Repeated exposures to anti-cholinesterase compounds induce profound enzyme inhibition and consequent neurotransmitter perturbations. When the result of exposure is an accumulation of neurotransmitter, as it is in the case of exposure to soman, target receptors may respond by compensating for the imbalance by regulating either their number or binding affinity. Recovery of function may further include alterations in receptor systems not themselves a direct target of the toxic compound. Rats were injected with soman (50 μg/kg, sc) three times weekly for 4 weeks and decapitated at intervals ranging from 1 hr to 6 days after the final injection. Synaptic membranes were prepared from hippocampus (HIP), striatum (STR), and cortex (COR) and kinetics of muscarinic cholinergic and GABAergic binding were assessed using [3H]QNB and [3H]muscimol, respectively, as ligands. Soman induced large and persistent decreases in the number of muscarinic receptors in HIP and COR with the surviving receptors being of higher affinity. The number of high affinity GABA receptors decreased in all three regions but returned to near baseline within 3 days after the final soman exposure. These effects are due either to a direct action of soman on the receptors, or a diaschitic response of the receptors to cholinergic excess. Muscarinic cholinergic receptors exhibit a persistent down regulation, and GABA receptors a transient response, to soman administration.  相似文献

During 1983–1989 about 1,200 infested plots in the Czech republic were investigated for the occurrence of new pathotypes of the potato cyst nematodes (PCN) Globoderu rostochiensis and G. pallida. Only eight populations of the pest were suspected of containing another pathotype than Ro1. Glasshouse pot trials were performed with those eight natural populations, two populations of Ro1, 10 potato cultivars resistant to Ro1 and 10 cultivars resistant to more pathotypes, G. pallida included. The multiplication rates (Pf/Pi) showed that four of the PCN populations were probably mixtures, with pathotype Ro1 prevailing. Three populations were suspected of containing G. pallida , but their morphological characters corresponded only with G. rostochiensis. One population corresponded with pathotype Ro5. The extent of the occurrence of pathotypes or virulence groups different from Ro1 can be estimated at 0.3–0.4% of infested plots.  相似文献

OBJECTIVE: To determine and compare the effects of caffeine and doxapram on cardiorespiratory variables in foals during isoflurane-induced respiratory acidosis. ANIMALS: 6 clinically normal foals (1 to 3 days old). PROCEDURES: At intervals of > or = 24 hours, foals received each of 3 IV treatments while in a steady state of hypercapnia induced by isoflurane anesthesia (mean +/- SD, 1.4 +/- 0.3% endtidal isoflurane concentration). After assessment of baseline cardiorespiratory variables, a low dose of the treatment was administered and variables were reassessed; a high dose was then administered, and variables were again assessed. Sequential low- and high-dose treatments included doxapram (loading dose of 0.5 mg/kg, followed by a 20-minute infusion at 0.03 mg/kg/min and then 0.08 mg/kg/min), caffeine (5 mg/kg and 10 mg/kg), and saline (0.9% NaCl) solution (equivalent volumes). RESULTS: Administration of doxapram at both infusion rates resulted in a significant increase in respiratory rate, minute ventilation, arterial blood pH, PaO(2), and arterial blood pressure. These variables were also significantly higher during doxapram administration than during caffeine or saline solution administration. There was a significant dose-dependent decrease in PaCO(2) and arterial bicarbonate concentration during doxapram treatment. In contrast, PaCO(2) increased from baseline values after administration of saline solution or caffeine. The PaCO(2) value was significantly lower during doxapram treatment than it was during caffeine or saline solution treatment. CONCLUSIONS AND CLINICAL RELEVANCE: Results indicated that doxapram restored ventilation in a dose-dependent manner in neonatal foals with isoflurane-induced hypercapnia. The effects of caffeine on respiratory function were indistinguishable from those of saline solution.  相似文献

