A variety of published physical measurements, computational algorithms, and structural modeling methods have been used to create a molecular model of 19 kDa alpha-zein (Z19). Zetaeins are water-insoluble storage proteins found in corn protein bodies. Analyses of the protein sequence using probability algorithms, structural studies by circular dichroism, infrared spectroscopy, small-angle X-ray scattering (SAXS), light scattering, proton exchange, NMR, and optical rotatory dispersion experiments suggest that Z19 has approximately 35-60% helical character, made up of nine helical segments of about 20 amino acids with glutamine-rich "turns" or "loops". SAXS and light-scattering experiments suggest that in alcohol/water mixtures alpha-zein exists as an oblong structure with an axial ratio of approximately 6:1. Furthermore, ultracentifugation, birefringence, dielectric, and viscosity studies indicate that alpha-zein behaves as an asymmetric particle with an axial ratio of from 7:1 to 28:1. Published models of alpha-zein to date have not been consistent with the experimental data, and for this reason the structure was re-examined using molecular mechanics and dynamics simulations creating a new three-dimensional (3D) structure for Z19. From the amino acid sequence and probability algorithms this analysis suggested that alpha-zein has coiled-coil tendencies resulting in alpha-helices with about four residues per turn in the central helical sections with the nonpolar residue side chains forming a hydrophobic face inside a triple superhelix. The nine helical segments of the 19 kDa protein were modeled into three sets of three interacting coiled-coil helices with segments positioned end to end. The resulting structure lengthens with the addition of the N- and C-terminal sections, to give an axial ratio of approximately 6 or 7:1 in agreement with recent experiments. The natural carotenoid, lutein, is found to fit into the core of the triple-helical segments and help stabilize the configuration. Molecular dynamics simulations with explicit methanol/water molecules as solvent have been carried out to refine the 3D structure.  相似文献

Polycyclic aromatic hydrocarbons (PAHs) are widespread environmental pollutants produced by incomplete combustion sources such as home heating, biomass burning, and vehicle emissions. PAH concentrations in soils are influenced by source inputs and environmental factors that control loss processes and soil retention. Many studies have found higher concentrations of these pollutants in soils within cities of temperate climates that have a centralized urban core. Less is known about the factors regulating PAH abundance in warm, arid urban ecosystems with low population densities but high traffic volumes. The relative importance of sources such as motor vehicle traffic load and aridland ecosystem characteristics, including temperature, silt, and soil organic matter (SOM) were explored as factors regulating PAH concentrations in soils near highways across the metropolitan area of Phoenix, AZ (USA). Highway traffic is high compared with other cities, with an average of 155,000 vehicles/day. Soils contained low but variable amounts of SOM (median 2.8?±?1.8% standard deviation). Across the city, median PAH concentrations in soil were low relative to other cities, 523?±?1,886 ??g/kg, ranging from 67 to 10,117 ??g/kg. Diagnostic ratio analyses confirmed that the source of PAHs is predominantly fuel combustion (i.e., vehicle emissions) rather than petrogenic, biogenic, or other combustion sources (coal, wood burning). However, in a multiple regression analysis including traffic characteristics and soil properties, SOM content was the variable most strongly related to PAH concentrations. Our research suggests that dryland soil characteristics play an important role in the retention of PAH compounds in soils of arid cities.  相似文献

Microorganisms play a major role in the degradation and detoxification of most soilapplied pesticides. Any interference with soil microbial activity may affect the persistence as well as the effectiveness of the pesticide. The objective of this study was to investigate the effect of granulated superphosphate fertilizers on terbutryn (2-(tert-butylamino)-4-(ethylamino)-6-(methylthio)-l,3,5-triazine) degradation and on its herbicidal activity. Concentrations exceeding 1% of superphosphate in the soil slowed down terbutryn degradation; a concentration of 3% completely inhibited terbutryn degradation for nearly 60 days. When terbutryn was impregnated on the surface of superphosphate granules, the concentration of the fertilizer that inhibited terbutryn degradation was reduced from 3% to 0.1%. Bioassays with mustard seedlings confirmed the results obtained by chemical analysis of terbutryn. The decrease in the rate of terbutryn degradation was not specific to superphosphate. The inhibition of degradation could be attributed mainly to the concentration of the salt in the soil solution, expressed as electrical conductivity values, and not to the pH of the soil or to the type of the salt. Terbutryn was found to be quite stable on the surface of the superphosphate granule. At the concentrations tested, superphosphate did not affect root growth. The optimal weight of the impregnated granule for extending terbutryn activity was 125 mg, containing 0.2% terbutryn. A greenhouse experiment confirmed the findings that fertilizers could serve as potential inhibitors of terbutryn degradation.  相似文献

