During coffee seed development, proteins are predominantly deposited in cotyledons and in the endosperm. Reserve proteins of the 11S family are the most abundant globulins in coffee seeds, acting as a nitrogen source during roasting and guaranteeing flavor and aroma. The aim of the present study was to compare the protein profiles of endosperm and zygotic embryos of coffee seeds. Proteins were extracted from whole seed as well as from embryo and endosperm, separately. Total proteins were analyzed by two-dimensional electrophoresis (2-DE) followed by identification by mass spectrometry (MS). The most abundant spots observed in the gels of coffee seeds were excised, digested with trypsin, and identified by MS as subunits of the 11S globulin. Spots with identical pI and molecular masses were also observed in the protein profiles of coffee endosperm and embryo, indicating that 11S protein is also highly expressed in those tissues. Peptide sequence coverage of about 20% of the entire 11S globulin was obtained. Three other proteins were identified in the embryo and endosperm 2-DE profiles as a Cupin superfamily protein, an allergenic protein (Pru ar 1), exclusive to the endosperm 2D map, and a hypothetical protein, observed only in the zygotic embryo profile. 相似文献

Cotton (Gossypium sp.) is a major crop grown under rainfed conditions in Vertisols and associated soils in semi-arid tropical (SAT) regions of Peninsular India. In recent years, cotton productivity has declined due to various biophysical factors including pest and diseases, seasonal water stress soil degradation and poor crop management practices. In this study, we compare two methods for evaluating the suitability of Vertisols for cotton in contrasting two agro-ecological regions viz., sub-humid moist (SHM) region and semi-arid dry(SAD) were characterized. Twelve cotton growing Vertisols (seven from SHM and five from SAD) were evaluated for their suitability for cotton cultivation using soil quality index (SQI) and modified Sys-FAO method. SQIs were calculated using the weighted additive index from transformed scores of selected indicators by principal component analysis. For Sys-FAO method both biophysical and soil characteristics were considered for suitability evaluation. We found that the soils of SHM region were moderately suitable for cotton cultivation with soil moisture as the major limiting factor, whereas the soils of SAD region are marginally suitable due to high exchangeable sodium percentage and poor hydraulic conductivity. Peripheral nerves, spinal cord, and brain of healthy adult dogs were analysed biochemically for sodium, potassium, calcium, magnesium, copper, zinc, iron, aluminium, silicon, total chloride, total tissue water and total lipid content. Flame atomic absorption spectrophotometry was used for elemental quantitation of the hydrochloric acid tissue extracts. Cerebrum and spinal cord had similar values for all parameters measured. Total sodium values were higher in nerves compared to central nervous system (CNS) (brain and spinal cord), while respective values for potassium were lower. Tissue levels of calcium, iron and silicon were comparable in brain, spinal cord and nerves. However, magnesium concentrations were two to three times higher in CNS than in nerves; and the reverse was true for aluminium levels. Uredospore production per day and per sporulation period was measured under near-optimal conditions. Pustule density influenced time and rate of pustule opening, size of pustules, time of maximum sporulation, length of the sporulation period and the time and rate of tissue necrotisation. Within limits total dry weight of spores per leaf per sporulation period was independent of pustule density; it roughly equalled the dry weight of the spore producing leaf. The longest sporulation period observed was 65 days; at low pustule densities secondary pustules replaced exhausted primary pustules. Infectivity of the spores was normal up to 46 days after inoculation. The long sporulation period was epidemiologicaly interpreted as a survival mechanism. 相似文献

A possible method of protecting biosphere reserve core zones is to encourage transition and buffer zone activities which are
compatible with the core and which provide sufficient returns to the human population to make entry into the core zone unnecessary.
The AMISCONDE project is promoting block plantations and coffee/tree agroforestry systems to do this, but did not analyze
their financial or economic feasibility. Unprofitable systems will not be adopted without subsidy and are not sustainable
after a project ends. Analyses show internal rates of return exceeding 30% for coffee/tree combinations both with and without
project subsidies. Coffee/tree systems have significantly higher returns than coffee without trees, reduce the risk from coffee
price fluctuations, and reduce the need for chemical fertilizers. Returns from a block plantation of cypress are a small fraction
of coffee and coffee/tree system returns, but still are positive and have the advantage of requiring much smaller initial
investments. The systems analyzed seem likely to be sustainable and to contribute to project conservation and development
objectives.

