Reasons for performing study: The flexion test is used routinely as part of lameness and prepurchase examinations. However, little is known about the mechanisms that cause a positive response to a flexion test. Objective: To determine which anatomical regions play a role in a positive outcome of a flexion test of the distal aspect of a forelimb in a nonlame horse. Methods: Eight clinically sound Dutch Warmblood horses were subjected to a standardised flexion test (force 250 N, time 60 s) inducing a consistent lameness. To discriminate between different areas of the distal aspect of a forelimb, effects of various nerve blocks on the outcome of the flexion test were investigated. Low palmar digital, palmar at the abaxial aspects of the base of the proximal sesamoids, high palmar, ulnar and low 4‐point nerve blocks were performed. Flexion test induced lameness was scored before and after each nerve block in separate sessions. Results: The low palmar digital nerve blocks and nerve blocks of the palmar nerves at the abaxial aspect of the base of the proximal sesamoid bones had no significant effect on the flexion test induced lameness score. The ulnar, high palmar and, most dramatically, the low 4‐point nerve blocks all caused a significant (P<0.05) reduction in the flexion test induced lameness score. Conclusions: Anatomical structures (soft tissue nor synovial structures) located distal to the metacarpophalangeal joint appear to contribute only minimally to the outcome of a positive flexion test of the distal aspect of a forelimb in a clinically nonlame horse. The structures in the region of, and including, the metacarpophalangeal joint appear to contribute most to a positive flexion test of the distal aspect of a forelimb in a nonlame horse. Potential relevance: The flexion test of the distal aspect of a forelimb may be sensitive for investigating the metacarpophalangeal joint region in horses free from lameness, but may be less relevant for structures distal to this region.  相似文献

Objective: To evaluate: (1) an arthroscopic technique for transection of the collateral sesamoidean ligament (CSL); and (2) the healing response using magnetic resonance (MR) and microscopic examination. Study Design: Experimental study. Animals: Adult horses (n=6). Methods: Six sound horses with normal front foot radiographic and MR examinations were used. Lameness examination was performed before surgery and monthly for 12 months. Front foot radiography was performed at 180 and 360 days after surgery. Front foot MR was performed before, and at 7, 90, 180, and 360 days after surgery. Arthroscopic CSL desmotomy was performed on 1 forelimb. Gross and microscopic examination was performed on the CSL from both forelimbs at 360 days after surgery. Lameness scores were compared over time using the nonparametric Friedman's test for paired groups. CSL measurements were compared using paired t‐tests with a 2‐tailed significance level of P<.05. Results: Radiographs remained normal throughout study period. Surgery resulted in lameness on the operated limb for up to 2 months, after which all horses returned to soundness. CSL transection was confirmed during arthroscopy and with MR examination 7 days after surgery. Gross and microscopic evaluation confirmed ligament healing. Conclusions: CSL desmotomy resulted in short‐term lameness after surgery followed by healing of the CSL confirmed by gross and microscopic analysis.  相似文献

Two major structural elements of a cell are the cytoskeleton and the lipid membranes. Actin and cholesterol are key components of the cytoskeleton and membranes, respectively, and are involved in a plethora of different cellular processes. This review summarizes and discusses the interaction of alphaherpesviruses with actin and cholesterol during different stages of the replication cycle: virus entry, replication and assembly in the nucleus, and virus egress. Elucidating these interactions not only yields novel insights into the biology of these important pathogens, but may also shed new light on cell biological aspects of actin and cholesterol, and lead to novel avenues in the design of antiviral strategies.  相似文献

Four different types of bedding materials (wheat straw, wood shavings, hemp shives, flax shives) and two roughages (hay, haylage) were treated using an air-driven particle separation technology. The airborne particle and mold generation of both treated and untreated samples were then analyzed under standardized laboratory conditions. In addition, samples of all the treated materials were stored for 8 weeks either in a pressed or incoherent form and then analyzed again for their ability to generate airborne particles. The airborne particle concentrations were detected online with the gravimetrically measuring analyzer tapered element oscillating microbalance (TEOM) 1400a that was equipped successively with different inlets to measure the particle fractions PM20, PM10, PM2.5, and PM1.0 (PM = Particulate matter). The particle separation resulted in a reduction in the airborne particle (PM20) generation in all materials: hay 49.16 to 22.79 mg/m3 (53.6%), haylage 28.57 to 25.04 mg/m3 (12.3%), wood shavings 141.68 to 15.04 mg/m3 (89.4%), wheat straw 143.08 to 22.97 mg/m3 (83.9%), flax 135.11 to 53.31 mg/m3 (60.5%), and hemp 63.67 to 17.64 mg/m3 (72.3%). The 8-week storage of the treated materials as compressed materials led to a renewed significant increase in the airborne particle (PM10) concentration in the haylage (+29.9%), wheat straw (+104.0%), wood shavings (+40.4%), and hemp shives (+30.7%). Storage of the incoherent materials caused a significant increase in these particles only in the wheat straw (+44.2%). The separation treatment reduced the mold production by 92.4% in the wood shavings, 88.0% in the wheat straw, and 85.8% in the hay.  相似文献

