Background
Doubled haploidy is a fundamental tool in plant breeding as it provides the fastest way to generate populations of meiotic recombinants in a genetically fixed state. A wide range of methods has been developed to produce doubled haploid (DH) plants and recent advances promise efficient DH production in otherwise recalcitrant species. Since the cellular origin of the plants produced is not always certain, rapid screening techniques are needed to validate that the produced individuals are indeed homozygous and genetically distinct from each other. Ideal methods are easily implemented across species and in crops where whole genome sequence and marker resources are limited.Results
We have adapted enzymatic mismatch cleavage techniques commonly used for TILLING (Targeting Induced Local Lesions IN Genomes) for the evaluation of heterozygosity in parental, F1 and putative DH plants. We used barley as a model crop and tested 26 amplicons previously developed for TILLING. Experiments were performed using self-extracted single-strand-specific nuclease and standard native agarose gels. Eleven of the twenty-six tested primers allowed unambiguous assignment of heterozygosity in material from F1 crosses and loss of heterozygosity in the DH plants. Through parallel testing of previously developed Simple Sequence Repeat (SSR) markers, we show that 3/32 SSR markers were suitable for screening. This suggests that enzymatic mismatch cleavage approaches can be more efficient than SSR based screening, even in species with well-developed markers.Conclusions
Enzymatic mismatch cleavage has been applied for mutation discovery in many plant species, including those with little or no available genomic DNA sequence information. Here, we show that the same methods provide an efficient system to screen for the production of DH material without the need of specialized equipment. This gene target based approach further allows discovery of novel nucleotide polymorphisms in candidate genes in the parental lines.232.

- This study aimed to develop an integrated analytical framework to identify candidate sites for surface water protection that is applicable at broad scales and in data scarce regions, using Zambia as a case study.
- In the Zambian Water Resources Management Act of 2011, Water Resource Protection Areas are defined as areas where special measures are necessary for the protection of a catchment, sub-catchment, aquifer, or geographical area. Three specific selection criteria are listed for the definition of Water Resource Protection Areas: (i) areas of high importance in providing water to users in a catchment; (ii) aquatic areas of high ecological importance; and (iii) areas that are particularly sensitive to human impact.
- In this project, each sub-catchment and river reach of Zambia was characterized for their importance regarding these three criteria. ‘Water provisioning’ was assessed by analysing patterns of runoff generation and human water use; ‘aquatic ecological importance’ was determined by conducting a freshwater biodiversity and ecosystem assessment using a systematic conservation planning approach; and ‘sensitive areas’ were identified by quantifying erosion potential and sediment transport. The work was supported by an assessment of free-flowing rivers in Zambia, i.e., those rivers where aquatic ecosystem functions and services are largely unaffected by changes to fluvial connectivity through dams and other infrastructure.
- Highly ranked sub-catchments were found in the Liuwa, Barotse, and Bangweulu floodplains and wetlands, and in the headwater regions of the upper Zambezi, Kafue, Chambeshi/Luapula, and Tanganyika catchments. The Luangwa was identified as the highest ranked candidate river for protection within Zambia.
- The resulting maps, data, and methods are intended to support national-scale efforts to prioritize areas for surface water protection, identify catchments and rivers with high conservation value, optimize decision making for infrastructure development, and inform concerted strategies to maintain and restore freshwater ecosystem services in Zambia.

Recent investigations on friction-welded wood-to-wood connections have shown interesting capacities of these adhesive-free joints for further development towards constructional elements. This paper addresses challenges and technical requirements for the enhancement of this technology from small specimens to samples of structural scale. Inhomogeneities in bonding quality and joint strength, herein referred to as scale effects, become relevant with an increase in size of the welded interface. It is assumed that the water vapour and smoke that evolves during the welding process by evaporation of moisture within the cell structure is a reason for these inhomogeneities. In order to achieve a better understanding of these effects, the influence of the water vapour on welded spruce (Picea abies) boards was investigated. It was shown that the vapour increases the internal gas pressure within the welding zone. Thus, it strongly influences the welding process and the quality of the joint. The results of these investigations lead to technical solutions, permitting a significant attenuation of the negative effects and improvement in quality and joint resistance. 相似文献

