The purpose of this research was to study changes in the salivary proteome of healthy pigs in stressful situations to identify any potential new salivary biomarker of stress. Three groups of animals were subjected to 3 stress models: snaring restraint followed by simulated sampling of vena cava blood; brief transport by road; and restriction of movement in a digestibility cage. Saliva was obtained from each animal before and 15 and 30 min after the induction of stress. The samples from the animals that showed the greatest increase in salivary cortisol concentration were pooled and run on 2-dimensional gels. Coomassie Brilliant Blue R-250 was used for spot detection and mass spectrometry for spot identification. Statistical analyses showed that 2 proteins had significant differences in expression before and after the induction of stress. These proteins were identified as odorant-binding protein and fragments of albumin. Further studies will be necessary to confirm the value of using these proteins as salivary biomarkers of stress in pigs. 相似文献

Previous reports on the spread of bovine virus diarrhoea virus (BVDV) from animals primarily infected with the agent are contradictory. In this study, the possibility of transmission of BVDV from calves simultaneously subjected to acute BVDV and bovine coronavirus (BCV) infection was investigated. Ten calves were inoculated intranasally with BVDV Type 1. Each of the 10 calves was then randomly allocated to one of two groups. In each group there were four additional calves, resulting in five infected and four susceptible calves per group. Virulent BCV was actively introduced in one of the groups by means of a transmitter calf. Two calves, susceptible to both BVDV and BCV, were kept in a separate group, as controls. All ten calves actively inoculated with BVDV became infected as shown by seroconversions, and six of them also shed the virus in nasal secretions. However, none of the other eight calves in the two groups (four in each) seroconverted to this agent. In contrast, it proved impossible to prevent the spread of BCV infection between the experimental groups and consequently all 20 study calves became infected with the virus. Following infection, BCV was detected in nasal secretions and in faeces of the calves and, after three weeks in the study, all had seroconverted to this virus. All calves, including the controls, showed at least one of the following clinical signs during days 3-15 after the trial started: fever (> or =40 degrees C), depressed general condition, diarrhoea, and cough. The study showed that BVDV primarily infected cattle, even when co-infected with an enteric and respiratory pathogen, are inefficient transmitters of BVDV. This finding supports the principle of the Scandinavian BVDV control programmes that elimination of BVDV infection from cattle populations can be achieved by identifying and removing persistently infected (PI) animals, i.e. that long-term circulation of the virus without the presence of PI animals is highly unlikely. 相似文献

A novel yeast named HQ-C-01 was isolated from activated sludge and identified as Pichia anomala based on the morphology and 18S rDNA sequence analysis. The HQ-C-01 strain degraded 95.2% of carbofuran when the insecticide
was used as the only C source and added at 50 mg/L in a mineral salts medium within 48 h. The optimal concentration, temperature,
and pH of medium for degradation of carbofuran were 50 mg/L, 30°C, and pH 7.5, respectively. Strain HQ-C-01 could also effectively
degrade other carbamate insecticides including carbaryl, indoxacarb, and fenobucarb, and the degradation rates were 99%, 85%,
and 67%, respectively. Gas chromatography–mass spectrometry analysis showed that the strain metabolized carbofuran to produce
benzofuranol as the intermediate metabolite, which was further degraded. Degradation of carbofuran added at 50 mg/kg of soil
was higher in yeast-inoculated soil than in the control. These results indicated that strain HQ-C-01 may potentially be used

This paper points out the usefulness of biopartitioning micellar chromatography (BMC) using capillary columns as a high-throughput primary screening tool providing key information about the oral absorption, skin permeability, and brain-blood distribution coefficients of 15 polyphenols (6 flavones, 2 flavonols, a flavanone, 2 flavan-3-ols, 3 phenolic acids, and a phloroglucinol) in a simple and economical way. For the compounds studied, except vicenin-2, rutin, chlorogenic acid, p-hydroxycinnamic acid, and 4-hydroxybenzoic acid, maximal oral absorption (>90%) can be expected, if there are not solubility problems or metabolic processes. On the other hand, the most retained compounds in BMC, that is, 5-hydroxyflavone, flavone, and flavanone, show the highest brain-blood distribution coefficients and skin permeability coefficients. 相似文献

