Water stress after flowering, one of the major factors limiting yields of pearl millet, affects both seed setting and grain filling and is a consequence of more/less water used prior to anthesis. However, whether genotypes have different sensitivities for seed setting and filling under drought, if exposed to similar stress intensity, is unclear. Experiments were conducted in two pairs of pearl millet genotypes, that is, PRLT2/89‐33 and H77/833‐2, 863B and 841B, contrasting for terminal drought tolerance, and two genotypes, ICMR 01046 and ICMR 01029 (IL‐QTLs), introgressed with a terminal drought tolerance QTL from PRLT2/89‐33 into H77/833‐2. Total seed weight, panicle number, 100‐seed weight, seed number and stover biomass were measured at different soil moistures and throughout grain filling. Sensitive H77/833‐2 had higher seed number and yield under well‐watered (WW) conditions than in PRLT2/89‐33 and IL‐QTLs. Upon increases in water stress intensity, H77/833‐2 suffered losses mostly in stover biomass (45 %) and seed number (60 %) at 0.3 FTSW whereas the biomass and seed number of PRLT2/89‐33 decreased little (20 % and 25 %). The 100‐seed weight of H77/833‐2 decreased only 20 % under stress. Tolerant 863B also maintained a higher seed number and biomass under water stress than 841B. Grain filling duration in PRLT2/89‐33 and IL‐QTLs was similar to that of H77/833‐2 under WW conditions but lasted longer than in H77833‐2 under water stress (WS). Similarly, seed growth of 863B was longer than 841B under WS. It is concluded that the higher seed yield of tolerant parents PRLT2/89‐33 and 863B, and of IL‐QTLs under WS was explained by the retention of a higher number of seeds than in sensitive lines, while the decrease in the 100‐seed weight was proportionally less than the decrease in seed number. Phenotype with lesser number and larger size of panicles and larger grain size, like genotypes PRLT2/89‐33 and 863B, withstood post‐anthesis water stress better. IL‐QTL inherited part of these characteristics, indicating a role for the terminal drought QTL in maintaining larger seed number and higher 100‐seed weight. The continuous stover biomass increase under WW in H77/833‐2, due to tillering, might indicate that tiller growth and grains are in competition for resources after anthesis, and this may relate to the relatively shorter grain‐filling period.  相似文献

OBJECTIVE: To evaluate the biological activity of dihydroartemisinin on canine osteosarcoma cell lines in vitro. SAMPLE POPULATION: 4 canine osteosarcoma cell lines. PROCEDURES: Cell viability assays were performed on canine osteosarcoma cell lines OSCA2, OSCA16, OSCA50, and D17 after 24, 48, and 72 hours of treatment with dihydroartemisinin at concentrations of 0.1 to 100 microM. Apoptosis was assessed by use of an ELISA for free nuclosomal DNA fragmentation and by western blot analysis for cleavage of caspase 3. Cell cycle analysis was performed by use of staining with propidium iodide and flow cytometry. Detection of reactive oxygen species (ROS) was conducted in the D17 cell line by use of 6-carboxy-2',7'-dihydrofluorescein diacetate and flow cytometry. RESULTS: The concentration of dihydroartemisinin required for 50% inhibition of cell viability (IC50) was achieved in all 4 canine osteosarcoma cell lines and ranged from 8.7 to 43.6 microM. Induction of apoptosis was evident as an increase in nucleosomal DNA fragmentation, cleavage of caspase 3, and an increase in the population in the sub G0/G1 phase of the cell cycle detected by flow cytometry. Exposure to dihydroartemisinin also resulted in a decrease in the G0/G1 population. Iron-dependent generation of ROS was detected in dihydroartemisinin-treated D17 cells; ROS generation increased in a dose-dependent manner. CONCLUSIONS AND CLINICAL RELEVANCE: Incubation with dihydroartemisinin resulted in biological activity against canine osteosarcoma cell lines, which included induction of apoptosis and arrest of the cell cycle. Clinical trials of dihydroartemisinin in dogs with osteosarcoma should be conducted.  相似文献

