Summary Molecular mapping is a promising strategy for studying and understanding traits with complex genetic control, such as partial resistance to oat crown rust. The objectives of this research were to develop molecular maps from the progenies of the cross UFRGS7 (susceptible) × UFRGS910906 (partially resistant) and to identify QTLs (quantitative trait loci) associated to partial resistance to oat crown rust in two generations of that population.DNA of 86 genotypes of the F2 and 90 genotypes of the F6 UFRGS7 × UFRGS910906 population were used to generate AFLP markers. Molecular maps were constructed using Mapmaker Exp. 3.0 and QTLs for partial resistance to oat crown rust were identified with Mapmaker/QTL software. Five hundred and fifty seven markers in the F2 and 243 markers in the F6 generations were identified. The F2 map integrated 250 markers in 37 linkage groups. The F6 map integrated 86 markers in 17 linkage groups.Five QTLs were identified for partial resistance to oat crown rust in the F2 generation and three QTLs in the F6. The QTL identified on F6 through the PaaaMctt340 AFLP marker showed consistency across two environments and two generations (F4 and F6), and appear to have potential for marker-assisted selection in oat. 相似文献

Breeders are encouraged to develop breeding approaches that strive to integrate food production into the healthy functioning of agro-ecosystems. In the case of legumes, this approach should preserve bee fauna by providing suitable floral resources within the crops themselves. In parallel, legume breeding for sustainable agriculture is linked to the development of environmental services. Foraging places and nesting sites for solitary and social bees are some of the ecological services provided for legumes. Crops with floral attractiveness and rewards for insects can be used to enhance pollinator conservation as well as crop yield and yield stability. We analyze how understanding crop-pollinator relationships (CPR) can contribute to the production of high-yielding and pollinator-friendly varieties by examining: (1) The status of knowledge on mating systems and floral traits; (2) The contribution of CPR understanding to plant breeding for both hybrid-seed production and open-pollinated population improvement. 相似文献

Selected metals were evaluated in surface sediments from Cadeia and Feitoria Rivers (Brazil), potentially affected by tanneries. Statistical factor analysis of sediment data allowed the identification of critical} metals and priority areas for biological monitoring, i.e., chromium and mercury at the lower course of Feitoria River. Non-piscivorous bottom-fish from the species Rineloricaria cadeae (violinha) were collected at reference and contaminated sites. Chromium was undetected} in fish tissues, and mercury accumulated in individuals living in contact with contaminated deposits. Mercury contents in fish were below World Health Organization quality standard for human consumption (0.5 μg/g). The ratio of total mercury content in fish (wet weight) to sediment (dry weight, <63 μm) was 0.5 and 0.7 at reference and contaminated sites, respectively. A risk assessment based on hazard index showed that maximum consumption of fish from contaminated site by human population should be 32 g/day, while at reference site the allowable amount could reach 140 g/day. 相似文献

Compounds with a (2H)-1,4-benzoxazin-3(4H)-one skeleton have attracted the attention of phytochemistry researchers since 2,4-dihydroxy-(2H)-1,4-benzoxazin-3(4H)-one (DIBOA) and 2,4-dihydroxy-7-methoxy-(2H)-1,4-benzoxazin-3(4H)-one (DIMBOA) were isolated from plants belonging to the Poaceae family. These compounds exhibit interesting biological properties, such as phytotoxic, antimicrobial, antifeedant, antifungal, and insecticidal properties. These chemicals, in addition to a wide variety of related compounds involved in their metabolism, detoxification mechanisms, and degradation on crop soils and other systems, have high interest and in some cases potential agronomic utility. This paper presents a complete review of the methods employed for their synthetic obtention in addition to some of the authors' own contributions to their chemistry. The degradation and phytotoxicity experiments carried out in ongoing research into the potential agronomic utility of these compounds required large amounts of them, which were obtained from natural sources. This paper presents a modified methodology to access DIMBOA from Zea mays cv. Apache and to obtain 2-O-beta-D-glucopyranosyl-2,4-dihydroxy-(2H)-1,4-benzoxazin-3(4H)-one (DIBOA-Glc) and DIBOA from Secale cereale L. New synthetic methodologies were employed for the obtention of the lactams 2-hydroxy-(2H)-1,4-benzoxazin-3(4H)-one and 2-hydroxy-7-methoxy-(2H)-1,4-benzoxazin-3(4H)-one and the malonamic acids N-(2-hydroxyphenyl)malonamic acid and N-(2-hydroxy-7-methoxyphenyl)malonamic acid. The aminophenoxazines 2-amino-7-methoxyphenoxazin-3-one and 2-acetamido-7-methoxyphenoxazin-3-one have been synthesized in the authors' laboratory by novel procedures. The distribution of six compounds with three different polyphenol skeletons have been studied in Rosmarinus officinalis: phenolic diterpenes (carnosic acid, carnosol, and 12-O-methylcarnosic acid), caffeoyl derivatives (rosmarinic acid), and flavones (isoscutellarein 7-O-glucoside and genkwanin), each showing a characteristic behavior and distribution during the vegetative cycle. Only in leaves were all six compounds present, and the highest accumulation rate was related with the young stages of development. Rosmarinic acid showed the highest concentrations of all the polyphenols in all organs. The distribution of this acid in leaves, flowers, and stems suggests that in the first stages of flower growth, levels were due to in situ biosynthesis, and in the last stages, the contribution of transport phenomena was increased. The antioxidant activity of six extracts with different polyphenolic composition was evaluated in aqueous and lipid systems. The results clearly suggest that rosemary extracts are excellent antioxidants in both aqueous and lipid systems. 相似文献

