A laboratory model, set to simulate the in vivo conditions of the porcine gastrointestinal tract, was used to study the small intestinal absorption of several mycotoxins and the effectiveness of Standard Q/FIS (a carbon/aluminosilicate-based product) in reducing mycotoxin absorption when added to multitoxin-contaminated diets. Mycotoxins were quickly absorbed in the proximal part of the small intestine at levels of 105 and 89% for fumonisins B1 and B2, respectively, 87% for ochratoxin A, 74% for deoxynivalenol, 44% for aflatoxin B1, and 25% for zearalenone. Addition of Standard Q/FIS to the diet (up to 2%, w/w) significantly reduced mycotoxin absorption, in a dose-dependent manner, up to 88% for aflatoxin B1, 44% for zearalenone, and 29% for the fumonisins and ochratoxin. Standard Q/FIS was ineffective in reducing deoxynivalenol uptake. These findings suggest that Standard Q/FIS can be used as a multitoxin adsorbent material to prevent the individual and combined adverse effects of mycotoxins in animals.  相似文献

Lipid transfer proteins (LTPs) are a family of low molecular mass (7-9 kDa) polypeptides, the members of which share 35-95% sequence homology. These proteins are widely distributed throughout the plant kingdom and are receiving attention for their biochemical characteristics and biological activity. LTPs are indeed studied in different research fields varying from allergy to food technology, and numerous molecules belonging to this class are progressively being identified and investigated. Proteins from pomegranate juice were fractioned by cation exchange chromatography and analyzed by SDS-PAGE. Two proteins were identified as putative LTPs on the basis of their molecular weights and their electrophoretic behaviors under reducing and nonreducing conditions. Finally, proteins were purified and characterized by mass spectrometry. This analysis confirmed that the two polypeptides are LTPs on the basis of an amino acid sequence common to LTPs from other plant sources and cysteine content. The two proteins, named LTP1a and LTP1b, showed similar molecular masses but different immunological profiles when immunodetected with rabbit antibodies specific for Pru p 3 and human IgE from a patient suffering from pomegranate allergy. The demonstration of the existence of two immunologically unrelated LTPs in pomegranate confirms the variability and the complexity of the plant LTP family. This should be taken into account when the role of these proteins as elicitors of allergies to fruits is investigated and could help to explain the contradictory literature data on pomegranate allergy.  相似文献

The phenolic compounds of Rumex induratus leaves were determined by HPLC-DAD-MS/MS-ESI. The results revealed a profile composed of 19 compounds: caffeoyl-hexoside, two p-coumaroyl-hexoside isomers, feruloyl-hexoside, sinapoyl-hexoside, 6-C-hexosyl-quercetin, 8-C-hexosyl-luteolin, 6-C-hexosyl-luteolin, 6-C-hexosyl-apigenin, 3-O-hexosyl-quercetin, 3-O-rutinosyl-quercetin, 7-O-hexosyl-diosmetin, 3-O-rutinosyl-isorhamnetin, 7-O-(acetyl)-pento-hexosyl-diosmetin, 6-C-hexosyl-genkwanin, and four unidentified O-glycosyl-C-glycosylflavones. The quantification of the identified phenolics by HPLC-DAD showed 6-C-hexosyl-luteolin as the main compound. Organic acid composition was determined by HPLC-UV, revealing a high content of oxalic acid. R. induratus was also investigated for its capacity to act as a scavenger of DPPH and superoxide radicals. Good antioxidative results were obtained against both radicals. Unlike other species of the genus Rumex, R. induratus did not present any anthraquinone derivative.  相似文献

The tumor suppressor programmed cell death protein 4 (PDCD4) inhibits the translation initiation factor eIF4A, an RNA helicase that catalyzes the unwinding of secondary structure at the 5' untranslated region (5'UTR) of messenger RNAs (mRNAs). In response to mitogens, PDCD4 was rapidly phosphorylated on Ser67 by the protein kinase S6K1 and subsequently degraded via the ubiquitin ligase SCF(betaTRCP). Expression in cultured cells of a stable PDCD4 mutant that is unable to bind betaTRCP inhibited translation of an mRNA with a structured 5'UTR, resulted in smaller cell size, and slowed down cell cycle progression. We propose that regulated degradation of PDCD4 in response to mitogens allows efficient protein synthesis and consequently cell growth.  相似文献

Impairment of water quality is a major concern for streams and rivers in the central USA. Total maximum daily loads (TMDLs) establish a watershed framework and set management targets to alleviate pollution from both point and nonpoint sources. For this study, we have used a hydrologic modeling approach to holistically examine the effect of land use management, urban development, and agricultural practices on sediment and nutrient loadings in an agricultural watershed. Annualized Agricultural Nonpoint Source (AnnAGNPS) simulation indicates that while point source dischargers contribute 8% of total nitrogen (TN) and 24% of total phosphorus (TP) loadings to the Marmaton River, agricultural nonpoint sources are the leading pollution source contributing 55% of TN and 49% of TP loading. Based on TMDL analysis and model simulation, 3% of the watershed area (3,244 ha) needs to be targeted to control TN loading whereas 1% of the total area (1,319 ha) is required for TP reduction management. Managing the TN areas alone can achieve a 57% reduction in the TP load required for the TMDL, whereas managing the targeted TP areas can only provide 30% of the required TN reduction. Areas required both TN and TP management comprise 469 ha. Targeting these areas can achieve approximately 22% of the required TN reduction and 29% of the required TP reduction. Overall, 4,094 ha will require management to achieve water quality goals. This study demonstrates that a modeling approach is needed to effectively address TMDL issues and help identify targeted areas for management.  相似文献

