ABSTRACT Bacterial apical necrosis of mango, elicited by Pseudomonas syringae pv. syringae, limits fruit production in southern Spain and Portugal. Examination of a collection of P. syringae pv. syringae isolates for copper resistance showed that 59% were resistant to cupric sulfate. The survey of a mango orchard revealed an increase in frequencies of copper-resistant bacteria after repeated treatments with Bordeaux mixture. These data suggest that selection of copper-resistant strains could be a major reason for control failures following management with copper bactericides. Most copper-resistant isolates harbored plasmids, although the majority of them contained a 62-kb plasmid that also was present in copper-sensitive strains. The 62-kb plasmids were differentiated by restriction enzyme analysis and hybridization to copABCD DNA. The most frequently found copper-resistant plasmid type (62.1) was transferable by conjugation. Southern blot hybridizations showed that genetic determinants partially homologous to copABCD were present in all the copper-resistant strains examined, and usually were associated with plasmids; these determinants were not detected in copper-sensitive strains. The selective pressure exerted by copper bactericide sprays on the diversity of copper resistance determinants in bacterial populations of mango is discussed. 相似文献

Several polymerase chain reaction (PCR) primers were designed from the internal transcribed spacer (ITS) regions of the rDNA genes of Rosellinia necatrix to develop a PCR-based identification method. Screening the primers against two isolates of R. necatrix and six other Rosellinia species resulted in the amplification of a single specific product from R. necatrix for most of the primer pairs. Two primer pairs (R2-R8 and R10-R7) confirmed their specificity when tested against 72 isolates of R. necatrix and 93 other fungi from different hosts and geographic areas. The R10 primer was modified to obtain a Scorpion primer for detecting a specific 112bp amplicon by fluorescence emitted from a fluorophore in a self-probing PCR assay. This assay specifically recognised the target sequence of R. necatrix over a large number of other fungal species. In conventional PCR, with primer pairs R2-R8 and R10-R7, 10-fold dilutions of R. necatrix DNA indicated a detection limit of 10pgul-1 using a single set of primers and 10fgl-1 in nested-PCR. For Scorpion-PCR, the detection limit was 1pgl-1 and 1fgl-1 in nested Scorpion-PCR, i.e. 10 times more sensitive than conventional PCR. A simple and rapid procedure for DNA extraction directly from soil was modified and developed to yield DNA of purity and quality suitable for PCR assays. Combining this protocol with the nested Scorpion-PCR procedure it has been possible to specifically detect R. necatrix from artificially inoculated soils in approximately 6h. 相似文献

The method of analysis employed in an in-vitro study on degradation of organochlorine compounds by meat starter micro-organisms was evaluated using the recommendations made by the Food and Drug Administration (FDA). Recoveries of the organochlorine pesticides α-, β- and γ-hexachlorocyclohexane, hexachlorobenzene, pp′DDE, dieldrin and the polychlorinated biphenyl PCB 153 from a mixture added to four different liquid media, tryptic soy broth, brain heart infusion, a commercial broth and a mineral salts medium, were determined. Recoveries were between 80 and 110% with all components, irrespective of the medium used, and there were no analytical interferences due to the reagent blank or to the matrix. The repeatability was very good with relative standard deviations in the range 2·5 to 11·1%. The concentration of each component of the mixture was determined by capillary GC analysis of extracts using an electron capture detector. © 1997 SCI. 相似文献

The occurrence and geographic distribution of longidorid nematode species inhabiting the rhizosphere of cultivated olive (cvs. Chemlali and Chétoui) in Tunisia were investigated. Morphological and morphometrical studies identified three Longidorus and six Xiphinema species, with frequencies of prevalence as following: Longidorus africanus (23.0 %), L. euonymus (4.5 %), L. glycines (13.7 %), Xiphinema conurum (13.7 %), X. italiae (36.4 %), X. meridianum (13.7 %), X. pachtaicum (18.2 %), X. robbinsi (9.1 %), and Xiphinema sp. (4.5 %). The three Longidorus species were reported for the first time in Tunisia, in addition to two species of Xiphinema (viz. X. meridianum and X. robbinsi). Molecular characterisation using D2-D3 expansion regions of 28S rRNA and ITS1-rRNA was carried out and Bayesian inference analysis was used to reconstruct phylogenetic relationships among these species and with other longidorids. Twenty-five new D2-D3 of 28S rRNA gene sequences were obtained in the present study, seven for Longidorus and 18 for Xiphinema spp., as well as 14 new ITS1 rRNA gene sequences (seven for Longidorus and seven for Xiphinema spp.). 相似文献