Trypsin inhibitors (TI), tannins, and lectins appear to have a role in preventing chronic diseases in humans. The genetic variability of these traits in common bean needs to be ascertained in order to increase levels through breeding. The variability of TI, tannin, and lectins was determined in five bean cultivars grown at five locations in Mexico. TI and tannins contents in colored beans that belong to the Jalisco race were higher (11.1-11.9 trypsin units inhibited (TUI)/mg and 29.0-38.1 mg catechin equivalent (CE)/g, respectively) than cultivars of the Durango race (7.9-8.3 TUI/mg and 16.8-19.9 CE/mg, respectively). Bayo Victoria, a Durango race cultivar, had three times more lectins than levels reported for soybean. Cultivar influenced TI and tannins contents (p < 0.001), whereas site affected lectins (p < 0.001). An increase in levels of TI and tannins could be enhanced through breeding. 相似文献

Fusarium mycotoxins are increasingly studied agronomically, chemically, and pathologically in the context of food safety, as a means of preventing new major health crises. Reliable mycotoxin techniques and sampling procedures are required for assessment of the effects of different sources of variation on grain mycotoxin content in agronomic experiments. Analyses were performed with the aim of formulating guidelines for grain sampling to increase the reliability of grain mycotoxin measurement in agronomic experiments. Two toxins in wheat samples, deoxynivalenol and nivalenol, were targeted. With a nested linear mixed model, it was estimated that the uncertainty of nivalenol determination was low (+/-15 microg/kg), whereas that for deoxynivalenol determination was higher (+/-38 microg/kg). It was also found that grinding of the grain decreased the variability of the results. Moreover, despite the heterogeneity in grain mycotoxin content across a given field, it was shown that heads can be harvested manually for agronomic experiments provided that sampling is representative (evenly distributed over the entire plot area). Finally, delaying the assay until after harvest was found to affect the results obtained and should therefore be avoided. 相似文献

Contamination of food products with pepsin resistant allergens is generally believed to be a serious threat to patients with severe food allergy. A sandwich type enzyme-linked immunosorbent assay (ELISA) was developed to measure pepsin resistant hazelnut protein in food products. Capturing and detecting rabbit antibodies were raised against pepsin-digested hazelnut and untreated hazelnut protein, respectively. The assay showed a detection limit of 0.7 ng/mL hazelnut protein or <1 microg hazelnut in 1 g food matrix and a maximum of 0.034% cross-reactivity (peanut). Chocolate samples spiked with 0.5-100 microg hazelnut/g chocolate showed a mean recovery of 97.3%. In 9/12 food products labeled "may contain nuts", hazelnut was detected between 1.2 and 417 microg hazelnut/g food. It can be concluded that the application of antibodies directed to pepsin-digested food extracts in ELISA can facilitate specific detection of stable proteins that have the highest potential of inducing severe food anaphylaxis. 相似文献

The differentiation between cacha?a and rum using analytical data referred to alcohols (methanol, propanol, isobutanol, and isopentanol), acetaldehyde, ethyl acetate, organic acids (octanoic acid, decanoic acid, and dodecanoic acid), metals (Al, Ca, Co, Cu, Cr, Fe, Mg, Mn, Ni, Na, and Zn), and polyphenols (protocatechuic acid, sinapaldehyde, syringaldehyde, ellagic acid, syringic acid, gallic acid, (-)-epicatechin, vanillic acid, vanillin, p-coumaric acid, coniferaldehyde, coniferyl alcohol, kaempferol, and quercetin) is described. The organic and metal analyte contents were determined in 18 cacha?a and 21 rum samples using chromatographic methods (GC-MS, GC-FID, and HPLC-UV-vis) and inductively coupled plasma atomic emission spectrometry, respectively. The analytical data of the above compounds, when treated by principal component analysis, hierarchical cluster analysis, discriminant analysis, and K-nearest neighbor analysis, provide a very good discrimination between the two classes of beverages. 相似文献

Microbial community structure in saltmarsh soils is stratified by depth and availability of electron acceptors for respiration. However, the majority of the microbial species that are involved in the biogeochemical transformations of iron (Fe) and sulfur (S) in such environments are not known. Here we examined the structure of bacterial communities in a high saltmarsh soil profile and discuss their potential relationship with the geochemistry of Fe and S. Our data showed that the soil horizons Ag (oxic–suboxic), Bg (suboxic), Cr1 (anoxic with low concentration of pyrite Fe) and Cr2 (anoxic with high concentrations of pyrite Fe) have distinct geochemical and microbiological characteristics. In general, total S concentration increased with depth and was correlated with the presence of pyrite Fe. Soluble + exchangable-Fe, pyrite Fe and acid volatile sulfide Fe concentrations also increased with depth, whereas ascorbate extractable-Fe concentrations decreased. The occurrence of reduced forms of Fe in the horizon Ag and oxidized Fe in horizon Cr2 suggests that the typical redox zonation, common to several marine sediments, does not occur in the saltmarsh soil profile studied. Overall, the bacterial community structure in the horizon Ag and Cr2 shared low levels of similarity, as compared to their adjacent horizons, Bg and Cr1, respectively. The phylogenetic analyses of bacterial 16S rRNA gene sequences from clone libraries showed that the predominant phylotypes in horizon Ag were related to Alphaproteobacteria and Bacteroidetes. In contrast, the most abundant phylotypes in horizon Cr2 were related to Deltaproteobacteria, Chloroflexi, Deferribacteres and Nitrospira. The high frequency of sequences with low levels of similarity to known bacterial species in horizons Ag and Cr2 indicates that the bacterial communities in both horizons are dominated by novel bacterial species. 相似文献
