The aim of the present study was to provide current data on the prevalence of anti-Yersinia-antibodies in Bavarian slaughtering pigs. The recomBlot Westernblot assay based on five recombinantly produced Yersinia Outer Proteins (YopD, YopE, YopH, YopM, V-antigen) is well evaluated for the use in the diagnosis of human yersiniosis and proved to be also a valuable tool for the detection of porcine antibodies. 965 out of 1002 meat juice samples collected in 53 Bavarian pig farms contained anti-Yop/V-Ag-antibodies. Only blots with three or more detectable signals were considered as positive. On the basis of these results a possible previous history of Yersinia infection was detected in 45.4% of the pigs and all of the pig farms. The high prevalence of anti-Yersinia-antibodies found in this investigation demonstrates the need for further epidemiological surveillance and animal studies to assess consumers' endangering.  相似文献

Compounds released by plant roots during growth can make up a high proportion of below-ground plant (BGP) carbon and nitrogen, and therefore influence soil organic matter turnover and plant nutrient availability by stimulating the soil microorganisms. The present study was conducted to examine the amount and fate of C (CdfR) and N rhizodeposits (NdfR), in this study defined as root-derived C or N present in the soil after removal of roots and root fragments, released during reproductive growth. BGP biomass of peas (Pisum sativum L.) and oats (Avena sativa L.) was successfully labelled in situ with a 13C-glucose-15N-urea mixture under field conditions using a stem feeding method. Pea plants were labelled at the beginning of flowering and harvested 36 and 52 days after labelling at pod filling (PP) and maturity (PM), respectively. Oat plants were labelled at grain filling and harvested 42 days after labelling at maturity (OM). CdfR was 24.2% (PP), 29.6% (PM) and 30.8% (OM) of total recovered plant C. NdfR was 32.1% (PP), 36.4% (PM) and 30.0% (OM) of total plant N. Due to higher N assimilation, amounts of NdfR were four times higher in peas in comparison with oats. The results for NdfR in peas were higher than results from other studies. The C-to-N ratio of rhizodeposits was lower under peas (17.3) than under oats (41.9) at maturity. At maturity, microbial CdfR at 0-30 cm soil depth was 37% of the microbial biomass C in peas and 59% in oats. Microbial NdfR was 15% of microbial N in peas and 5% in oats. Furthermore, inorganic NdfR was 34% in peas and 9% in oats at 0-30 cm at maturity. These results show that rhizodeposits of peas provide a more easily available substrate to soil microorganisms, which are incorporated to a greater extent and turned over faster in comparison with oats. Beside the higher amounts of N released from pea roots, this process contributes to the higher N-availability for subsequent crops.  相似文献

Different types of transmissible spongiform encephalopathies (TSEs) affect sheep and goats. In addition to the classical form of scrapie, both species are susceptible to experimental infections with the bovine spongiform encephalopathy (BSE) agent, and in recent years atypical scrapie cases have been reported in sheep from different European countries. Atypical scrapie in sheep is characterized by distinct histopathologic lesions and molecular characteristics of the abnormal scrapie prion protein (PrP(sc)). Characteristics of atypical scrapie have not yet been described in detail in goats. A goat presenting features of atypical scrapie was identified in Switzerland. Although there was no difference between the molecular characteristics of PrP(sc) in this animal and those of atypical scrapie in sheep, differences in the distribution of histopathologic lesions and PrP(sc) deposition were observed. In particular the cerebellar cortex, a major site of PrP(sc) deposition in atypical scrapie in sheep, was found to be virtually unaffected in this goat. In contrast, severe lesions and PrP(sc) deposition were detected in more rostral brain structures, such as thalamus and midbrain. Two TSE screening tests and PrP(sc) immunohistochemistry were either negative or barely positive when applied to cerebellum and obex tissues, the target samples for TSE surveillance in sheep and goats. These findings suggest that such cases may have been missed in the past and could be overlooked in the future if sampling and testing procedures are not adapted. The epidemiological and veterinary public health implications of these atypical cases, however, are not yet known.  相似文献

The melanocortin 1 receptor (MC1R) regulates pigmentation in humans and other vertebrates. Variants of MC1R with reduced function are associated with pale skin color and red hair in humans of primarily European origin. We amplified and sequenced a fragment of the MC1R gene (mc1r) from two Neanderthal remains. Both specimens have a mutation that was not found in approximately 3700 modern humans analyzed. Functional analyses show that this variant reduces MC1R activity to a level that alters hair and/or skin pigmentation in humans. The impaired activity of this variant suggests that Neanderthals varied in pigmentation levels, potentially on the scale observed in modern humans. Our data suggest that inactive MC1R variants evolved independently in both modern humans and Neanderthals.  相似文献

