Timothy (Phleum pratense L.) is an important forage grass used for pasture, hay and silage in regions with cool and humid growth seasons. Harvesting conditions could reduce its nutritive value, particularly with extended wilting periods. To understand how daytime or night‐time wilting influences the nutritive value of timothy, this study investigated the metabolism of non‐structural carbohydrates, including fructan and starch, together with total soluble protein and amino acid patterns in timothy plants harvested at two maturity stages (heading and anthesis) and wilted under controlled conditions for 24 h at two temperatures (15°C, 20°C) and two light regimes (darkness, light) by simulating different wilting management practices. Correlation analysis with the whole dataset showed that soluble protein, glucose and starch contents declined in plant tissues concomitantly with water loss, while amino acid, sucrose and fructose contents increased. Transient increase in amino acid content suggests that the decrease in protein content was due to proteolysis during wilting. Sucrose and fructose contents generally increased in plant tissues harvested at anthesis and wilted in light whereas they were unaffected in plants wilted in darkness. Fructan content remained stable. Fructan exohydrolase (FEH) and soluble acid invertase (INV) activities were well preserved during the first 12 h of wilting and might facilitate the fermentation process at the beginning of ensiling by supplying fructose from fructans and hexoses from sucrose to the fermentive bacteria.  相似文献

栉江瑶(Atrina pectinata)为我国重要的海洋经济双壳贝类,近年来,其养殖生产活动备受养殖企业关注,为发展和优化其人工繁殖技术,本研究探究了人工海水中铵离子浓度和pH的变化对栉江瑶新鲜精子游泳运动的激活作用,定量描述了高度活化状态下精子曲线运动速率(VCL)、直线运动速率(VSL)、平均路径运动速率(VAP)和鞭毛摆动频率(BCF)的运动学特征,并对精子ATP含量、ATP酶与超氧化物歧化酶(SOD)活性进行了记录。结果显示,简单提高海水pH可略微提高栉江瑶精子运动率,但无法使精子进入高度活化状态;含有铵离子的碱化海水可有效激发精子游泳运动,3 mmol/L氨海水激活效果最佳。经3 mmol/L氨海水激活后,精子活力等级(MI)在21 min内一直保持在≥4的状态,在激活的前3 min内精子运动率都在80%以上,VCL>56 μm/s,VSL>17 μm/s,VAP>30 μm/s,BCF>6 Hz。精子ATP含量在激活5 min后降低至初始含量的30.29% [(128.80± 66.92) μmol/g prot],随后,无显著变化(P>0.05)。精子Na+-K+-ATP酶和Ca2+-Mg2+-ATP酶活性在运动过程中较为稳定,其中,Na+-K+-ATP酶活性较低[(0.62±0.03) U/mg prot],Ca2+-Mg2+-ATP酶活性较高[(6.08± 0.04) U/mg prot]。精子SOD活性在15 min内逐步降低至[(1.23±0.73) U/mg prot],随后维持稳定。本研究可为栉江瑶精子激活机制深入研究提供基础,助力发展栉江瑶人工繁育技术。  相似文献

Blood sera, skeletal muscle, and cardiac muscle from 24 bovine carcasses condemned for eosinophilic myositis by US Department of Agriculture meat inspection veterinarians were compared with similar specimens from 35 random carcasses passed for human consumption. Fluorescence values determined by using a fluorometric immunoassay system were used to measure the specific antibodies to Sarcocystis cruzi. The values were significantly elevated in carcasses condemned for eosinophilic myositis as compared to carcasses passed for human consumption. The elevated fluorescence values appeared to be more than coincidental, suggesting that S. cruzi may be a causative agent of eosinophilic myositis. Microscopic examination of affected muscle revealed lesions typical of eosinophilic myositis. Lesions were characterized by extensive multifocal areas of myofiber hyaline degeneration, necrosis with sarcoplasmic fragmentation, mineralization of occasional myofibers, and atrophy of fibers with varied stages of fibrosis. Inflammatory cell exudates were predominantly eosinophils, with some macrophages and lymphocytes, and extravasated erythrocytes within, and adjacent to, affected myofibers. Affected muscles contained more S. cruzi than unaffected muscle from passed carcasses. However, a distinct cause and effect relationship could not be determined between the parasite and the presence of eosinophilic myositis.  相似文献

Portions of the placenta and kidneys from a 5-month-old bovine fetus were examined histologically. The placental cotyledonary villi were necrotic and protozoa were found in lesions. The organisms were in trophoblasts, periodic acid Schiff-negative and did not react in an immunoperoxidase test using anti-Toxoplasma gondii serum. Individual zoites were approximately 5 x 2 microns and the biggest collection was 45 x 35 microns. The organism in the present case was structurally distinct from Sarcocystis spp. and Toxoplasma gondii, but resembled Neospora caninum.  相似文献

