The pharmacokinetics of propofol were investigated in two groups of five Scottish blackface sheep undergoing surgery for the implantation of subcutaneous tissue pouches. After premedication with acepromazine and papaveretum, anaesthesia was induced with either propofol at 4 mg kg−1 intravenously (group 1) or with a mixture of propofol at 3 mg kg−1 and ketamine at 1 mg kg −1 intravenously (group 2). Anaesthesia was maintained with a variable infusion rate of either propofol alone (group 1) or propofol and ketamine (group 2). Both regimens produced satisfactory conditions for superficial surgery of the body surface. The mean (SD) duration of anaesthesia was 64·8 (3·1) minutes for group 1 and 60 (0) minutes for group 2; the mean total dose of propofol given to the sheep in group 1 was 801 (84) mg, and the sheep in group 2 received 470 (46) mg of propofol and 267 (30) mg of ketamine. The mean elimination half-life of propofol was 56·6 (13·1) minutes in group 1 and 50·3 (21·4) minutes in group 2; the mean volume of distribution at steady state was 1037 (0480) litre kg−1 in group 1 and 1·515 (0939) litre kg−1 in group 2; the mean body clearance was 85·4 (28·0) ml kg−1 min−1 in group 1 and 1280 (35·0) ml kg−1 min−1 in group 2; the mean residence time corrected for a bolus injection was 12·1 (4·2) minutes in group 1 and 11·9 (6·6) minutes in group 2; for the infusion, the mean residence time was 72·1 (4·2) minutes in group 1 and 69·9 (7·9) minutes in group 2. There were wide variations in the blood propofol concentrations reached in individual sheep by using this standard dosing regimen. All the sheep recovered quickly from anaesthesia; the mean times to extubation, sternal recumbency and standing for the animals in group 1 were 2·8 (0·4) 6·3 (1·2) and 10·9 (1·6) minutes from the end of the infusion, and the times for group 2 were 5·3 (0·9), 11·2 (1·7) and 15·1 (2·2) minutes.  相似文献

The auscultatory method was used to obtain indirect systolic and diastolic pressures in 13 dogs anesthetized with either halothane or sodium pentobarbital (30 mg/kg of body weight). Korotkoff sounds were obtained, using a 1-cm (diameter) piezoelectric element cemented to the inner surface of a pediatric cuff (width 5.5 cm) which was placed on a shaved thoracic limb (membrum thoracicum). The signal from the piezoelement was amplified by a differential amplifier (30 to 200 Hz) and a commercially available audio amplifier. Indirect pressure (I) was compared with direct pressure (D) in the brachial, femoral, or carotid artery. The linear regression lines and correlation coefficients (r) for the data were as follows: systolic, I = 0.94 (D) + 1.1, r = 0.98; diastolic, I = 0.99 (D) + 3.2, r = 0.99. The quality of the Korotkoff sounds and the accuracy of the determinations were best in the halothane-anesthetized dogs. These results indicate that indirect auscultatory systolic and diastolic pressures are in excellent agreement with the directly measured pressures.  相似文献

The transmission of a malignant catarrhal fever-like syndrome to sheep is reported. Fetal sheep between 40 and 66 days gestation were inoculated intravenously with viable cells either from a red deer with clinical malignant catarrhal fever or from rabbits with the disease. Of the 21 fetuses in the experiment only five were born live and of these four developed clinical signs similar to malignant catarrhal fever in other species and died or were killed 10, 16, 47 and 175 days after birth. The fifth lamb remained unaffected. Histology of the four affected lambs revealed a generalised lymphoproliferation, with T-dependent areas of lymphoid tissues being affected, and an overall paucity of immunoglobulin containing cells. In addition arteritis and interstitial infiltration of many organs by lymphoid cells was present. The infectious agent was not reisolated in rabbits from lambs or passed to red deer housed in the same pen and it is thus considered possible that gene expression of the putative virus was incomplete.  相似文献

Cats were vaccinated with one of the three preparations: purified feline immunodeficiency virus (FIV) incorporated into immune stimulating complexes (ISCOMs), recombinant FIV p24 ISCOMs, or a fixed, inactivated cell vaccine in quil A. Cats inoculated with the FIV ISCOMs or the recombinant p24 ISCOMs developed high titres of antibodies against the core protein p24 but had no detectable antibodies against the env protein gp120 or virus neutralising antibodies. In contrast, all of the cats inoculated with the fixed, inactivated cell vaccine developed anti-env antibodies and four of five had detectable levels of neutralising antibody. However, none of the vaccinated cats were protected from infection after intraperitoneal challenge with 20 infectious units of FIV. Indeed there appeared to be enhancement of infection after vaccination as the vaccinated cats become viraemic sooner than the unvaccinated controls, and 100% of the vaccinated cats became viraemic compared with 78% of the controls. The mechanism responsible for this enhancement remains unknown.  相似文献

