Summary Effects on yield and quality associated with the glabrous leaf, frego bract, okra leaf and nectariless genes (singly and combined) were studied under two insecticide spray regimes (heavy and light) in field experiments repeated over three seasons. Under the heavy spray regime there were few consistent differences between mutant genotypes and the normal except for low yields associated with the glabrous leaf, nectariless, frego bract genotype and the okra leaf genotype. Under the light spray regime, genotypes containing the okra leaf gene and/or the nectariless genes were associated with higher yields than the normal and genotypes containing glabrous leaf and/or frego bract genes were associated with lower yields. Positive epistatic interactions occurred in a number of instances including the okra leaf gene in the glabrous hair, normal bract background under heavy spraying, and with all backgrounds under light spraying, and for glabrous leaf in both the okra normal and okra frego backgrounds under heavy spraying. Nectariless also interacted positively with the normal leaf, normal hair, normal bract background under light spraying. Pronounced negative epistatic interactions only occurred in the light spray regime, and included glabrous leaf in all backgrounds, and frego bract in all backgrounds except the normal leaf, normal hair. The mutant genes had only minor effects on lint quality. The implications of the results for breeding and pest management are diseussed.  相似文献

The emergence of the infectious disease chytridiomycosis is a major factor responsible for amphibian extinctions in pristine habitats. However, some populations coexist with Batrachochytrium dendrobatidis (Bd), the agent of chytridiomycosis, and others go extinct when Bd arrives. Variation of pathogen severity among populations may be explained by differences of antimicrobial skin peptides and anti-Bd skin bacteria. Previous work showed that a population of the frog Rana sierrae had a high proportion of individuals with at least one species of cultured anti-Bd skin bacteria and was coexisting with Bd for more than 6 years. A population of the closely related sister species Rana muscosa had a significantly lower proportion of individuals with anti-Bd bacteria, and the following year, it went extinct due to Bd. We extended previous work to include another sampling of the R. sierrae population coexisting with Bd and found that, although the anti-Bd bacterial community somewhat differed, both populations had a high proportion of individuals with antifungal bacteria. We also included a population of R. muscosa that was naïve of Bd and predicted to go extinct once Bd emerged since that was the fate of neighboring populations. However, the naïve R. muscosa population had a high proportion of antifungal bacteria, which suggested that the population might survive if Bd emerged. Two years later, the population was surviving with Bd endemic. Variation in the proportion of individuals with anti-Bd skin bacteria appears to explain why some populations persist when Bd emerges in a population.  相似文献

The fatty acids from a series of milk-chocolate-based confectionery samples were analyzed as methyl esters by GC to determine the presence and amount of conjugated linoleic acid (CLA). A single peak corresponding to the 9-cis,11-trans isomer and ranging from less than 0.1% to nearly 0.2% of the total fatty acids, corresponding to up to 0.3 mg per g of chocolate, was observed. One of the chocolate extracts and a milk extract were subjected to silver ion HPLC and GC-MS in order to confirm the identity of the major isomer and tentatively identity minor isomers.  相似文献

The development and implementation of a population supplementation and restoration plan for any endangered species should involve an understanding of the species’ habitat requirements prior to the release of any captive bred individuals. The freshwater pearl mussel, Margaritifera margaritifera, has undergone dramatic declines over the last century and is now globally endangered. In Northern Ireland, the release of captive bred individuals is being used to support wild populations and repatriate the species in areas where it once existed. We employed a combination of maximum entropy modelling (MAXENT) and Generalized Linear Mixed Models (GLMM) to identify ecological parameters necessary to support wild populations using GIS-based landscape scale and ground-truthed habitat scale environmental parameters. The GIS-based landscape scale model suggested that mussel occurrence was associated with altitude and soil characteristics including the carbon, clay, sand, and silt content. Notably, mussels were associated with a relatively narrow band of variance indicating that M. margaritifera has a highly specific landscape niche. The ground-truthed habitat scale model suggested that mussel occurrence was associated with stable consolidated substrates, the extent of bankside trees, presence of indicative macrophyte species and fast flowing water. We propose a three phase conservation strategy for M. margaritifera identifying suitable areas within rivers that (i) have a high conservation value yet needing habitat restoration at a local level, (ii) sites for population supplementation of existing populations and (iii) sites for species reintroduction to rivers where the mussel historically occurred but is now locally extinct. A combined analytical approach including GIS-based landscape scale and ground-truthed habitat scale models provides a robust method by which suitable release sites can be identified for the population supplementation and restoration of an endangered species. Our results will be highly influential in the future management of M. margaritifera in Northern Ireland.  相似文献

