The present investigation deals with purification and thermal characterization of an acid invertase produced by Fusarium solani in submerged culture. The maximum enzyme activity (9.90 U mL(-1)) was achieved after 96 h of cultivation at pH 5.0 and 30 degrees C in a basal medium containing molasses (2%) as the carbon and energy source supplemented with 1% peptone. Invertase was purified by ammonium sulfate fractionation and column chromatography on DEAE-cellulose and Sephadex G-200. The purified enzyme was proven to be homogeneous by sodium dodecyl sulfate polyacrylamide gel electrophoresis. The molecular mass of the enzyme was 65 kDa. The optimum pH and temperature for activity were 2.6 and 50 degrees C, respectively. The Km value for sucrose was 3.57 mM with an activation energy of 4.056 kJ mol(-1). Enthalpies of activation (DeltaH) were decreased while entropies (DeltaS) of activation increased at higher temperatures. The effects of alpha-chymotrypsin and 4 M urea were tetraphasic with periodic gain and loss of enzyme activity. A possible explanation for the thermal inactivation of invertase at higher temperatures is also discussed.  相似文献

Much effort has been invested in identifying molecular markers in wheat (Triticum aestivum L.) linked to quantitative trait loci (QTL) that confer resistance to Fusarium head blight (FHB) caused by Fusarium graminearum Schwabe [teleomorph Gibberella zeae (Schwein) Petch]. Even after several generations of crossing and selection by many wheat breeding programs, resistance of the Chinese spring wheat cultivar ‘Sumai 3’ (PI 481542) remains among the most effective. It therefore seems that undocumented resistance QTL present in Sumai 3 were not detected in various mapping studies. Using an extremely susceptible Tibetan landrace (‘Y1193-6’; unknown pedigree) in the creation of a mapping population with Sumai 3, the objective of this research was to identify undocumented resistance QTL in Sumai 3. This was accomplished through collecting disease index (DI) and Fusarium damaged kernel (FDK) phenotypic values along with 305 Diversity Array Technology (DArT) and 52 Simple Sequence Repeat (SSR) marker genotypes on 160 F2:6 recombinant inbred lines (RILs). Disease response evaluations were based on four (two greenhouse and two field) experiments where spray inoculation methods were used. Three QTL were identified on chromosome arms 3BS, 6BL and 2DS explaining 26.1, 10.7 and 18.9% of the phenotypic variation for DI, respectively. The same QTL were also significantly associated with reduced FDK scores and explained 28.0, 11.0 and 23.0% of phenotypic variation. Lines within the mapping population were placed in eight categories with respect to their various QTL combinations. Lines with no QTL were the most susceptible, whereas those with the Sumai 3-derived 3BS and 6BL QTL combined with the 2DS QTL from Y1193-6 were the most resistant. Though the 3BS and 6BL QTL are well-documented, the 2DS resistance QTL, which was contributed by the susceptible parent, confers increased susceptibility when derived from Sumai 3. In this study no new FHB QTL from Sumai 3 was discovered, but results suggest that Sumai 3 contains a QTL for susceptibility on chromosome arm 2DS. Selection against this QTL may potentially increase resistance levels among Sumai 3-derived populations.  相似文献

The anti-tick efficacy of combined aqueous herbal extracts of Azadirachta indica leaves, Nicotiana tabacum leaves, Calotropis procera flowers and Trachyspermum ammi seeds was evaluated using adult immersion test, larval packet test and ear bag method. The extract exhibited lethal effects on egg laying (index of egg laying=0.371404±0.00435), hatching (22.35%) and total larval mortality at 50 mg ml(-1) and reduced tick intensity on the infested calves (18 detached out of 35 at 45% (w/w) suspension, topically applied). The herbal extract exerted dose- and time-dependent response against all the developmental stages of Rhipicephalus (Boophilus) microplus considered in this study, thus justified their use in the traditional system of Pakistan.  相似文献

