The elaboration of novel techniques for flavonoid intracellular tracing would elucidate the compounds' absorption and bioavailability and assist molecular and pharmacological approaches, as they are promising candidates for drug development. This study exploited the properties of quercetin (3,3',4',5,7-pentahydroxyflavone), found in high concentrations in the majority of edible plants. Through the use of UV-vis spectroscopy, confocal microscopy, and HPLC-ESI-MS, native quercetin, at physiologically relevant concentrations, was found to exhibit a specific fluorescence (488 nmex/500-540 nmem) upon internalization. This fluorescence shift is due to a non-covalent binding to intracellular targets (probably proteins) and compatible with the settings applied in confocal microscopy. This property provides a valuable, selective alternative technique for quercetin tracing in cellular systems, permitting the quantitative evaluation of its transit at pharmacologically relevant concentrations and the validation of a number of already described molecular functions.  相似文献

Anthocyanins (Anths) in grape (Vitis vinifera L.) berries harvested at véraison from Pinot Noir and Pinot Meunier cultivars were assessed nondestructively by measuring chlorophyll fluorescence (ChlF) excitation spectra. With increasing Anth content, less excitation light was transmitted to the deeper Chl layers, and thus the ChlF signal decreased proportionally. By applying Beer-Lambert's law, the logarithm of the ratio between the fluorescence excitation spectra (log FER) from a green and a red berry gave the in vivo absorption spectrum of Anths, which peaked at about 540 nm. Absolute quantitative nondestructive determination of Anths for each berry was obtained by the log FER calculated for two excitation wavelengths, 540 and 635 nm (absorbed and not-absorbed by Anths, respectively) of ChlF at 685 nm. Over a range of skin colors going from green to purple, the relationship between the log [ChlF(635)/ChlF(540)] and the Anth concentration of berry extracts was fairly well fitted (r 2 = 0.92) using a power function. Reflectance spectra on the same berry samples were also measured, and Anth reflectance indices, which were originally developed for apples and table grapes, were derived. The log FER Anth index was superior to the reflectance-ratio-based index, but was as good as the color index for red grapes (CIRG) calculated from the whole visible reflectance spectrum. The proposed log FER method, applied by means of suitable portable devices, may represent a new, rapid, and noninvasive tool for the assessment of grape phenolic maturity in vineyards.  相似文献

In the last few years, landscape researchers have sought to understand temporal and spatial patterns of landscape changes in order to develop comprehensive models of land cover dynamics. To do so, most studies have used similar methods to quantify structural patterns, usually by comparing various landscape structural indices through time. Whereas the necessity for complementary approaches which might provide insights into landscape dynamics at some finer scale relevant to local managers has been expressed, few studies have proposed alternative methodologies. Moreover, the important relationship between the physical constraints of the landscape and land use dynamics has been seldom emphasized. Here we propose a methodological outline which was applied to the study of a rural landscape of Southern Quebec, Canada, to detect spatial and temporal (1958 to 1993) patterns of land cover changes at field, patch and landscape level. We then relate these patterns to the underlying physical structure of landscape elements using GIS and canonical correspondence analyses. We use the different geomorphological deposit types as stable discriminant factors which may constrain land use.Canonical correspondence analyses showed relations of land use and land use changes to the physical attributes of the landscape elements, whereas spatial analyses revealed very dynamic patterns at finer spatial and temporal scales. They highlighted the fact that not only the physical attributes of the landscape elements but also their spatial configuration were important determinants of land use dynamics in this area. Thus more land use changes occurred at the boundary between geomorphological deposit types than in any other locations. This trend is apparent for specific small-size changes (e.g. forest to crop), but not for the large-size ones (e.g. abandoned land to forest). Although land use changes are triggered by socioeconomic forces in this area, these changes are nevertheless constrained by the underlying physical landscape structure. A thorough comprehension of historical changes will enhance our capability to predict future landscape dynamics and devise more effective landscape management strategies.  相似文献