A stolbur‐type phytoplasma is the putative pathogen of grapevine yellows disease that causes economic damage to vineyards in most growing areas around the world. The pathogen is known to be transmitted to vines by two planthoppers, Hyalesthes obsoletus and Reptalus panzer; the latter is found in Europe but has not yet been observed in Israel. The establishment of a vector–pathogen–plant relationship requires that the pathogen and the vector meet on a shared host plant. This does not happen in the ecosystem examined here, where two different principal host plants for the obligate pathogen and its vector exist: the pathogen is established on vines, while its vector, H. obsoletus, develops on Vitex agnus‐castus. The present study verified that: (i) the vector cannot complete its life cycle on vines; (ii) V. agnus‐castus does not grow in the immediate vicinity of vines, and does not harbour the pathogen; and (iii) the pathogen is not vertically transmitted from mother to offspring. Moreover, in a thorough search of plants in vine growing areas, no other plants were found that host both the vector and the pathogen. However, it was found that the planthopper can acquire the phytoplasma from infected vines. Nonetheless, this does not prove the ability of the planthopper to further transmit the pathogen to vines and does not explain the presence of the vector on the non‐preferred vines. Thus, the enigma of the pathogen–vector–host triangle in this system remains unresolved.  相似文献

The symbiotic fungus Fusarium sp., carried in mycacangia located in the anterior region of the female head, is responsible for the typical wilt symptoms inflicted on avocado (Persea americana Miller) trees. The objective of this study was to evaluate the seroprevalence and identify the strains of swine influenza virus (SwIV), as well as the seroprevalence of porcine parvovirus (PPV), transmissible gastroenteritis virus (TGEV), porcine reproductive and respiratory syndrome virus (PRRSV), porcine respiratory coronavirus (PRCV), porcine circovirus type 2 (PCV-2), and classical swine fever virus (CSFV) in pigs in Trinidad and Tobago (T&T). Blood samples (309) were randomly collected from pigs at farms throughout T&T. Serum samples were tested for the presence of antibodies to the aforementioned viruses using commercial ELISA kits, and the circulating strains of SwIV were identified by the hemagglutination inhibition test (HIT). Antibodies against SwIV were detected in 114 out of the 309 samples (37%). Out of a total of 26 farms, 14 tested positive for SwIV antibodies. HI testing revealed high titers against the A/sw/Minnesota/593/99 H3N2 strain and the pH1N1 2009 pandemic strain. Antibodies against PPV were detected in 87 out of the 309 samples (28%), with 11 out of 26 farms testing positive for PPV antibodies. Antibodies against PCV-2 were detected in 205 out of the 309 samples tested (66%), with 25 out of the 26 farms testing positive for PCV-2 antibodies. No antibodies were detected in any of the tested pigs to PRRSV, TGEV, PRCV, or CSFV.  相似文献

Bacterial communities in sediments underneath milkfish cages and adjacent off‐cage areas in Bolinao, Lingayen Gulf, West Philippine Sea were characterized using PCR‐denaturing gradient gel electrophoresis to assess influence of the fish farming to the sediments. The sediments in the fish cage (FC) area showed anoxic conditions [redox potential of ?165 to ?213 mV and acid‐volatile sulphide‐sulphur (AVS‐S) of 0.55–1.88 mg g?1 (dry weight)] compared to the relatively oxic off‐cage area [redox potential of 72–81 mV and AVS‐S of 0.09–0.14 mg g?1 (dry weight)]. The composition of the microbial populations revealed influence of the fish feed sedimentation with higher dominance of putative sulphate‐reducing bacteria (SRB) in the FC area. Compositions of the bacterial taxa showed dominance of the Bacteroidetes group and Deltaproteobacteria. The results suggest that the fish feeds created a new suitable environment, in which reductive microorganisms such as SRB, ferric‐ion‐reducing bacteria and nitrate‐reducing bacteria can thrive. Organic load from the feed and quantity of its leftover constituents may be the key factors that influence the structure of the bacterial community in the sediments of the milkfish farms.  相似文献

