Current management of the grizzly bear (Ursus arctos) population in Yellowstone National Park and surrounding areas requires annual estimation of the number of adult female bears with cubs-of-the-year. We examined the performance of nine estimators of population size via simulation. Data were simulated using two methods for different combinations of population size, sample size, and coefficient of variation of individual sighting probabilities. We show that the coefficient of variation does not, by itself, adequately describe the effects of capture heterogeneity, because two different distributions of capture probabilities can have the same coefficient of variation. All estimators produced biased estimates of population size with bias decreasing as effort in creased. Based on the simulation results we recommend the Chao estimator for model M h be used to estimate the number of the female bears with cubs of the year; however, the estimator of Chao and Shen may also be useful depending on the goals of the research.  相似文献

The dependence of global green tea metabolome on plucking positions was investigated through (1)H nuclear magnetic resonance (NMR) analysis coupled with multivariate statistical data set. Pattern recognition methods, such as principal component analysis (PCA) and orthogonal projection on latent structure-discriminant analysis (OPLS-DA), were employed for a finding metabolic discrimination among fresh green tea leaves plucked at different positions from young to old leaves. In addition to clear metabolic discrimination among green tea leaves, elevations in theanine, caffeine, and gallic acid levels but reductions in catechins, such as epicatechin (EC), epigallocatechin (EGC), epicatechin-3-gallate (ECG), and epigallocatechin-3-gallate (EGCG), glucose, and sucrose levels were observed, as the green tea plant grows up. On the other hand, the younger the green tea leaf is, the more theanine, caffeine, and gallic acid but the lesser catechins accumlated in the green tea leaf, revealing a reverse assocation between theanine and catechins levels due to incorporaton of theanine into catechins with growing up green tea plant. Moreover, as compared to the tea leaf, the observation of marked high levels of theanine and low levels of catechins in green tea stems exhibited a distinct tea plant metabolism between the tea leaf and the stem. This metabolomic approach highlights taking insight to global metabolic dependence of green tea leaf on plucking position, thereby providing distinct information on green tea production with specific tea quality.  相似文献

Carbohydrate digestion by α-glucosidase and subsequent glucose uptake at the brush border are critical for postprandial blood glucose control. Any specific inhibitors are useful as hyperglycemia modulating agents. In this study, it was postulated that an array of active components in mulberry leaf extract (MLE) may provide higher potency in inhibiting intestinal glucose absorption compared to the single component 1-deoxynojirimycin (DNJ), which is recognized as a promising inhibitor of intestinal glucose absorption. Both MLE and DNJ were active in inhibiting α-glucosidase. However, in Caco-2 cells, only MLE showed significant inhibition of 2-deoxyglucose uptake, whereas DNJ was ineffective. For glucose loading, co-administration of MLE resulted in potent inhibitions of glucose responses compared to those by DNJ in Sprague Dawley (SD) rats, but this was not found for maltose loading. These novel findings add evidence that the unabsorbed phytochemicals in MLE compete with glucose for intestinal glucose transporters, but DNJ itself does not. We also evaluated the timing of MLE consumption. By administering MLE for 30 min before glucose loading, the incremental area under the curve (IAUC) was significantly lowered in the rats, as compared to a simultaneously administered group. Similarly, cellular glucose uptake was significantly reduced in Caco-2 cells following pretreatment.  相似文献

A technique has been developed to extract soil phenolics by their adsorption onto resin, the separation of resin from soil, and desorption from the resin followed by high performance liquid chromatography (HPLC) analysis. Four representative phenolic acids, p-hydroxybenzoic, p-hydroxycinnamic, p-methoxybenzoic and 2,4-dimethoxybenzoic acids, were added to a subtropical soil (66 nmol g?1 soil) and recovered by extraction with Amberlite XAD-4 resin. The recovery efficiency of the added phenolics was greater using a flotation method to separate resin from soil than by a filtration method. The recoveries of p-hydroxybenzoic acid and p-hydroxycinnamic acid decreased with increasing extractant pH. Recoveries of p-methoxybenzoic acid and 2,4-dimethoxybenzoic acid increased slightly with increasing pH. The addition of NaCl to the XAD-4 resin extractant generally increased the recovery of phenolics by filtration but not by flotation. The recoveries of phenolics from the soil by extraction with XAD-4 resin were much greater than by alkaline extraction at pH 11.  相似文献