“Effective microorganisms” (EM) are a poorly defined mixture of supposedly beneficial microorganisms that are claimed to enhance microbial turnover in compost and soil. In Costa Rica, EM are used to produce organic compost (bokashi) from banana residues (Musa ssp.). Given the scarcity of scientific data about the effects of EM on the mineralization of plant residues, this study aimed at investigating the effects of EM addition on the decomposition of banana residues during Bokashi production. To this end, the following non‐EM treatments were compared to EM Bokashi: Bokashi produced with water (W), with molasses (M) as an EM additive, and with sterilized EM (EMst). Subsequently, the effects of the resulting Bokashi treatments on the growth of young banana plants were evaluated. Compared with non‐EM controls, the effect of EM on the mineralization of banana material was negligible. Dry‐matter losses of the composts with different EM treatments were similar, with about 78% over 5 weeks. Ergosterol concentration was highest in EM Bokashi (77 µg (g dry soil)–1) and lowest in EMst Bokashi (29 µg (g dry soil)–1). Microbial biomass carbon (Cmic) and microbial biomass nitrogen (Nmic) were both lowest in EM (Cmic = 3121 µg g–1; Nmic = 449 µg g–1), while Cmic was highest in Bokashi produced with molasses (3892 µg g–1) and Nmic was highest in EMst (615 µg g–1). Treatment effects on adenylate concentrations and adenylate energy charge were negligible. Application of all Bokashi variants to young banana plants significantly increased shoot growth under greenhouse conditions compared to plants grown in a control soil without amendments. However, these effects were similar for all Bokashi treatments, even if EM Bokashi increased the K concentrations in banana leaves significantly compared to Bokashi produced with EMst and the control. Bokashi produced with only molasses and EM Bokashi decreased the number of root nematodes under greenhouse conditions compared to the control. Overall, the results confirmed the expected influence of composting on the degradation of organic material and the effect of compost application on plant growth. Hower, under the conditions of this study, EM showed no special effects in this, except for increasing the K concentrations in the leaves of young banana plants.  相似文献

A field experiment with winter oilseed rape was conducted near Göttingen (northern Germany) in the growing season 1998–99. Twelve varieties were compared at two nitrogen (N) application rates (0, 206 kg N ha–1) regarding shoot dry matter production, shoot N content, shoot N uptake (beginning of shooting, beginning of flowering), and seed yield. Canopy reflectance was measured one week before the beginning of shooting and one week before flowering at different wavelengths between 550 and 940 nm using a spectral reflectance sensor, type FAL II. The vegetation indices red edge inflection point (RIP), soil adjusted vegetation index (SAVI), and normalized vegetation index (NDVI) introduced in literature as indicators of growth parameters were calculated from the reflectance values and compared with growth parameters. The results showed that the measuring angle (–45° to 45°) had a significant influence on the calculated vegetation indices. Therefore, the measuring angle should be kept constant during the measurements. Nitrogen fertilization led to an increase of all vegetation indices. The results revealed significant differences between the tested varieties, indicating problems regarding an accurate estimation of the N status of the crop. Consistent differences in the vegetation indices appeared between varieties with extremely different growth parameters. However, no general relationship between growth parameters and vegetation indices of the 12 varieties could be found. Therefore, variety‐specific calibrations are necessary. The indices RIP and SAVI resulted in similar statements and seem to be good parameters regarding N effects and variety differences, whereas NDVI could resolve variety differences in the fertilized treatments only imperfectly.  相似文献

The availability of O2 is one of the most important factors controlling the chemical and biological reactions in soils. In this study, the effects of different aeration conditions on the dynamics of the emission of trace gases (CO2, N2O, CH4) and the leachate composition (NO3, DOC, Mn, Fe) were determined. The experiment was conducted with naturally structured soil columns (silty clay, Vertisol) from a well aerated forest site. The soil monoliths were incubated in a microcosm system at different O2 concentrations (0, 0.001, 0.005, 0.01, 0.05, and 0.205 m3 m‐3 in the air flow through the headspace of the microcosms) for 85 days. Reduced O2 availability resulted in a decreased CO2 release but in increased N2O emission rates. The greatest cumulative N2O emissions (= 1.6 g N2O‐N m‐2) were observed at intermediate O2 concentrations (0.005 and 0.01 m3 m‐3) when both nitrification and denitrification occurred simultaneously in the soil. Cumulative N2O emissions were smallest (= 0.05 g N2O‐N m‐2) for the aeration with ambient air (O2 concentration: 0.205 m3 m‐3), although nitrate availability was greatest in this treatment. The emission of CH4 and leaching of Mn and Fe were restricted to the soil columns incubated under completely anoxic conditions. The sequence of the reduction processes under completely anoxic conditions complied with the thermodynamic theory: soil nitrate was reduced first, followed by the reduction of Mn(IV) and Fe(III) and finally CO2 was reduced to CH4. The re‐aeration of the soil columns after 85 days of anoxic incubation terminated the production of CH4 and dissolved Fe and Mn in the soil but strongly increased the emission rates of CO2 and N2O and the leaching of NO3 probably because of the accumulation of DOC and NH4+ during the previous anoxic period.  相似文献

