Type material of the proteocephalidean cestodes Manaosia bracodemoca Woodland, 1935 and Paramonticellia itaipuensis Pavanelli et Rego, 1991 (both monotypic genera) as well as recently collected material of the latter species are redescribed. A close similarity between both species was observed: the shape of scoleces is identical, both possess a globular scolex with hidden suckers, a well-developed circular, horseshoe-shaped musculature surrounding suckers; the measurements of both taxa are also similar (scolex, suckers, young proglottides). Both cestodes possess a medullary ovary crossing the dorsal muscle layer and ending in the dorsal cortex, the testes are in the dorsal cortex and the cortical vitellarium is laterally situated. Both species parasitize the same fish host. Therefore Paramonticellia is considered a junior synonym of Manaosia and Paramonticellia itaipuensis becomes a junior synonym of Manaosia bracodemoca. An emended diagnosis of Manaosia, which is a monotypic genus, is provided.  相似文献

Ethoxylates of rapeseed oil and of methylated rapeseed oil were synthesized and tested as adjuvants for 2,4-D and phenmedipham. Provided they had less than 6 units of ethylene oxide (EO), 1.0 to 10 g litre(-1) ethoxylates in water induced droplet spreading on barley leaves. In an acetone-based medium all derivatives strongly promoted the foliar uptake of 2,4-D, with no clear influence of the ethoxylation degree. In the same medium there was a negative influence of ethoxylate chain length on the foliar uptake of phenmedipham. In a water-based medium, phenmedipham applied with rapeseed oil emulsified with ethoxylated (20 EO) rapeseed oil displayed uptake rates close to a commercial preparation. The same was true for phenmedipham applied with ethoxylated (2 EO) methylated rapeseed oil. In bioassays, phenmedipham prepared with methylated rapeseed oil emulsified with ethoxylated (20 EO) rapeseed oil was as efficacious on barley as a commercial formulation. The same was true for phenmedipham prepared with ethoxylated (2 EO) methylated rapeseed oil. However, neither rapeseed oil nor methylated rapeseed oil emulsified with ethoxylated (2 EO) methylated rapeseed oil conferred good efficacy to phenmedipham. Hence, ethoxylated rapeseed oil derivatives are promising adjuvants or formulants for herbicides.  相似文献

Proteinase inhibitor activity had increased sharply in melon seedlings infected by Colletotrichum lagenarium 3 days after inoculation. The activity was associated with heat stable proteins and was effective against the protease produced by the fungus as well as against trypsin. Treatment of healthy melon leaves with an elicitor of ethylene isolated from the pathogen, resulted in a three-fold increase in proteinase inhibitor activity after 24 h. Ethylene production increased early in elicitor-treated leaves and may be involved in the elicitation of proteinase inhibitor activity. In the presence of aminoethoxyvinylglycine, an inhibitor of ethylene biosynthesis, both elicitor-induced ethylene and, to a lesser extent, elicitor-induced proteinase inhibitor activity were inhibited. In contrast, 1-aminocyclopropane-1-carboxylic acid, the direct precursor of ethylene, triggers proteinase inhibitor activity. It is concluded that ethylene is involved in the elicitation of proteinase inhibitor activity, but its exact role remains to be defined.  相似文献

Phoma macdonaldii, the causal agent of black stem disease of sunflower (Helianthus annuus), also attacks roots and collars of the plants, resulting in early death. Totally resistant lines do not exist for infection of the aerial parts, but tolerant lines have been characterised. This paper presents a study on colonisation of a partially resistant and a susceptible sunflower line by P. macdonaldii. The fungus was transformed with a constitutively expressed reporter gene encoding the jellyfish green fluorescent protein via Agrobacterium tumefaciens, and colonisation of sunflower roots by this transformed strain was studied by various microscopy techniques including confocal and scanning electron microscopy. The results show that penetration of the fungus into the root occurred through natural fissures or through the epidermis and was similar in both lines. In contrast, the colonisation rate of the stele was reduced in the partially resistant line, and the morphology of the fungal hyphae was also affected. The effect on hyphal morphology was strongest in the stele, indicating a localised production of defence compounds in this line.  相似文献

