This study investigated the effects of skim milk on the quality and fertility of boar spermatozoa under long‐term chilled preservation. Semen samples were stored in Modena solution supplemented with 0 (control) to 50 mg/mL skim milk at 5°C for 4 weeks; spermatozoa stored with 7.5 and 15 mg/mL of skim milk (7.5‐SM and 15‐SM groups, respectively) exhibited significantly higher motility indices than those of the control group up to 3 weeks (P < 0.05), and the 7.5‐SM group showed improved motility indices even after 4 weeks (P < 0.05). In vitro fertilization using spermatozoa in the 7.5‐SM and 15‐SM groups stored at 5°C for 2 weeks showed significantly higher fertilization rates of spermatozoa and the development rates to blastocyst than the control group (P < 0.05), and the 7.5‐SM group showed similar rates of fertilization and blastocyst formation in the fresh non‐stored spermatozoa group. After artificial insemination using spermatozoa stored for 2 weeks in the 7.5‐SM group, healthy piglets were obtained. Boar spermatozoa can be stored at 5°C in a Modena solution containing skim milk. Supplementation of 7.5 mg/mL skim milk improves boar spermatozoa motility and fertility even after liquid preservation at 5°C for 2 weeks.  相似文献

We investigated the prevalence and risk factors associated with Cryptosporidium oocysts shedding in pigs in Central Vietnam. A total of 740 single fecal samples collected from diarrheic and non-diarrheic pigs on 89 farms were screened by the modified Ziehl-Neelsen staining method. Prevalence at the animal and the farm levels were 18.1% (134/740) and 71.9% (64/89), respectively. Risk factors for the infection were identified using univariate and multivariate logistic regression analysis. The results revealed that age, sanitary condition and topography were significantly associated with oocyst shedding (P<0.05). Pre-weaned piglets were at the highest risk for infection, followed by post-weaners, sows and finishing pigs. Good sanitary conditions showed positive effects in decreasing oocysts shedding. Topographically, Cryptosporidium was more common in mountainous zone than that in coastal delta zone. There was an association between the occurrence of diarrhea and the level of Cryptosporidium oocyst excretion within infected pigs. This is the first epidemiological investigation of prevalence and risk factors of Cryptosporidium in pigs in Vietnam.  相似文献

The aims of this study were to investigate the prevalence of natural Fasciola infections in both the definitive hosts (cattle) and the intermediate hosts (Lymnaea snails) in central Vietnam. A total of 1,075 fecal samples, randomly collected from cattle in Binh Dinh, Khanh Hoa, and Phu Yen provinces, were examined for Fasciola eggs by a sedimentation method. The overall prevalence of Fasciola was 45.3?%. A subset of the animals (235) was also screened for antibodies against Fasciola by an enzyme-linked immunosorbent assay. Overall, 46.3?% of these animals were shedding Fasciola eggs while 87.2?% were Fasciola seropositive. A lower prevalence of Fasciola was observed in calves ??2?years of age (37.6?%) compared to that in cattle >2?years of age (53.7?%) (p?<?0.05). The prevalence in the rainy season (50.8?%) was significantly different to that in the dry season (38.1?%) (p?<?0.05). Of the 3.269 Lymnaea viridis and 1.128 Lymnaea swinhoei examined, 31 (0.95?%) and seven (0.62?%), respectively, were found to be infected with Fasciola. This appears to be the first epidemiological survey of the prevalence of Fasciola in cattle and snails in these three provinces in central Vietnam.  相似文献

We investigated the temporal changes in creep and stress relaxation behavior in both microscopic crystalline cellulose and macroscopic strain of wood specimen using Japanese cypress (Chamaecyparis obtusa Endl.) to understand the viscoelastic properties of wood cell walls. Specimens 600 µm in thickness were observed by the X-ray diffraction and submitted to tensile load. The crystal lattice strain of (004) plane and macroscopic strain of specimen were continuously detected during creep and stress relaxation tests. It was found that the creep compliance based on macroscopic strain showed a gradual increase after instantaneous deformation due to loading and then the parts of creep deformation remained as permanent strain after unloading. On the other hand, crystal lattice strain showed a different behavior for macroscopic strain; it kept a constant value after instantaneous deformation due to loading and then increased gradually after a certain period of time. These differences between macroscopic and microscopic levels were never found in the stress relaxation tests in this study. Relaxation modulus at the macroscopic level only showed a decreasing trend throughout the relaxation process. However crystal lattice strain kept a constant value during the macroscopic relaxation process. In addition, the microfibril angle (MFA) of wood cell wall has a role of mechanical behavior at microscopic level; crystal lattice strains were smaller with increasing MFA at both creep and relaxation processes. Creep compliance and stress relaxation modulus at the macroscopic level decreased and increased with increasing MFA, respectively. Our results on the viscoelastic behavior at microscopic level evidenced its dependency on MFA.  相似文献