Our understanding of how mineral nutrition affects productivity and composition of bioenergy crops grown on marginal lands remains fragmented and incomplete despite world‐wide interest in using herbaceous biomass as an energy feedstock. Our aim was to determine switchgrass (Panicum virgatum L.) biomass production and maize (Zea mays L.) grain yield on marginal soils used previously to evaluate the effect of soil phosphorus (P) and potassium (K) fertility on alfalfa (Medicago sativa L.) forage production. Grain yield of maize was reduced on P‐ and/or K‐limited plots that also impaired alfalfa forage yield, whereas switchgrass biomass yields were high even in plots possessing very low available P (4 mg kg–1) and K (< 70 mg kg–1) levels. Linear‐plateau regression models effectively described the relationship of soil test P and K to tissue P and K concentrations, and tissue P and K concentrations accurately predicted removal of P and K in harvest biomass. However, neither soil‐test P and K, nor tissue P and K concentrations were effective as diagnostics for predicting switchgrass biomass yield nor could soil tests and their change with cropping predict nutrient removal. Concentrations of cellulose, hemicellulose, lignin, and ash were not influenced by P and K nutrition. Predicted bio‐ethanol production was closely associated with biomass yield whereas high biomass K concentrations reduced estimated bio‐oil production per hectare by as much as 50%. Additional research is needed to identify diagnostics and managements to meet the bioenergy production co‐objectives of having high yield of biomass with very low mineral nutrient concentrations (especially K) while sustaining and improving the fertility of marginal soils.  相似文献

We compared N fluxes in a 150-year-old Fagus sylvatica coppice and five adjacent 25-year-old plantations of Fagus sylvatica, Picea abies, Quercus petraea, Pinus laricio and Pseudotsuga menziesii. We measured net N mineralization fluxes in the upper mineral horizon (A1, 0–5 cm) for 4 weeks and gross N mineralization fluxes for two days. Gross rates were measured during the 48-h period after addition of 15NH4 and 15NO3. Mineralization was measured by the 15NH4 dilution technique and gross nitrification by 15NO3 production from the addition of 15NH4, and by 15NO3 dilution. Net and gross N mineralization was lower in the soil of the old coppice, than in the plantations, both on a soil weight and organic nitrogen basis. Gross nitrification was also very low. Gross nitrification measured by NO3 dilution was slightly higher than measured by 15NO3 production from the addition of 15NH4. In the plantations, gross and net mineralization and nitrification from pool dilution were lowest in the spruce stand and highest in the beech and Corsican pine stands. We concluded that: (1) the low net mineralization in the soil of the old coppice was related to low gross rate of mineralization rather than to the concurrent effect of microbial immobilisation of mineral N; (2) the absence of nitrate in the old coppice was not related to the low rate of mineralization nor to the absence of nitrifyers, but most probably to the inhibition of nitrifyers in the moder humus; (3) substituting the old coppice by young stands favours nitrifyer communities; and (4) heterotrophic nitrifyers may bypass the ammonification step in these acid soils, but further research is needed to check this process and to characterize the microbial communities.  相似文献

Nineteen commercial hexaploid wheat varieties were crossed with the maize F1 hybrid ‘Seneca 60’. Fertilization frequencies ranged from 32.1 % to 47.5 % of pollinated florets (mean 39.5 %) in the 14 winter wheat varieties and from 40.7 % to 51.4 % (mean 47.8 %) in the five spring wheat varieties. In some cases only an endosperm was formed and the frequencies of embryo formation were therefore slightly lower, being 28.2 % to 45.9 % (mean 36.4 %) for winter wheats and 39.8 % to 48.6 % (mean 45.1 %) for spring wheats. Mean values were significantly higher in the spring wheats but no significant variation was found between varieties within the spring or winter categories. In the five spring wheats the mean yield of embryos, and hence the potential yield of haploid plants, was 3.4-fold higher than with the tetraploid Hordeum bulbosum clone PB179. For the 14 winter wheats the figure was 10.9-fold higher. These differences were highly significant (p < 0.001) in all varieties. A single 2,4-D treatment given to spikes one day after pollination with maize enabled embryos to be recovered from all 19 varieties. A total of 311 embryos were recovered from 950 florets (an average of 7.3 embryos per spike) of which 191 germinated, giving an average yield of one haploid plant for every 5.0 florets pollinated (4.4 haploid plants per spike).  相似文献