Ecdysone, the major steroid hormone of Drosophila melanogaster, is known for its role in development and reproduction. Flies that are heterozygous for mutations of the ecdysone receptor exhibit increases in life-span and resistance to various stresses, with no apparent deficit in fertility or activity. A mutant involved in the biosynthesis of ecdysone displays similar effects, which are suppressed by feeding ecdysone to the flies. These observations demonstrate the importance of the ecdysone hormonal pathway, a new player in regulating longevity.  相似文献

Nucleotide sequences for the nuclear genes encoding chloroplast (GapA and GapB) and cytosolic (GapC) glyceraldehyde-3-phosphate dehydrogenases (GAPDHs) from Arabidopsis thaliana were determined. Comparison of nucleotide sequences indicates that the divergence of chloroplast and cytosolic GAPDH genes preceded the divergence of prokaryotes and eukaryotes. In addition, some intron-exon junctions are conserved among GapB, GapC, and chicken GAPDH genes. These results provide evidence at the molecular level to support the idea that introns existed before the divergence of prokaryotes and eukaryotes.  相似文献

Recent attention has been given to the influence of dietary factors on health and mental well-being. Oxidative stress is associated with many diseases including neurodegenerative disorders. Dietary flavonoids exert cardioprotective, chemopreventive, and neuroprotective effects. The biological activities of flavonoids have been attributed to their antioxidant, anti-inflammatory, and signaling properties. A clear understanding of the mechanisms of action, as either antioxidants or signaling molecules, is crucial for the application of flavonoids as interventions in neurodegeneration and as brain foods. Citrus flavonoids exert little adverse effect and have low or no cytotoxicity to healthy, normal cells. The main citrus flavonoids can also traverse the blood-brain barrier; hence, they are promising candidates for intervention in neurodegeneration and as constituents in brain foods. In this review, we discuss the bioactivity, multiple neuroprotection mechanisms, and antioxidant and signaling properties of citrus flavonoids. Receptor-mediated neuroprotective actions and parallel signaling pathways are also explored. Finally, the induction of cellular defense proteins against oxidative stress and neurotoxicity by hesperetin, a main and widespread citrus flavonoid, are also discussed. It is suggested that citrus fruits, which are rich in abundant sources of hesperetin and other flavonoids, are promising for the development of general food-based neuroprotection and brain foods.  相似文献

AIM: To identify if there was a significant difference in the average daily liveweight gain (ADG) between Thoroughbred foals weaned using two weaning protocols commonly used in New Zealand.

METHODS: Sixteen Thoroughbred foals were blocked for sex and age, randomly allocated into progressive (187, SD 33 days; three colts, five fillies) or abrupt (182, SD 28 days; four colts, four fillies) weaning groups, and weighed every second day for 2 weeks either side of weaning, then fortnightly from birth to 480 (SD 31) days old. ADG was calculated to examine the short-term (10 days before weaning, 5 and 10 days post weaning) and long-term (0–6 and 6–16 months of age) effect of the two weaning treatments.

RESULTS: ADG was 1.10 (SD 0.091) kg/day before weaning (0–6 months of age) and 0.59 (SD 0.06) kg/day from weaning to 480 (SD 31) days old. At the start of weaning, liveweights of the progressive and abrupt weaning groups were 276.5 (SD 40.3) kg and 257 (SD 15) kg, respectively (p=0.23). For the 5-day period during weaning, irrespective of treatment, there was a significant decrease in ADG of –0.29 (SD 0.49) kg/day and –0.15 (SD 0.30) kg/day for progressive and abrupt weaning, respectively. There was no significant difference in ADG between weaning methods at any measurement period (short or long term) during and after weaning. However, there was large variation between foals in ADG in the 10 days after the weaning process, which may indicate variation in individual foals' responses to being weaned, rather than the weaning treatment.

CONCLUSIONS: Weaning, irrespective of method, was associated with a decrease in ADG in the first week after weaning. The method of weaning had no effect on post-weaning ADG either short term, 10 days after weaning, or long term up to 480 days of age. Practically, it may be more important to consider maturity and liveweight as criteria for weaning foals rather than age alone.  相似文献