The examination of the type series of Ophiotaenia Gallardi (Johnston, 1911) (syn. Proteocephalus gallardi Johnston, 1911) revealed that it is a mixture of two species of different genera. Lectotype of Ophiotaenia gallardi is designated and the species is redescribed on the basis of it, conspecific paralectotypes and additional materials. The remaining part of the type series belongs to Vandiermenia gen. n. (Acanthotaeniinae), with V Beveridgei sp. n. as the type- and only species. The new genus differs from all other acanthotaeniine genera, i.e. Rostellotaenia Freze, 1963, Acanthotaenia von Linstow, 1903 and Kapsulotaenia Freze, 1963, by the presence of cortical uterine stem and paramuscular vitelline follicles, particular structure of the internal longitudinal musculature (absent laterally and more developed than in the three above-mentioned genera) and testes limited in two fields separated medially. Type series of Ophiotaenia mjobergi (Nybelin, 1917) (syn. Crepidobothrium mjobergi Nybelin, 1917), O. amphiboluri (Nybelin, 1917) (syn. Crepidobothrium amphiboluri Nybelin, 1917), O. striata (Johnston, 1914) (syn. Acanthotaenia striata Johnston, 1914) and O. longmani Johnston, 1916 are revised and compared with Ophiotaenia gallardi. Australotaenia hylae (Johnston, 1912) comb. n. is proposed for Ophiotaenia hylae Johnston, 1912. Australotaenia gen. n. differs from the remaining genera of the subfamily Acanthotaeniinae by (1) the Type 2 of the formation of the uterus (sensu de Chambrier et al. 2004) (all the other acanthotaeniines have the Type 1 of uterine development), (2) the cortical position of the uterine stem (all the other genera have medullary uterine stem) and (3) the morphology of the internal longitudinal musculature, which is composed of few well-developed bundles of fibres (in contrast to the other genera). The new genus also differs from ?by eggs not in clusters, the presence of two testicular fields (versus one in Vandiermenia) and the structure of the longitudinal internal musculature with only 8-10 bundles (versus formed by numerous bundles and with the presence of secondary muscles in Vandiermenia). Ophiotaenia sp. sensu de Chambrier (2004), a parasite of Litoria moorei, is described as Australotaenia grobeli sp. n., which can be distinguished from Australotaenia hylae by the smaller number of testes (46-76 versus 74-106), greater cirrus-sac length/width of proglottis ratio (27-33% versus 17-19%) and the smaller ovary width/proglottis width ratio (55-63% versus 68-71%).  相似文献

Many studies already investigated marker-assisted selection (MAS) efficiency but mainly in biparental populations. Connected multiparental populations address a broader diversity and confer a gain of power for QTL detection which must be beneficial for MAS. Our objective was to compare multiparental connected designs to biparental populations taken separately for MAS and phenotypic selection. We first detected QTL for flowering time and grain yield in an experimental maize design involving four parental inbred lines crossed to produce six different biparental populations and confirmed the advantage of multiparental connected designs over biparental populations for QTL detection. Based on these results we performed stochastic simulations to evaluate the expected efficiency of four generations of MAS and phenotypic selection. Different parameters were considered: trait heritability, genetic architecture and whether QTL were assumed to be known or have to be detected. Genetic gains were higher in the multiparental design than on average over the biparental populations considered separately, especially when favourable alleles were equally distributed among parental lines. When QTL detection was included in the simulation process, we found that type I error risk considered for declaring QTL as significant should be adapted to the design. Type I error risks leading to the best response were higher for the biparental populations than for the multiparental design. Besides addressing a broader diversity, multiparental designs increase the power of QTL detection, which reinforces their superiority over biparental designs for MAS. Application of MAS to multiparental designs therefore appears promising to accelerate genetic gain in plant breeding programs.  相似文献

Context

Herbicide treatments in viticulture can generate highly contrasting mosaics of vegetated and bare vineyards, of which vegetated fields often provide better conditions for biodiversity. In southern Switzerland, where herbicides are applied at large scales, vegetated vineyards are limited in extent and isolated from one another, potentially limiting the distribution and dispersal ability of organisms.

