A karyotype study was performed on 15 populations of Eremostachys laciniata Bunge from Iran in order to quantify the extent of cytological variation for use in breeding programmes. Ten mitotic root tip cells at the metaphase stage were prepared from each population by the squash method. The chromosomes of suitable mitotic cells were counted and various parameters, including long and short arm lengths, chromosome lengths, total length of the haploid chromosome complement, arm ratios, and the centromeric index, were measured. All 15 populations were diploid with 2n = 2x = 22 and were differentiated by their karyotypic parameters. All 15 E. laciniata populations occupied Class 1A of Stebbins’ karyotype classification, indicating the presence of a primitive symmetrical karyotype. The size of the mitotic chromosomes was medium, and mean chromosome lengths ranged from 3.84 to 4.77 μm. The shortest chromosome lengths were observed in the Ajabshir population and the longest in the Heydarabad population. The Heydarabad population also had the longest total chromosome length and the Ajabshir population had the shortest total chromosome length. The centromeric indices of the chromosome complements varied from 39.3% to 43.7%. Metacentric chromosomes were the most common in all populations, whereas submetacentric chromosomes were rare. The A1 index varied from 0.215 (Areshtanab) to 0.337 (Til). The A2 index ranged from 0.116 (Til) to 0.143 (Kaleybar), and the karyotype asymmetry index varied between 0.051 and 0.097. The results of a principal component analysis (PCA) showed that the main elements of the first three main principal components accounted for 97% of the total variance. Cluster analysis by Ward’s method indicated that all 15 populations could be classified into three clusters. Finally, the results of the karyotypic parameters showed that the Til, Heydarabad, and Marand populations had higher levels of karyotypic heterogeneity and may be useful when developing breeding programmes.  相似文献

Apple breeding is usually done to produce higher quality apples. We crossed two late ripening high quality apple cultivars (i.e., Golden Smoothie and Red Spur) as maternal parents with two Iranian domestic early ripening apple cultivars (i.e., Golab-e Kohanz and Shafi Abadi) as pollen parents in 2005 to breed new early ripening apple cultivars. We then measured qualitative and quantitative traits of 360 off spring during 2010 and 2011. Results of recorded traits on the 5th and 6th year of growth of the progeny showed that during two evaluation years, flowering and fruit ripening time, titrable acidity, SSC/acid ratio, and fruit weight had the highest coefficients of phenotypic variations. Progenies were classified in three groups: early, mid, and late ripening. Some of the selected progeny were better than their early ripening parental cultivars being earlier in the ripening, with larger fruit size and better texture and flavor. Genotypes A31, A32, I5, I23, and I57 were in the early ripening category, B11 and A34 were in the middle, and B77 and E21 were in the late ripening categories. These characteristics were considered to present an improvement.  相似文献

To find out more about the biochemical aspects of seed-softness trait in pomegranate (Punica granatum L.), some of the phenylpropanoid pathway enzymes as well as their products were evaluated in the various soft- and hard-seeded genotypes during fruit development. Results showed that peroxidase (POD) activity decreased in all studied genotypes during fruit growth, while increasing at the last stages only in the hard-seeded genotypes. Although laccase (LAC) activity did not show high differences during fruit ripening and in different genotypes, but its general trend was increasing. Phenylalanine ammonia-lyase (PAL) activity decreased during the fruit development and approximately diminished for the hard-seeded genotypes. All genotypes had lignin components in their seeds and amount of lignin increased during the growth season. Total phenolic decreased drastically and antioxidant capacity increased gradually showing an opposite trend and probable competitive effect for the same substrate during fruit development. Considerable differences that were observed in the POD activity at the last sampling stage, and higher activity that was observed in POD than LAC, may indicate that POD is more important for polymerization of lignin in the pomegranate seed and this enzyme might have some role in the development of seed softness trait in pomegranate. Also, this is the first report about lignin detection in the seeds of soft-seeded pomegranate genotypes, and represents a different mechanism (such as monolignol conformation in the final polymer) from lignin content for development of the soft-seeded pomegranate genotype.  相似文献