Plants have developed numerous strategies for responding to abiotic and biotic stresses. In particular, the microbiota surrounding plants may have a positive effect on plant stress responses. One is the reaction to rhizobacteria, which can lead to induced systemic resistance. Gram-negative soil bacteria that produce N-acyl homoserine lactones (AHL), for example, Ensifer meliloti, induce a primed state in plants that is part of the inducible resistance phenomenon. Observing Rhopalosiphum padi feeding behaviour on a priming sensitive barley genotype, treated with the AHL-producing E. meliloti strain expR?+?ch, using electrical penetration graph technique showed decreased ingestion of food. Aphids appear to overcome this effect within the eight-hour observation period, possibly explaining the absence of differences of reproduction. Reproduction was observed for a period of 14 days on primed and control-treated plants. Long-term observations over a period of 40 days after aphid infestation showed a lower aphid biomass in contrast to a control group, interpreted as delayed population growth, and an increase in the biomass of barley plants. Priming-related genotypic effects of the defence response to aphids were observed, with no beneficial effects on the plant genotype when its sensitivity to priming was low. Previously, an AHL-priming sensitive barley genotype showed enhanced resistance against fungi when primed with the expR?+?ch strain of E. meliloti. The present study reports the same effect against R. padi. These findings suggest that sensitivity to AHL-priming may represent a new approach for plant breeding, targeting multiple pests in parallel by induced plant resistance.

Vegetable‐production systems often show high soil mineral‐N contents and, thus, are potential sources for the release of the climate‐relevant trace gas N2O from soils. Despite numerous investigations on N2O fluxes, information on the impact of vegetable‐production systems on N2O emissions in regions with winter frost is still rare. This present study aimed at measuring the annual N2O emissions and the total yield of a lettuce–cauliflower rotation at different fertilization rates on a Haplic Luvisol in a region exposed to winter frost (S Germany). We measured N2O emissions from plots fertilized with 0, 319, 401, and 528 kg N ha–1 (where the latter three amounts represented a strongly reduced N‐fertilization strategy, a target value system [TVS] in Germany, and the N amount fertilized under good agricultural practices). The N2O release from the treatments was 2.3, 5.7, 8.8, and 10.6 kg N2O‐N ha–1 y–1, respectively. The corresponding emission factors calculated on the basis of the total N input ranged between 1.3% and 1.6%. Winter emission accounted for 45% of the annual emissions, and a major part occurred after the incorporation of cauliflower residues. The annual N2O emission was positively correlated with the nitrate content of the top soil (0–25 cm) and with the N surpluses of the N balance. Reducing the amount of N fertilizer applied significantly reduced N2O fluxes. Since there was no significant effect on yields if fertilization was reduced from 528 kg N ha–1 according to “good agricultural practice” to 401 kg N ha–1 determined by the TVS, we recommend this optimized fertilization strategy.  相似文献

Sarcoptic mange occurs in many parts of the world and is common in populations of domestic and wild canids, including red foxes (Vulpes vulpes). In recent years, an indirect antibody enzyme-linked immunosorbent assay (ELISA), with higher sensitivity and specificity than traditional diagnostic methods, has been successfully applied in the diagnosis of sarcoptic mange in dogs. The same ELISA has also demonstrated specific antibodies to Sarcoptes scabiei in experimentally infected red foxes. The aim of this study was to evaluate the indirect ELISA when used to detect antibodies to S. scabiei in field sera from Swedish red foxes. One cohort of both infected and non-infected red foxes (cohort 1; n = 88), and one cohort of apparently non-infected foxes (cohort 2; n = 67) were examined for skin lesions and presence of S. scabiei by thorough visual examination at autopsy and skin scrapings. Samples of blood-tinted body liquid from the abdomen or thorax cavity were collected and analysed by the indirect ELISA. The relative sensitivity and specificity of the ELISA at different cut-offs (OD values) were estimated by comparing the test results to the infection status as determined by examination and skin scrapings. The highest combination of relative sensitivity and specificity, calculated based on cohort 1, was 95.4 and 100.0%, respectively. These estimates were constant for cut-offs 0.150-0.225, which included the cut-off based on the mean plus three standard deviations of test results from cohort 2 (0.165). It is concluded that this test can be useful in diagnosis and epidemiological studies of S. scabiei infection in red foxes.  相似文献
