Exogenous DNA Uptake of Boar Spermatozoa by a Magnetic Nanoparticle Vector System

The sperm‐mediated gene transfer method is applicable to transgenesis in many species that use spermatozoa for reproduction recently, which has been shown various results. In the current study, we show that transgenic porcine embryos can be efficiently produced by employing a simple transfection method that uses magnetic nanoparticles (MNPs). The complexes formed between plasmid DNA and MNPs were bounded on ejaculated boar spermatozoa at a higher efficiency compared to methods using DNA alone or lipofection. Using confocal microscopy, rhodamine fluorophore‐labelled MNPs were detected on external surfaces of the spermatozoa membrane, which were bounded on zona pellucida of in vitro maturated oocyte during in vitro fertilization. Electron microscopy revealed that clusters of MNPs were detected in inside of plasma membrane and nucleus of the spermatozoa head. Additionally, we found that magnetofected boar spermatozoa could be fertilized with oocytes in vitro and that the resulting gene of green fluorescent protein was detected in fertilized eggs by genomic PCR analysis. Taken together, these results suggest that MNPs can be used to efficiently introduce a transgene into embryo via spermatozoa.     

不同的干燥工艺对荷花蜂花粉功能成分的影响

探讨了5种干燥工艺对荷花蜂花粉功能成分的影响。微波干燥的干燥效率最高,60%火力处理540s可使水分从31.41%降到6.42%;干燥方式对荷花蜂花粉蛋白质影响较小;微波干燥后总黄酮提取率最高,为13.20mg/g;真空冷冻干燥后维生素C和过氧化氢酶含量最高,分别为48.89mg/100g和29.86mg/g·min。总的来说,真空冷冻干燥对荷花蜂花粉的功能成分影响最小;常压恒温干燥对功能成分影响较小、工艺简单、对设备要求低,宜于大规模应用于实际生产中。     

家蚕母种继代蛾区选择方法对种性维持的影响

为了有效保持生产实用家蚕品种的优良性状,在家蚕母种继代繁育中采用5种选择方法选择继代蛾区,探讨种性维持效果。分析8年12代及F1代的成绩表明:所有性状以保持种性为主,采用选留平均数上下1个标准差范围内蛾区的稳定化选择和以茧丝质指标在平均数-0.5~1.0标准差范围内选择,并结合健康性指标选留高值的综合选择,有利于家蚕母种的种性维持。     

A case of perosomus elumbis in a Holstein calf

Perosomus elumbis is an occasionally found congenital anomaly of unknown etiology and is characterized by partial or complete agenesis of lumbar, sacral and coccygeal vertebrae and ankylosis of the hindlimbs. A 2-day-old female Holstein calf presented nearly normal forelimbs but flexure and ankylosis of the hindlimbs. The vertebrae and pelvic malformations and agenesis were radiographed and then necropsied. Mild ankylosis of the hindlimbs, absence of cauda equina, left scoliosis in state of fusion of T11 and T12 and complete fusion of L4 and L5, narrowed pelvic canal and misshapen ilium were confirmed. However, abnormal development or agenesis was not observed in the urogenital and intestinal system in this calf.     

应用单引物PCR指纹技术分析Vc-獭兔的分子遗传结构

采用1条小卫星引物M13(5’GAGGGTGGCGGTTCT3’)和2条微卫星引物(GTG)5、(GACA)4，对Vc—Ⅰ系、Vc—Ⅱ系獭兔、日本大耳白兔(JWR)和青紫蓝(QZL)4个品系各8只，总计32个个体的遗传结构进行了DNA指纹分析。结果，3条引物的扩增产物都呈现良好的多态性和稳定性；对每个品系混合DNA池进行分析，计算出了4个品系间及32个个体间的共有带率及遗传距离指数，通过聚类分析，得出各品系间和个体间欧式遗传距离，并绘制出4个品系间及各品系32个个体问的亲缘关系树状聚类图，从而初步建立了4个品系兔的SPAR指纹图谱。     

Aspergillus niger pulmonary infection in a dog

A 6-month-old male golden retriever was presented with fever, bloody-watery diarrhea and mild cough. Parvovirus and Isospora canis infection was confirmed and successfully treated. Two weeks later, the dog had severe cough and mucopurulent nasal discharge. Aspergillus niger was cultured from endotracheal washings on blood agar at 37 degrees C. Treatment with itraconazole for about 10 weeks resolved the clinical signs.     

