The mechanisms of resistance in barley to fusarium head blight (FHB), caused by Gibberella zeae are complex. Metabolomics technology was explored to phenotype resistance. Spikelets of barley genotypes with contrasting
levels of resistance to FHB, mock inoculated or with the pathogen, were extracted with aqueous methanol and the metabolites
were analyzed using liquid chromatography and hybrid mass spectrometry. Peaks were de-convoluted using XCMS and annotated
using CAMERA and IntelliXtract bioinformatics tools. A t-test, of a total of 1608 purified peaks, selected 626 metabolites with significant treatment effects, of which 161 were identified
as resistance related (RR) metabolites. A total of 53 metabolites, that are RR or pathogenicity related (PR), were assigned
with putative compound names. These mainly belonged to three metabolic pathways: fatty acid (jasmonic acid, methyl jasmonate,
9,10- dihydro-isojasmonate, linolenic acid, linoleic acid, traumatic acid), phenylpropanoid (p-coumaric acid, caffeyl alcohol, dimethoxy-4-phenylcoumarin, rosmarinic acid, diphyllin, 5-methoxypodophyllotoxin) and flavonoid
(naringenin, catechin, quercetin, and alpinumisoflavone). A few PR/RR metabolites significantly reduced mycelial growth of

A specific assay has been developed for a blood-borne non-A, non-B hepatitis (NANBH) virus in which a polypeptide synthesized in recombinant yeast clones of the hepatitis C virus (HCV) is used to capture circulating viral antibodies. HCV antibodies were detected in six of seven human sera that were shown previously to transmit NANBH to chimpanzees. Assays of ten blood transfusions in the United States that resulted in chronic NANBH revealed that there was at least one positive blood donor in nine of these cases and that all ten recipients seroconverted during their illnesses. About 80 percent of chronic, post-transfusion NANBH (PT-NANBH) patients from Italy and Japan had circulating HCV antibody; a much lower frequency (15 percent) was observed in acute, resolving infections. In addition, 58 percent of NANBH patients from the United States with no identifiable source of parenteral exposure to the virus were also positive for HCV antibody. Betacyanins are reddish to violet pigments that can be found in red pitahaya (Hylocereus polyrhizus) and red spinach (Amaranthus dubius). This study investigated the impact of sub-fractionation (solvent partitioning) on betacyanin content in both plants. Characterization of betacyanins and evaluation of their antimicrobial activities were also carried out. Betanin was found in both plants. In addition, isobetanin, phyllocactin and hylocerenin were found in red pitahaya whereas amaranthine and decarboxy-amaranthine were found in red spinach. Sub-fractionated red pitahaya and red spinach had 23.5 and 121.5 % more betacyanin content, respectively, than those without sub-fractionation. Sub-fractionation increased the betanin and decarboxy-amaranthine content in red pitahaya and red spinach, respectively. The betacyanin fraction from red spinach (minimum inhibitory concentration [MIC] values: 0.78–3.13 mg/mL) demonstrated a better antimicrobial activity profile than that of red pitahaya (MIC values: 3.13–6.25 mg/mL) against nine Gram-positive bacterial strains. Similarly, the red spinach fraction (MIC values: 1.56–3.13 mg/mL) was more active than the red pitahaya fraction (MIC values: 3.13–6.25 mg/mL) against five Gram-negative bacterial strains. A random-primed complementary DNA library was constructed from plasma containing the uncharacterized non-A, non-B hepatitis (NANBH) agent and screened with serum from a patient diagnosed with NANBH. A complementary DNA clone was isolated that was shown to encode an antigen associated specifically with NANBH infections. This clone is not derived from host DNA but from an RNA molecule present in NANBH infections that consists of at least 10,000 nucleotides and that is positive-stranded with respect to the encoded NANBH antigen. These data indicate that this clone is derived from the genome of the NANBH agent and are consistent with the agent being similar to the togaviridae or flaviviridae. This molecular approach should be of great value in the isolation and characterization of other unidentified infectious agents. 相似文献

Measurements of the hot plasma environment during the Ulysses flyby of Jupiter have revealed several new discoveries related to this large rotating astrophysical system. The Jovian magnetosphere was found by Ulysses to be very extended, with the day-side magnetopause located at approximately 105 Jupiter radii. The heavy ion (sulfur, oxygen, and sodium) population in the day-side magnetosphere increased sharply at approximately 86 Jupiter radii. This is somewhat more extended than the "inner" magnetosphere boundary region identified by the Voyager hot plasma measurements. In the day-side magnetosphere, the ion fluxes have the anisotropy direction expected for corotation with the planet, with the magnitude of the anisotropy increasing when the spacecraft becomes more immersed in the hot plasma sheet. The relative abundances of sulfur, oxygen, and sodium to helium decreased somewhat with decreasing radial distance from the planet on the day-side, which suggests that the abundances of the Jupiter-derived species are dependent on latitude. In the dusk-side, high-latitude region, intense fluxes of counter-streaming ions and electrons were discovered from the edge of the plasma sheet to the dusk-side magnetopause. These beams of electrons and ions were found to be very tightly aligned with the magnetic field and to be superimposed on a time- and space-variable isotropic hot plasma background. The currents carried by the measured hot plasma particles are typically approximately 1.6 x 10(-4) microamperes per square meter or approximately 8 x 10(5) amperes per squared Jupiter radius throughout the high-latitude magnetosphere volume. It is likely that the intense particle beams discovered at high Jovian latitudes produce auroras in the polar caps of the planet. 相似文献