Cultivated Chinese wheat germplasm has been a valuable genetic resourcein international plant breeding. Landraces endemic to China are a geneticresource that is distinct from other wheat germplasm. Patterns of geneticdiversity among cultivated Chinese accessions and relationship to othergermplasm pools are unknown, despite the proven value and potential novelty. Theobjective of this work was to determine the level of genetic diversity withinimproved Chinese germplasm in the context of several other wheat germplasmpools. We analyzed a set of improved accessions cultivated from the 1940s to the1990s in Shaanxi province, China, using six simple sequence repeat (SSR) primerpairs and 30 restriction fragment length polymorphism - probe enzymecombinations (RFLP-PEC) previously used to characterize 21 geographically basedgermplasm pools. Shaanxi germplasm consists of three groups based on foreignintroductions from Italy, Australia, Denmark, and Russia. There was a decreasein genetic diversity among Shaanxi accessions cultivated in the 1970s and 1980sto the 1990s, and accession classifications based on primary decade ofcultivation were found to be significantly undifferentiated. The analysis of themean genetic distance among 22 geographically based pools of germplasm suggestsseveral regions are significantly undifferentiated. A vast majority of the totalamount of variation was found within pools; therefore, pools appear to belargely differentiated based on small differences in band relative frequency andfew if any unique bands. Previous studies have identified some Chinese landracepools as morphologically and genetically unique. The Shaanxi pool does not havethe same unique morphological or genetic features, nor is it more similar to thelandrace pools than other improved germplasm pools.  相似文献

Exotic weeds are invading rangelands of the western United States at unprecedented rates. Understanding plant-soil relationships and competitive interactions of invasive weeds is crucial in long-term control strategies. In a greenhouse experiment, we investigated the influence of soil nutrient depletion on plant growth and plant competition between the exotic invasive weeds, Lepidium latifolium (invading wetlands) and Bromus tectorum (invading a multitude of habitats). Plants were grown individually and in combination until L. latifolium flowered, then roots and aboveground mass were harvested. Soil in individual pots was homogenized, subsamples collected for nutrient analyses, and the soil was re-planted to the same species, grown, and harvested twice more for a total of three growth cycles. As nutrient supplying capacity of the soil declined through growth cycles, aboveground mass of L. latifolium decreased significantly (PА.05) and growth potential of B. tectorum surpassed that of L. latifolium. Only bicarbonate-extractable soil ortho-P positively correlated with plant mass of L. latifolium. A separate experiment demonstrated that L. latifolium has a narrow window of soil water potentials for optimal growth; greatest growth at -20 kPa with significantly declining growth at saturation and -400 kPa. Our data suggest that L. latifolium primarily invades wetlands because the high soil water content reduces tortuosity and allows efficient transport of nutrients to this sparsely rooted species. When soil moisture and/or the nutrient supplying capacity of the soil declines, plants with greater root density can out-compete L. latifolium. Monocultural stands of L. latifolium may be self-limiting in time as available nutrients, particularly P, are biocycled to drier upper soil layers.  相似文献

Plants tolerant to polluted environments were selected, based on several criteria, to restore a coastal forest ecosystem severely damaged by air pollutants discharged from an industrial complex. In addition, a restoration plan was prepared synthesizing these results and the diagnostic ecological indicators in the area for which restoration is required. Pollution-tolerant plants of 11 tree and subtree species, 10 herb species and one shrub species were selected from field surveys in the vicinity of two representative industrial complexes in Korea, Ulsan and Yeocheon. Nine species were selected for tolerance to SO2 fumigation and six species were selected for tolerance to Al3+. Growth and photosynthetic responses of sample plants transplanted into polluted and unpolluted sites showed that 15 species out of the 26 sample plants showed a disposition for tolerance. Most of these are endemic plants and they are composed of diverse species in structure and function. This result implies that these tolerant species could play important roles in the restoration of the study area, which has several specific features. On the other hand, results from transplant tests indicate that a field survey is the most reasonable method for selection of tolerant plants to restore a pollution-damaged ecosystem, as was shown in another restoration program. Results of ecological analysis on vegetation map indicate that the spatial range within the first ridge is the sector for which restoration is required. This sector was classified into four zones on the basis of topographic conditions: lower and upper slopes of both slopes facing and opposite the pollution source. Guidelines for soil amelioration and arrangement of tolerant plants were prepared considering the degree of vegetation degradation, leaf damage of major plant species and soil pollution in each zone under the restoration plan.  相似文献