There is a lack of scientific evidence for objective evaluation of neck and back musculoskeletal sensitivity in horses, although pressure algometry has been described as an objective tool to quantify musculoskeletal responses by mechanical nociceptive threshold (MNT) values. This study aimed to evaluate the use of pressure algometry for objectively quantifying the effect of diagnostic palpation applied by physiotherapists on the musculoskeletal function of the equine neck and back. The inter-examiner repeatability of animal physiotherapists was tested, and their subjective clinical scores for the vertebral column area were objectively compared with MNT values measured at the same locations to investigate the potential clinical implementation of the pressure algometer in daily equine rehabilitation practice. Six adult Dutch Warmblood riding school mares were randomly assigned to an experimental or a control group. The MNT of all horses was measured on 35 predefined sites on the vertebral column in the morning and in the evening of the same day. In the experimental group (n = 3), neck and back surface “temperature”, “pain”, “muscle tone”, and passive “mobility” were scored through palpation by three certified physiotherapists and related to MNT measurements at the same vertebral column locations. Agreement between the physiotherapists was determined from Spearman's rank correlation coefficients (P < .05). These correlation coefficients showed a significant agreement between the scores of individual physiotherapists and with objective MNT measurements. The three physiotherapists agreed best in their subjective gradings of “pain”, but less for “temperature” and “muscle tone”, and least for “mobility”. There was also a significant difference in MNT between individual horses. The physiotherapeutic diagnostic intervention did not significantly alter the MNT of the experimental group compared with the control group. There was a significant difference, however, between morning (7.4 kg/cm2) and evening (6.9 kg/cm2) MNT-measurements within the combined group (n = 6, P < .05). In conclusion, a pressure algometer proved to be a useful tool to objectively monitor the palpation of individual Warmbloods by individual physiotherapists. The correlation of their scores to the objective MNT measurements elucidated that there were differences on which scale (“pain”, “temperature”, “muscle tone”, “mobility”) they merely relied upon in their palpation. Significant effects of physiotherapeutic diagnostic palpation on MNT, however, were not found. The lower MNT of the horses at the second trial in the evening could be a sensitization of the measurement location because of bruising, a learning effect of the horses, or a diurnal fluctuation. The use of pressure algometry has both a potential to quantify clinical neck and back musculoskeletal sensitivity in horses possibly leading to dysfunction, as well as to objectively evaluate treatment results. Repeated measurements on the same day and on the same location along the vertebral column may influence absolute MNT values. The algometer can be used with success provided that the operator has proper and frequent training.  相似文献

Background: Nosocomial salmonellosis is often assumed to occur because infection control and surveillance practices are inadequate, but published evidence is lacking to support the related contention that rigorous application of these practices can impact the severity of outbreaks. Objective: Describe active surveillance, early recognition, and intensive mitigation efforts used in an effort to control an outbreak of nosocomial Salmonella enterica serotype Newport infections without hospital closure. Animals: Large animals hospitalized at a referral hospital. Methods: This prospective outbreak investigation was initiated when Salmonella Newport infections were detected among hospitalized animals by active surveillance. Data were analyzed to identify temporal and spatial patterns for epidemic spread of Salmonella in the hospital. Mitigation efforts were aggressively adjusted in response to surveillance data. Genetic relatedness of isolates was investigated by pulsed‐field gel electrophoresis. Results: Of 145 large animals sampled, 8 (5.6%) were infected with the Salmonella strain associated with this outbreak, and all but 1 shed Salmonella in the absence of or before the onset of disease. This strain was recovered from 14.2% (42/295) of environmental samples (ENV samples), indicating that widespread environmental contamination had occurred. Isolates of Salmonella Newport obtained from infected animals and the environment were genetically indistinguishable, confirming clonal dissemination. Conclusions and Clinical Importance: Active surveillance allowed early detection of nosocomial Salmonella transmission and hospital contamination. Use of aggressive interventions was followed by cessation of transmission. Active surveillance can allow earlier recognition and mitigation compared with programs by only sampling of clinically affected animals.  相似文献

Objective – To determine whether older dogs will have a more pronounced pro‐inflammatory response and blunted anti‐inflammatory response to pathogen‐associated molecular patterns (PAMPs) compared with younger dogs. Design – Prospective. Setting – University teaching hospital. Animals – Thirty‐eight privately owned sexually altered dogs of various ages. Interventions – Blood was collected for HCT, WBC count, plasma biochemical analysis, and whole blood culture. Whole blood was stimulated with lipopolysaccharide (LPS) or, lipoteichoic acid or, peptidoglycan or, addition of phosphate‐buffered saline. Tumor necrosis factor (TNF), interleukin (IL)‐6, and IL‐10 production from whole blood were compared among young, middle aged, and geriatric dogs. Measurements and Main Results – LPS, lipoteichoic acid, and peptidoglycan stimulated significant TNF, IL‐6, and IL‐10 production from canine whole blood compared with phosphate‐buffered saline. Whole blood from geriatric dogs had a blunted IL‐10 response to LPS stimulation and middle‐aged dogs had increased LPS‐induced TNF production compared with the other groups. Conclusion – PAMPs from gram‐positive and gram‐negative bacteria stimulate TNF, IL‐6, and IL‐10 production from canine whole blood. The inflammatory mediator response to PAMPs from gram‐negative bacteria alters with age and may be one factor contributing to mortality in geriatric dogs with sepsis.  相似文献

Phytochemical investigation of the methanol extract of Tinospora cordifolia aerial parts led to the isolation of four new and seven known compounds. The structures of two new aporphine alkaloids, N-formylasimilobine 2-O-β-d-glucopyranosyl-(1 → 2)-β-d-glucopyranoside (tinoscorside A, 1) and N-acetylasimilobine 2-O-β-d-glucopyranosyl-(1 → 2)-β-d-glucopyranoside (tinoscorside B, 2), a new clerodane diterpene, tinoscorside C (3), and a new phenylpropanoid, sinapyl 4-O-β-d-apiofuranosyl-(1 → 6)-O-β-d-glucopyranoside (tinoscorside D, 6) were determined by extensive spectroscopic methods including FTICR-MS and 1D and 2D NMR.  相似文献