Rangelands and hayfields provide a large portion of remaining surrogate habitat for many species of declining grassland birds in North America. We compared late-cut hayfields and continuously grazed pastures at low to moderate cattle densities for providing suitable breeding habitat in eastern Canada for the nationally threatened Bobolink (Dolichonyx oryzivorus). To examine the quality of both habitats, we conducted point counts and monitored 87 nests during the 2015 and 2016 breeding seasons. Bobolink abundance and daily survival rate (DSR) of nests were modeled sequentially by habitat and sex as a function of vegetation structure, prey availability, and agricultural management. Year and habitat were the strongest predictors of abundance. When analyzed separately for pastures and hayfields, vegetation height was most important for female abundance in pastures while pasture size was most important for males. Nests in hayfields had significantly higher daily survival (DSR = 0.98 ± 0.01) than nests in pastures (DSR = 0.94 ± 0.01). Nesting success was highest in hayfields with taller vegetation, while in pastures, no microhabitat variable showed a clear relationship with DSR. Within pastures, cattle stocking densities of ≤ 1 animal units (AU) · ha? 1 were not related to DSR. This study provides evidence that late-cut hay is of highest quality, but that small-scale beef farms with low to moderate stocking densities are suitable targets for conservation efforts of Bobolinks in eastern North America.  相似文献

The objective of this study was to evaluate different concentrations of growth hormone (GH) on the development of bovine preantral follicles cultured included in the ovarian tissue (in situ) on the rates of morphologically normal, viable, primordial and developing follicles, as well as the oocyte and follicle diameter and ultrastructural analysis. Ovarian fragments collected from cows with no cross‐breeds defined were cultured in situ for 1 and 7 days in minimal essential medium (α‐MEM+) supplemented with different concentrations of recombinant human GH (0, 10, 25, 50 ng/ml). The ovarian fragments non‐cultured (control) and cultured were processed for classic histology, mechanical isolation and electron transmission microscopy (MET). The parameters underwent anova (Tukey′s and Dunnett′s tests) and chi‐square test (χ2). After 7 days of culture, the treatment with 50 ng/ml GH showed no differences with fresh control (p > 0.05) and had greater effectiveness than in the 0, 10 and 25 ng/ml GH concentrations of the morphologically normal follicles. Regarding the primordial follicles, a reduction was observed in the 50 ng/ml GH concentration concomitant with the significant increase in developing follicles, differing from both the fresh control and the other GH concentrations tested. In addition, 50 ng/ml GH showed a larger follicle and oocyte diameter when compared to the other treatments cultured. Similar structures were ultrastructurally observed in the control group, 50 ng/ml GH. Follicles cultured in 10 ng/ml GH showed nuclear invagination, vacuoles and lesioned basal membrane. Hence, it is concluded that 50 ng/ml GH is the most effective concentration for the development of preantral follicles cultured in situ.  相似文献

Raccoons are common in urban and rural environments and can carry a wide range of bacteria, including Salmonella, that can negatively affect human and livestock health. Although previous studies have reported that raccoons shed a variety of Salmonella serovars in their faeces, it is unknown whether Salmonella is carried on raccoon paws. Our objective was to compare the prevalence of Salmonella on the paws and in the faeces of raccoons in south‐western Ontario. Raccoons were sampled in a repeat cross‐sectional study on five swine farms and five conservation areas from May to October 2012. A total of 416 paired faecal and paw samples were collected from 285 individual raccoons. Salmonella was detected in 18% (75/416; 95% CI, 14–22%) and 27% (111/416; 95% CI, 22–31%) of paw and faecal samples, respectively. Salmonella was detected only on paws in 8% (35/416; 95% CI, 5.9–11.5%), only in faeces in 17% (71/416; 95% CI, 13.6–21.0%) and on both paws and in faeces in 10% (40/416; 95% CI, 7.0–12.9%) of raccoon captures. Multilevel logistic regression models were used to examine associations between the presence of Salmonella and age (adult, juvenile), sex (male, female), location type (swine farm, conservation area), sample type (faeces, paw) and season (May–July and August–October). Random intercepts were included to account for clustering by individual animal and location. Significant differences, that varied by sample type and season, were noted in the prevalence of Salmonella carriage between sexes. Raccoons can carry Salmonella serovars known to infect humans and livestock on their paws and/or in their faeces and therefore have the potential to mechanically and biologically disseminate Salmonella among livestock facilities and human recreational areas.  相似文献