In order to evaluate wheat response to yellow rust, 25 advanced, promising and commercial bread wheat cultivars were tested as seedlings in greenhouse conditions in Karaj, Iran, and as adult plants in field conditions at four locations. Five pathotypes of yellow rust, 14E176A+, 134E142A+, 6E210A+, 4E128A- and 64E146A+ prevailing in field test locations, were used in the seedling tests. The results showed that some of the cultivars have seedling or overall resistance to the pathotypes and some have adult plant resistance. Cultivars M-70-4 and MV17 were resistant to all pathotypes as seedlings and showed good adult plant resistance. The main aim of the present study was to assess the effects of chronic high stocking density on liver proteome of rainbow trout. Rainbow trout juveniles (42.6 ± 2.3 g average body weight) were randomly distributed into six tanks at two stocking densities (low stocking density (LD) = 20 kg m?3 and high stocking density (HD) = 80 kg m?3). Both treatments were performed in triplicate tanks for a period of 60 days. High stocking density caused a reduction in the growth performance compared with LD fish. Lysozyme activity increased with stocking density, while serum complement activity presented the opposite pattern. Serum cortisol and total protein levels did not show significant differences (P > 0.05) between experimental groups. The fish reared at high stocking density showed significantly lower osmolality and globulin values but higher albumin level. The HD group had significantly higher activities of catalase, glutathione peroxidase and superoxide dismutase, and malondialdehyde content in the liver when compared to the LD group. Comparative proteomics was used to determine the proteomic responses in livers of rainbow trout reared at high stocking density for 60 days. Out of nine protein spots showing altered abundance (>1.5-folds, P < 0.05), eight spots were successfully identified. Two proteins including apolipoprotein A-I-2 precursor and mitochondrial stress-70 protein were found to increase in HD group. The spots found to decrease in the HD group were identified as follows: 2-peptidylprolyl isomerase A, two isoforms of glyceraldehydes-3-phosphate dehydrogenase, an unnamed protein product similar to fructose-bisphosphate aldolase, 78 kDa glucose-regulated protein, and serum albumin 1 protein.  相似文献

The objective of this work was to study the relationship between female size (weight) and variables of egg and larval stage of Persian sturgeon Acipenser persicus . In this study, 19 female breeders were captured in Caspian Sea and fertilized by routine methods. Positive significant correlations ( P <0.05) were established between female weight and ovulated eggs per female, time of second mitosis division and volume of yolk-sac at hatching. There was not significant correlation ( r =0.33, P =0.161) between female weight and egg diameter. Female weight was not affected weight of larvae at hatching time ( r =0.37, P =0.119), as well as larval length ( r =−0.14, P =0.558) and larval weight at the end of the experiment (48 hours after first feeding) ( r =0.16, P =0.491). Mortality rate during yolk-sac absorption was higher with increased female weight but their correlation was not significant ( r =0.40, P =0.076). During the first feeding stage, mortality rate was 13.39% and there was no significant correlation between mortality rate in this period and female weight ( r =−0.12, P =0.613). Conclusively, as a result female size influenced fecundity, time of second mitosis division and yolk-sac volume at hatching time without affecting mortality rate during yolk-sac absorption and first feeding stage in Persian sturgeon. Thus, smaller female broods do not cause more mortality than larger ones in larval production and they can be used in reproduction procedure.  相似文献

The aim of this study was to evaluate the long‐term effects (7‐month experiment) of diets consisting of fish oil (Kilka fish) and vegetable oil (rapeseed) on the reproductive performance of sterlet sturgeon (Acipenser ruthenus) broodstock. Forty‐five broodstock (990.3 ± 20.05 g) were randomly allocated to three different diet treatments. Three experimental diets were formulated with graded levels of fish oil (100% FO), vegetable oil (100% VO), and a combination of fish and vegetable oil (50% FO + 50% VO). At the end of the 7‐month feeding trial period, the weight gain and final weight were changed significantly different between the treatments (p < 0.05). Broodstock fed the FO + VO diet had higher growth than those fed the only FO or VO diets (p < 0.05). The highest germinal vesicle migration percentage was observed in FO + VO treatment (p < 0.05). The DHA/EPA, DHA/ARA and EPA/ARA ratios in oocyte exhibited a significant difference in the different treatments (p < 0.05). This study indicates that nutrition of broodstock with diet including FO + VO (p < 0.05) can positively affect the growth performance of larvae compared with only FO or VO diets. Furthermore, the high levels of 18:1n‐9, AL and ALA contents in oocytes from broodstock fed VO and the lowest ALA content in oocytes from broodstock fed FO underlined the important role of broodstock diets in the reproductive process and embryonic and/or larval developments of sterlet.  相似文献

In this study, the effect of maternal age on prepupae diapause of progeny in sexual (Wolbachia free) and asexual (Wolbachia infected) Trichogramma brassicae was studied at two temperatures. Maternal generation (G1) of both populations was reared at 10L:14D photoperiod, 14 and 20 °C, separately. Mated females from G1 were supplied with eggs of Ephestia kuehniella, each 48 h from one day after emergence to death. The tendency to diapause in progeny (G2) was determined by rearing at 10 °C. According to the results, reproductive modes, maternal temperature and age have significant effect on the mean number of eggs laid by G1 and the diapause percentage of G2. Offspring whose maternal generation was reared at 20 °C had higher diapause percentage than those reared at 14 °C. Diapause percentage in sexual population was higher than the asexual, which demonstrates a negative effect of Wolbachia on this process. Wolbachia infection modifies the effect of maternal age on the diapause percentage in the offspring at 20 °C but not at 14 °C. The result can be due to lower Wolbachia density at 14 °C and greater Wolbachia density at 20 °C.  相似文献