An endophytic bacterium, wild poplar strain B (WPB), isolated from stems of wild cottonwood (Populus trichocarpa) was identified to Burkholderia vietnamiensis by analyzing the recA and rDNA genes. Phylogenetic analysis of the nifHDK cluster indicates that the WPB isolate shares high sequence similarity with known B. vietnamiensis strains. The nitrogenase activity of WPB was determined by a 15N2 incorporation assay and an acetylene reduction assay. WPB was also monitored for production of indole-3-acetic acid (IAA), a phytohormone which can promote plant growth, when incubated with l-tryptophan. In addition, its plant growth promotion capacity was assessed by inoculating the WPB strain onto Kentucky bluegrass in nitrogen-free medium. Compared to uninoculated control plants, the plants inoculated with WPB gained more dry weight (42%, p = 0.01) and more nitrogen content (37%, p = 0.04) in 50 days.  相似文献

Cover crops may influence soil carbon (C) sequestration and microbial biomass and activities by providing additional residue C to soil. We examined the influence of legume [crimson clover (Trifolium incarnatum L.)], nonlegume [rye (Secale cereale L.)], blend [a mixture of legumes containing balansa clover (Trifolium michelianum Savi), hairy vetch (Vicia villosa Roth), and crimson clover], and rye + blend mixture cover crops on soil C fractions at the 0–150 mm depth from 2001 to 2003. Active fractions of soil C included potential C mineralization (PCM) and microbial biomass C (MBC) and slow fraction as soil organic C (SOC). Experiments were conducted in Dothan sandy loam (fine-loamy, kaolinitic, thermic, Plinthic Kandiudults) under dryland cotton (Gossypium hirsutum L.) in central Georgia and in Tifton loamy sand (fine-loamy, siliceous, thermic, Plinthic Kandiudults) under irrigated cotton in southern Georgia, USA. Both dryland and irrigated cotton were planted in strip tillage system where planting rows were tilled, thereby leaving the areas between rows untilled. Total aboveground cover crop and cotton C in dryland and irrigated conditions were 0.72–2.90 Mg C ha−1 greater in rye + blend than in other cover crops in 2001 but was 1.15–2.24 Mg C ha−1 greater in rye than in blend and rye + blend in 2002. In dryland cotton, PCM at 50–150 mm was greater in June 2001 and 2002 than in January 2003 but MBC at 0–150 mm was greater in January 2003 than in June 2001. In irrigated cotton, SOC at 0–150 mm was greater with rye + blend than with crimson clover and at 0–50 mm was greater in March than in December 2002. The PCM at 0–50 and 0–150 mm was greater with blend and crimson clover than with rye in April 2001 and was greater with crimson clover than with rye and rye + blend in March 2002. The MBC at 0–50 mm was greater with rye than with blend and crimson clover in April 2001 and was greater with rye, blend, and rye + blend than with crimson clover in March 2002. As a result, PCM decreased by 21–24 g CO2–C ha−1 d−1 but MBC increased by 90–224 g CO2–C ha−1 d−1 from June 2001 to January 2003 in dryland cotton. In irrigated cotton, SOC decreased by 0.1–1.1 kg C ha−1 d−1, and PCM decreased by 10 g CO2–C ha−1 d−1 with rye to 79 g CO2–C ha−1 d−1 with blend, but MBC increased by 13 g CO2–C ha−1 d−1 with blend to 120 g CO2–C ha−1 d−1 with crimson clover from April 2001 to December 2002. Soil active C fractions varied between seasons due to differences in temperature, water content, and substrate availability in dryland cotton, regardless of cover crops. In irrigated cotton, increase in crop C input with legume + nonlegume treatment increased soil C storage and microbial biomass but lower C/N ratio of legume cover crops increased C mineralization and microbial activities in the spring.  相似文献