Objective-To determine whether a novel optimized plasmid carrying the porcine growth hormone-releasing hormone (GHRH) wild-type cDNA administered at a lower dose was as effective at eliciting physiologic responses as a commercial GHRH plasmid approved for use in Australia. Animals-134 gilts. Procedures-Estrus was synchronized and gilts were bred. Pregnant gilts were assigned to 2 treatment groups (40 gilts/group) or 1 untreated control group (24 gilts). Gilts in one of the treatment groups received the commercial GHRH plasmid, whereas gilts in the other treatment group received a novel optimized GHRH plasmid; both plasmids were administered IM in the right hind limb, which was followed by electroporation. Sow and litter performance were monitored for the 3 gestations after treatment. Results-A significant increase in insulin-like growth factor-I concentrations, decrease in perinatal mortality rate, increase in the number of pigs born alive, and increase in the weight and number of pigs weaned were detected for both groups receiving the GHRH-expressing plasmids, compared with values for the control group. Additionally, there was a significant decrease in sow attrition in GHRH-treated females, compared with attrition in the control group, during the 3 gestations after treatment. Conclusions and Clinical Relevance-Both of the GHRH plasmids provided significant benefits for sow performance and baby pig survivability for pregnant and lactating sows and their offspring during the 3 gestations after treatment, compared with results for untreated control gilts. Use of a novel optimized plasmid reduced the effective plasmid dose in these large mammals.  相似文献

A total of 250 one-day-old Ross 308 male chicks were used to investigate the effects of Fe-methionine (Fe-met) and iron sulfate (FeSO4.7H2O) on performance, mineral concentration, meat quality, and oxidative stability of breast. The feeding experiment was performed with five dietary groups including group I (basal diet [BD] + 80 mg/kg FeSO4), group II (BD + 60 mg/kg FeSO4 + 20 mg/kg Fe-met), group III (BD + 40 mg/kg FeSO4 + 40 mg/kg Fe-met), group IV (BD + 20 mg/kg FeSO4 + 60 mg/kg Fe-met), and group V (BD + 80 mg/kg Fe-met). The results showed that Fe and Cu content in the breast muscle increased by dietary supplementation of organic and inorganic iron from II to V groups. Dietary groups of II, IV, and V increased L* value and b* value of breast meat. Additionally, redness of breast meat enhanced linearly and quadratically by dietary supplementation of 80 mg/kg Fe as FeSO4. In addition, dietary supplementation Fe from II to V groups led to a significant increase in pHu at 24-h postmortem while it was negatively correlated with L* value of breast muscle. This study demonstrates that replacing FeSO4 with Fe-Met could improve breast meat yield of broilers. Although 80 mg of Fe-Met improved the iron content of meat, it showed detrimental effects on the lipid oxidation of breast meat.  相似文献

Multiple seeded cocklebur has been found in the last decade in Texas, and described as a biotype of Xanthium strumarium L with up to 25 seeds per bur instead of the usual two. The multiple seeded bur typically produces up to nine seedlings, causing concern that it may be harder to control than normal seeded common cocklebur. The efficacies of a series of fungal and conventional commercial herbicides have been compared in the greenhouse on seedlings of multiple seeded cocklebur from Texas (MSC-TX) and normal common cockleburs from Texas (NCC-TX), Arkansas (NCC-AR), Illinois (NCC-ILL) and two from Mississippi (NCC-MS#1, NCC-MS#2). Three measures of herbicidal activity (reductions in plant height and dry weight, and mortality) were used. The fungal herbicide Alternaria helianthi (Hansf) Tubaki & Nishihara at 1 x 10(5) conidia ml(-1) + 2 g liter(-1) Silwet L-77 with an 8-h dew period was an effective herbicide with all biotypes, as were the commercial chemical herbicides chlorimuron (14.8 g ha(-1)), imazaquin (29.6 g ha(-1)), sodium hydrogen methylarsonate (MSMA; 279.1 g ha(-1)) and imazethapyr (39.5 g ha(-1)). The membrane-disrupting organic arsenical MSMA was effective with all biotypes, whereas commercial chemical herbicides which act by inhibiting branched-chain amino acid synthesis (chlorimuron, imazaquin and imazethapyr) were less effective against normal seeded common cocklebur biotypes with short stature. These studies showed that multiple seeded cocklebur was at least as susceptible to the biological agent A helianthi and to the conventional commercial herbicides studied as were normal seeded cockleburs, suggesting that existing methods should be adequate to control this novel biotype.  相似文献

