Surface meltwater that reaches the base of an ice sheet creates a mechanism for the rapid response of ice flow to climate change. The process whereby such a pathway is created through thick, cold ice has not, however, been previously observed. We describe the rapid (<2 hours) drainage of a large supraglacial lake down 980 meters through to the bed of the Greenland Ice Sheet initiated by water-driven fracture propagation evolving into moulin flow. Drainage coincided with increased seismicity, transient acceleration, ice-sheet uplift, and horizontal displacement. Subsidence and deceleration occurred over the subsequent 24 hours. The short-lived dynamic response suggests that an efficient drainage system dispersed the meltwater subglacially. The integrated effect of multiple lake drainages could explain the observed net regional summer ice speedup. 相似文献

DNA is thought to behave as a stiff elastic rod with respect to the ubiquitous mechanical deformations inherent to its biology. To test this model at short DNA lengths, we measured the mean and variance of end-to-end length for a series of DNA double helices in solution, using small-angle x-ray scattering interference between gold nanocrystal labels. In the absence of applied tension, DNA is at least one order of magnitude softer than measured by single-molecule stretching experiments. Further, the data rule out the conventional elastic rod model. The variance in end-to-end length follows a quadratic dependence on the number of base pairs rather than the expected linear dependence, indicating that DNA stretching is cooperative over more than two turns of the DNA double helix. Our observations support the idea of long-range allosteric communication through DNA structure. 相似文献

The present study was aimed at optimization, production and partial purification of lipases from Pseudomonas aeruginosa, Candida albicans and Aspergillus flavus. Various nutritional and physical parameters affecting lipase production such as carbon and nitrogen supplements, pH, temperature, agitation speed and incubation time were studied. Refined sunflower oil (1% v/v) and tryptone at a pH of 6.2 favored maximum lipase production in Pseudomonas at 30 degrees C and 150 rpm, when incubated for 5 days. In C. albicans refined sunflower oil (3% v/v) and peptone resulted in maximum lipase production at pH 5.2, 30 degrees C and 150 rpm, when incubated for 5 days. In A. flavus coconut oil (3% v/v) and peptone yielded maximum lipase at pH 6.2, 37 degrees C, 200 rpm after an incubation period of 5 days. The lipases were partially purified by ammonium sulphate precipitation and dialysis. In P. aeruginosa enzyme activity of the dialyzed fraction was found to be 400 U mL-' and for C. albicans 410 U mL(-1). The dialysed lipase fraction from A. flavus demonstrated an activity of 460 U mL(-1). The apparent molecular weights of the dialyzed lipases were determined by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE). The dialyzed lipase fraction obtained from P. aeruginosa revealed molecular weights of 47, 49 and 51 kDa, whereas, lipases from C. albicans and A. flavus demonstrated 3 bands (16.5, 27 and 51 kDa) and one band (47 kDa), respectively. Yield losses of different crops due to the attack of various classes of insects are a worldwide problem. Sucking type homopteran pests causing damage to many crop species are not controlled by commonly known insecticidal proteins, namely, Bacillus thuringiensis delta-endotoxin (Bt). This study describes the purification of mannose-binding lectins from three different monocotyledonous plants (Allium sativum, Colocasia esculenta, and Diffenbachia sequina) and their effects on a homopteran insect, the red cotton bug. All of them had a detrimental effect on the growth and development of the insect, where A. sativum bulb lectin showed the highest mortality of all, in particular. The same bulb lectin not only affected the growth and fecundity of the insect but also imparted drastic changes in the color, weight, and size, even on the second generation of the insects which have been reared on artificial diet supplemented with a sublethal dose of the lectin. Thus, this finding opens up a possibility of using this lectin as an important component in crop management. 相似文献