- Giant guitarfishes (Glaucostegidae) and wedgefishes (Rhinidae) are some of the most threatened marine taxa in the world, with 15 of the 16 known species exhibiting global population declines and categorized as Critically Endangered according to the International Union for Conservation of Nature (IUCN) Red List of Threatened Species. The recent inclusion of all species in Appendix II of the Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES) necessitates more rigorous enforcement by regulatory authorities.
- Challenges in regulating the trade of giant guitarfish and wedgefish products due to difficulties in visual identification of processed products and labelling issues impede enforcement. The aim of this study is to characterize the diversity and origins of associated traded products that were commercially available in Singapore, one of the world's top importers and re-exporters of shark and ray products.
- A total of 176 samples of elasmobranch products were obtained between June and December 2019 from fishery ports and various retailers in Singapore. By applying cytochrome c oxidase subunit I gene barcoding, 31 elasmobranch species were detected, with 55% of the species considered threatened (Critically Endangered, Endangered, or Vulnerable) based on the IUCN Red List and 35% of species listed in CITES Appendix II. Four species of giant guitarfishes and wedgefishes were commercially available to consumers in fresh forms of whole fish, fillet, and fin, as well as dried and cooked meats.
- DNA barcoding has proven to be an effective tool for identifying elasmobranch products that are impossible to recognize visually and would aid enforcement of CITES trade regulations. This work underscores the urgent need to step up enforcement of marine wildlife regulations and draw public attention to the elasmobranch trade.

ABSTRACT Fusarium head blight (FHB) or scab is a destructive disease of barley in many countries. A better understanding of the interrelationships between plant traits and FHB resistance should help in the development of effective and efficient breeding strategies for FHB-resistant cultivars. Recent mapping studies indicate that many of the quantitative trait loci (QTL) for FHB resistance coincide with the QTL for plant height, heading date, and spike characteristics. Therefore, a study was conducted to investigate the relationship of morphological and physiological traits to FHB infection and deoxynivalenol (DON) accumulation in a barley doubled-haploid (DH) population derived from a Léger x CI9831 cross. Approximately 190 DH lines were grown at Ottawa (Ontario) for 2 years, Charlottetown (Prince Edward Island) for 1 year, and Hangzhou (Zhejiang) for 2 years. The field plots were inoculated with Fusarium graminearum at each location. FHB incidence was positively correlated with DON content. Resistance to FHB was associated with two-row spike, purple lemma, long glume awn, tall stature, and resistance to lodging, but it was not associated with long rachilla hairs, rough lemma awn, or heading date. Two-row spike was associated with tall stature and resistance to lodging. These associations as well as its spike characteristics helped reduce FHB infection and DON accumulation in two-row lines compared with six-row lines. The association between long glume awn and FHB resistance could be due to genetic linkages. Therefore, trait associations should be taken into consideration when breeding for FHB resistance and interpreting data from FHB experiments. 相似文献

This study was conducted to determine the effects of coconut water (CW) and activated charcoal (AC) on multiplication of Phalaenopsis gigantea protocorms. The protocorms used for this study were obtained by germinating seeds in vitro. Protocorms with trimmed and untrimmed bases were cultured on XER basal medium containing 0, 10, 15 or 20% (v/v) CW; and 0, 1, 2 or 2.5 g AC l−1. Trimmed protocorms exhibited the highest percentage of proliferation on a medium containing 15% (v/v) CW and 2.5 g AC l−1 (56.82 ± 38.86%) with an average of 4.24 ± 2.89 protocorms formed per protocorm. Untrimmed protocorms cultured on a medium containing 20% (v/v) CW without AC produced the highest percentage of new protocorms (6.93 ± 6.28%) with an average of 0.72 ± 0.57 per protocorm. When CW was added to a medium singly, 10% (v/v) CW induced a higher degree of proliferation on trimmed protocorms (5.68 ± 10.14%) with an average 0.50 ± 0.84 new protocorms per protocorm. Untrimmed protocorms proliferate to a much lower extent (2.57 ± 2.74%) with an average of 0.72 ± 0.57 protocorms per protocorm when cultured on a similar medium. A high concentration of CW enhanced proliferation on untrimmed protocorms, but increased mortality of trimmed protocorms. The addition of CW with AC to media increased protocorm proliferation and survival of both trimmed and untrimmed protocorms. When cultured on all media, trimmed protocorms produced a higher number of new protocorms (an average 0.5–7.0) as compared to untrimmed protocorms (0.3–1.9). Comparative studies showed that trimmed protocorms produced up to 10 times more new protocorms than untrimmed ones. Altogether this study showed that trimmed protocorms cultured on a medium containing CW and AC can be used for high-frequency multiplication of P. gigantea seedlings. 相似文献

The emphasis on cow records in Malaysian dairy extension programmes reflects the importance of herd fertility in the economics of dairying. Manual record keeping has not been able to make an impact on management due to difficulties experienced in quality control of the data and in analysing the data to produce useful information for farm managers. Computerised recording systems have been in use in Malaysia since 1985, both on government farms and in the small-holder dairy sector. The aim of both systems is firstly to improve farm efficiency by the provision of information to managers and extension workers and secondly to provide information for departmental planning purposes. The systems used in Malaysia are outlined, and the results over the first three years of operation are summarised. 相似文献