Gonadotropin (GTH) hormones are glycoprotein which stimulates gonadal maturation in vertebrates. Follicle stimulating hormone is involved in initiation of gametogenesis and regulation of gonadal growth. FSHβ has been cloned and characterized from the brain of Catla catla. The FSHβ full‐length of cDNA sequence of 523 bp comprised 3, 394 and 128 bp of 5′‐UTR, open reading frame (ORF) 3′‐UTR respectively. The coding region of C. catla FSHβ encoded a peptide of 130 amino acids. Phylogenetic analysis of C. catla FSHβ deduced amino acid sequence showed high similarity with Gobiocypris rarus followed by goldfish, Carassius auratus. The qPCR result shows that FSHβ mRNA is mainly expressed in pituitary while moderate and low expression was observed in testis and ovary respectively. Chitosan‐nanoconjugated kisspeptin‐10 (CK‐10) of particle size 125 nm, polydispersity index of 0.335 to 0.65 and zeta potential of ?34.95 mV were synthesized and evaluated at against naked kisspeptin‐10 for their reproductive hormonal profile. Treatment of fish with CK‐10 showed controlled and sustained surge of the reproductive hormones (FSH & LH) with peak at 12 h. The hormone levels of naked kisspeptin‐10 treated fish decline after 6 h. The sustained release of this CK‐10 will help in reducing maturation age, synchronization of ovulation and spawning in fish. This is the first report on use of chitosan‐nanoconjugated kisspeptin‐10 (CK‐10) for reproduction in fish.  相似文献

Purpose  

A large amount of nitrogen (N) fertilizers has been broadcasted over soil surface for reliable crop production. Unfortunately, the broadcasted N vulnerable to volatilization and leaching can lead to serious environmental problems. As a new approach to mitigate N loss of broadcasted fertilizers, massive intercalation of urea into montmorillonite (MMT) was recently proposed to innovatively enhance the urea use efficiency. This study aims to determine leaf litter preference, consumption rate, growth rate, food conversion efficiency, and quality of fecal pellets of two endemic pill millipedes (Arthrosphaera dalyi and Arthrosphaera davisoni) of the Western Ghats of India by laboratory microcosm experiments. Among seven combinations of three plantation leaf litters offered in 4-day trial, top three preferred combinations were selected for 4-week trial. In 4-week trial, preference of mixed litter diet was higher than single litter diet, which resulted in enhanced growth as well as food conversion efficiency of millipedes. Among Hopea , Pongamia , and Areca litters, A. dalyi preferred Hopea + Pongamia, and its consumption was significantly correlated with contents of organic carbon (P < 0.05; r = –0.97) and nitrogen (P < 0.01; r = 0.99), while growth rate with phosphorus content (P < 0.05; r = 0.97) and food conversion efficiency with contents of organic carbon (P < 0.05; r = 0.98) and calcium (P < 0.01; r = –0.99). Among Areca , Elettaria , and Coffea litters, Areca + Elettaria+ Coffea was most preferred by A. davisoni, which was significantly correlated with organic carbon content (P < 0.05; r = 0.98) and food conversion efficiency with calcium content (P < 0.0001; r = 0.99). The food conversion efficiency, however, was the highest in millipedes fed with Areca  +  Elettaria. The present study demonstrated increased nitrogen and phosphorus contents and decreased phenolic content and C/N ratio in fecal pellets of pill millipedes fed with plantation litter, and thus, these millipedes play an important role in leaf litter mineralization and soil enrichment in plantations Western Ghats.  相似文献

In the present study, a comparative assessment of 2,4,6-T (2,4,6-Trichlorophenol) degradation by different AOPs (Advanced Oxidation Processes – UV, UV/ H2O2, Fenton, UV/Fenton and UV/TiO2) in the laboratory scale is performed. The effects of different reactant concentrations and pH are assessed. 2,4,6-T removal, Total Organic Carbon mineralization (TOC) and dechlorination are monitored. Of all the AOPs, UV/Fenton process is more effective in degrading 2,4,6-T. The optimum conditions obtained for the best degradation with UV/Fenton are: pH?=?3, Fe+2 concentration of about 5 ppm, and peroxide concentration of 100 ppm for an initial 100 ppm of 2,4,6 T concentration at room temperature. In these conditions, a pseudo first-order rate constant is evaluated. Phytochemical investigation of Caralluma adscendens var. gracilis and Caralluma pauciflora (Asclepiadaceae) whole plant extracts allowed to isolate one pregnane glycoside and two pregnanes characterized as 12β,20-O-dibenzoyl-5α,6-dihydrosarcostin β-oleandropyranosyl-(1→4)-β-cymaropyranosyl-(1→4)-β-digitoxypyranosyl-(1→4)-β-cymaropyranosyl-(1→4)-β-cymaropyranoside (1), 12β-O-benzoyl-3β,11α,14β,20R-pentahydroxy-pregn-5-ene (2), and 11α-O-benzoyl-3β,12β,14β,20R-pentahydroxy-pregn-5-ene (3), respectively. Their structural characterization was obtained on the basis of extensive NMR spectral studies. Three known pregnane glycosides along with lupeol and β-sitosterol were also isolated and characterized.  相似文献
