To prevent, detect, and protect against forest fires, forest personnel need to define rules for determining forest fire risk. In Portugal, all municipalities must annually produce forest fire risk(FFR) maps. To produce more reliable FFR maps more easily, we developed an open source model using the Modeler plugin of SEXTANTE in the program QGIS version 2.0 Dufour. The model provides all the maps involved in the FFR model(susceptibility map, hazard map, vulnerability map, economic value map,and potential loss map) and was produced according to Portuguese Forest Authority's(AFN, Autoridade Florestal Nacional) rules for determining the FFR. This model was tested for the Portuguese municipality Santa Maria da Feira, where 40 % of the total municipality area falls in the category ‘‘very high' or ‘‘high' fire risk. The ‘‘very high'fire risk area is mainly classified as broad-leaved forest and has the steepest slopes(15 %). The distance of burned areas to roads was also analyzed; the proportion of burned areas increased with increasing distance to the main roads.In addition, 92.6 % of the ‘‘high' and ‘‘very high' risk zones were located in areas with lower elevation. These results confirmed that forest fire is strongly influenced not only by environmental factors but also by anthropogenic factors. The procedure implemented here was compared with our open source application already available in QGIS and also to the same procedure implemented in GIS proprietary software. Although the results were obviously the same, the model developed here presents several advantages over the other two approaches. Besides being faster,it is easy to change the model parameters according to user needs(i.e., to the rules of different countries), and can be modified and adapted to other variables and other areas to create risk maps for different natural phenomena(e.g.,floods, earthquakes, landslides). It has been postulated that osmotic effects on plant tissue are mediated by abscisic acid (ABA). Hybrid larch (Larix kaempferi (Lambert) Carr. x L. decidua Mill.) plantlets, transformed with the ABA-inducible wheat Em promoter associated with the Gus reporter gene, were axenically inoculated with two ectomycorrhizal fungi: Cenococcum geophilum Fr., considered tolerant to water stress, and Laccaria bicolor (Marie) Orton, considered less tolerant to drought. The mycorrhizal and non-mycorrhizal transgenic plantlets were subjected to osmotic stress by adding polyethylene glycol (PEG) to the culture medium. In the presence of PEG, L. bicolor and C. geophilum reduced shoot water potential and turgor potential, but increased host osmotic potential. Treatment of plantlets with PEG induced a significant increase in endogenous ABA concentrations. Laccaria bicolor and C. geophilum behaved similarly and significantly decreased the ABA response of plantlets to PEG treatment. Moreover, inoculation with either fungus regulated the ABA response of the plantlets even when the fungus was separated from the host by a cellophane sheet that prevented mycorrhiza formation. Although the wheat Em promoter was inducible in larch plantlets, it was not regulated by endogenous ABA. Induction of the wheat Em promoter in larch plantlets depended on organ type, with maximum induction in the root apex. Ovine and caprine stockbreeding have been gaining attention in developing countries as an attractive investment. On these animals, infectious diseases of the reproductive tract, such as leptospirosis, can compromise the production leading to economic losses. The present study aimed to determine the risk factors associated with incidental leptospirosis and its influence on the reproductive parameters of ewes and goats of Espírito Santo state, Brazil. A total of 737 animals distributed on 24 herds/flocks were studied, and an overall prevalence of 10.9 % seroreactive animals was observed. Serogroup Icterohaemorrhagiae was the most frequent in goats (97.0 %) as well as in ewes (78.3 %). Regarding risk factors related to leptospirosis, the presence of waterholes and the semi-intensive breeding system were the most important associated to seroreactivity. Besides, there was an observed association between seroreactivity and reproductive failures (P?<?0.05). Moreover, seroreactive ewes (relative risk (RR)?=?1.3) and goats (RR?=?1.9) presented more chances to have abortions than seronegative animals. Furthermore, seroreactive ewes presented 11.6 more chances of having premature births when compared to the seronegative ones. It can be concluded that Leptospira infection, mainly those caused incidental strains (such as Icterohaemorrhagiae serogroup), is a significant factor to reduce the productivity of small ruminants’ herds/flocks in the studied region, and environmental measures must be considered on control programs.
In the present study a technique was developed with the aim of guaranteeing the composition and security of fish meals, since it allows verification of whether these meals contain land animal remains. The method is based on polymerase chain reaction (PCR) and length polymorphism, followed by a restriction fragment length polymorphism (RFLP). Specific primers for every species were designed and calibrated, generating exclusively a PCR product with a specific size when DNA for each species was present in the sample. This technique allows the detection of land animal remains in fish meals, specifically cow, chicken, pig, horse, sheep, and goat. The identity of the PCR products can be confirmed by RFLP analysis using only one restriction enzyme. The selected restrictase generated one characteristic restriction profile for every species included in this study. The detection limit of this method was calculated by using mixtures of fish meals in different proportions and meal that exclusively contained remains of one of these land species studied. The analytical strategy herein proposed was applied to fish and meat meals, giving good results, both in the analyzed standards and in commercial samples. 相似文献
Protein fractions from transgenic Bt and non-transgenic maize varieties, extracted by the Osborne solvent fraction procedure, were characterized for the first time by perfusion and monolithic RP-HPLC in very short analysis times. Albumins and globulins from different transgenic Bt maizes as well as from their non-transgenic isogenic varieties were eluted in four peaks using perfusion RP-HPLC, whereas prolamins and glutelins were separated in seven peaks. Monolithic RP-HPLC enabled the separation of maize proteins in a large number of peaks showing 6 and 10 main peaks for albumins and globulins, respectively. Prolamins migrated at retention times higher than 5 min as seven peaks, whereas glutelins were separated in three main peaks appearing at retention times higher than 6.0 min. Moreover, chromatograms of the whole protein extracts showed 8 and 11 components for perfusion and monolithic RP-HPLC, respectively. A comparison of the chromatograms of the whole protein extracts relative to transgenic and non-transgenic varieties evidenced quantitative differences on the percentages of area, mainly for peaks 2 and 3 by perfusion RP-HPLC and for peaks 3 and 7 by monolithic RP-HPLC. A discriminant analysis based on these proteic profiles was carried out to classify and predict transgenic Bt maize lines, achieving 100% correct classification using perfusion RP-HPLC. 相似文献