The effect of Gd-DTPA on the development in NMR relaxation of skeletal rabbit muscles post-mortem was investigated by dynamic low-field (0.47 T) relaxation measurements from 4 min post-mortem and until 23 h post-mortem. Twelve rabbits were included in the study, and half of the animals were administered 0.2 mmol of Gd-DTPA iv 15 min before sacrifice, while the other half was administered an isotonic salt solution. A significant effect of Gd-DTPA treatment corresponding to a 25% reduction in the T(1) relaxation time was observed. T(2) relaxation was decomposed into two components reflecting intra- and extracellular components (T(2)()alpha and T(2)()beta, respectively), and Gd-DTPA treatment was found to affect both components. However, around 150 min post-mortem a dramatic increase in the difference between control and Gd-DTPA-treated rabbits was observed in the relaxation time of the intracellular water population (T(2)()alpha). Electrical stimulation of the muscles resulted in a significantly earlier onset of the increased effect of Gd-DTPA on the T(2)()alpha population. The increased effect of Gd-DTPA treatment on the T(2)()alpha component is believed to reflect leakage of water from the muscle cells due to membrane destabilization, known to be promoted by electrical stimulation. Accordingly, the present study demonstrates how Gd-DTPA can be used for probing membrane integrity in post-mortem muscles known to be of importance for subsequent water distribution and final water-holding capacity.  相似文献

The isoflavone compositions and concentrations in the leaf, flower, petiole, and stem of 13 red clover cultivars were studied using high-performance liquid chromatography coupled with a diode array and a mass spectrometric detector with negative electrospray ionization. Different cultivars showed significantly different concentrations of individual and total isoflavones. The leaf contained the highest overall concentration, followed by the stem, petiole, and flower. Biochanin A and formononetin were the predominant isoflavones in all cultivars and all parts, along with eight other minor aglycones, daidzein, genistein, glycitein, irilone, orobol, pratensein, pseudobaptigenin, and prunetin, and four minor malonylglycosides, genistein-7-glucoside-6' '-malonate, orobol-7-glucoside-6' '-malonate, formononetin-7-glucoside-6' '-malonate, and biochanin A-7-glucoside-6' '-malonate. The isoflavone compositions and concentrations were also found to be different between red clover parts harvested at the early bud stage and the late flowering stage. Sample storage and handling prior to analysis were also found to be important. Samples in this study were kept at -5 degrees C for a few days before being freeze-dried and were found to contain mainly the aglycones of isoflavones. This may actually be an advantage in that "natural" and more bioactive isoflavones can be obtained without using chemical hydrolysis. Findings in this study therefore provide important information for developing isoflavone-rich red clovers and for optimizing harvest timing and choosing the right part of the red clover plant.  相似文献

The effect of spraying exogenous plant hormone methyl jasmonate (MeJA) upon radish sprout (Raphanus sativus L.) was investigated in aspects of total phenolic content (TPC), isothiocyanate content, antioxidant activity of the radish extract, and enzymatic activities of phenylalanine ammonia lyase (PAL) and myrosinase. The MeJA treatment significantly increased the TPC that resulted in the increased DPPH* (2,2-diphenyl-1-picrylhydrazyl) free radical scavenging capacity. In addition, the PAL activity also increased by 60% at 24 h after MeJA treatment. However, the same treatment decreased the amount of 4-methylthio-3-butenylisothiocyanate (MTBITC), a major isothiocyanate in radish sprout and the activity of myrosinase, an enzyme related to produce isothiocyanates.  相似文献