Thirty-six isolates of Fusarium oxysporum originated from Eruca vesicaria and Diplotaxis tenuifolia together with eight reference strains belonging to the formae speciales raphani, matthioli and conglutinans, typical on the Brassicaceae family, were tested for pathogenicity on two species of rocket plants (E. vesicaria L., syn. E. sativa, cv. ‘Rucola coltivata’; and D. tenuifolia cv. ‘Winter’) cultivated in the glasshouse. The results showed that different isolates were slightly, moderately or highly virulent. The strains were examined for differences in the nucleotide sequence of the ribosomal DNA (rDNA) intergenic spacer (IGS) region, about 2.5 kb long. The phylogenetic (neighbor-joining) analysis performed on the isolates enabled identification of four different groups, named I, II, III and IV. Thirty-one isolates out of 36 clustered in group I and were genetically similar to F. oxysporum f.sp. raphani. By considering the pathogenicity of the strains included in Group I, a partial host specialization could be observed: the average disease index of the isolates from D. tenuifolia was higher on wild rocket, whereas the average disease index of the isolates from E. vesicaria was higher on cultivated rocket. Moreover, isolates from cultivated rocket showed, on average, a higher degree of aggressiveness than the isolates from wild rocket. Concerning Group I, the sequence analysis confirmed the homogeneity of the population, with only five parsimony-informative SNPs and five haplotypes. Twenty-six out of 31 isolates belonged to haplotype 1. Groups II and III were genetically similar to strains of F. oxysporum f.sp. matthioli. Three other strains, not pathogenic or with a medium level of virulence, clustered together in Group 4, but their sequence was distant from that of other formae speciales. The pathogenicity and IGS analysis confirmed the presence of virulence variation and genetic diversity among the F. oxysporum isolates studied. To our knowledge, this is the first report of differentiation of formae speciales of F. oxysporum on rocket plants by IGS analysis.  相似文献

The bacterial canker of kiwifruit caused by Pseudomonas syringae pv. actinidiae is a severe threat to kiwifruit production worldwide. Many aspects of P. syringae pv. actinidiae biology and epidemiology still require in-depth investigation. The infection by and spread of P. syringae pv. actinidiae in xylem and phloem was investigated by carrying out artificial inoculation experiments with histological and dendrochronological analyses of naturally diseased plants in Italy. We found that the bacterium can infect host plants by entering natural openings and lesions. In naturally infected kiwifruit plants, P. syringae pv. actinidiae is present in the lenticels as well as in the dead phloem tissue beneath the lenticels, surrounded by a lesion in the periderm which appears to indicate the importance of lenticels to kiwifruit infection. Biofilm formation was observed outside and inside plants. In cases of advanced stages of P. syringae pv. actinidiae infection, neuroses of the phloem occur, which are followed by cambial dieback and most likely by infection of the xylem. Anatomical changes in wood such as reduced ring width, a drastic reduction in vessel size, and the presence of tyloses were observed within several infected sites. In the field, these changes occur only a year after the first leaf symptoms are observed suggesting a significant time lapse between primary and secondary symptoms. It was possible to study the temporal development of P. syringae pv. actinidiae-induced cambial dieback by applying dendrochronology methods which revealed that cambial dieback occurs only during the growing season.  相似文献

Psyttalia concolor (Szépligeti) (Hymenoptera: Braconidae) is a koinobiont larval–pupal endoparasitoid of a large number of tephritids. P. concolor can be commonly reared on fully grown Ceratitis capitata (Wiedemann) (Diptera: Tephritidae) larvae. In our laboratory, over 180 consecutive generations have been completed on C. capitata. In this study, we tested the possibility that the uninterrupted mass-rearing of P. concolor on C. capitata may influence its host location and parasitisation behaviours when used against Bactrocera oleae (Rossi) (Diptera: Tephritidae), the fruit fly commonly targeted in mass release of P. concolor in olive-groves. Choice and no-choice tests were used with naïve and experienced P. concolor females exposed to B. oleae and/or C. capitata larvae. The study showed no significant difference in oviposition behaviours and host acceptance when provided either C. capitata or B. oleae. However, the study did show that experience on a host plus a substrate complex could influence host preferences, since females that had previous oviposition experience on a given host species searched and chose significantly more when the same host species was presented, both in choice and no-choice conditions. Therefore, within the context of the pest management of fruit flies, it could be very useful to incorporate procedures in insectary-propagated fruit fly parasitoids designed to provide adequate training (i.e. oviposition experience) on the fruit fly species needing to be controlled.  相似文献

The aim of this study was to compare the arterial vascularization of the pelvic limb between southern caracara (Caracara plancus) and great egret (Ardea alba) by dissection and radiographic examinations. Five specimens of caracaras (three males and two females), and seven great egrets (five males and two females) were used. Barium sulphate and latex suspension were injected into the left ventricle of the birds. The radiographs were taken with the pelvic limbs in the ventrodorsal, dorsoplantar, mediolateral and lateromedial recumbency. Thereafter, the material was fixed in a 10% solution of formaldehyde and dissected. The pelvic limb received its arterial supply from two main vessels, the ischiatic and external iliac arteries. The ischiatic artery presented to be the principal artery of pelvic limb in the caracara and great egret. Several branches arised from the ischiatic and external iliac arteries were described. No gender differences were observed in both species. The caracara and great egret showed arteries similar to those reported for the ostrich and domestic fowl. According to the results of this study, it is suggested that the caracara has a pelvic limb with more arterial branches and larger arterial diameter than the great egret, which is probably related to the specific behaviour of these birds, since the caracara is a bird that exercise more their pelvic limbs to capture its prey when compared with the great egret.  相似文献