Purpose
Remediation of metal contaminated soil with biochar is attracting extensive interest in recent years. Understanding the significance of variable biochar properties and soil types helps elucidating the meticulous roles of biochar in immobilizing/mobilizing metals/metalloids in contaminated soils.Materials and methods
Six biochars were produced from widely available agricultural wastes (i.e., soybean stover, peanut shells and pine needles) at two pyrolysis temperatures of 300 and 700 °C, respectively. The Pb-, Cu-, and Sb-contaminated shooting range soils and Pb-, Zn-, and As-contaminated agricultural soils were amended with the produced biochars. The mobility of metals/metalloids was assessed by the standard batch leaching test, principal component analysis and speciation modeling.Results and discussion
The changes in soil properties were correlated to feedstock types and pyrolysis temperatures of biochars based on the principal component analysis. Biochars produced at 300 °C were more efficient in decreasing Pb and Cu mobility (>93 %) in alkaline shooting range soil via surface complexation with carboxyl groups and Fe-/Al-minerals of biochars as well as metal-phosphates precipitation. By contrast, biochars produced at 700 °C outperformed their counterparts in decreasing Pb and Zn mobility (100 %) in acidic agricultural soil by metal-hydroxides precipitation due to biochar-induced pH increase. However, Sb and As mobility in both soils was unfavorably increased by biochar amendment, possibly due to the enhanced electrostatic repulsion and competition with phosphate.Conclusions
It is noteworthy that the application of biochars is not equally effective in immobilizing metals or mobilizing metalloids in different soils. We should apply biochar to multi-metal contaminated soil with great caution and tailor biochar production for achieving desired outcome and avoiding adverse impact on soil ecosystem.96.

Purpose
The main objective of this study was to examine the potential of using hyperspectral image analysis for prediction of total carbon (TC), total nitrogen (TN) and their isotope composition (δ13C and δ15N) in forest leaf litterfall samples.Materials and methods
Hyperspectral images were captured from ground litterfall samples of a natural forest in the spectral range of 400–1700 nm. A partial least-square regression model (PLSR) was used to correlate the relative reflectance spectra with TC, TN, δ13C and δ15N in the litterfall samples. The most important wavelengths were selected using β coefficient, and the final models were developed using the most important wavelengths. The models were, then, tested using an external validation set.Results and discussion
The results showed that the data of TC and δ13C could not be fitted to the PLSR model, possibly due to small variations observed in the TC and δ13C data. The model, however, was fitted well to TN and δ15N. The cross-validation R2 cv of the models for TN and δ15N were 0.74 and 0.67 with the RMSEcv of 0.53% and 1.07‰, respectively. The external validation R2 ex of the prediction was 0.64 and 0.67, and the RMSEex was 0.53% and 1.19 ‰, for TN and δ15N, respectively. The ratio of performance to deviation (RPD) of the predictions was 1.48 and 1.53, respectively, for TN and δ15N, showing that the models were reliable for the prediction of TN and δ15N in new forest leaf litterfall samples.Conclusions
The PLSR model was not successful in predicting TC and δ13C in forest leaf litterfall samples using hyperspectral data. The predictions of TN and δ15N values in the external litterfall samples were reliable, and PLSR can be used for future prediction.97.
Context
Common indicators are needed to monitor biodiversity loss and the implications for the sustainable provision of ecosystem services (ES). A plethora of different sets of indicators may hinder the identification of major endpoints for large-scale assessments of biodiversity and ecosystem services (BES).Objectives
We aim to describe the main challenges of indicators for BES assessment and provide suggestions for establishing a comprehensive indicator system.Methods
An extensive literature review was conducted in this study. We review the main challenges of indicators for BES assessment and propose corresponding improvements from our perspectives of theory and practical applications.Results
The main theoretical challenges of existing indicators include inconsistent definitions and classifications of ES, misunderstanding of linkages among biodiversity, ecosystem structure, functions and services, and practical problems relate to such issues as indicator representativeness, data availability, and uncertainty. Our suggested improvements include clarifying the main terms, concepts and classification of indicators, establishing a robust conceptual framework with clear interactions among different components and indicators, selecting indicators based on ecosystem properties, streaming existing data into one platform, and strengthening validation of proxies. The steps for constructing a comprehensive indicator system for BES assessment are summarized.Conclusions
Clear definitions of key terms are indispensable to classify indicators and construct a conceptual framework. Improved understanding of the relations among indicators of biodiversity, ecosystem functions, and ES across multiple scales can guide the development of the indicator system. The integrated indicator system is an important tool for BES assessment to support decision making for sustainable development.98.

