Cyclolinopeptides are a group of naturally occurring hydrophobic cyclic peptides found in flaxseed and flax oil that have immunosuppressive activity. This study describes the measurement of flaxseed cyclolinopeptide concentrations using an internal standard HPLC method. In addition, the concentration of cyclolinopeptides in the seed of Canadian flax cultivars grown at two locations over two years is reported. The data are consistent with the formation of flaxseed cyclolinopeptides from two ribosome-derived precursors. Each precursor protein includes the sequences corresponding to three cyclolinopeptides from which those cyclolinopeptides are presumably derived by precursor processing. The concentrations of cyclolinopeptides C and E, which are encoded by the same gene sequence, are highly correlated, and the concentrations of cyclolinopeptides D, F, and G, which are encoded by a second gene sequence, are also highly correlated. The strong correlation between the cyclolinopeptides arising from the same gene may prove to be important in understanding how peptide concentration is controlled. Additional research may lead to approaches to improve flax either as a platform for peptide production or as a source of oil with improved drying properties and flavor.  相似文献

Enteropathogenic Escherichia coli (EPEC) is a major cause of infantile diarrhea in developing countries. However, sporadic outbreaks caused by this microorganism in developed countries are frequently reported recently. As an important zoonotic pathogen, EPEC is being monitored annually in several countries. Hallmark of EPEC infection is formation of attaching and effacing (A/E) lesions on the small intestine. To establish A/E lesions during a gastrointestinal tract (GIT) infeciton, EPEC must thrive in diverse GIT environments. A variety of stress responses by EPEC have been reported. These responses play significant roles in helping E. coli pass through GIT environments and establishing E. coli infection. Stringent response is one of those responses. It is mediated by guanosine tetraphosphate. Interestingly, previous studies have demonstrated that stringent response is a universal virulence regulatory mechanism present in many bacterial pathogens including EPEC. However, biological signficance of a bacterial stringent response in both EPEC and its interaction with the host during a GIT infection is unclear. It needs to be elucidated to broaden our insight to EPEC pathogenesis. In this review, diverse responses, including stringent response, of EPEC during a GIT infection are discussed to provide a new insight into EPEC pathophysiology in the GIT.  相似文献

水稻稻瘟病是世界范围内水稻主要病害之一。掌握水稻广谱抗病基因型及病原菌的致病特征特性,对培育抗病品种、指导品种布局具有重要意义。通过抗稻瘟病单基因系水稻品种与稻瘟病菌互作,鉴定出Pi19、Pi20、Pi12、Pi9、Piz5、Piz-t为吉林省水稻广谱抗病基因型,筛选出2014-17-1,2014-66-1等可用于抗性评价标准菌株,建立了一套以IRBL9-W(Pi9)、IRBL12-M([Pi12(t)]、IRBLta-CT2(Pib)、IRBLz-Fu(Piz)、IRBL5-M[Pi5(t)]、IRBLkm-Ts(Pik-m)、IRBLta2-Pi(Pita-2)这7个抗稻瘟病单基因系品种组成的生理小种鉴定体系,优势小种不显著。  相似文献

Six new β-resorcylic acid derivatives (1–5 and 7) were isolated from a halophyte-associated fungus, Colletotrichum gloeosporioides JS0419, together with four previously reported β-resorcylic acid lactones (RALs). The relative and absolute stereochemistry of 1 was completely established by a combination of spectroscopic data and chemical reactions. The structures of the isolated compounds were elucidated by analysis of HRMS and NMR data. Notably, compounds 1–3 had a β-resorcylic acid harboring a long unesterified aliphatic side chain, whereas the long aliphatic chains were esterified to form macrolactones in 4–9. Among the isolated compounds, monocillin I and radicicol showed potent antifungal activities against Cryptococcus neoformans, comparable to clinically available antifungal agents and radicicol showed weak antifungal activity against Candida albicans. These findings provide insight into the chemical diversity of fungal RAL-type compounds and their pharmacological potential.  相似文献

The Korean rockfish, Sebastes schlegeli, is a valuable and intensively exploited species in Korea. We discuss the genetic diversity and genetic structure of four Korean rockfish populations using eight microsatellite loci. In total, 161 different alleles from 138 individuals were observed. Average allele number per locus ranged from 2.5 to 23 and allelic richness varied from 13.38 to 14.63 within a population. Despite a long history of stocking practices, we found very high levels of polymorphism (mean heterozygosity = 0.810), which is comparable to other congeneric species. No significant difference in genetic diversity and molecular genetic variance (FST and RST) was observed among four local samples (P > 0.1). Little indication of contemporary inbreeding (FIS= 0.051) or population structure (K = 1) was detected. This absence of differentiation may reflect high levels of gene flow along the coast of Korea. Our study demonstrates that rockfish in Korea should be managed as a single unit. Currently, the species does not appear to be genetically threatened, but the potential for a rapid loss of genetic diversity remains. This information on the genetic characteristics of Korean rockfish populations has important implications for fisheries management and conservation efforts, and will aid in the sustainable exploitation of the fishing resources and the preservation of biodiversity.  相似文献

