Starch is the major component of wheat (Triticum aestivum L.) grain and is composed of two large glucan molecules, amylose and amylopectin. The ratio between the two polymers types influences the water absorbing properties of starch upon heating, and thus affects the end-use of grain and purified starch. In this study, we evaluated the starch swelling power (SSP) values in seven wheat populations developed from crosses involving low-SSP lines. Analysis of starch produced by the F2 generation plants showed that the largest SSP variation (11.4–16.2) and lowest SSP mean (13.9) was obtained for a population derived from doubled haploid lines SM1028 (SSP = 14.5) and VK306 (SSP = 13.6). The population of 360 lines was advanced by single seed descent to the following generations for further studies. Starch analysis of grain produced by F4 generation lines in two field locations during 2006 and in a greenhouse environment during 2005 showed that SSP values were relatively stably inherited. The average broad-sense heritability was 73% and significant (P < 0.001) genotype × genotype and genotype × environment interactions were seen. Starches with the highest and lowest SSP values were inversely related to amylose concentration determined by high pressure liquid chromatography (HPLC)–size exclusion chromatography (SEC) of debranched starch. Developed lines with the lowest SSP values surpassed 40% in apparent amylose concentration. The study illustrates that screening for SSP in early generations can be used to develop wheat lines with desired starch swelling characteristics.  相似文献

In present article fabrication and characterization of unfilled and granite powder filled carbon epoxy composites are reported. Addition of carbon fiber shows positive effect on mechanical performance of the composites. However, incorporation of granite powder has negative hybridizing effect on the properties such as tensile strength, flexural strength and inter-laminar shear strength. The storage modulus evaluated at 30 °C is in close agreement with flexural modulus of composites. Further, successful attempt is made for numerical simulation of actual geometry of wind turbine blade. The results obtained from numerical analysis are comparable with experimental results.  相似文献

Salvadora oleoides Decne. is a pharmaceutically important plant. Owing to poor seed formation, viability and, germination, and to anthropogenic disturbances, this species is on the verge of extinction. A reproducible micropropagation protocol to increase the population through tissue culture has been standardized and the results are reported here. Callus tissues were initiated from young leaves and stem explants. Leaf calluses proliferated with 1.5 mg/L BAP and 0.9 mg/L 2, 4-D with additives and continuous slow proliferation up to 15 weeks on 0.5 mg/L BAP and additives with 200 mg/L activated charcoal.Direct shoot initiation took place from stem node explants after 12 days; 4–5 shoots per node were produced in 30 days. Shoot clumps elongated and grew further on MS media supplemented with 2 mg/L BAP, 0.2 mg/L NAA and additives, which generated 20–23 shoots. The elongated shoots induced tap roots with 4 mg/L NAA and200 mg/L activated charcoal in 12 days. In vitro raised plants produced secondary roots when transferred to pots containing vermiculite maintained at 28–35 ℃. The plantlets successfully acclimatised in pots containing soil in natural conditions.  相似文献

The amount of (1 → 3),(1 → 4)-β-d glucan (β-glucan) accumulated in cell walls of barley (Hordeum vulgare L.) kernel is an important determinant for grain end-use. Grain β-glucan concentration is affected by environmental and genetic factors and usually varies from 3 to 6%. In this study, we have analyzed the β-glucan trait in a doubled-haploid (DH) population of 170 lines grown in three separate field trials. Most of the DH lines showed β-glucan values that ranged from that of the low β-glucan parent (cultivar CDC Bold; 3.3%) to that of the high β-glucan parent (breeding line TR251; 5.4%). Eighty-eight lines of the DH population were genotyped using simple sequence repeat (SSR), amplified fragment length polymorphism (AFLP) and diversity array technology (DArT) markers, which were subsequently integrated into a barley genetic map spanning 1059 cM. Interval mapping and multiple-QTL-mapping (MQM) of quantitative trait loci (QTL) from the three trials indicated seven genomic regions associated with low grain β-glucan concentration. For all putative QTLs, the low β-glucan concentration was contributed by alleles from CDC Bold except for two loci on chromosomes 5H that were derived from TR251. A major QTL located to the centromere region of chromosome 7H was identified by both mapping methods for all three trials. The 7H QTL explained up to 39% of the β-glucan concentration and genetic markers associated with the locus may be used to aid selection of high and low β-glucan barley lines.  相似文献

