Dietary phytoestrogens may be involved in the occurrence of chronic diseases. Reliable information on the phytoestrogen content in foods is required to assess dietary exposure and disease risk in epidemiological studies. However, existing analyses have focused on only one class of these compounds in plant-based foods, and there is only little information on foods of animal origin, leading to an underestimation of intake. This is the first comprehensive study of phytoestrogen content in animal food. We have determined the phytoestrogen content (isoflavones: biochanin A, daidzein, formononetin, genistein, and glycitein; lignans: secoisolariciresinol and matairesinol; coumestrol; equol; enterolactone; and enterodiol) in 115 foods of animal origin (including milk and milk-products, eggs, meat, fish, and seafood) and vegetarian substitutes using liquid chromatography-mass spectrometry (LC-MS) with (13)C-labeled internal standards. Phytoestrogens were detected in all foods analyzed; the average content was 20 microg/100 g of wet weight (isoflavones, 6 microg/100 g; lignans, 6 microg/100 g; equol, 3 microg/100 g; and enterolignans, 6 microg/100 g). In infant soy formula, 19 221 microg/100 g phytoestrogens were detected (compared to 59 microg/100 g in non-soy formula). Our study shows that all foods analyzed contained phytoestrogens and most foods (except for fish, seafood, and butter) contained mammalian phytoestrogens (enterolignans and equol). This is the first comprehensive study of phytoestrogen content of foods of animal origin and will allow for a more accurate estimation of exposure to dietary phytoestrogens. 相似文献

Production of sponge-derived bioactive compounds in vitro has been proposed as an alternative to wild harvest, aquaculture, and chemical synthesis to meet the demands of clinical drug development and manufacture. Until recently, this was not possible because there were no marine invertebrate cell lines. Recent breakthroughs in the development of sponge cell lines and rapid cell division in improved nutrient media now make this approach a viable option. We hypothesized that three-dimensional (3-D) cell cultures would better represent how sponges function in nature, including the production of bioactive compounds. We successfully cultured sponge cells in 3-D matrices using FibraCel® disks, thin hydrogel layers, and gel microdroplets (GMDs). For in vitro production of bioactive compounds, the use of GMDs is recommended. Nutrients and sponge products rapidly diffuse into and out of the 3-D matrix, the GMDs may be scaled up in spinner flasks, and cells and/or secreted products can be easily recovered. Serum and seminal plasma concentrations or activities of acid phosphatase (AP), prostate specific antigen (PSA), and canine prostate specific esterase (CPSE) were measured in normal dogs, dogs with benign prostatic hyperplasia (BPH), dogs with bacterial prostatitis, and dogs with prostatic carcinoma to determine if these assays would be of value in differentiating dogs with prostatic carcinoma from normal dogs, and dogs with other prostatic disorders. In addition, tissue sections of prostatic adenocarcinomas were stained with antiprostatic AP, anti-CPSE, and anti-PSA antibodies to determine if these would be suitable immunohistochemical markers of prostatic carcinoma. Prostate-specific antigen was not detected in canine serum or seminal plasma. Serum and seminal AP activities did not differ significantly between normal dogs and those with prostatic diseases, or among dogs with different prostatic disorders. Serum CPSE activities were significantly higher in dogs with BPH than in normal dogs. Mean serum CPSE activities in dogs with BPH, bacterial prostatitis, and prostatic carcinoma were not significantly different from each other. Slight to moderate immunohistochemical staining of canine prostatic adenocarcinomas was noted for prostatic AP and PSA; most tumors did not stain for CPSE. These results show that proteins of prostatic origin