Legumes form symbiotic associations with both mycorrhizal fungi and nitrogen-fixing soil bacteria called rhizobia. Several of the plant genes required for transduction of rhizobial signals, the Nod factors, are also necessary for mycorrhizal symbiosis. Here, we describe the cloning and characterization of one such gene from the legume Medicago truncatula. The DMI1 (does not make infections) gene encodes a novel protein with low global similarity to a ligand-gated cation channel domain of archaea. The protein is highly conserved in angiosperms and ancestral to land plants. We suggest that DMI1 represents an ancient plant-specific innovation, potentially enabling mycorrhizal associations.  相似文献

Studies were carried out in the glass house using sand culture to find out the specific ion effect on the accumulation of abscisic acid in wheat. Seeds were sown in sixteen different solutions of chloride, sulphate, nitrate, carbonate salts of sodium, potassium, calcium and magnesium used each at ‐2 bar osmotic potential. The leaf samples taken after 25 days of sowing indicated that osmotic as well as salt stress increased significantly the accumulation of abscisic acid. The accumulation of abscisic acid was significantly less under potassium and calcium salt ions as compared to that under sodium and magnesium salts ions. Overall effect of anions showed that accumulation of abscisic acid was more under PEG (osmotic stress and chloride salts but it was less under sulphate and nitrate salts and was the least under carbonate salts. The accumulation of abscisic acid was exceptionally high under potassium dihydrogen phosphate. In general, the accumulation of abscisic acid‐a growth inhibitor varied not only with the type of the cation but also depended on the accompanying anion indicating differential growth response of plants under different types of saline conditions having dominance of different salt ion(s).  相似文献

Salinity toxicity is a worldwide agricultural and eco-environmental problem. The intent of this study was to determine the salt tolerance of Piriformospora indica and strains of Azospirillum, isolated from non-saline and saline soil, as well as to determine their affect on the tolerance of wheat to soil salinity. In this study, an experiment was conducted to investigate the salt stress tolerance abilities of the endophytic fungi, P. indica, and Azospirillum strains, isolated from non-saline and saline soil, at five NaCl levels (0, 0.1, 0.2, 0.3, 0.4, 0.5 mol L?1). Additionally, a greenhouse experiment was conducted to test the effects of these selected microorganisms under increasing salinity levels on seedling growth, solute accumulation (proline and sugars), and photosynthetic pigments (Chl a, b, ab) of seedling wheat. Azospirillum strains were isolated in Iran from the root of field-grown maize from non-saline soil with an EC = 0.7 dS m?1 and from saline soil with an EC = 4.7 dS m?1. Plants were irrigated with non-saline water–tap water with an electrical conductivity water (ECw) value of 0.2 dS m?1, as well as low, moderate and severe saline water-irrigation with saline water with an ECw of 4 dS m?1, 8 dS m?1 and 12 dS m?1, respectively. The upper threshold of P. indica salinity tolerance was 0.4 mol L?1 NaCl in both liquid and solid broth medium. The upper thresholds of the salt adapted and non-adapted Azospirillum strains were 0.2 and 0.4 mol L?1 NaCl, respectively. The results indicated a positive influence of the organisms on salinity tolerance, more with the saline-adapted Azospirillum strains than the non-adapted strains. P. indica was more effective than the Azospirillum strains. These results could be related to a better water status, higher photosynthetic pigment contents and proline accumulation in wheat seedlings inoculated with P. indica. The benefits of both isolates and P. indica depended on two factors: water salinity and growth stage of the host plant. Inoculation with the two isolates increased salinity tolerance of wheat plants; the saline-adapted Azospirillum strains showed better performance with respect to improved fresh and dry weights at 80 and 100 days after sowing under both non-saline and saline conditions. When compared to plants inoculated with non-saline-adapted Azospirillum strains, those inoculated with adapted Azospirillum strains had much better performance with respect to the presence of photosynthetic pigment (Chl a, b and ab) and proline accumulation. Overall, these results indicate that the symbiotic association between P. indica fungus and wheat plants improved wheat growth, regardless of the salinity. It is concluded that the mechanisms for protecting plants from the detrimental effects of salinity by P. indica fungus and Azospirillum strains may differ in their salinity tolerance and influence the uptake of water, photosynthetic pigment contents and proline accumulation in wheat seedlings.  相似文献

Recent studies on the protection afforded by moderate wine consumption against cardiovascular diseases have focused mainly on the activity of red wine in view of its high content of antioxidants, especially polyphenols. White wine lacks polyphenols, but it contains other compounds such as hydroxycinnamic acids (caffeic acid) and monophenols (tyrosol), which are known to have antioxidant properties. Therefore, this study was designed to examine the effect of white wine in myocardial ischemic-reperfusion injury. The experimental rats were gavaged with white wine (Soave Suavia "Le Rive" 2004) at a dosage of 6.5 mL/(kg.rat.day) for 30 days. Rats were divided into four groups: control sham (CS), wine-treated sham (WS), control ischemia (I)/reperfusion (R) (CIR), and wine + IR (WIR). All the rats in both IR groups underwent 30 min occlusion of the left anterior descending coronary artery followed by 8, 24 h, and 30 days of reperfusion (R). Significant reduction in infarct size (21 vs 39%, n = 6), cardiomyocyte (274 vs 384 counts/100 HPF, n = 6), and endothelial cell apoptosis (387 vs 587 counts/100 HPF) was observed in WIR as compared with CIR after 24 h of reperfusion. Echocardiography demonstrated significant increased fractional shortening (32 vs 22%) and ejection fraction (60 vs 44%) following 30 days of reperfusion in WIR rats compared to CIR ( n = 6). In addition, increased phosphorylation of AKT, Foxo3a, and eNOS were found in WS and WIR, as compared to their respective controls. The gel-shift analysis demonstrated significant upregulation of DNA binding activity of NF-kappaB in the white wine-treated groups. This report demonstrated for the first time that the white wine mediated cardioprotection in ischemic reperfused myocardium is through the PI-3kinase/Akt/FOXO3a/e-NOS/NF-kappaB survival pathway.  相似文献