A 3-yr study used 16 cows and their spring-born calves (yr 1) and 48 first-calf heifers (yr 2, n = 24; yr 3, n = 24) and their spring-born calves in a completely randomized design. All cows and heifers were Angus x Beefmaster, and calves were sired by Angus bulls. Cow-calf pairs were assigned randomly to one of two management systems: 1) an early-weaning system, in which steer and heifer calves were weaned at 108 d of age and fed a postweaning growing diet (EW), or 2) a normal weaning system, in which calves were weaned at 205 d without supplementation (NW). Before early weaning and within each management system, calves and their dams were maintained in two 1.4-ha, endophyte-infected tall fescue pastures for 35 d (yr 1) or 14 d (yr 2 and 3). Early-weaned calves and cow-calf pairs were then randomly allotted to 1.4-ha, endophyte-infected tall fescue pastures with two (yr 1) or three (yr 2 and 3) calves or cow-calf pairs per pasture (four pastures per management system). Cow weights and BCS changes and calf gains were measured from early to normal weaning. Dietary intakes and nutrient digestibilities by EW and NW calves were determined during two periods of yr 1 and three periods of yr 2 and 3. Total gains and BCS changes were greater (P < 0.01) for cows that produced EW calves in all years. Calf ADG from early to normal weaning did not differ (P = 0.32). Similar to ADG, BW of calves at normal weaning were not different (P = 0.11). Forage intake was greater (P < 0.01) by NW calves during Periods 2 and 3 of yr 1 and Periods 1 and 2 of yr 2 and 3; however, total DM and CP intakes were greater (P < 0.01) for EW calves in Periods 2 and 3 of each year. Intakes of NDF tended (P = 0.11) to be greater by EW calves across all years. Estimates of CP and NDF digestibilities were higher (P < 0.01) for EW calves during yr 1 and 2; however, all components of the diet consumed by NW calves in yr 3 were more digestible (P < 0.05) than those consumed by EW calves. These results show the condition of cows with EW calves was improved by early weaning and gains by calves weaned at 108 d to pasture plus a commercial grower diet were comparable to those by calves continuing to nurse dams until weaned at 205 d.  相似文献

Enalapril is an angiotensin converting enzyme (ACE) inhibitor that is frequently used in human, feline and canine patients with cardiac disease. Its use has been associated with impotence in human patients. The purpose of this study was to evaluate if enalapril (0.5 mg/kg PO, q24h) is likely to alter behavior in stallions and to assess its effect on ACE activity at the standard dose used in dogs and cats. Twelve pony stallions were evaluated by physical examination and echocardiography followed by treatment with enalapril (n = 6) or placebo (n = 6) for 2 months. After one month, blood was drawn and stored to evaluate ACE activity in the 2 groups. At the end of the study, repeat physical examination and echocardiography were performed. Physical examination, echocardiographic indices, and reproductive performance were unchanged and there was no suppression of ACE activity. Results of this study suggest that enalapril (0.5 mg/kg PO, q24h) is either poorly absorbed in the horse or is inadequately converted to the active form of the drug, enalaprilat.  相似文献

为了研究不同面粉对鲜白盐面条颜色的影响,在澳大利亚商业面粉中添加一定量的淀粉、面筋粉和麦芽,配制了8种面粉,用其制成鲜白盐面条后,用Minolta在0和24h时分别测量其L*、a*、b*值。经分析,在0和24h时,8种面条的L*、a*、b*值都分别达到显著或极显著水平;在24h后,所有面条的颜色变差,其L*值降低,a*、b*值升高;经比较,在添加了淀粉的面条颜色较好,其中以面条CK 9%S(在面粉中添加9%淀粉)的颜色最好。结果建议育种家在优质面条小麦育种中,应注意选出的品种蛋白质含量不应高,而淀粉含量应较高和面条颜色稳定性好的抗穗发芽品种。  相似文献

Obstruction of critical blood vessels due to thrombosis or embolism is a leading cause of death worldwide. Here, we describe a biomimetic strategy that uses high shear stress caused by vascular narrowing as a targeting mechanism--in the same way platelets do--to deliver drugs to obstructed blood vessels. Microscale aggregates of nanoparticles were fabricated to break up into nanoscale components when exposed to abnormally high fluid shear stress. When coated with tissue plasminogen activator and administered intravenously in mice, these shear-activated nanotherapeutics induce rapid clot dissolution in a mesenteric injury model, restore normal flow dynamics, and increase survival in an otherwise fatal mouse pulmonary embolism model. This biophysical strategy for drug targeting, which lowers required doses and minimizes side effects while maximizing drug efficacy, offers a potential new approach for treatment of life-threatening diseases that result from acute vascular occlusion.  相似文献
