This paper points out the usefulness of biopartitioning micellar chromatography (BMC) using capillary columns as a high-throughput primary screening tool providing key information about the oral absorption, skin permeability, and brain-blood distribution coefficients of 15 polyphenols (6 flavones, 2 flavonols, a flavanone, 2 flavan-3-ols, 3 phenolic acids, and a phloroglucinol) in a simple and economical way. For the compounds studied, except vicenin-2, rutin, chlorogenic acid, p-hydroxycinnamic acid, and 4-hydroxybenzoic acid, maximal oral absorption (>90%) can be expected, if there are not solubility problems or metabolic processes. On the other hand, the most retained compounds in BMC, that is, 5-hydroxyflavone, flavone, and flavanone, show the highest brain-blood distribution coefficients and skin permeability coefficients. 相似文献

To determine the role of the spleen in the pathogenesis of canine visceral leishmaniasis (CVL), we analyzed cellular immunophenotypic profiles of 52 dogs naturally infected with Leishmania infantum, clinically classified as follows: asymptomatic dogs-I (AD-I), seronegative/PCR+; asymptomatic dogs-II (AD-II), seropositive/PCR+; oligosymptomatic dogs (OD) and symptomatic dogs (SD). Seven non-infected dogs (CD) were included as a control group. AD-II presented higher levels of CD8+ T splenocytes and lower TCD4+/TCD8+ ratio in comparison with CD. OD and SD showed lower percentages of CD21+ as compared with AD-II. All seropositive dogs presented lower levels of CD45RA+ than CD. Regardless of the stimuli used, the proliferation index from splenocytes in vitro was inversely correlated with clinical status. After LSA stimulation, there was a higher percentage of specific CD8+ T in AD-II than CD and non-stimulated culture. In contrast, splenocytes from SD under in vitro LSA stimulation induced decreased MHC-II+ expression in comparison with all groups, and non-stimulated culture. In conclusion, the role of CD8+ T splenocytes seems to be important for an effective immunological response, a hallmark of asymptomatic CVL, whereas the pronounced loss of MHC-II expression upon LSA stimulation is a biomarker of symptomatic CVL. 相似文献

This study was aimed at describing post-fire mushroom production in a Mediterranean ecosystem dominated by Pinus pinaster Ait. in the northwest of Spain and assessing the results by classificatory models. During the autumn periods of 2003–2006, fruit bodies from 115 fungal taxa were collected in burned and unburned areas and were further grouped into the following categories: saprotrophic/mycorrhizal; and edible/non-edible. After wildfires, a significant reduction in the number of fungal species and fruit body biomass production was observed. Based on this relevant information, the first simple classificatory model for this aim is provided. Nine alternative models based on classifications according to combinations of edibility and functional groups were fitted, and four fruiting body biomass production classes were defined as possible responses. As explanatory factors, time after fire and climatic variables significantly related to fruit body production were included. The best predictive results were obtained for edible and edible-mycorrhizal models, for which the correct classification rate of production classes was between 92 and 85 %. Moreover, the models obtained were applied to analyse the effect of time after fire on fungal production. Mycorrhizal and edible fungal production after fire was classified into the lowest class, whereas saprotrophic and non-edible species followed a contrary trend. The classificatory models can be useful to optimise management and harvest of these increasingly appreciated non-timber forest resources. 相似文献

ObjectiveTo evaluate the effects of intravenous (IV) or intramuscular (IM) hyoscine premedication on physiologic variables following IV administration of medetomidine in horses.Study designRandomized, crossover experimental study.AnimalsEight healthy crossbred horses weighing 330 ± 39 kg and aged 7 ± 4 years.MethodsBaseline measurements of heart rate (HR), cardiac index (CI), respiratory rate, systemic vascular resistance (SVR), percentage of patients with second degree atrioventricular (2oAV) block, mean arterial pressure (MAP), pH, and arterial partial pressures of carbon dioxide (PaCO2) and oxygen (PaO2) were obtained 5 minutes before administration of IV hyoscine (0.14 mg kg?1; group HIV), IM hyoscine (0.3 mg kg?1; group HIM), or an equal volume of physiologic saline IV (group C). Five minutes later, medetomidine (7.5 μg kg?1) was administered IV and measurements were recorded at various time points for 130 minutes.ResultsMedetomidine induced bradycardia, 2oAV blocks and increased SVR immediately after administration, without significant changes in CI or MAP in C. Hyoscine administration induced tachycardia and hypertension, and decreased the percentage of 2oAV blocks induced by medetomidine. Peak HR and MAP were higher in HIV than HIM at 88 ± 18 beats minute?1 and 241 ± 37 mmHg versus 65 ± 16 beats minute?1 and 192 ± 38 mmHg, respectively. CI was increased significantly in HIV (p ≤ 0.05). Respiratory rate decreased significantly in all groups during the recording period. pH, PaCO2 and PaO2 were not significantly changed by administration of medetomidine with or without hyoscine.Conclusion and clinical relevanceHyoscine administered IV or IM before medetomidine in horses resulted in tachycardia and hypertension under the conditions of this study. As part of an ongoing inventory of the helminth parasites of freshwater fishes in Mexico, 570 individual fish were collected between Apr 2008 and Oct 2011 in 26 localities along the Cuatro Ciénegas region in Coahuila State, northern Mexico. Seventeen species of hosts, mostly corresponding to Nearctic freshwater elements, were studied. A total of 8324 individual worms were collected during this survey, representing 25 species of helminths, of which 9 were digeneans, 3 monogeneans, 3 acanthocephalans, 9 nematodes and 1 cestode. Most of the records in this checklist represent new host or locality records. The information provided in this checklist may be helpful for our understanding of the biodiversity and historical biogeography of this host–parasite system, because in the Cuatro Ciénegas region occur a Nearctic freshwater fish fauna, along with Neotropical and endemic elements, and from a biogeographical point of view, this may represent a transitional area. 相似文献