The effect of Spirulina on iron status was assessed based on hemoglobin, packed cell volume, serum iron, total iron binding capacity and ferritin levels of rats during pregnancy and lactation. Rats were fed 5 different kinds of diets (casein, Spirulina, wheat gluten, Spirulina + wheat gluten, Spirulina without additional vitamins and minerals) each providing 22 percent protein. Diets containing Spirulina alone or in combination with wheat gluten resulted in significantly higher iron storage and hemoglobin contents than casein and wheat gluten diets during the first half of pregnancy and lactation. Wheat gluten diet result in the smallest increase in hemoglobin levels and iron stores compared to other diets. The values of serum iron and iron binding capacity remained unchanged with different diets. Spirulina appears to be effective in improving the iron status of rats during pregnancy and lactation. 相似文献

A granulosis virus strain infecting Pieris brassicae (PbGV) was isolated from the dry temperate region of northwestern Himalayas as a potential microbial agent for its management.
The effect of different botanicals (having insecticidal action against P. brassicae) on the bioefficacy of PbGV was evaluated under laboratory conditions using leaf disc bioassays on cabbage for improving
the insecticidal performance of the PbGV. The synergistic action of different botanical extracts was evident in terms of reduction
in LC50 values against different botanical extracts. Among different extracts, petroleum-ether extract of neem seed kernel (NSK)
when combined with PbGV resulted in maximum reduction of LC50 value (4.39 × 102 occlusion bodies [OBs] ml−1) followed by methanolic extract (7.38 × 102 OBs ml−1) and aqueous extract (9.36 × 103 OBs ml−1) as compared with PbGV alone (1.85 × 104 OBs ml−1) for 2nd instar larvae of the test insect. These trends were found analogous in cases of 3rd and 4th instars of P. brassicae with different solvent extracts of NSK. The other botanicals evaluated, viz., Eupatorium and Artemesia, also resulted in reduction of LC50 values for 2nd, 3rd and 4th instars as compared with PbGV alone when different extracts were combined with virus for bioassays.
The studies suggest that the PbGV in combination with botanical pesticides could be more useful as a bio-pesticide against
cabbage butterfly (P. brassicae) in IPM programs. 相似文献

The physicochemical and gel properties of myofibrillar protein (MFP) from sin croaker fish were studied during ice storage for 18 days. Significant changes in the trends of solubility, Ca2+ ATPase enzyme activity, surface hydrophobicity, and water holding capacity of extracted MFP were observed by the 6th day of storage. The Ca2+ ATPase enzyme activity reduced significantly (p < 0.05) by the 6th day of storage. However, sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) did not show a remarkable change in the concentration of myosin heavy chain. Surface hydrophobicity increased almost four times from its original value of 18.98 µg; whereas, water holding capacity showed a fluctuating trend during storage. The emulsion capacity of the MFP was in the range of 0.89- to 1.92-mL oil/mg protein during storage. The gel strength value (313.45 g.cm) of heat-induced gel prepared from fresh minced meat decreased significantly (p < 0.05) by the 6th day of ice storage. Texture profile analysis revealed that changes in hardness and gumminess were concurrent to steeply reducing breaking force up to the 6th day. The histological observation showed gradually increasing gaps between muscle fibers. The histological observations and physicochemical quality indicated that the sin croaker fish can be used for producing good quality surimi when stored for up to 6 days in ice. 相似文献