The objectives of this study were to establish the method of evaluating wood mechanical properties by acoustic nondestructive testing at standing trees and at logs of a Chinese fir (Cunninghamia lanceolata (Lamb.) Hook.) plantation, and to compare three acoustic nondestructive methods for evaluating the static bending modulus of elasticity (MOE), modulus of rupture (MOR), and compressive strength parallel-to-grain (σc) of plantation wood as well. Fifteen Chinese fir plantation trees at 36 years of age were selected. Each tree was cut into four logs, for which three values of dynamic modulus of elasticity, i.e., E sw, of the north and south face based on stress waves to assume the measuring state of the standing tree, E fr, longitudinal vibration, and E us, ultrasonic wave, were measured in the green condition. After log measurements, small specimens were cut and air-dried to 12% moisture content (MC). Static bending tests were then performed to determine the bending MOE and MOR, and compressive tests parallel-to-grain were made to determine σc. The bending MOE of small clear specimens was about 7.1% and 15.4% less than E sw and E us, respectively, and 11.3% greater than E fr. The differences between the bending MOE and dynamic MOE of logs as determined by the three acoustic methods were statistically significant (P < 0.001). Good correlation (R = 0.77, 0.57, and 0.45) between E sw, E fr, and E us and static MOE, respectively, were obtained (P < 0.001). It can be concluded that longitudinal vibration may be the most precise and reliable technique to evaluate the mechanical properties of logs among these three acoustic nondestructive methods. Moreover, the results indicate that stress wave technology would be effective to evaluate wood mechanical properties both from logs and from the standing tree.  相似文献

Rumen microorganisms produce various fibrolytic enzymes and degrade lignocellulosic materials into nutrient sources for ruminants; therefore, the characterization of fibrolytic enzymes contributing to the polysaccharide degradation in the rumen microbiota is important for efficient animal production. This study characterized the fibrolytic isozyme activities of a rumen microbiota from four groups of housed cattle (1, breeding Japanese Black; 2, feedlot Japanese Black; 3, lactating Holstein Friesian; 4, dry Holstein Friesian). Rumen fluids in all cattle groups showed similar concentrations of total volatile fatty acids and reducing sugars, whereas acetic acid contents and pH were different among them. Predominant genera were commonly detected in all cattle, although the bacterial compositions were different among cattle groups. Zymograms of whole proteins in rumen fluids showed endoglucanase activities at 55 and 57 kDa and xylanase activity at 44 kDa in all cattle. Meanwhile, several fibrolytic isozyme activities differed among cattle groups and individuals. Treponema, Succinivibrio, Anaeroplasma, Succiniclasticum, Ruminococcus, and Butyrivibrio showed positive correlations with fibrolytic isozyme activities. Further, endoglucanase activity at 68 kDa was positively correlated with pH. This study suggests the characteristics of fibrolytic isozyme activities and their correlations with the rumen microbiota.  相似文献

The binding of ferritin to heme has been well studied using commercial horse spleen apoferritin, which is almost entirely composed of the L subunit, suggesting that mammalian ferritins bind heme. The present study revealed that both mammalian holoferritins (commercial horse spleen ferritin and purified horse spleen, bovine spleen and canine liver ferritins with L/H subunit ratios of 4.0, 1.1, and 2.3, respectively) and their apoferritins bound biotinylated hemin; apoferritins had higher binding activity than holoferritins, except for canine holo- and apoferritins, which showed the same binding. Bovine ferritin H subunit homopolymers expressed by a baculovirus expression system showed heme binding and had higher binding activity to biotinylated hemin than the L subunit homopolymer expressed by the same system. These bindings were inhibited by heme but not by iron-free or Zn-protoporphyrin IX (Zn-PPIX). Purified chicken liver holoferritin was found to be composed of only H subunits and showed the highest binding activity with biotinylated hemin compared with mammalian holoferritins. The binding of chicken liver holoferritin to biotinylated hemin was also inhibited by heme but not by PPIX or Zn-PPIX. These results indicate that mammalian and avian ferritins bind heme and that the H subunit preferentially recognizes heme.  相似文献