二维薄层色谱法分析分枝杆菌脂质成分

为探讨卡介苗巴氏德菌株(Bacillus Calmette-Guérin-pasteur,BCG-pasteur)和耻垢分枝杆菌(Mycobacterium smegma-tis,mc^2155)两种菌株的脂质成分,且通过比较脂质成分差异为研发新型牛结核病脂类疫苗佐剂提供试验基础。本文通过石油醚,氯仿,甲醇,丙酮等有机溶剂萃取BCG-pasteur和mc^2155两种菌株脂质,采用薄层色谱法分析其非极性脂质成分的差异。结果显示,BCG-pasteur中脂质成分富含三酰甘油,甲硫酚二角酰亚胺,PAT家族蛋白,甘油二酯,游离脂肪酸,游离分枝菌酸,葡萄糖单霉菌酸酯以及硫脂I类和硫脂III类等物质;mc^2155脂质成分除了硫脂I类,其他成分均有表达,但相对于BCG-pasteur其含量明显减少。本萃取方法简便稳定,适用于分枝杆菌的非极性脂质提取,为分枝杆菌脂质成分的分析提供基础;本试验通过薄层色谱法分析显示卡介苗和耻垢分枝杆菌细胞壁脂质成分差异明显,表明不同结核分枝杆菌的细胞壁脂质成分存在明显差异,从而可以引起不同的免疫反应,可为新型牛结核病疫苗提供方向。     

Screening for reproductive biomarkers in Bactrian camel via iTRAQ analysis of proteomes

Bactrian camel is an ancient and precious species of livestock; that is, unique resources exist in the desert and have important economic and scientific value. In recent years, the number of Bactrian camels has declined sharply. Due to its long reproductive cycle and seasonal oestrus, the mechanism of oestrus is unknown. To identify candidate biomarkers of reproduction, we performed a comprehensive proteomic analysis of serum from Bactrian camel in oestrus and non-oestrus, using isobaric tags for relative and absolute quantitation (iTRAQ) coupled with tandem mass spectrometry. We identified 359 proteins, of which 32 were differentially expressed: 11 were up-regulated and 21 were down-regulated in samples from camels in oestrus. We validated the differential expression of a subset of these proteins using qPCR and Western blot. Gene ontology annotation identified that the differentially expressed proteins function in cellular processes, metabolic processes and immune system processes. Notably, five of the differentially expressed proteins, PCGF5, histone H1.2, RBP4, FOLR1 and ANTXR2, are involved in reproductive regulatory processes in other animals. KEGG enrichment analysis demonstrated significant enrichment in several cardiac-related pathways, such as ‘dilated cardiomyopathy’, ‘hypertrophic cardiomyopathy’, ‘cardiac muscle contraction’ and ‘adrenergic signalling in cardiomyopathy’. Our results suggest that candidate biomarker (PCGF5, histone H1.2, RBP4, FOLR1 and ANTXR2) discovery can aid in understanding reproduction in Bactrian camels. We conclude that the profiling of serum proteomes, followed by the measurement of selected proteins using more targeted methods, offers a promising approach for studying mechanisms of oestrus.     

新冠疫情对牧区半牧区草原畜牧业的影响及建议

基于全国13个省(区)牧区和半农半牧区的线上调研数据,定量分析了新冠疫情发生对农牧民生活和生产的影响。结果表明:疫情对牧区半牧区生产生活带来一定影响,但整体来看影响低于东南沿海地区。储备饲草料习惯、家畜饲养规模和家庭净收入对农牧民抵御疫情影响的能力具有显著影响。牧区半牧区发展在此次抗击新冠疫情中表现出一定的适应能力,是我国牧区政策、牧区地缘优势和农牧民生产生活习惯共同作用的结果。针对牧区存在的风险与发展短板,从完善牧区政策和市场条件、创新生产模式和社会化服务模式及加强基础建设等方面提出建议,以期为牧区半牧区草原畜牧业平稳有序发展提供科技支撑。     

活禽交易对十堰市公共卫生安全的影响及对策

本文针对十堰市活禽养殖和交易的现状,结合工作实际,分析了活禽交易方式对公共卫生安全、禽流感防控、环境污染等方面的影响,以及活禽交易存在的问题及产生的原因,从政策建议、防疫条件、检疫监管、疫情监测预警、市场准入、可追溯体系建设、动物经纪人管理等方面提出了措施和建议,为进一步加强禽流感防控工作和动物产品质量安全管理提出了有益的探讨。