The system of the paranasal sinuses morphologically represents one of the most complex parts of the equine body. A clear understanding of spatial relationships is needed for correct diagnosis and treatment. The purpose of this study was to describe the anatomy and volume of equine paranasal sinuses using three‐dimensional (3D) reformatted renderings of computed tomography (CT) slices. Heads of 18 cadaver horses, aged 2–25 years, were analyzed by the use of separate semi‐automated segmentation of the following bilateral paranasal sinus compartments: rostral maxillary sinus (Sinus maxillaris rostralis), ventral conchal sinus (Sinus conchae ventralis), caudal maxillary sinus (Sinus maxillaris caudalis), dorsal conchal sinus (Sinus conchae dorsalis), frontal sinus (Sinus frontalis), sphenopalatine sinus (Sinus sphenopalatinus), and middle conchal sinus (Sinus conchae mediae). Reconstructed structures were displayed separately, grouped, or altogether as transparent or solid elements to visualize individual paranasal sinus morphology. The paranasal sinuses appeared to be divided into two systems by the maxillary septum (Septum sinuum maxillarium). The first or rostral system included the rostral maxillary and ventral conchal sinus. The second or caudal system included the caudal maxillary, dorsal conchal, frontal, sphenopalatine, and middle conchal sinuses. These two systems overlapped and were interlocked due to the oblique orientation of the maxillary septum. Total volumes of the paranasal sinuses ranged from 911.50 to 1502.00 ml (mean ± SD, 1151.00 ± 186.30 ml). 3D renderings of equine paranasal sinuses by use of semi‐automated segmentation of CT‐datasets improved understanding of this anatomically challenging region. 相似文献

An 11‐year‐old Brown Swiss cow was referred to the Farm Animal Department of the Veterinary Teaching Hospital in Zurich, Switzerland, because of lateral recumbency due to puerperal hemolytic anemia. The animal had developed enophthalmos due to dehydration at the time of presentation. Two days after hospitalization, the cow showed blepharospasm and epiphora of the right eye. Ophthalmic examination of the right eye revealed a fluorescein‐positive, paraxial, superficial corneal ulcer with focal edema, and mild superficial neovascularization. White corneal stromal infiltrates were seen at the edges of the ulcer bed. After initial topical treatment with an antibiotic ointment (Neomycin 3.5 mg/g, Bacitracin 250 IU/g) three times a day, an increase in corneal infiltrates was noted on re‐examination 2 days later. Several fluorescein‐negative, punctate, stromal, white opacities were seen dorsal to the ulcer. Cytology demonstrated the presence of fungal hyphae. Topical treatment with 2% miconazole ointment and 0.36% K‐EDTA eye drops six times daily and four times daily, respectively, from the second day and continued antibiotics three times daily resolved the clinical symptoms within 6 days. The leaching model PESTRAS was used to estimate sorption and degradation values for bentazone from three lysimeter datasets using the inverse modelling package PEST. Investigations were undertaken to assess the influence on calibration results of (1) values attributed to uncertain parameters not included in the calibration, and (2) starting values supplied to the inverse modelling package. Automatic calibrations with different realistic values for the Freundlich exponent n(f) yielded different combinations of K(om) and DT50. Similarly, the supply of different starting values for K(om) and DT50 revealed that different combinations of these two parameters equally calibrated PESTRAS for two of the three lysimeters. Examination of the error surface, ie the forward running of the model for different combinations of K(om) and DT50 values, and the calculation of the goodness-of-fit to the experimental data, was found useful for identifying those instances where non-uniqueness in the calibration is likely to occur. Although the derivation of sorption and degradation values through inverse modelling is expected to offer significant benefits over laboratory determinations, care should be exercised when examining values derived through this approach. Research is needed to identify data requirements for robust estimation of sorption and degradation parameters through calibration of pesticide fate models against leaching data. 相似文献