Objectives

We tested the separate and interactive effects of habitat amount and fragmentation on invertebrate abundance using a multi-scale framework, along with additional environmental factors. We identified which variables at which scales were most important in predicting patterns of invertebrate abundance.

Methods

We used a factorial design to sample across a gradient of habitat amount (area of vegetated vineyards, measured as percentage of landscape PLAND) and fragmentation (number of vegetated patches, measured as patch density PD). Using 10 different spatial scales, we identified the factors and scales that most strongly predicted invertebrate abundance and tested potential interactions between habitat amount and fragmentation.

Results

Habitat amount (PLAND index) was most important in predicting invertebrate numbers at a field scale (50 m radius). In contrast, we found a negative effect of fragmentation (PD) at a broad scale of 450 m radius, but no interactive effect between the two.

Conclusions

The spatial scales at which habitat amount and fragmentation affect invertebrates differ, underpinning the importance of spatially explicit study designs in disentangling the effects between habitat amount and configuration. We showed that the amount of vegetated vineyards has more influence on invertebrate abundance, but that fragmentation also contributed substantially. This suggests that efforts for augmenting the area of vegetated vineyards is more beneficial for invertebrate numbers than attempts to connect them.

Because of their highly polymorphic shell patterns, Cepaea land snails have been the subject of numerous studies in ecological genetics. Here, we investigated the spatial structure of polychromatism in Cepaea hortensis in agricultural landscapes with zones from low to high hedgerow densities. Our main purpose was to search for a relationship between landscape composition and spatial structuring of chromatism. Despite significant spatial heterogeneity in the three landscapes sampled, only the high hedgerow density landscape showed a significant spatial structuring of shell polymorphism. In order to understand this result, an investigation of daily movement patterns in relation to habitat form was carried out on a mark-release experiment under semi-artificial conditions. This experiment revealed a strong influence of a linear corridor on snail dispersal. In the field, spatial heterogeneity of shell polymorphism, related to the effects of genetic drift, was shaped by restricted dispersal in narrow corridors. In the more enclosed one, i.e. where hedgerow density was the highest, the significant spatial structure we detected involved a balance between local genetic drift and environmentally mediated gene flow. This isolation-by-distance pattern resulted from direct gene exchange through fields between neighbouring populations. When applying landscape distances based on hedgerow length, no significant spatial correlation with polychromatism was found. In the more fragmented sites, random genetic drift seemed to be the prevailing force and, at the scale of the whole sampled area, selective pressures potentially interfere with these genetic drift-dispersal events.  相似文献

Background

Efficient high throughput screening systems of useful mutants are prerequisite for study of plant functional genomics and lots of application fields. Advance in such screening tools, thanks to the development of analytic instruments. Direct analysis in real-time (DART)-mass spectrometry (MS) by ionization of complex materials at atmospheric pressure is a rapid, simple, high-resolution analytical technique. Here we describe a rapid, simple method for the genetic discrimination of intact Arabidopsis thaliana mutant seeds using metabolic profiling by DART-MS.

Results

To determine whether this DART-MS combined by multivariate analysis can perform genetic discrimination based on global metabolic profiling, intact Arabidopsis thaliana mutant seeds were subjected to DART-MS without any sample preparation. Partial least squares-discriminant analysis (PLS-DA) of DART-MS spectral data from intact seeds classified 14 different lines of seeds into two distinct groups: Columbia (Col-0) and Landsberg erecta (Ler) ecotype backgrounds. A hierarchical dendrogram based on partial least squares-discriminant analysis (PLS-DA) subdivided the Col-0 ecotype into two groups: mutant lines harboring defects in the phenylpropanoid biosynthetic pathway and mutants without these defects. These results indicated that metabolic profiling with DART-MS could discriminate intact Arabidopsis seeds at least ecotype level and metabolic pathway level within same ecotype.

Conclusion

The described DART-MS combined by multivariate analysis allows for rapid screening and metabolic characterization of lots of Arabidopsis mutant seeds without complex metabolic preparation steps. Moreover, potential novel metabolic markers can be detected and used to clarify the genetic relationship between Arabidopsis cultivars. Furthermore this technique can be applied to predict the novel gene function of metabolic mutants regardless of morphological phenotypes.  相似文献
