Forty percentage of UK peatlands have been drained for agricultural use, which has caused serious peat wastage and associated greenhouse gas emissions (carbon dioxide (CO2) and methane (CH4)). In this study, we evaluated potential trade-offs between water-table management practices for minimizing peat wastage and greenhouse gas emissions, while seeking to sustain romaine lettuce production: one of the most economically relevant crop in the East Anglian Fenlands. In a controlled environment experiment, we measured lettuce yield, CO2, CH4 fluxes and dissolved organic carbon (DOC) released from an agricultural fen soil at two temperatures (ambient and +2°C) and three water-table levels (−30 cm, −40 cm and −50 cm below the surface). We showed that increasing the water table from the currently used field level of −50 cm to −40 cm and −30 cm reduced CO2 emissions, did not affect CH4 fluxes, but significantly reduced yield and increased DOC leaching. Warming of 2°C increased both lettuce yield (fresh leaf biomass) and peat decomposition through the loss of carbon as CO2 and DOC. However, there was no difference in the dry leaf biomass between the intermediate (−40 cm) and the low (−50 cm) water table, suggesting that romaine lettuce grown at this higher water level should have similar energetic value as the crop cultivated at −50 cm, representing a possible compromise to decrease peat oxidation and maintain agricultural production.  相似文献

Landscape genetics integrates theory and analytical methods of population genetics and landscape ecology. Research in this area has increased in recent decades, creating a plethora of options for study design and analysis. Here we present a practical toolbox for the design and analysis of landscape genetics studies following a seven-step framework: (1) define the study objectives, (2) consider the spatial and temporal scale of the study, (3) design a sampling regime, (4) select a genetic marker, (5) generate genetic input data, (6) generate spatial input data, and (7) choose an analytical method that integrates genetic and spatial data. Study design considerations discussed include choices of spatial and temporal scale, sample size and spatial distribution, and genetic marker selection. We present analytical methods suitable for achieving different study objectives. As emerging technologies generate genetic and spatial data sets of increasing size, complexity, and resolution, landscape geneticists are challenged to execute hypothesis-driven research that combines empirical data and simulation modeling. The landscape genetics framework presented here can accommodate new design considerations and analyses, and facilitate integration of genetic and spatial data by guiding new landscape geneticists through study design, implementation, and analysis.  相似文献

Feline immunodeficiency virus (FIV) is a naturally occurring lentivirus of domestic cats, and is the causative agent of feline AIDS. Similar to human immunodeficiency virus (HIV), the pathogenesis of FIV involves infection of lymphocytes and macrophages, and results in chronic progressive immune system collapse and death. Neuropathologic correlates of FIV infection have not yet been elucidated, and may be relevant to understanding HIV-associated neurologic disease (neuroAIDS). As in HIV, FIV strains have been shown to express differential tendencies towards development of clinical neuroAIDS. To interrogate viral genetic determinants that might contribute to neuropathogenicity, cats were exposed to two well-characterized FIV strains with divergent clinical phenotypes and a chimeric strain as follows: FIV(PPR) (PPR, relatively apathogenic but associated with neurologic manifestations), FIV(C36) (C36, immunopathogenic but without associated neurologic disease), and Pcenv (a chimeric virus consisting of a PPR backbone with substituted C36 env region). A sham inoculum control group was also included. Peripheral nerve conduction velocity, CNS imaging studies, viral loads and hematologic analysis were performed over a 12 month period. At termination of the study (350 days post-inoculation), brain sections were obtained from four anatomic locations known to be involved in human and primate lentiviral neuroAIDS. Histological and immunohistochemical evaluation with seven markers of inflammation revealed that Pcenv infection resulted in mild inflammation of the CNS, microglial activation, neuronal degeneration and apoptosis, while C36 and PPR strains induced minimal neuropathologic changes. Conduction velocity aberrations were noted peripherally in all three groups at 63 weeks post-infection. Pcenv viral load in this study was intermediate to the parental strains (C36 demonstrating the highest viral load and PPR the lowest). These results collectively suggest that (i) 3' C36 genomic elements contribute to viral replication characteristics, and (ii) 5' PPR genomic elements contribute to CNS manifestations. This study illustrates the potential for FIV to provide valuable information about neuroAIDS pathogenesis related to genotype and viral kinetics, as well as to identify strains useful to evaluation of therapeutic intervention.  相似文献

