A microplate agglutination test (MAT) was compared with the tube agglutinin test (TAT), a standard test for the diagnosis of Brucella canis, in terms of the sensitivity and specificity. The results showed that MAT was more sensitive, simpler to perform and easier to read the results than TAT. On top of that the MAT allows us to handle a larger number of samples at once. Using this method we conducted sero-surveillance of the prevalence of B. canis in dogs kept in an Animal Shelter located in Kanagawa Prefecture. Twelve of 485 (2.5%) showed seropositive against B. canis. These results indicate that B. canis infection in dogs is still occurring in Japan.  相似文献

Granulocyte colony-stimulating factor (G-CSF) is widely used to mobilize peripheral blood stem cells, and expected to restore cardiac function for patients with coronary artery diseases as a consequence of progression of atherosclerosis. Safety issues related to the administration of G-CSF to these patients, however, are still under study. The animal model for atherosclerosis was produced by feeding miniature swine a high-cholesterol diet for 3 months. G-CSF (5 or 10 microg/kg/day) was given to the animal model by daily subcutaneous injections for 10 days and 20 main arteries were evaluated pathologically. In addition, the general toxicological effects were studied on clinical signs, body weight, hematology, blood chemistry and pathology. In the G-CSF-treated groups, a variety of changes related to the major pharmacological activity of G-CSF including an increase in white blood cell (WBC) counts were observed. In many arteries, atherosclerotic lesions similar to Type I-V of the proposed classification by the American Heart Association were observed. No effects of the G-CSF treatment were seen on the histopathological findings, incidence, severity or distribution of atherosclerotic lesions. In addition, no infiltration of neutrophils to the lesions was observed. These findings suggest that the administration of G-CSF causes neither exacerbation or modification of atherosclerotic lesions nor adverse changes despite that a sufficient increase in WBC counts could be achieved in the peripheral blood.  相似文献

The aim of our study was to investigate the effects of suckling on reproductive performance and metabolic status of obese (mean body condition score of more than 4.0 on a scale of 1-5) maternal Japanese Black cows during early postpartum period. We used 7 postpartum Japanese Black cattle. Four cows were suckled ad libitum (suckled) until completion of their first artificial insemination (AI), while 3 cows were not suckled at all because they were separated from their calves immediately after parturition (non-suckled). Body weight and plasma concentrations of metabolites and hormones were measured from wk 1 to 9 postpartum. Ovarian activity was detected using plasma progesterone concentration, and all cows received their first AI after application of the Ovsynch protocol at approximately 4 months postpartum. Although body weights of non-suckled cows increased during experimental period (P<0.05), those of suckled cows remained unchanged. Plasma concentrations of glucose of non-suckled cows were higher at wk 2 postpartum (P<0.05) and their levels of non-esterified fatty acid tended to be lower at wk 1 and 2 postpartum compared with suckled cows (P<0.1); however, these differences between groups were not observed with progression of postpartum period. In addition, plasma insulin concentrations of non-suckled cows were higher than those of suckled cows during experimental period (P<0.05). During sampling period (wk 0 to 9 postpartum), onset of normal ovarian cycle was observed in all non-suckled and 2 of 4 suckled cows, and it was delayed in other 2 suckled cows compared with non-suckled cows; however, 3 suckled cows conceived at the first AI after application of the Ovsynch protocol; none of non-suckled cows conceived at this time. Overall, we suggest that suckling seems to reduce increase of body weight after parturition, although it does not improve obesity, and influences conception despite delay in resumption of normal ovarian cyclicity in obese Japanese Black cows.  相似文献

