Teeth of a North American ictidosaur, Pachygenelus milleri, n. sp., found in the Upper Triassic Dockum Group of Texas, indicate that it is very similar to Pachygenelus monus of South Africa and Chaliminia musteloides of South America. The presence of a Gondwana element in the Northern Hemisphere attests to the ease of dispersal of the Late Triassic vertebrates through Pangea. Ictidosaurs are small, highly advanced, carnivorous cynodonts that display a mosaic of reptilian and mammalian features in the masticatory apparatus. Metarhizium anisopliae infects mosquitoes through the cuticle and proliferates in the hemolymph. To allow M. anisopliae to combat malaria in mosquitoes with advanced malaria infections, we produced recombinant strains expressing molecules that target sporozoites as they travel through the hemolymph to the salivary glands. Eleven days after a Plasmodium-infected blood meal, mosquitoes were treated with M. anisopliae expressing salivary gland and midgut peptide 1 (SM1), which blocks attachment of sporozoites to salivary glands; a single-chain antibody that agglutinates sporozoites; or scorpine, which is an antimicrobial toxin. These reduced sporozoite counts by 71%, 85%, and 90%, respectively. M. anisopliae expressing scorpine and an [SM1](8):scorpine fusion protein reduced sporozoite counts by 98%, suggesting that Metarhizium-mediated inhibition of Plasmodium development could be a powerful weapon for combating malaria. 相似文献

A study was carried out on a silty clay loam soil (Typic Haplustept) to evaluate the effect of farmyard manure (FYM) vis‐à‐vis fertilizer and irrigation application on the soil organic C content and soil structure. The fertilizer treatments comprised of eight different combinations of N and FYM and three water regimes. The results indicated that the application of FYM and increasing N rate increased soil organic carbon (SOC) content. Addition of FYM also increased the percentage of large sized water stable aggregates (> 5 mm) and reduced the percentage of smaller size aggregates. This was reflected in an increase in the mean weight diameter (MWD) and improved soil structure. The organic carbon content in macroaggregates (> 1 mm) was greater compared to microaggregates, and it declined with decrease in size of microaggregates. This difference in organic C content between macro‐ and microaggregates was more with higher N dose and FYM treated plots. The effect of residual FYM on MWD and organic C content of the soil after wheat harvest was not significant. The effect was less in deeper layers compared to surface layers of the soil. MWD was significantly correlated with the SOC content for the top two layers. 相似文献

