a potentially varied outplanting performance. Previous research has focused
on specific seedling quality attributes, obviating the integral effect of nursery culture on overall quality. We studied growing
regime, seedling quality, and field performance in nine holm oak stocklots produced in commercial nurseries during two consecutive
growing years. Results proved variations in field performance were related to stocklot quality and, hence, to the growing
regime practised. This dependence on stock quality may vary with planting site weather: in the drier year, survival was related
to attributes like height, water status and K concentration, while, in the second, milder year, only growth performance was
related to nutrient concentrations, plant size and water status. Results indicated the following quality standards for height:
12–17 cm, diameter: 3.5–4.8 mm, shoot and root weights: 1.3–1.6 and 2.8–4.7 g, respectively, N–P–K foliar concentrations:
over 10–0.9–3.7 mg g−1, respectively and in water status parameters: EMX < 5 MPa and SWDTL > 15%. These attributes can be adjusted using nursery cultural practices in order to meet seedling quality standards for

Live aboveground biomass (AGB) is an important source of uncertainty in the carbon balance from the tropical regions in part due scarcity of reliable estimates of live AGB and its variation across landscapes and forest types. Studies of forest structure and biomass stocks of Neotropical forests are biased toward Amazonian and Central American sites. In particular, standardized estimates of aboveground biomass stocks for the Brazilian Atlantic forest are rarely available. Notwithstanding the role of environmental variables that control the distribution and abundance of biomass in tropical lowland forests has been the subject of considerable research, the effect of short, steep elevational gradients on tropical forest structure and carbon dynamics is not well known. In order to evaluate forest structure and live AGB variation along an elevational gradient (0–1100 m a.s.l.) of coastal Atlantic Forest in SE Brazil, we carried out a standard census of woody stems ≥4.8 cm dbh in 13 1-ha permanent plots established on four different sites in 2006–2007. Live AGB ranged from 166.3 Mg ha−1 (bootstrapped 95% CI: 144.4,187.0) to 283.2 Mg ha−1 (bootstrapped 95% CI: 253.0,325.2) and increased with elevation. We found that local-scale topographic variation associated with elevation influences the distribution of trees >50 cm dbh and total live AGB. Across all elevations, we found more stems (64–75%) with limited crown illumination but the largest proportion of the live AGB (68–85%) was stored in stems with highly illuminated or fully exposed crowns. Topography, disturbance and associated changes in light and nutrient supply probably control biomass distribution along this short but representative elevational gradient. Our findings also showed that intact Atlantic forest sites stored substantial amounts of carbon aboveground. The live tree AGB of the stands was found to be lower than Central Amazonian forests, but within the range of Neotropical forests, in particular when compared to Central American forests. Our comparative data suggests that differences in live tree AGB among Neotropical forests are probably related to the heterogeneous distribution of large and medium-sized diameter trees within forests and how the live biomass is partitioned among those size classes, in accordance with general trends found by previous studies. In addition, the elevational variation in live AGB stocks suggests a large spatial variability over coastal Atlantic forests in Brazil, clearly indicating that it is important to consider regional differences in biomass stocks for evaluating the role of this threatened tropical biome in the global carbon cycle. 相似文献

An accurate characterization of tree carbon (TC), forest floor carbon (FFC) and soil organic carbon (SOC) in tropical forest plantations is important to estimate their contribution to global carbon stocks. This information, however, is poor and fragmented. Carbon contents were assessed in patula pine (Pinus patula) and teak (Tectona grandis) stands in tropical forest plantations of different development stages in combination with inventory assessments and soil survey information. Growth models were used to associate TOC to tree normal diameter (D) with average basal area and total tree height (HT), with D and HT parameters that can be used in 6–26 years old patula pine and teak in commercial tropical forests as indicators of carbon stocks. The information was obtained from individual trees in different development stages in 54 patula pine plots and 42 teak plots. The obtained TC was 99.6 Mg ha−1 in patula pine and 85.7 Mg ha−1 in teak forests. FFC was 2.3 and 1.2 Mg ha−1, SOC in the surface layer (0–25 cm) was 92.6 and 35.8 Mg ha−1, 76.1 and 19 Mg ha−1 in deep layers (25–50 cm) in patula pine and teak, respectively. Carbon storage in trees was similar between patula pine and teak plantations, but patula pine had higher levels of forest floor carbon and soil organic carbon. Carbon storage in trees represents 37 and 60% of the total carbon content in patula pine and teak plantations, respectively. Even so, the remaining percentage corresponds to SOC, whereas FFC content is less than 1%. In summary, differences in carbon stocks between patula pine and teak trees were not significant, but the distribution of carbon differed between the plantation types. The low FFC does not explain the SOC stocks; however, current variability of SOC stocks could be related to variation in land use history. 相似文献

The chemical composition of oleo-gum-resin from Ferula gummosa collected in the northern part of Iran has been studied. The fraction of oleo-gum-resin soluble in ethanol (ca 67 wt.%) is composed by three major fractions: (i) monoterpenes and monoterpenoids (ca 15 wt.% fraction), (ii) sesquiterpenes and sesquiterpenoids (ca 30 wt.%) and (iii) triterpenes and triterpenoids (ca 55 wt.%). The major families of terpenes and terpenoids were identified employing gas chromatography coupled with mass spectrometry detector (GC-MS). Almost 25 wt.% of oleo-gum-resin was insoluble in ethanol and, according to wet chemistry analyses, assigned to arabinogalactane structurally associated with protein complex (AGP). This arabinogalactan possessed the molecular weight of ca 30 kDa, as revealed by size exclusion chromatography, and the main backbone was constituted by β-(1 → 3)/β-(1 → 3,6)-linked d-galactopyranosyl residues ramified predominantly by terminal α-l-arabinofuranosyl and β-d-glucuronopyranosyl residues, as assessed by 1D/2D 1H NMR. 相似文献