The efficacy of emamectin benzoate as an oral treatment of sea lice, Lepeophtheirus salmonis (KrÒyer), infestations in Atlantic salmon, Salmo salar L., was evaluated in a dose titration study and two dose confirmation studies. Replicated groups of salmon with induced infestations of sea lice were given emamectin benzoate on pelleted feed at doses of 0, 25, 50 and 100 μg kg−1 biomass day−1 for seven consecutive days. Sea lice were counted at 7, 14 and 21 days from the start of treatment, and comparisons made with control fish given the same diet without emamectin benzoate. Total numbers of sea lice were significantly reduced at all doses in all three studies when compared to control fish. There was no significant difference between doses of 50 and 100 μg kg−1, but the 50 μg kg−1 dose resulted in significantly fewer lice than the 25 μg kg−1 dose. Emamectin benzoate was highly effective in reducing numbers of preadult and adult lice and prevented the maturation of chalimus to motile stages. The optimum therapeutic dose was selected as 50 μg kg−1 day−1 for seven days. Treatment reduced the incidence of epidermal damage by sea lice and, in one study, survival of treated fish was 48% higher than in control groups. No fish mortalities or adverse effects were attributed to treatment with emamectin benzoate at any of the doses tested.  相似文献

Objective of investigation

Chickpea is a major global food legume for which seed weight and plant growth habit are important yield and harvestability components for plant breeding. This study tested seed weight and plant growth habit inheritance and identified quantitative trait loci (QTL).

Experimental material

A 10 nested association mapping (NAM) populations of chickpea were created from crosses between ‘Gokce’, a cultivar and wild crop relative accessions of Cicer reticulatum and Cicer echinospermum. Families were then developed to the F2:4 generation.

Method of investigation

A 10 families were grown at the Field Experiment Station, Harran University near Şanlıurfa, Turkey during 2019.

Data collection

A 100-seed weight and prostrate or erect growth habit was scored in the field. Two families were genotyped for 60 single-nucleotide polymorphisms (SNP).

Result and conclusions

A 100-seed weight showed polygenic control, and three QTLs were found. Growth habit was controlled by one or two QTLs. The two traits were significantly correlated for five populations. The crop wild relatives of chickpea contain variations at novel loci affecting seed weight compared to the literature.  相似文献

A total of 90 genotypes of Lycopersicon species were tested forresistance to the Tomato leaf curlgeminivirus (ToLCV) by agroinoculation andthe vector whitefly (Bemisia tabaciGenn.) inoculation techniques underinsect-proof glasshouse conditions. Therate of infection in the inoculated plantswas determined by detection of the viralDNA in individual plants by the nucleicacid spot hybridization (NASH). Of the 38cultivars and 11 breeding lines of L.esculentum Mill. tested, none was highlyresistant or resistant while three andseven were moderately resistant whenexposed to the cloned virus DNAs byagroinoculation and whitefly inoculationrespectively. On the other hand, among the38 commercial cultivars screened, 16(42.1%) were highly susceptible in vectorinoculations and 31 (81.6%) inagroinoculation. Among the exoticcollection (EC) accessions six were highlyresistant, eleven resistant to whiteflyinoculation and none was highly susceptiblein either of the two tests, indicating thepresence of resistance among the ECaccessions. A higher degree of resistancewas observed in other species of Lycopersicon. While only one accession ofL. cheesmanii Riley was tested, itcould not be infected by either of the twomethods. L. pimpinellifolium (Jusl.)Mill. genotype EC 251580 was similarlyresistant. In L. peruvianum (L.)Mill., five EC accessions could not beinfected by whitefly inoculation, withthree of these being resistant and twomoderately resistant in agroinoculation.This study demonstrates the importance ofthe agroinoculation technique in the virusresistance screening programs andidentifies several good sources ofresistance to the Tomato leaf curlvirus in Lycopersicon species.  相似文献

Abstract

The inevitable large‐scale plant germplasm exchanges between countries for crop improvement have led to strict plant quarantine inspections, safeguards, and sometimes restrictions. The paper describes the procedures and techniques adopted for the safe exchange by the International Crops Research Institute for the Semi‐Arid Tropics (ICRISAT) plant germplasm in India. Safeguards for the imported germplasm include intermediate quarantine for groundnut (Arachis sp.) cuttings, fumigation, dry seed inspection, and microscopic examination followed by seed treatment and growing of healthy crops in a quarantine isolation area before releasing seeds of disease free progeny to the scientists. Similarly, inspection of crops before collection of seeds followed by fumigation, microscopic examination, enzyme‐linked immunosorbent assay (ELISA) test for groundnut viruses, seed treatment, and finally good packing have been found effective for export of healthy seeds.  相似文献
