In order to investigate the infection status of avian leukosis virus (ALV) and avian reticuloendotheliosis virus (REV) in the major local breeds of Qinzhou,Guangxi,totally 953 samples of egg white,cloaca swab and serum of Ma duck,Shitou goose,Tiejiao-Ma chicken,turkey and pigeon were collected from the representing flocks and detected by the commercial ELISA kits.ALV was isolated for the ALV p27 positive samples by culturing on DF-1 cells,and gp85 gene was sequenced.The results showed that the detections of ALV were negative in the samples except those of Tiejiao-Ma chicken,while REV antibody was found positive in Ma duck,Tiejiao-Ma chicken and turkey.The nucleotide sequences of gp85 gene of two isolates shared 94.5% identity with each other,and shared 86.9% to 94.9% with reference strains.The amino acid sequences of gp85 gene of two isolates shared 91.5% identity with each other,and shared 84.0% to 91.6% with reference strains.There were many variable sites in the hyper variable region hr1 and hr2,and the vr2 and vr3 variable regions were relatively conservative.Phylogenetic tree analysis showed that the two isolates shared the highest homology with SCAU11-XG strain.  相似文献

The study was conducted to explore the potential different characters between Blue-shelled chicken and White leghorn.Global genome microRNA was combined the identified microRNA with complementary lab-predicted microRNA.Then the two breed chicken's SNP data got by GGRS were mapped to the microRNA and focused on SNP that deliberately located in mature-microRNA.Bioinformatics method was adopted for target prediction on microRNA which had SNPs.By further gene enrichment analysis,the study found these genes enriched in 22 GO terms,10 KEGG pathways,and 3 IPA important networks.And they enriched in traits which associated with growth,such as mTOR signaling pathways,Wnt signaling pathways,growth hormone receptor networks and insulin-like growth factor Ⅰ receptor networks.And they also enriched in some laying traits,such as oocyte meiotic signaling pathways and progesterone mature oocytes signaling pathways.The methods and the results might provide references for further studies.  相似文献

本试验利用禽用开放式呼吸测热装置进行能量代谢试验,通过间接测热法结合替代法测定不同类型玉米在产蛋期蛋鸡饲粮中的表观代谢能和净能。选用34周龄产蛋期海兰褐蛋鸡180只,随机分为6组,每组30只。试验选用1种玉米-豆粕型基础饲粮和5种待测饲粮。待测饲粮由5种待测玉米(3种2018年10月收获的正常玉米和2种2016年收获储存3年的陈化玉米),分别以50%比例替代基础饲粮构成。试验鸡在舍内笼养,预试期7 d,正试期27 d,其中正试期分为3期,每期9 d(适应3 d、呼吸测热3 d、绝食测热3 d)。每期试验中,从每组中选择4只试验鸡,称重后分别放入呼吸测热装置的12个代谢室(每个代谢室2只),每2个代谢室对应1种饲粮,测定气体交换和排泄物总量,呼吸测热的同时进行消化代谢试验。结果表明:与基础饲粮相比,5种玉米待测饲粮的表观代谢能显著提高(P<0.05),3种正常玉米待测饲粮的净能显著高于基础饲粮和2种陈化玉米待测饲粮(P<0.05);2种陈化玉米的表观代谢能和净能显著低于3种正常玉米(P<0.05)。本试验中,3种正常玉米的表观代谢能分别为16.19、15.85、16.17 MJ/kg,2种陈化玉米的表观代谢能分别为15.12和15.06 MJ/kg;3种正常玉米的净能分别为12.39、12.57、12.25 MJ/kg,2种陈化玉米的净能分别为11.29和12.05 MJ/kg。  相似文献