The bovine growth hormone gene (bGH) possesses three haplotypes, A, B and C, that differ by amino acid mutations at positions 127 and 172 in the fifth exon: (leucine 127, threonine 172), (valine 127, threonine 172) and (valine 127, methionine 172) respectively. The correlation between meat quality or carcass weight and these haplotypes was investigated in Japanese black cattle. Altogether, 940 bGH haplotypes were compared with respect to six carcass traits: carcass weight, longissimus muscle area, rib thickness, subcutaneous fat thickness, beef marbling score and beef colour. The frequency of the B haplotype was higher (0.421) than that of A (0.269) and C (0.311). High carcass weight and low beef marbling were associated with haplotype A (p < 0.05 and p < 0.01 respectively), whereas beef marbling was increased by haplotype C (p < 0.05). Estimated regression coefficient of the A haplotype substitution effect for carcass weight and beef marbling score were 5.55 (13.1% of the phenotypic SD) and -0.31 (17.0%) respectively. That of the C haplotype for beef marbling score was 0.20 (11.0%). The other traits showed no relationship to the haplotypes examined. The results of this investigation suggest that information pertaining to bGH polymorphisms in Japanese black cattle could be used to improve the selection of meat traits.  相似文献

Eugenol, which has been used as an anesthetic for fish, was administered to Japanese flounder, Paralichthys olivaceus (127 ± 50.8 g), by bath treatment at concentrations of 0.025, 0.125, and 0.25 mL/L seawater for 10 min and intramuscular injection at 40 μL/fish to investigate the pharmacokinetic characteristics of eugenol in the plasma of Japanese flounder. In the bath treatment test, plasma eugenol concentration increased with increasing eugenol concentration up to 0.125 mL/L and reached steady state within 5 min. After a 10‐min bath treatment in 0.25 mL/L eugenol, plasma eugenol concentration was about 58.4 µg/mL. After transfer into running seawater, plasma eugenol concentration decreased biphasically with half‐lives of 0.0296 h (α‐phase) and 0.289 h (β‐phase). The AUC0000→0800 was about 16.5 µg h/mL. In administration by intramuscular injection, plasma eugenol concentration increased rapidly after administration and decreased biphasically with half‐lives of 0.0329 h (α‐phase) and 8.08 h (β‐phase). The AUC0000→0800 was about 52.5 µg h/mL. In both methods of administration, Japanese flounder with average weight of 127 g were effectively anesthetized when plasma eugenol concentrations were between 2.19 and 4.88 µg/mL.  相似文献

Pull-down tests were conducted for apple trees grafted in an orchard and for support posts in order to discuss the failure mode in windthrow damage, the effect of rootstock types on the uprooting strength, and the reinforcing effect of support posts on young trees. The test samples were ten mature trees (20–23 years old) of three varieties grafted on two types of rootstocks, three young trees (7 years old) grafted on dwarf rootstocks, and nine support posts of larch logs and steel pipes. The failure modes were found to be uprooting for mature trees and rootstock breakage for young trees. The uprooting strength of apple trees was considerably smaller than the uprooting strength of forest trees reported in previous studies. However, the windthrow resistance of the apple trees with respect to the critical wind velocity was estimated to be sufficient despite their low uprooting strength because of their small height. Young trees should be supported with wooden posts for a few years after grafting so that they acquire sufficient windthrow resistance. The mechanical properties of 613 small clear specimens of 35 species (11 ring-porous hardwoods, 19 diffuse-porous hardwoods, and 5 softwoods) were evaluated. The aim of the study was to examine indexes of wood quality that are easy to measure and that exhibit a high correlation with bending performance and hardness that are essential properties of hardwood products. The modulus of rigidity, dynamic modulus of elasticity, bending properties (modulus of elasticity, modulus of rupture, stress at the proportional limit, absorbed energy, Tetmajer’s modulus), dynamic energy absorption by an impact bending test, compressive strength parallel to the grain, shear strength, partial bearing strength, and Brinell’s hardness were measured. A high correlation was found between dynamic modulus of elasticity and static modulus of elasticity. Bending stress at the proportional limit was found to be approximately equivalent to the compressive strength parallel to the grain. Static energy absorption correlated with dynamic energy absorption. Tetmajer’s modulus was found to be closely related to the ratio of the initial stiffness within the elastic range to the secant modulus at the maximum load. A high correlation was observed between Brinell’s hardness and partial bearing strength. The difference in the regression coefficients obtained for these correlations between the species groups was small. We investigated the curing behavior of an emulsion polymer isocyanate adhesive for wood with different ethylene-vinyl acetate (EVA) emulsions, because it was found that the ethylene/vinyl acetate weight composition (E/VA ratio) of EVA could control the volume of gas emitted from the adhesive. EVA with higher VA content reduced the volume of gas emitted at the initial stage, which was caused by the extent of the reaction between the NCO groups and water. The reaction scheme of NCO could not be determined because the broad infrared (IR) absorptions due to CaCO3 and water masked the information of the NCO products in IR spectra. Examination of the degree of swelling suggested that the adhesive films stored for 3 weeks had the same degree of cross-linking structure in spite of the variation of EVA. This result was supported by IR spectroscopy, which showed the same amounts of NCO remained in these films. All the adhesives prepared in this study had satisfactory bond performance. The findings indicate that the E/VA ratio affects the curing behavior of the adhesive in the initial curing stage, whereas the cross-linking structure and the bond performance were independent of the E/VA ratio after storage for 3 weeks. Purpose