appear in the serum of dogs as a result of prostatic pathology, especially BPH. Canine prostatic adenocarcinoma does not appear to be associated with significant increases in CPSE or AP activities, possibly because of down-regulation of these enzymes by prostatic carcinoma cells. Pulmonary arterial pressure (PAP) is an indicator of resistance to blood flow through the lungs and when measured at high altitude is a reliable predictor of susceptibility of an animal to brisket disease, a noninfectious cardiac pulmonary condition. (Co)-variance components for PAP, birth weight, and adjusted 205-d weaning weight were estimated from 2,305 spring-born, registered Angus cattle from a Colorado ranch at an elevation of 1,981 m. A single measure of PAP was collected after weaning on animals born from 1984 to 2003. The same licensed veterinarian measured every animal. Multitrait animal models with and without PAP maternal effects were fitted for a pedigree including 132 sires and 793 dams. The interaction of year x sex was a significant fixed effect (P <0.05) for PAP, but age of dam was not. Age at PAP testing was a significant (P <0.1) linear covariate for PAP, and scores increased 0.012 +/- 0.007 mmHg X d(-1) of age. Heritability of PAP direct was 0.34 +/- 0.05. Maternal heritability converged to a boundary at 0.0, and the model with maternal genetic effects for PAP was not significantly better than a model with only direct effects. Phenotypically, PAP was uncorrelated with birth or weaning weights. Genetically, PAP appeared to have positive, unfavorable relationships with direct effects for birth (0.49 +/- 0.12) and weaning weight (0.50 +/- 0.18). Positive correlations imply sires whose offspring exhibited resistance to brisket disease had lower weights and gains. A model that evaluated PAP in females and males as different traits had heritability estimates for each sex of 0.38 +/- 0.07 and 0.46 +/- 0.09, respectively, with a genetic correlation of 0.64 +/- 0.12 between the sexes and was not significantly better than the model assuming homogeneity by sex and a unit genetic correlation between sexes. The results suggest that PAP is moderately heritable in spring-born Angus cattle acclimatized and tested at high altitude, and selection for low PAP scores would be effective. OBJECTIVE: To test the hypotheses that kinematic data of the sagittal motion of canine hind limbs during walking obtained with a 2-dimensional (2-D) system correlate well with those obtained with a 3-dimensional (3-D) system and that the data obtained with the 2-D system are repeatable. ANIMALS: 6 adult dogs with no evidence of lameness. PROCEDURES: Hind limb motions of 6 walking dogs were recorded via 2-D video and 3-D optoelectric systems simultaneously. Five valid trials were digitized, and 5 data sets (2-D 60 Hz, 3-D 180 Hz, 3-D sagittal 180 Hz, 3-D 60 Hz, and 3-D sagittal 60 Hz) of a complete gait cycle were created for each dog. In sagittal data sets, 3-D data were reduced to exclude coordinates for mediolateral orientation. Temporospatial parameters; angles of hip, stifle, and tarsal joints; and coefficients of variation of angular measurements of each dog were calculated for each data set. Accuracy of the 2-D analysis was determined by calculating mean absolute differences and estimating agreement between the 2-D and 3-D 180-Hz data sets. RESULTS: Values of joint angles and angular excursions measured with the 2-D system were repeatable and agreed with respective values obtained with the 3-D system. Reduction of the sampling rate had a greater impact on values of kinematic variables obtained with the 3-D system than did elimination of data on mediolateral orientation. 利用农杆菌介导法将BCL和RIP 2个与细胞凋亡有关的基因分别导入了小麦品种扬麦10号和Bobwhite,转化效率分别为1.60%和1.25%。Southern检测结果表明,细胞凋亡基因已稳定整合到小麦染色体上,多数植株为单拷贝整合。Northern检测结果表明,细胞凋亡基因能以小麦遗传背景高水平转录为RNA。转基因植株T1代分离比例为2.11~2.33∶1,表明外源基因在后代中能够稳定遗传。BCL转基因植株和RIP转基因植株对赤霉病均表现出一定抗性,其中BCL-20、BCL-21、RIP-8、RIP-18等15个T1代植株的小穗发病率为5.6%~16.1%,可望进一步培育抗小麦赤霉病新材料。 相似文献