AIM To explore the expression and mechanisms of circular RNA hsa_circ_087631 in the patients with primary biliary cholangitis (PBC). METHODS RT-qPCR was used to detect the expression of hsa_circ_087631 in PBC patients and healthy controls. Hsa-miR-346-overexpressing lentiviral vector pLenti-EF1a-EGFP-F2A-Puro-CMV-MCS was constructed and transfected into human acute T cell leukemia Jurkat cells, and then the expression levels of hsa_circ_087631, Bcl-6 mRNA and interleukin-21 (IL-21) mRNA were detected by RT-qPCR. Dual-luciferase reporter assay was performed to identify the interactions between hsa_circ_087631 and hsa-miR-346. RESULTS The expression of hsa_circ_087631 in the PBC patients was significantly increased compared with the healthy subjects. Hsa-miR-346-overexpressing lentiviral vector pLenti-EF1a-EGFP-F2A-Puro-CMV-MCS was successfully constructed. The expression of hsa-miR-346 was significantly increased after the hsa-miR-346-overexpressing lentiviral vector was transfected into the Jurkat cells, while the expression levels of hsa_circ_0087631, Bcl-6 mRNA and IL-21 mRNA were significantly decreased. After wild-type or mutant hsa_circ_087631 vector and hsa-miR-346 mimics were transfected into 293T cells, the results of dual-luciferase reporter assay showed that hsa-miR-346 significantly decreased the luciferase activity of wild-type hsa_circ_087631 (P<0.01), but the regulation did not change significantly after mutation of the predicted binding site. CONCLUSION Peripheral blood hsa_circ_087631 level is elevated in the PBC patients. The hsa_circ_087631/hsa-miR-346/Bcl-6 signaling may take effect in human T cells. Hsa-miR-346 significantly reduces the expression of hsa_circ_087631, but it may not be regulated by predicted binding sites.  相似文献

In order to investigate the effect of AIR on inflammatory reaction infected by Brucellamelitensis (16M), the AIR domain of Tecpr1 gene of murine macrophages RAW264.7 were knocked down (I-A), overexpressed (O-A) and reversed (OA-IA). Using the chlorine fluorescein (DCFH-DA) as a probe, we detected the variation of ROS production and mitochondria distribution by confocal laser scanning microscopy. We observed the expression changes of NLRP3, ASC and Caspase-1 by qRT-PCR and the expression changes of IL-18,IL-1β and Caspase-1 in host cells by ELISA. The results showed that 16M could stimulate RAW264.7 cells to produce ROS by time-dependent pathway, and I-A group and O-A group showed more abnormal accumulation of mitochondrial. The results of qRT-PCR and ELISA suggested that it had effect on the expression levels of NLRP3, ASC,Caspase-1 and IL-18, IL-1β and Caspase-1 in cells of different groups. Those results indicated that with AIR gene deletion, the release amount of ROS changed, mitochondrial clustered abnormally, and AIR was closely related to the activation of inflammasomes and induction of inflammatory reactions.  相似文献

This experiment was conducted to study the effects of dietary lactic acid bacteria (LAB) and fructooligosaccharides (FOS) on growth performance, nutrient digestibility and serum biochemical indices of Arctic foxes (Alopex lagopus) during early growth period. Sixty male post-litter Arctic foxes with the average body weight of (1.16±0.15) kg and at the age of (45±3) days were randomly assigned to 4 groups with 3 replicates per group and 5 foxes per replicate. The foxes were fed the following diets, respectively: basal diet (control group), basal diet+3×109 CFU / kg LAB (LAB group), basal diet+5 g / kg FOS (FOS group) and basal diet+3×109 CFU / kg LAB+5 g / kg FOS (LAB+FOS group). The experiment lasted for 30 days. The results showed as follows: 1) on the days 1 to 15, the diarrhea rate of foxes in the LAB group and LAB+FOS group was significantly lower than that in the control group and FOS group (P<0.05). On the days 16 to 30, the average daily gain of foxes in the LAB+FOS group was significantly higher than that in the control group and FOS group (P<0.05), but had no significant difference with LAB group (P>0.05); the diarrhea rate of foxes in the LAB+FOS group was significantly lower than that in the control group and FOS group (P<0.05), but had no significant difference with LAB group (P>0.05); compared with the FOS group, the feed to gain ratio of foxes in the LAB+FOS group was significantly decreased (P<0.05). 2) The dry matter digestibility of foxes in the LAB + FOS group was significantly higher than that in the control group and FOS group (P < 0.05), but had no significant difference with LAB group (P>0.05). There were no significant differences in nitrogen intake, fecal nitrogen, urine nitrogen, nitrogen deposition, net protein utilization and protein biological value among all groups (P>0.05). 3) Serum total protein (TP) content of foxes in the LAB+FOS group was significantly higher than that in the FOS group (P<0.05), but had no significant difference with control group and LAB group (P>0.05). Serum albumin (ALB) content of foxes in the LAB group was significantly higher than that in the control group and FOS group (P<0.05), but had no significant difference with LAB+ FOS group (P>0.05). Serum urea nitrogen (UN) content of foxes in the LAB+FOS group was significantly decreased compared with the control group (P < 0. 05). In conclusion, the combined supplementation of 3 × 109 CFU / kg LAB and 5 g / kg FOS in the diet is beneficial to reduce diarrhea rate, improve dry matter digestibility, improve body immune level, and promote the growth of Arctic foxes during early growth period. © 2023 Chinese Journal of Urology.