In the southern United States, corn production encounters moisture deficit coupled with high‐temperature stress, particularly during the reproductive stage of the plant. In evaluating plants for environmental stress tolerance, it is important to monitor changes in their physical environment under natural conditions, especially when there are multiple stress factors, and integrate this information with their physiological responses. A low‐cost microcontroller‐based monitoring system was developed to automate measurement of canopy, soil and air temperatures, and soil moisture status in field plots. The purpose of this study was to examine how this system, in combination with physiological measurements, could assist in detecting differences among corn genotypes in response to moisture deficit and heat stress. Three commercial hybrids and two inbred germplasm lines were grown in the field under irrigated and non‐irrigated conditions. Leaf water potential, photosynthetic pigments, cell membrane thermostability (CMT) and maximum quantum efficiency of photosystem II (Fv/Fm) were determined on these genotypes under field and greenhouse conditions. Variations observed in air and soil temperatures, and soil moisture in plots of the individual corn genotypes helped explain their differences in canopy temperature (CT), and these variations were reflected in the physiological responses. One of the commercial hybrids, having the lowest CT and the highest CMT, was the most tolerant among the genotypes under moisture deficit and heat stress conditions. These results demonstrated that the low‐cost microcontroller‐based monitoring system, in combination with physiological measurements, was effective in evaluating corn genotypes for drought and heat stress tolerance.  相似文献

Urban waste disposal occurred on fenland to the west of Manchester, England, between 1900 and 1964. The reclaimed fenland, Chat Moss, is now used for mixed arable farming. A total of 1.92 Mt of waste including privy midden, street sweepings, clinkers and slaughterhouse refuse was incorporated into the moss resulting in a modified topsoil with raised pH and reduced organic matter content compared with the subsoil. Elevated levels of potentially toxic elements (PTEs) are observed in the topsoil beyond the typical depth of atmospheric contamination; Cd and As concentrations exceed soil guideline values (SGVs) at 1.8 and 43 mg/kg, respectively. Sequential extraction indicates that waste‐derived Pb, Zn and Ni remain predominantly in the residual fraction, whereas Cu was mainly organically bound. Arsenic was predominately found in oxide and organic matter fractions with Cd in carbonate, oxide, organic matter and residual fractions. Pot trials indicated limited uptake of PTEs by vegetables grown on the waste‐amended soil, with the exception of Cd uptake by lettuce (0.22 mg/kg FW) and Pb uptake by radish (0.16 mg/kg FW), which exceeded current EU limits of 0.2 and 0.1 mg/kg FW, respectively. Hazard quotients (HQs) identified no risks to adults from consumption of vegetables grown in these soils with the exception of lettuce consumption with a HQ of 1.4. Risks to children were slightly greater with HQs >1 for Cd in lettuce, spinach, carrots and onion, As in lettuce, parsley and onion and for Zn in spinach.  相似文献

Systemic changes in gene expression have been analysed in potato plants following root infection with the cyst nematode Globodera rostochiensis. Root invasion by the pathogen induced major changes to occur in the composition of proteins extracted in intercellular fluid from the leaf apoplastic space. The changes involved the disappearance of innate leaf proteins and the accumulation of novel gene products. Analysis by SDS-PAGE indicated that the leaf polypeptides induced on nematode invasion are not cultivar or nematode pathotype-specific. Aspirin treatment of leaves induced a similar pattern to that induced systemically by nematodes, but several gene products were specific for the latter method of induction. In particular, two abundant [125I]-Con A binding polypeptides of high Mr accumulated in intercellular fluid only on aspirin treatment. The results are discussed in the context of the role of the extracellular leaf proteins and their modulation by systemic signals.  相似文献

Integration of reticuloendotheliosis virus (REV) into the genome of fowl poxvirus (FPV) has been reported recently. With a view to determine whether this event had occurred in the past, we screened by polymerase chain reaction (PCR) for the presence of REV provirus in the DNAs of nine avian poxviruses, some of which had been lyophilized 50 yr ago. For REV, 5' long terminal repeat (LTR) and REV envelope sequences were amplified, whereas for FPV, the major envelope antigen gene and the region flanking REV sequences were amplified. In six of seven FPV strains examined, the specific PCR amplicons were obtained for both REV provirus and FPV sequences. One isolate in which presence of REV 5' LTR and envelope was not detected by PCR, a LTR remnant was detected by Southern hybridization. Interestingly, no REV sequence was detected in either canary poxvirus or pigeon poxvirus genome. These observations indicate that REV integration in the FPV genome is not a recent phenomenon but probably occurred prior to 1949.  相似文献