The determination of the occurrence and level of cocoa shells in cocoa products and chocolate is an important analytical issue. The recent European Union directive on cocoa and chocolate products (2000/36/EC) has not retained the former limit of a maximum amount of 5% of cocoa shells in cocoa nibs (based on fat-free dry matter), previously authorized for the elaboration of cocoa products such as cocoa mass. In the present study, we report a reliable gas-liquid chromatography procedure suitable for the determination of the occurrence of cocoa shells in cocoa products by detection of fatty acid tryptamides (FATs). The precision of the method was evaluated by analyzing nine different samples (cocoa liquors with different ranges of shells) six times (replicate repeatability). The variations of the robust coefficient of variation of the repeatability demonstrated that FAT(C22), FAT(C24), and total FATs are good markers for the detection of shells in cocoa products. The trueness of the method was evaluated by determining the FAT content in two spiked matrices (cocoa liquors and cocoa shells) at different levels (from 1 to 50 mg/100 g). A good relation was found between the results obtained and the spiking (recovery varied between 90 and 130%), and the linearity range was established between 1 and 50 mg/100 g in cocoa products. For total FAT contents of cocoa liquor containing 5% shells, the measurement uncertainty allows us to conclude that FAT is equal to 4.01 +/- 0.8 mg/100 g. This validated method is perfectly suitable to determine shell contents in cocoa products using FAT(C22), FAT(C24), and total FATs as markers. We estimated forest biomass carbon storage and carbon density from 1949 to 2008 based on nine consecutive forest inventories in Henan Province,China.According to the definitions of the forest inventory,Henan forests were categorized into five groups: forest stands,economic forests,bamboo forests,open forests,and shrub forests.We estimated biomass carbon in forest stands for each inventory period by using the continuous biomass expansion factor method.We used the mean biomass density method to estimate carbon stocks in economic,bamboo,open and shrub forests.Over the 60-year period,total forest vegetation carbon storage increased from34.6 Tg(1 Tg = 1×10~(12)g) in 1949 to 80.4 Tg in 2008,a net vegetation carbon increase of 45.8 Tg.By stand type,increases were 39.8 Tg in forest stands,5.5 Tg in economic forests,0.6 Tg in bamboo forests,and-0.1 Tg in open forests combine shrub forests.Carbon storageincreased at an average annual rate of 0.8 Tg carbon over the study period.Carbon was mainly stored in young and middle-aged forests,which together accounted for 70–88%of the total forest carbon storage in different inventory periods.Broad-leaved forest was the main contributor to forest carbon sequestration.From 1998 to 2008,during implementation of national afforestation and reforestation programs,the carbon storage of planted forest increased sharply from 3.9 to 37.9 Tg.Our results show that with the growth of young planted forest,Henan Province forests realized large gains in carbon sequestration over a 60-year period that was characterized in part by a nation-wide tree planting program. 相似文献

Ocular ultrasonography in horses represents a valuable imaging diagnostic tool for the diagnosis of intraocular and periocular diseases, particularly when cornea or lens opacities preclude ophthalmoscopy of deeper structures. The authors studied normal and pathological aspects of the eye. Nineteen horses with opacities of the dioptric structures underwent an ultrasound examination. This technique allowed the diagnosis of a posterior synechia (1), cataracts (8), vitreous opacities (6), retinal detachment (3), and a foreign body (1). Ultrasonography provided helpful information about the structure and vascular pattern when the conventional ophthalmic evaluation was unable to achieve a correct diagnosis. 相似文献