The rearing of fish in cages could greatly contribute to increasing fish production in existing inland waters. Although it presents numerous advantages, its limitations and possible inconveniences must be borne in mind. Well-developed technology is available mainly from the Democratic Republic of Germany, The Netherlands, Japan, the U.S.A., the U.S.S.R. and the Lower Mekong region. Research is currently pursued in numerous other countries, where the use of floating cages in fish culture is becoming increasingly diversified.The most popular fish species used in intensive cultural systems are Cyprinus carpio, Ictalurus punctatus and Salmo gairdneri. In the semi-intensive Asian systems, Barbus spp., Pangasius spp. and murrels are among the favourite cage-reared fishes. Tilapia aurea and Tilapia nilotica have also proved to be well adapted to cage culture. Polyculture is only rarely practised.Floating cages seem to be the best suited for fish culture in most cases. Net cages are the simplest in design and the cheapest to build. The escape of fish, however, always remains an eventuality which might lead to choosing more expensive designs. Maintenance of cages must be regular. For flexibility of exploitation, cages should not be too large. Their location within the water body can play an important role in keeping production high.At high stocking rates, artificial feeding of a well-adapted diet is a necessity. Optimum conversion rates require a well-adjusted daily ration, possibly fractioned. Mechanized feed distribution is to be encouraged as a labour-saving practice. Less intensive systems rely more on locally available wastes, such as trash fish, but the feeding rate then depends mostly on feed prices.Best growth is generally achieved by C. carpio and I. punctatus. Attainable maximum production is as high as 35 kg/m3/month under intensive management. However, generally, production averages from 5 to 30 kg/m3/month for these two species. S. gairdneri is less productive. T. aurea and T. nilotica have both a very good potential. Production in less intensive systems varies greatly according to management practices. 相似文献

- Marine aquaculture, and its fast-growing development, has the potential for wider environmental, ecosystem, and biodiversity impacts. This study assesses the impact of fish farming on a bottlenose dolphin (Tursiops truncatus) population within the Gulf of Ambracia (western Greece) between 2007 and 2018. Two different study areas were defined in order to investigate differences on occurrence, abundance, behaviour, and seasonal fluctuations between them: a ‘control area’, in the north-western side of the Gulf; and a ‘fish farm area’, in the south west, where the highest density of fish farm cages is found.
- A total of 169 daily surveys were dedicated to the control area and 74 days were dedicated to the fish farm area, yielding 104 and 37 sightings, respectively. Both the probability of detecting dolphins (U = 6,763.000, P = 0.002) and the group sizes (U = 578.000, P < 0.001) were smaller around fish farms.
- Seasonality analyses were restricted to 2007–2008, as this was the period with year-round effort. Results revealed that dolphins were more frequently seen around fish farms in the winter (Kruskal–Wallis test, P = 0.036).
- From the 40 identified individuals that were re-sighted at least 10 times during the study period, 21 used the fish farm area less than expected according to sampling effort, and 10 of them were never observed in that area (binomial test, P < 0.05).
- The results revealed an impact of fish farms on the distribution of bottlenose dolphins in the Gulf of Ambracia. This information should be taken into consideration when defining ecosystem-based management measures within the management plan, which is currently in preparation for this Natura 2000 site.

Tanjungides A (1) (Z isomer) and B (2) (E isomer), two novel dibrominated indole enamides, have been isolated from the tunicate Diazona cf formosa. Their structures were determined by spectroscopic methods including HRMS, and extensive 1D and 2D NMR. The stereochemistry of the cyclised cystine present in both compounds was determined by Marfey’s analysis after chemical degradation and hydrolysis. We also report the first total synthesis of these compounds using methyl 1H-indole-3-carboxylate as starting material and a linear sequence of 11 chemical steps. Surface-associated marine bacteria are an interesting source of new secondary metabolites. The aim of this study was the isolation and identification of epiphytic bacteria from the marine brown alga, Bifurcaria bifurcata, and the evaluation of the antioxidant and antimicrobial activity of bacteria extracts. The identification of epiphytic bacteria was determined by 16S rRNA gene sequencing. Bacteria extracts were obtained with methanol and dichloromethane (1:1) extraction. The antioxidant activity of extracts was performed by quantification of total phenolic content (TPC), 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and oxygen radical absorbance capacity (ORAC). Antimicrobial activities were evaluated against Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis, Salmonella enteritidis, Staphylococcus aureus, Saccharomyces cerevisiae and Candida albicans. A total of 39 Bifurcaria bifurcata-associated bacteria were isolated and 33 were identified as Vibrio sp. (48.72%), Alteromonas sp. (12.82%), Shewanella sp. (12.26%), Serratia sp. (2.56%), Citricoccus sp. (2.56%), Cellulophaga sp. (2.56%), Ruegeria sp. (2.56%) and Staphylococcus sp. (2.56%). Six (15.38%) of the 39 bacteria Bifurcaria bifurcata-associated bacteria presented less than a 90% Basic Local Alignment Search Tool (BLAST) match, and some of those could be new. The highest antioxidant activity and antimicrobial activity (against B. subtilis) was exhibited by strain 16 (Shewanella sp.). Several strains also presented high antimicrobial activity against S. aureus, mainly belonging to Alteromonas sp. and Vibrio sp. There were no positive results against fungi and Gram-negative bacteria. Bifurcaria bifurcata epiphytic bacteria were revealed to be excellent sources of natural antioxidant and antimicrobial compounds. 相似文献