Alternative hosts are an important way of phytopathogenic bacteria survival between crop seasons, constituting a source of inoculum for the following crops. Bacterial wilt, caused by Curtobacterium flaccumfaciens pv. flaccumfaciens (Cff), is one of the most important diseases for common bean, and little information is available about the host range of the bacterium. In this study, we assessed possible alternative hosts for Cff, especially those cultivated during winter, in rotation systems with common bean. Plants of barley, black oat, canola, radish, ryegrass, wheat and white oat, were assessed under field and greenhouse conditions. Cff colonized epiphytically all plant species and endophytically black oat, ryegrass, wheat and white oat plants assessed in the greenhouse assays. Under field conditions, Cff colonized all plant species by except radish. All bacterial strains re-isolated from the plants were pathogenic to common bean and identified as Cff by PCR with specific primers. Based on our results, the cultivation of bean crop in succession with barley, black oat, canola, ryegrass, wheat and white oat should not be recommended, mainly in areas with a history of bacterial wilt occurrence. Mustard clubroot, caused by Plasmodiophora brassicae, is a serious disease that affects Brassica juncea var. tumida Tsen, a mustard plant that is the raw material for a traditional fermented food manufactured in the Chongqing Municipality, People’s Republic of China. To find antagonistic bacteria for P. brassicae, 124 bacteria were obtained from the rhizosphere soil of B. juncea var. tumida grown in Fuling, Chongqing. Isolates were preliminarily chosen by evaluating the inhibition rate of the P. brassicae resting spore germination. The biocontrol effects of three antagonistic bacteria against clubroot on B. juncea var. tumida were evaluated in a greenhouse experiment. B18 showed the highest control efficiency, at 63.4% in the greenhouse test. In a field trial, B18 was also effective in controlling clubroot, but only at a 49.7% efficiency rate. According to 16S rDNA sequence analysis, strain B18 had a 100% sequence similarity with type strain Zhihengliuella aestuarii DY66T (EU939716). Based on morphological, cultural, physiological and biochemical characteristics, the DNA G + C content, polar lipids, fatty acids, cell wall analysis, as well as DNA–DNA hybridization, strain B18 was identified as Z. aestuarii B18. Thus, the isolate B18 might have a potential biocontrol application for clubroot. We report for the first time that Z. aestuarii B18 can control clubroot. 相似文献

Recent data on the epidemiology of the common mycotoxigenic species of Fusarium, Alternaria, Aspergillus and Penicillium in infected or colonized plants, and in stored or processed plant products from the Mediterranean area are reviewed. Emphasis is placed on the toxigenicity of the causal fungal species and the natural occurrence of well known mycotoxins (aflatoxins, ochratoxins, fumonisins, trichothecenes, zearalenone, patulin, Alternaria-toxins and moniliformin), as well as some more recently described compounds (fusaproliferin, beauvericin) whose toxigenic potential is not yet well understood. Several Fusarium species reported from throughout the Mediterranean area are responsible of the formation of mycotoxins in infected plants and in plant products, including: Fusarium graminearum, F. culmorum, F. cerealis, F. avenaceum, F. sporotrichioides and F. poae, which produce deoxynivalenol, nivalenol, fusarenone, zearalenone, moniliformin, and T-2 toxin derivatives in wheat and other small grains affected by head blight or scab, and in maize affected by red ear rot. Moreover, strains of F. verticillioides, F. proliferatum, and F. subglutinans, that form fumonisins, beauvericin, fusaproliferin, and moniliformin, are commonly associated with maize affected by ear rot. Fumonisins, were also associated with Fusarium crown and root rot of asparagus and Fusarium endosepsis of figs, caused primarily by F. proliferatum. Toxigenic A. alternata strains and associated tenuazonic acid and alternariols were commonly found in black mould of tomato, black rot of olive and citrus, black point of small cereals, and black mould of several vegetables. Toxigenic strains of A. carbonarius and ochratoxin A were often found associated with black rot of grapes, whereas toxigenic strains of A. flavus and/or P. verrucosum, forming aflatoxins and ochratoxin A, respectively, were found in moulded plant products from small cereals, peanuts, figs, pea, oilseed rape, sunflower seeds, sesame seeds, pistachios, and almonds. Finally, toxigenic strains of P. expansum and patulin were frequently found in apple, pear and other fresh fruits affected by blue mould rot, as well as in derived juices and jams. 相似文献

The addition of species of Trichoderma to compost is a widespread technique used to control different plant diseases. The biological control activity of these species is mainly attributable to a combination of several mechanisms of action, which may affect the microbiota involved in the suppressiveness of compost. This study was therefore performed to determine the effect of inoculation of Trichoderma harzianum (T. harzianum) on compost, focusing on bacterial community structure (16S rRNA) and chitinase gene diversity. In addition, the ability of vineyard pruning waste compost, amended (GCTh) or not (GC) with T. harzianum, to suppress Fusarium wilt was evaluated. The addition of T. harzianum resulted in a high relative abundance of certain chitinolytic bacteria as well as in remarkable protection against Fusarium oxysporum comparable to that induced by compost GC. Moreover, variations in the abiotic characteristics of the media, such as pH, C, N and iron levels, were observed. Despite the lower diversity of chitinolytic bacteria found in GCTh, the high relative abundance of Streptomyces spp. may be involved in the suppressiveness of this growing media. The higher degree of compost suppressiveness achieved after the addition of T. harzianum may be due not only to its biocontrol ability, but also to changes promoted in both abiotic and biotic characteristics of the growing media. 相似文献

The potential role of 10 frequently occurring weed species found amongst Spanish lettuce crops as host plants for the two
viruses associated with the lettuce big-vein disease, Lettuce big-vein associated virus (LBVaV) and Mirafiori lettuce big-vein virus (MLBVV), was studied. The results showed that both viruses can infect naturally growing Sonchus oleraceus (common sowthistle) plants, the unique susceptible species detected among the analysed weeds. The sequences of the coat protein
(CP) genes of the LBVaV and MLBVV isolates recovered from S. oleraceus plants were determined. Phylogenetic studies revealed a very close relationship between the CP sequences from these weed
isolates and those from Spanish lettuce. Moreover, we showed that S. oleraceus can act as a source of lettuce infection by means of Olpidium brassicae, the vector fungus of both viruses. 相似文献