A greenhouse pot experiment was conducted to study the effect of application of three levels of phosphorus (0, 50 and 100 ppm) and three levels of zinc (0, 5 and 10 ppm) in all possible combinations on yield and tissue concentrations of zinc, copper, iron and manganese by rice at two different growth stages (35 and 90 days after transplantation). The results showed that application of both phosphorus and zinc increased the dry matter yield of shoot and root at both the growth stages. Phosphorus or zinc application was found to influence the concentration of zinc, copper, iron and manganese in shoot and in root but these influences varied with the growth stages of the plant as well as with the level of zinc and phosphorus application. The results further showed that phosphorus or zinc application influenced the absorption of zinc/phosphorus, iron, manganese and copper by rice plants and their translocation from root to shoot. Plasma concentrations of beta-endorphin were assayed in 10 sheep during the conscious state and in relation to electrical stunning. There was a 2 fold increase after stunning, and a further elevation following recovery. Of 200 genotypes of cauliflower screened for their reaction to blackrot disease (Xanthomonas campestris (Pam.) Dowson) ‘MGS-2⊕-3⊕’, ‘Pua Kea’ and ‘SN 445’, derived from Indian cauliflower cultivars, possessed a high degree of resistance. Neither ‘Snowball’ nor its relatives ‘Erfurt’ and ‘Alpha’ showed any resistance. For the plant breeder, soaking the seed in a bacterial suspension is the most useful screening-technique; less useful were the spraying of whole plants with a suspension and the severing of a leaf-tip beneath a suspension. OBJECTIVE: To characterize patterns of removal and evaluate the associations among culling because of lameness and sow productivity traits among culled gilts and sows. DESIGN: Cross-sectional study. SAMPLE POPULATION: Data from a convenience sample of 11 farms pertaining to the removal of 51,795 gilts and sows from January 1991 to December 2002. Mean culling and mortality (death and euthanasia) rates for all inventoried gilts and sows ranged from 23% to 50% and 4.7% to 9.5%, respectively. PROCEDURE: An analysis of categories of removal (cull, death, or euthanasia) and reasons for removal of gilts and sows was performed. Multivariate logistic regression was used to determine associations among culling because of lameness and sow productivity traits among culled gilts and sows. RESULTS: Among sows that were removed, the proportion of parity > or = 1 sows that died (both death and euthanasia) was > 3 times the proportion of parity > or = 1 sows that were culled within 20 days after farrowing. Among lame sows that were removed, the proportion of parity > or = 1 sows that died (death and euthanasia) was higher than the proportion of parity > or = 1 sows that were culled within 20 days after farrowing. Among sows that were removed, the proportion of sows that died (deaths and euthanasia) was higher during lactation than nonlactation. This was also observed among lame sows that were removed. CONCLUSIONS AND CLINICAL RELEVANCE: The proportion of death among removed sows, especially lame sows, was higher during lactation than nonlactation. Non-methylated CpG motifs, present in viral and bacterial DNA, are one of many pathogen-associated molecular patterns (PAMP) recognized by the mammalian innate immune system. Recognition of this PAMP occurs through a specific interaction with toll-like receptor 9 (TLR9) and this interaction can induce cytokine responses that influence both innate and adaptive immune responses. Previous investigations determined that both the flanking sequences in synthetic CpG oligodeoxynucleotides (CpG ODN) and the cellular pattern of TLR9 expression can influence species-specific responses to CpG ODN. Therefore, the structure, function and cellular distribution of bovine TLR9 were compared with what is known for mice and human. Analysis of the bovine TLR9 gene revealed greater sequence homology between cattle and humans than cattle and mice Similar CpG motifs induced optimal activation of both human and bovine leukocytes and these motifs were distinct from those which activated mouse leukocytes. Functional analyses with CpG ODN stimulated bovine blood leukocytes revealed that class A CpG ODN were more potent inducers of interferon-alpha (IFN-alpha) than class B CpG ODN. Furthermore, magnetic activated cell sorting of bovine blood leukocyte subpopulations implicated dendritic cells but not monocytes in the regulation of CpG ODN-induced IFN secretion. Thus, the cellular pattern of CpG ODN-induced responses in cattle shared many similarities with human leukocytes. Collectively, these analyses revealed substantial conservation of TLR9 structure and TLR9 function in blood leukocytes of humans, cattle and other domestic species. 相似文献

The pathogenic mechanisms involved in tropical theileriosis, caused by the tick-borne protozoan parasite Theileria annulata, are unclear. Pathology is associated with the schizont stage of the parasite, which resides within bovine macrophages. Breed-specific differences in pathology have been observed in cattle, several Bos indicus breeds are relatively resistant to tropical theileriosis whilst Bos taurus cattle are highly susceptible. Infected cells express pro-inflammatory cytokines and it has been hypothesized that these cytokines play a major role in the pathology of the disease. Therefore, using quantitative RT-PCR we investigated the expression of the key candidates, interleukin 1 beta (IL-1beta), IL-6 and tumour necrosis factor alpha (TNF-alpha), in T. annulata low passage infected cell lines derived ex vivo from experimental infection of resistant and susceptible cattle. mRNA for each cytokine was detected in all cell lines investigated at levels higher than those observed in resting monocytes. However, the analyses did not identify any breed-specific differences. Therefore, these results are not consistent with the hypothesis that differential regulation of infected cell derived pro-inflammatory cytokines (IL-1beta, IL-6 and TNF-alpha) accounts for the breed-related differences in resistance and susceptibility to T. annulata infection. Other, currently unknown mechanisms may be of greater importance. 相似文献