Objective: Evaluate the effect of marker placement on kinematics of the canine stifle in 3 distinct hindlimb models. Study Design: In vivo biomechanical study. Animals: Normal adult mixed‐breed dogs (n=5). Methods: Ten retroreflective markers were affixed to the skin on the right rear leg of each dog to establish normal stifle kinematics. Four additional markers were placed around the greater trochanter (GT), 2 cm cranial, caudal, dorsal, and ventral to evaluate single marker placement variability on kinematic model data. Dogs were walked and trotted 5 times through the calibrated space. Sagittal flexion and extension angle waveforms were acquired during each trial with 3 models that were produced simultaneously during each gait. The GT marker was reassigned to 1 of the 4 additional locations (cranial, caudal, dorsal, and ventral) to alter the kinematic model. Comparison of sagittal flexion and extension angle waveforms was performed with Generalized Indicator Function Analysis. Results: Each model provided consistent equivalent sagittal flexion–extension data. Analysis revealed statistically significant differences between all GT locations. The differences were greatest in the cranial and caudal locations for all models. Conclusions: Deviation of the GT marker in the cranial/caudal direction from an anatomically normal position produces a greater degree of difference than deviation in a dorsal/ventral direction.  相似文献

Three cats were evaluated at a veterinary teaching hospital for congestive heart failure (CHF) secondary to hyperviscosity syndrome from plasma cell neoplasia. All cats had severe hyperproteinemia due to hyperglobulinemia. Multiple myeloma or plasma cell neoplasia was diagnosed based on cytopathology and post mortem examination. The cats presented with signs of CHF including acute collapse, tachypnea, increased respiratory effort, and pulmonary crackles. All cats had heart murmurs and echocardiographic signs consistent with hypertrophic cardiomyopathy. An enlarged left atrium was found in all cats and two of three cats also had spontaneous echocardiographic contrast. Plasmapheresis (centrifugal plasma exchange) was performed on all three cats by the removal of whole blood and the infusion of a balanced electrolyte solution while the whole blood was centrifuged and separated. The RBCs were then washed before being readministered to the patient. Plasmapheresis alleviated the clinical signs of CHF (tachypnea) in all three cats. Plasmapheresis should be considered in cases of CHF secondary to hyperviscosity syndrome to rapidly alleviate clinical signs associated with heart failure while diagnosis of the underlying cause is made and appropriate therapy implemented.  相似文献

The median infectious dose (ID(50)) of porcine reproductive and respiratory syndrome (PRRS) virus isolate MN-184 was determined for aerosol exposure. In 7 replicates, 3-week-old pigs (n=58) respired 10l of airborne PRRS virus from a dynamic aerosol toroid (DAT) maintained at -4°C. Thereafter, pigs were housed in isolation and monitored for evidence of infection. Infection occurred at virus concentrations too low to quantify by microinfectivity assays. Therefore, exposure dose was determined using two indirect methods ("calculated" and "theoretical"). "Calculated" virus dose was derived from the concentration of rhodamine B monitored over the exposure sequence. "Theoretical" virus dose was based on the continuous stirred-tank reactor model. The ID(50) estimate was modeled on the proportion of pigs that became infected using the probit and logit link functions for both "calculated" and "theoretical" exposure doses. Based on "calculated" doses, the probit and logit ID(50) estimates were 1 × 10(-0.13)TCID(50) and 1 × 10(-0.14)TCID(50), respectively. Based on "theoretical" doses, the probit and logit ID(50) were 1 × 10(0.26)TCID(50) and 1 × 10(0.24)TCID(50), respectively. For each point estimate, the 95% confidence interval included the other three point estimates. The results indicated that MN-184 was far more infectious than PRRS virus isolate VR-2332, the only other PRRS virus isolate for which ID(50) has been estimated for airborne exposure. Since aerosol ID(50) estimates are available for only these two isolates, it is uncertain whether one or both of these isolates represent the normal range of PRRS virus infectivity by this route.  相似文献

This study was designed to determine the ability of tibial plateau leveling osteotomy (TPLO) to eliminate cranial tibial translation (CTT) through a loaded range of motion. Twenty-four large-breed canine cadaver limbs were compared. Each limb was placed in a custom designed jig at 120° of stifle extension under an axial load of 20% body weight. A force of approximately 10 N/s mimiced the action of the quadriceps muscle and allowed the limb to move from 120° to maximal extension. Positional data were acquired using electromagnetic motion-tracking sensors. Each limb was tested under normal, cranial cruciate ligament (CrCL)-deficient, and TPLO-treated conditions. Cranial tibial translation significantly increased after transection of the CrCL. The TPLO failed to normalize CTT within the CrCL deficient stifle; however, values trended towards intact values throughout the range of motion. The TPLO was more effective at higher angles of flexion. These altered biomechanics may help explain the continued progression of osteoarthritis in TPLO repaired stifles. This loaded model may serve as a method for future evaluation of other surgical techniques.  相似文献