A cross-sectional study was conducted from March to August 2006 in dairy herds in Fars province, southern Iran to determine the herd-level risk factors for infection with Mycobacterium avium subspecies paratuberculosis (MAP). Statistical analysis using multivariable logistic regression showed that contamination of udders of periparturient cows with manure (OR = 6.4, P = 0.02) and history of having suspected cases of Johne's disease in the herd (OR = 6.7, P = 0.04) were significantly associated with the herd infection status. No relationship between breed, herd size and other management practices with the infection status of the herd were found in this study. Implementing high sanitary measures in the farm, particularly with respect to manure handling and cleaning could be considered as one of the important aspects in controlling disease in the region as well as in the future educational effort.  相似文献

Subclinical ketosis (SCK) is simply a condition marked by increased levels of circulating ketone bodies without the presence of the clinical signs of ketosis. Subclinical ketosis can cause economic losses through decreased milk production and association with preparturient diseases. Limited information is available regarding the prevalence of SCK in dairy herds in Southwestern Iran. The objectives of this study were (i) determination of the cutoff point of nonesterified fatty acids (NEFAs) and glucose concentrations for diagnosis of SCK using receiver operating characteristic (ROC) analysis, and (ii) determination of prevalence of subclinical ketosis in apparently healthy dairy cattle in Southwestern Iran. From October to December 2009, a total of 100 clinically healthy multiparous Holstein cows (3-8 years old) were randomly selected from 16 dairy herds around Kazerun, Fars Province, Iran. The cows had two-six lactations, with body weight ranging from 500 to 650 kg. Blood samples for each cow were taken at 2, 4 and 6 weeks post parturition and 3-4h after the morning feeding. The optimal cutoff point was set, by the ROC method, to >0.26 mmol/L for NEFA, and < 2.26 mmol/L for glucose with corresponding 82.54% sensitivity and 91.89% specificity for NEFA and 44.44% sensitivity and 78.38% specificity for glucose. Cows with BHB concentrations higher than 1200 μmol/L were classified as having SCK. In 2, 4 and 6 weeks post parturition 63%, 68% and 59% of the tested cows were subclinically ketotic. Overall, 97% of tested cows (97/100) were considered subclinically ketotic in at least one sample period. Thirty percent of tested cows (30/100) suffered from subclinical ketosis in all of the 2, 4 and 6 weeks postpartum. The results suggest that, a cut-off point of 0.26 mmol/L for NEFA concentrations can be used during early lactation for diagnosis of subclinical ketosis and making management decisions for prevention and treatment. Glucose cannot be a good criterion for diagnosis of SCK and it does not appear to be useful for monitoring subclinical ketosis.  相似文献

A wheat (Triticum aestivum L.) recombinant inbred line (RIL) population was used to identify quantitative trait loci (QTL) associated with yield, yield components, and canopy temperature depression (CTD) under field conditions. The RIL population, consisting of 118 lines derived from a cross between the stress tolerant cultivar ‘Halberd’ and heat stress sensitive cultivar ‘Karl92’, was grown under optimal and late sown conditions to impose heat stress. Yield and yield components including biomass, spikes m?2, thousand kernel weight, kernel weight and kernel number per spike, as well as single kernel characteristics were determined. In addition, CTD was measured during both moderate (32–33 °C) and extreme heat stress (36–37 °C) during grain-filling. Yield traits showed moderate to high heritability across environments with a large percentage of the variance explained by genetic effects. Composite interval mapping detected 25 stable QTL for the 15 traits measured, with the amount of phenotypic variation explained by individual QTL ranging from 3.5 to 27.1 %. Two QTL for both yield and CTD were co-localized on chromosomes 3BL and 5DL and were independent of phenological QTL. At both loci, the allele from Halberd was associated with both higher yield and a cooler crop canopy. The QTL on 3BL was also pleiotropic for biomass, spikes m?2, and heat susceptibility index. This region as well as other QTL identified in this study may serve as potential targets for fine mapping and marker assisted selection for improving yield potential and stress adaptation of wheat.  相似文献