To explore the mechanisms in the deposition and release of phosphorus (P) in the sediment of a shallow eutrophic lake using preserved samples, we investigated the vertical and temporal changes in P, manganese (Mn), sulfur (S), iron (Fe), aluminum (Al), calcium (Ca), and magnesium (Mg) in the sediment samples and the phosphate in the sediment pore water samples over a period of 6 years.

Materials and methods

The upper 15 cm of sediment from Lake Kasumigaura in Japan was collected monthly from 2003 to 2008 from the center of the lake. Sediment cores were divided into seven depth segments and were acid-digested for an elemental analysis via inductively coupled plasma atomic emission spectroscopy. Phosphate concentrations of the sediment pore water were determined using the molybdenum blue method. A multiple regression analysis was conducted by setting the P content as the response variable and Mn, S, Fe, Al, Ca, and Mg as explanatory variables.

Results and discussion

The results of the multiple regression analysis demonstrated that P co-precipitates with Fe and Al oxides and accumulates on the sediment surface. The vertical distributions of Mn and S suggest that Mn reduction occurs within the 0–1-cm-depth layer of the sediment and that iron sulfide is actively formed in the 6–10-cm-depth layer of the sediment. These findings imply that the layer in which ferric oxides are reduced to ferrous ions is present near the 1–6-cm-depth layer of the sediment. This layer corresponds to the layer in which the maximum phosphate concentration of the sediment pore water often occurred (the 2–6-cm-depth layer). These results indicate that vertical distributions of mineral elements are useful for assessing P dynamics in sediments.

Conclusions

The lake sediments record the dynamics of P in the sediment. Our analytical approach using long-term observation data demonstrated that the accumulation and release of P associated with a change in the redox state can be assessed based on the vertical distributions of mineral elements in the lake sediments.

We have shown that dietary bluefin tuna skin (TUS) protects against carbon tetrachloride (CCl4)-induced hepatic damage in mice. The CCl4-induced necrotic area was decreased in mice fed a TUS-containing diet. Consistent with the decreased necrotic area, dietary TUS markedly lowered the elevated serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities and the thiobarbituric acid-reactive substance (TBARS) formation induced by CCl4 injection. TUS diets also decreased phosphorylation of inhibitory kappa B-?? and blocked the translocation of nuclear factor-kappa B to the nucleus. TUS is composed mainly (80.7?%) of type I collagen, and our results revealed that dietary tuna collagen peptides (TUCP) attenuated the increased hepatic necrotic area, serum AST and ALT activities, and liver TBARS levels induced by CCl4, similar to TUS, thus enabling us to attribute the hepatoprotective action of TUS in CCl4-intoxicated mice to tuna collagen. Therefore, TUS and TUCP may be potential food resources that are capable of alleviating hepatitis symptoms.  相似文献
