Membrane transport proteins that transduce free energy stored in electrochemical ion gradients into a concentration gradient are a major class of membrane proteins. We report the crystal structure at 3.5 angstroms of the Escherichia coli lactose permease, an intensively studied member of the major facilitator superfamily of transporters. The molecule is composed of N- and C-terminal domains, each with six transmembrane helices, symmetrically positioned within the permease. A large internal hydrophilic cavity open to the cytoplasmic side represents the inward-facing conformation of the transporter. The structure with a bound lactose homolog, beta-D-galactopyranosyl-1-thio-beta-D-galactopyranoside, reveals the sugar-binding site in the cavity, and residues that play major roles in substrate recognition and proton translocation are identified. We propose a possible mechanism for lactose/proton symport (co-transport) consistent with both the structure and a large body of experimental data.  相似文献

A single spray of solutions of 0.005M H3BO3, 0.0025M CuSO4, and 0.0025 MnCl2, on the upper surface of the first true leaf of cucumber plants 2 h before inoculation with a conidial suspension of Sphaerotheca fuliginea, induced systemic protection against powdery mildew in leaves 2 and 3 without causing any damage on the induced leaf (first leaf). A similar level of systemic protection was observed when plants were induced by micronutrients, 2, 24 and 72 h before challenge with S. fuliginea. The level of protection induced by various concentrations varied from solution to solution. In general, the systemic protection induced by K2HPO4 was similar to that by the microelements. Spraying of a 1:1 mixture of phosphate and micronutrient solutions did not improve the systemic protection over that obtained with each of the solutions alone. Increasing the inoculum concentration of S. fuliginea increased the number of powdery mildew colonies produced on both induced and non-induced plants and has relatively affected the systemic protection on induced plants. A single foliar spray of micronutrient solutions, as a prophylactic treatment, on the upper surface of all the leaves of 3-leaf stage cucumber plants significantly inhibited powdery mildew development. A single spray of MnCl2 on leaf 1 elevated peroxidase activity in the soluble fraction and caused an enhancement of -1,3-glucanase content in the ionically bound fractions of leaf 2 of non-inoculated plants. Forty-eight hours after inoculation, the level of both fractions of the enzymes increased in non-treated plants and decreased (-1,3-glucanase) or remained unchanged (peroxidase) in treated (induced) plants as compared to non-treated plants. The possible mechanism for this protection, and the use of microelements and phosphate solutions as inducers for systemic protection and as agents for disease control are discussed.  相似文献

In the present study, the hemagglutinating activity of 9 reference strains (serovars A-I) of Ornithobacterium rhinotracheale was investigated by using fresh erythrocytes from 15 different species: chicken (broiler, rooster, hen), turkey, pigeon, quail, duck, Harris hawk (Parabuteo unicinctus), house finch (Carpodacus mexicanus), cow, sheep, horse, dog, rabbit, pig, human (groups A, B, AB, and O), and rainbow trout (Oncorhynchus mykiss). All 9 strains agglutinated rabbit erythrocytes. None of the strains was able to agglutinate hen, cow, horse, or rainbow trout erythrocytes. The number of positive reactions among the remaining species varied. Results indicate that the use of rabbit erythrocytes is better suited for testing the hemagglutinating activity of O. rhinotracheale.  相似文献

Although the application of sequencing-by-synthesis techniques to DNA extracted from bones has revolutionized the study of ancient DNA, it has been plagued by large fractions of contaminating environmental DNA. The genetic analyses of hair shafts could be a solution: We present 10 previously unexamined Siberian mammoth (Mammuthus primigenius) mitochondrial genomes, sequenced with up to 48-fold coverage. The observed levels of damage-derived sequencing errors were lower than those observed in previously published frozen bone samples, even though one of the specimens was >50,000 14C years old and another had been stored for 200 years at room temperature. The method therefore sets the stage for molecular-genetic analysis of museum collections.  相似文献

Endophytic bacteria were isolated from the juice of a 6 month-old sugarcane cv. NiF-8, which was collected from Miyako, Okinawa and planted in a field lysimeter in Tsukuba, Ibaraki, Japan. The most probable number of N2-fixing endophytes was 4.5 × 105 cells per gram of fresh weight sample and 21 isolates have a positive reaction for ARA in an N2-free semi-solid medium with 10% crystallized sugarcane sugar and 0.5% sugarcane juice. Analyses of some of the biochemical properties of the N2-fixing isolates indicated that 13 isolates were putative strains of Acetobacter diazotrophicus, 4 isolates showed similar characteristics to those of Herbaspirillum seropedicae, and 4 isolates consisted of Herbaspirillum rubribalbicans-like strains. This study confirmed the existence of N2-fixing endophytic bacteria in the Japanese sugarcane cv. NiF-8.  相似文献

In the present study, Staphylococcus (S.) hyicus strains isolated in Russia (n = 23) and Germany (n = 17) were investigated for the prevalence of the previously described genes sheta and shetb. Sheta was detected in 16 S. hyicus strains. Sheta-positive strains were mainly found among strains isolated from exudative epidermitis, and frequently together with the exfoliative toxin-encoding genes exhD and exhC. Partial sequencing of sheta in a single S. hyicus strain revealed an almost complete match with the sheta sequence obtained from GenBank. None of the S. hyicus strains displayed a positive reaction with the shetb-specific oligonucleotide primer used in the present study. According to the present results, the exotoxin encoding gene sheta seems to be distributed among S. hyicus strains in Russia and Germany. The toxigenic potential of this exotoxin, which does not have the classical structure of a staphylococcal exfoliative toxin, remains to be elucidated.  相似文献
