Summary Gelprotein or SDS-insoluble gel-forming glutenin was isolated from wheat flour by extraction with an aqueous 1.5% SDS solution. Remarkable intervarietal differences were observed both in amount and subunit composition of these proteins.The amount of gelprotein and the SDS-sedimentation volume both proved to be good parameters for the bread-making quality of wheat cultivars. A high correlation was observed between amount of gelprotein and SDS-sedimentation volume. The amount of gelprotein was therefore tentatively assumed to be the essential basis of the SDS-sedimentation test.The subunit composition of the gelprotein was studied by SDS-PAGE after reduction of SS bonds by mercaptoethanol. It was found that the average bread-making quality of wheat cultivars and progeny of the cross Atlas 66 x Atys which possessed subunits 3 and 10, coded for by chromosome 1D, was significantly higher than that of wheat samples possessing subunit 2 and 11, their allelic counterparts.  相似文献

Bolting and flowering of Aster novi-belgii seedlings as well as correlations between characteristics within seedling populations and between seedlings and their vegetatively propagated offspring were investigated in order to improve breeding efficiency. In seedlings of A. novi-belgii, flower induction was observed without vernalization. Seedlings initially formed a rosette, but bolting was a prerequisite for development of a normal flowering shoot. Bolting was promoted by long photoperiods as compared to short days. The shortest time from sowing to anthesis was recorded when plants were grown under long photoperiods until the main stem was at least 5 cm long, and then moved to short photoperiods. A large variation in time to anthesis, plant height, branching ability, and flower size and color was found among the seedlings within all crosses. Magnitude and significance of correlations between some characteristics varied among crosses. The more consistent correlations seem mainly to be due to natural courses of plant growth and development. High correlations between seedling characteristics and their vegetatively propagated offspring were only found for flower hue. Lower, but significant correlations were found for flower color intensity, plant height and branching ability. There were no significant correlations for the time-to-anthesis between the two groups of plants. This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献

Summary Visual assessment, black & white image analysis and colour image analysis of the severity of powdery mildew (Sphaerotheca fuliginea) infection on leaves of cucumber (Cucumis sativus) were compared. The extent of infection with powdery mildew was expressed as the number of spores per cm2. Statistical treatment of the data showed colour image analysis to be superior to the other two methods of evaluation. Major veins, light reflections and variations in the colour of leaves can degrade the accuracy of colour image analysis somewhat. These problems are, however, surmountable. Summary Several independent tests have shown that climate and weather during sexual reproduction influence the adaptive properties of the Picea abies progenies. This phenomenon is expressed in seed orchards established by moving parent trees, propagated as grafts, from north to south, from high to low elevation, or from outdoor to indoor greenhouse conditions. The progenies exhibit delayed flushing in the spring, later growth cessation of leader shoots in the summer, delayed bud-set, higher frequency of lammas shoots and delayed development of frost hardiness in the autumn compared to progenies reproduced in the colder native environment. The altered performance is persistent. We have found no effect on progenies of photoperiod and temperature treatments given to the males during meiosis and pollen production. However, when crosses were made in early spring (March), inside a heated greenhouse (short day, high temperature), the progenies are less frost hardy during cold acclimation than progenies from identical crosses performed in late spring (May; long day high temperature) in the greenhouse. The most hardy offspring were from crosses performed under outdoor conditions in May (long day, low temperature). These results indicate that some stages in reproduction, such as female meiosis, pollen tube growth, syngamy and early embryo development, are sensitive to temperature and/or photoperiod which then alter the phenotypic performance of the offspring. The most likely explanation is the existence of a regulatory mechanism affecting the expression of genes controlling adaptive traits. If this is true, it must have implications for the genetic interpretation of provenance differences in Norway spruce.  相似文献