The minke whale is the last of the great whale species to be hunted in significant numbers. Effective management must include an understanding of how genetic diversity is divided and distributed among putative local populations, and as for many migratory species, this is complicated for the minke whale by large-scale seasonal movement among geographic regions. The problem is that the geographic identity of breeding populations is not known, and instead these whales are predictably found and hunted where different breeding stocks may mix on seasonal feeding grounds. Here we use microsatellite DNA and mtDNA markers to investigate minke whale population structure across the species’ range in the North Atlantic. We found no evidence of geographic structure comparing putative populations in recognized management areas, though some limited structure had been indicated in earlier studies. However, using individual genotypes and likelihood assignment methods, we identified two putative cryptic stocks distributed across the North Atlantic in similar proportions in different regions. Some differences in the proportional representation of these populations may explain some of the apparent differentiation between regions detected previously. The implication would be that minke whales range extensively across the North Atlantic seasonally, but segregate to some extent on at least two breeding grounds. This means that established stock boundaries in the North Atlantic, currently used for management, should be re-considered to ensure the effective conservation of genetic diversity.  相似文献

There is evidence that colonisation by mycorrhizal fungi can protect host plants from toxic concentrations of heavy metals. The mechanism by which protection is provided by the fungus for any particular metal is poorly understood. Rice (Oryza sativa L.) plants were inoculated with Glomus mosseae and grown for 4 weeks to ensure strong colonisation. The plants were then exposed to low to toxic concentrations of copper (Cu) and the uptake and distribution were examined. The effect of mycorrhizal colonisation on the cell wall composition and Cu binding capacity of roots was also investigated. Mycorrhizal plants showed moderate reductions in Cu concentrations in roots but large reductions in shoots. In roots, mycorrhizal plants accumulated more Cu in cell walls but much less in the symplasm compared to non-mycorrhizal plants. The differences in cell wall binding of Cu could be partly explained by changes in the composition of the cell wall. The mechanistic basis for the reduced Cu accumulation and the potential beneficial consequences of mycorrhizal associations on plant growth in Cu toxic soil are discussed.  相似文献

The potential of combining solid-phase microextraction (SPME) with gas chromatography and chemometric data analysis to differentiate between pure strawberry samples (Fragraria ananassa) and strawberry samples adulterated with 10, 40, and 70% (v/v) apple purée was investigated. The method involved the extraction of aroma volatiles from the headspace of the purée samples using a SPME fiber followed by GC analysis with flame ionization detection. The principal component analysis (PCA) data matrix consisted of the relative percent peak areas of 37 compounds deemed to be significant in the differentiation of the samples on the basis of adulteration. The PCA results clearly showed that differentiation of the adulterated and unadulterated samples was possible, particularly at the higher levels of adulteration. Partial least-squares regression (PLSR) using a dummy set of Y variables (set to 0 for unadulterated and 1 for adulterated samples) resulted in clear discrimination between unadulterated purées and those containing 40 and 70% (v/v) apple. PLSR using a second set of Y variables, consisting of the actual level of adulteration, enabled quantification of apple purée with a standard error of prediction of 11.6%, implying a minimum detectable level of 25% (v/v) apple. GC-MS analysis enabled identification of the compounds with the greatest influence on sample differentiation. These compounds were identified as hexanoic acid, 2-hexenal, and alpha-farnesene, all of which are key aroma compounds in apples.  相似文献

Concern has been expressed over the status of the bottlenose dolphin population that uses the Moray Firth and adjacent waters in NE Scotland. Consequently, part of this population's range has been proposed as a Special Area of Conservation. Efforts to manage and monitor the status of this population require information on the level of genetic diversity within the population and its genetic relatedness to neighbouring populations to appropriately designate units for management and monitoring. Here we examine mitochondrial genetic diversity within the NE Scotland population, and compare this to other regions around the UK and Ireland. Sequence analysis of 549bp of the mitochondrial DNA control region identified eight unique haplotypes in a sample of 29 individuals. Analysis of molecular variance suggests that the Moray Firth population is genetically more closely related to Welsh animals than to its nearest neighbour population in west Scotland. Furthermore, measures of within-population genetic diversity were markedly lower in the Moray Firth than any other sampled region. The low levels of mtDNA genetic variability observed and its apparent geographic isolation provide further support for the precautionary approach currently being applied to the management of this population, despite the lack of direct evidence of harm.  相似文献

Arid rangelands host a variety of drought-tolerant wildlife species, many of them requiring conservation efforts for the survival of their populations. The development of drinking water sources for people and livestock forms one of the main development interventions in these rangelands. However, the impact of availability of permanent drinking water on wildlife remains unknown. In this study we analyzed the distribution of wildlife and livestock in northern Kenya in relation to distance to permanent water. Livestock were concentrated in areas close to permanent water, while wildlife were frequently farther away from water; their distributions were inversely correlated. In addition, wildlife assemblages were more diverse farther from water. These results suggest that livestock and human activities related to water points negatively affect the distribution of wildlife.  相似文献

The species Magicicada septendecim and Magicicada cassini of the 17-year cicada produce sound by sequentially buckling a series of stiff ribs embedded in a flexible tymbal. Each such collapse excites a damped oscillation in a resonant cavity.