Comfort is one of the most important aspects of clothing. Thermal comfort is related to fabric’s ability to maintain skin temperature and allow transfer of perspiration produced from the body. Properties like thermal resistance, air permeability, water vapor permeability, and liquid water permeability are suggested as critical for thermal comfort of clothed body. In this study the fabrics developed from the EliTe compact yarns are compared with the fabrics made from normal yarns. The thickness of the fabrics made from EliTe® compact yarns is also slightly less than the fabrics made from normal yarns. Fabrics made from EliTe® compact yarns have shown greater air permeability as compared to the fabrics made from normal yarns. It is observed that, thermal resistivity values of the fabrics developed from EliTe® compact yarns are lower than the fabrics made from normal yarns indicating they are cooler fabrics compared to normal fabrics. Fabrics developed from the EliTe® compact yarns have shown slightly higher values of MVTR (moisture vapor transmission rate) as compared to the fabrics made from the normal yarns. We analyzed data from a long-term rice–wheat cropping sequence to evaluate the effects of integrated nutrient management (INM) on yield trends and sustainability, nutrient balance and soil fertility of the system. After 30th cycle, grain yield of both the crops significantly declined under control and highest rice and wheat grain yields were obtained when 50% N supplied through green manure and farm yard manure, respectively. The magnitude of yield slope under INM was found considerably higher than 100% recommended fertilizer (RDF). Sustainable yield index (SYI) for both the crops were found lower in control but considerably higher with the 100% RDF and under INM. The soils under all the treatments suffered an apparent loss of K and N (except where organics replacing 50% N). Correlation study also reveals K is the sole factor for the yield sustainability as apparent K balance was negatively correlated with SYI and yield slope. 相似文献

Rhizobia diversity is considered as one of the most useful resources for bioprospecting due to their symbiotic nitrogen-fixing ability with members of Leguminosae. The highly conserved nature of the nitrogenase reductase gene (nifH) makes it an ideal molecular tool to determine the potential for biological nitrogen fixation in any environment. In the present investigation, 250 rhizobial strains were isolated from legumes belonging to different geographical locations of Chhattisgarh, India. Genetic diversity of the nitrogenfixing bacterial community was analyzed using the nifH gene-specific primer. The polymorphism was found among the nitrogen-fixing population of different sources and origin but not in same source of rhizobia. Further, the symbiotic plasmid DNA was characterized on the basis of size and copy number of plasmids. The plasmid number varying from one to three in different rhizobial isolates had a size greater than 23 kb, while in some rhizobial isolates plasmids were absent. In addition, to examine the role of ascorbate in respiratory protection, the clear black spot margin of ascorbate was observed in the endodermis region of the nodule whereas scarcely dispersed in the infected region. Therefore, our findings demonstrated that knowing the rhizobial nifH gene diversity along with copy number of the plasmid is important for strain identification, deciding its fertility, productivity standards, and potential of biological nitrogen fixation across the geographical region. 相似文献

Field experiments were conducted during rainy seasons of three consecutive years (2008–2010) to study the effect of green leaf manuring on dry matter partitioning and productivity of lowland rice (Oryza sativa L.). Green leaves of five indigenous agroforestry tree species viz., Erythrina indica, Acacia auriculiformis, Alnus nepalensis, Parkia roxburghii, and Cassia siamea were treated at 10 t ha?1 on fresh weight basis in rice fields and compared with recommended N–P2O5–K2O (80:60:40 kg ha?1) and control treatments. During 2008–2009 year, yield attributes and rice yield were greater in NPK plots as compared to the green-leaf manured ones. However, in the third year, green leaf manuring (except that of Alnus) surpassed even the recommended N–P2O5–K2O treatment in terms of dry matter production and yield; better response was however observed with Erythrina. The soil available N after final harvest increased by ca. 14–20 % in Alnus and Erythrina treated plots as compared to the control. Over all, it could be said that management of plant residues can have long-term implications apart from the desired maintenance of soil organic matter and improving crop yield. 相似文献

The exchange of elite breeding materials across regions is an important way in which multinational maize breeding programmes access new genetic variation, improve efficiency and reduce costs. Our objectives were to examine whether CIMMYT's breeding programmes for tropical and subtropical environments in Mexico and Eastern and Southern Africa (ESA) can effectively share materials. Sets of selected and unselected lines were evaluated for per se and testcross performance in multiple environments in Mexico and ESA for grain yield, days to anthesis and plant height. Genotypic correlations between performance in Mexico and ESA as testcross and line per se were high (≥ 0.72) for all experiments, and indirect selection efficiency ranged from 67 to over 100% for all traits. Lines selected in ESA or Latin America performed equally well in each region, indicating selection was for broad rather than regional adaptation. Thus, breeding programmes of CIMMYT in both Mexico and ESA can benefit tremendously by exchanging breeding materials and test results, and elite selections from each region should be fast‐tracked for evaluation in the other. 相似文献
