Black rot of crucifers is one of the most important diseases of wild rocket (Diplotaxis tenuifolia L. (D.C.)) caused by the seedborne pathogen Xanthomonas campestris pv. campestris. From 2005, it frequently affected this cultivation in the south of Italy, leading to heavy crop losses. In the present work, we aimed to describe the physiological and molecular characteristics of twenty X. campestris pv. campestris strains isolated from plants and seeds. Ten Xanthomonas spp. strains contaminating seeds were identified on the basis of molecular characterization and in vivo pathogenicity on a discriminating host range. Some of seed-borne isolates were ascribed to the species Xanthomonas campestris pv. raphani and X. campestris pv. incanae, indicating the occurrence of non-host pathogenic Xanthomonas on wild rocket seeds. As well as the presence of pathogenic bacteria, even non-pathogenic Xanthomonas spp. strains were detected on the seeds, underlying the importance of identifying them to evaluate the suitability of lots intended for sowing. A phylogeny using 69 Gyrase B (gyrB) sequences retrieved from the literature, was also carried out, highlighting species relatedness. Overall, this study provides a comprehensive framework for Xanthomonas species affecting wild rocket in Southern Italy.
Shiga toxin-producing Escherichia coli (STEC) are an important group of emerging pathogens, with ruminants recognised as their main natural reservoir. The aim of this work was to establish the prevalence of non-O157 STEC in free-ranging wild ruminants in the Extremadura region of Spain and to characterise them phenogenotypically. Faecal samples were collected from 243 wild ruminants, including Cervus elaphus, Capreolus capreolus, Dama dama and Ovis musimon and were examined for STEC using both phenotypic (Vero cells) and genotypic (PCR and PFGE) methods.Shiga toxin-producing Escherichia coli were isolated from 58 (23.9%) of the samples and a total of 65 isolates were characterised. A PCR method indicated that 11 (16.9%) strains carried the stx1 gene, 44 (67.7%) carried the stx2 gene and 10 (15.4%) carried both these genes. The ehxA gene was detected in 37 (57%) of the isolates but none contained either the eae or saa genes. The isolates were from a total of 12 ‘O’ serogroups, although 80% were restricted to the O2, O8, O128, O146, O166 and O174 serogroups. The most commonly isolated STEC bacteria, which were from the O146 serogroup, exhibited a high degree of polymorphism as indicated by PFGE. Shiga toxin-producing Escherichia coli isolates of serogroups O20, O25, O166, O171, O174 and O176 had not previously been found in wild ruminants. Intake of polyunsaturated fatty acids (PUFA) could modify follicular dynamics and reduce the length of the interval from calving to first ovulation (ICOV) in dairy cows, associated with changes in metabolites (i.e. cholesterol) or hormones (i.e. IGF-I). To test this hypothesis, 48 cows were stratified in two groups (primiparous or multiparous) and within each group were randomly assigned to three treatments to begin at calving: 0 kg (SS0), 0.7 kg (SS0.7) and 1.4 kg (SS1.4) of whole sunflower seeds (WSS) per cow per day. The experimental period lasted for 60 days after calving and the diets, which consisted of direct grazing of improved pastures, whole-plant wheat silage and concentrates, were designed to be isocaloric and isoproteic (1.6 Mcal NEL/kg DM, 16.7% CP). Ovaries were examined by ultrasound three times per week from day 8 postpartum (PP) until first PP ovulation. Body condition score (BCS) was determined and blood samples were collected weekly from calving to characterize the metabolic profiles. In treatments SS0.7 and SS1.4, 7/8 and 6/8 primiparous cows ovulated the first follicular wave PP, compared with only 1/8 primiparous cow in treatment SS0, while there were no treatment differences in multiparous cows. The ICOV was 44, 21 and 19 days for the primiparous, and 22, 21 and 25 days for the multiparous cows, in treatments SS0, SS0.7 and SS1.4, respectively. Treatments had no effect on BCS, plasma IGF-I and metabolic profiles except that NEFA and urea were increased by WSS supplementation. Intake of PUFA (WSS up to 1.4 kg/day or 6.7% of the diet) during early lactation reduced the ICOV in primiparous grazing dairy cows in early lactation but not in multiparous cows, and while the precise mechanism for these results is unclear, it was not related to changes in the plasma concentrations of IGF-I or cholesterol. 相似文献
We conducted an immunological assay of blood samples taken from 85 swine‐specialist veterinarians attending the Congress of the Mexican Association of Swine Specialist Veterinarians in Mexico in 2011. Serum samples were assayed for Porcine rubulavirus (PorPV), Encephalomyocarditis virus (EMCV) and Leptospira spp. antibodies. Using a hemagglutination inhibition test, we registered 2.3% and 27% seropositivity for PorPV and EMCV, respectively. Using viral neutralization tests, we registered 5.8% and 47% seropositivity for PorPV and EMCV, respectively. For Leptospira spp., we registered a seropositivity of 38.8%. The variables (sex, age, years of exposure, number of visited farms, biosecurity level and region) showed no significant effect (P > 0.05) on the seropositivity for EMCV, PorPV and Leptospira spp. except for number of visited farms on HI seropositivity for EMCV (P < 0.05; odds ratio: 1.38). The data obtained provide information on the epidemiology of emerging diseases with zoonotic potential in occupational risk groups. 相似文献