A manually operated high-pressure hot-water washing system consisting of a boiler, hot-water mixing tank, contact loop, heat exchanger, spray mixing tank, high-pressure hot-water washing manifold, low-pressure fresh water rinse manifold, and pressure pump was constructed and installed in a packingline. The system developed 20–50 °C washing water at pressures up to 980 kPa. ‘d’Anjou’ pears (Pyrus communis L.), shortly after harvest, and after storage for 3 and 4 months in regular air (RA) or for 4, 7 and 8 months in controlled atmosphere (CA) at −1 °C were washed through the packingline with different wetting agents (0.1% Silwet, 0.01 and 0.1% Defoamer, and water), water pressures (regular and high-pressure (210–980 kPa)), water temperatures (control (tap water, 4–22 °C), 40 °C, and 50 °C), and brushes (soft and firm), respectively. The effect of the washing conditions on fruit quality was investigated after 1 month of storage at −1 °C to simulate shipping condition, and then again after 1 week at 20 °C to simulate marketing condition. Hot-water caused severe heat scald. When nozzle temperature was 50 °C, the incidence of heat scald increased to over 50% for the fruit stored in RA for 3 months. Combined with hot-water, 540 kPa high-pressure washing increased the incidence of friction discoloration. There were lower incidences of friction discoloration and heat scald for fruit stored in CA for 7 months, in comparison to that in RA for 3 months. However, those fruit did not ripen properly as indicated by a high extractable juice content. Fruit washed at harvest had minor incidences of friction discoloration regardless of different brushes, water pressures, and wetting agents. Fruit washed after storages in either 4 months RA or 4 or 8 months CA suffered a high incidence of friction discoloration including scuffing symptoms and pressure marking. The firm brushes caused a higher incidence of friction discoloration mainly because of scuffing symptoms. However, no differences were found between different water pressures and wetting agents with respect to friction discoloration. Fruit stored for 4 months RA suffered 26–28% friction discoloration in comparison to 16–18% in CA stored fruit with firm brush washing. Extended CA storage increased friction discoloration even with soft brush washing. The results suggest that a washing system with high-pressure spray, <30 °C warm water, wetting agent Defoamer and rotating soft brushes were significantly effective in removing surface pests and decay control without causing internal or external damage of fruit.  相似文献

Decline of grassland diversity throughout Europe within the last decades is threatening biological diversity and is a major conservation problem. There is an urgent need to determine the underlying factors that control vascular plant species richness and composition in managed grasslands. In this study, 117 grasslands were sampled using standardised methods. Explanatory variables were recorded for each grassland site, reflecting the local field management, site-specific environmental conditions and large-scale spatial trends. Using variation partitioning methods, we determined the pure and shared effects of these three sets of explanatory variables on the plant species richness and composition in grasslands. Most of the explained variation in plant species richness was related to the joint effect of local field management and environmental variables. However, the applied variation partitioning approach revealed that the pure effect of spatial variables contributed relatively little to explaining variation in both the plant species richness and species composition. The largest fractions of explained variation in plant species composition were accounted for by the pure effects of environmental and local field management variables. Moreover, the results revealed that the main mechanisms by which these sets of explanatory variables affect plant species vary according to the type of management regime under study. From our findings we could conclude that particularly a reduction of nitrogen fertilisation on meadows and grazing at a low stocking rate on pastures can help to conserve biodiversity.  相似文献

Genotoxic compounds can act at various levels in the cell (causing gene, chromosome, or genome mutations), necessitating the use of a range of genotoxicity assays designed to detect these different types of mutations. The production of melanoidins during the processing and cooking of foods is associated with changes in their nutritional character, and the discovery of mutagenic substances in pyrolyzed protein and amino acids has raised concern about the safety of these foods. The aim of this work was to test melanoidin fractions in three different in vitro assays (Ames test, Vitotox test, and micronucleus test). These melanoidin fractions were produced from the condensation of glucose with glycine and their separation was conducted by dialysis. The crude reaction mixture (before dialysis) and both the LMW and HMW fractions obtained by dialysis showed no genotoxicity in these assays, despite being tested at concentrations much higher than those naturally found in food products. The LMW fraction, however, showed toxicity at these high concentrations. The volatile fraction produced in this reaction showed genotoxicity only in the Vitotox test, at high concentrations.  相似文献