Enrichment of soil organic carbon (SOC) stocks through sequestration of atmospheric CO2 in agricultural soils is important because of its impacts on adaptation to and mitigation of climate change while also improving crop productivity and sustainability. In a long‐term fertility experiment carried out over 27 y under semiarid climatic condition, we evaluated the impact of crop‐residue C inputs through rainfed fingermillet (Eleusine coracana [L.] Gaertn.) cropping, fertilization, and manuring on crop yield sustainability and SOC sequestration in a Alfisol soil profile up to a depth of 1 m and also derived the critical value of C inputs for maintenance of SOC. Five treatments, viz., control, farmyard manure (FYM) 10 Mg ha–1, recommended dose of NPK (50 : 50 : 25 kg N, P2O5, K2O ha–1), FYM 10 Mg ha–1 + 50% recommended dose of NPK, and FYM 10 Mg ha–1 + 100% recommended dose of NPK imposed in a randomized block design replicated four times. Application of FYM alone or together with mineral fertilizer resulted in a higher C input and consequently built up a higher C stock. After 27 y, higher profile SOC stock (85.7 Mg ha–1), C build up (35.0%), and C sequestration (15.4 Mg C ha–1) was observed with the application of 10 Mg FYM ha–1 along with recommended dose of mineral fertilizer and these were positively correlated with cumulative C input and well reflected in sustainable yield index (SYI). For sustenance of SOC level (zero change due to cropping) a minimum quantity of 1.13 Mg C is required to be added per hectare per annum as inputs. While the control lost C, the application of mineral fertilizer served to maintain the priori C stock. Thus, the application of FYM increased the C stock, an effect which was even enhanced by additional amendment of mineral fertilizer. We conclude that organic amendments contribute to C sequestration counteracting climate change and at the same time improve soil fertility in the semiarid regions of India resulting in higher and more stable yields.  相似文献

Rabi-summer-produced (dry season, February–May) groundnut ( Arachis hypogaea L.) cv. GG 2 was dried using three different methods, viz. windrow (W), windrow-shade (WS) and DOR (Directorate of Oilseeds Research) methods and stored in polyethylene lined gunny bags with or without desiccant (CaCl2 or silica gel, 10gkg−1 pods). The seeds were analysed for the retentation of seed viability, seedling vigour, membrane integrity and Held emergence during storage, and crop stand and pod yield after 12 months of storage. The drying and storage method significantly affected the getminability, seedling vigour and field emergence. Both root length and seedling biomass were adversely affected due to windrow drying (W) The quick loss of viability in the W treatment was associated with an increase in electrical conductivity of the seed leachate. The retention of higher seed viabiliry and vigour in the seeds dried by DOR method and stored with CaCl2 (DOR C) was due to lower drying remperarures and lower pod moisture during the initial storage period. The DOR C method was found to be an effective drying and storage method for retaining acceptable seed viability and vigour till the following rabi-summer sowing of groundnut.  相似文献

The effect of a chitosan coating on antifungal activity and rate of respiration, chitinase and β, 1-3 glucanase activities with reference to papaya variety ‘Rathna’ was investigated. One percent chitosan, extracted from locally available prawn waste, was selected as the effective concentration to inhibit spore germination via a series of experiments on potato dextrose agar. Rate of respiration and the concentration of CO2 in the internal cavity of chitosan-treated and untreated papaya were tested via gas chromatography. Chitinase and β,1-3 glucanase activities were tested in peel samples using gel diffusion and spectrophotometric assays, respectively. Complete inhibition of spore germination was observed in-vitro at treatments of 1% chitosan and above. This concentration significantly (P < 0.05) reduced both disease incidence and severity on inoculated fresh papaya. Significant (P < 0.05) decrease was observed in rate of respiration while internal CO2 concentration of the fruit increased (P < 0.05) with the chitosan treatment. Chitinase and β,1-3 glucanase activities of papaya variety Rathna subjected to chitosan treatment were much higher than in the untreated control. Chitosan shows antifungal activity to the anthracnose disease causing fungus and stimulates the defense response on the papaya peel by increasing the chitinase and β,1-3 glucanase activities. The antifungal activity of chitosan could be attributed to the induction of elicitation activity due to these defense enzymes. It also forms a semi-permeable coating around the fruit and extends storage life of papaya by reducing the rate of respiration and delaying ripening.  相似文献

This study was carried out to investigate the effect of supplementing culture medium with different concentrations of taurine and melatonin, on buffalo oocyte in vitro meiotic maturation and embryo development. In experiment 1, oocytes were matured in vitro and the cleaved embryos were cultured in the same following seven culture medium; (i) control (TCM 199 + 10% SS); (ii) control + 0.5 m m taurine; (iii) control + 1 m m taurine; (iv) control + 3 m m taurine; (v) control + 5 μ m melatonin; (vi) control + 10 μ m melatonin and (vii) control + 50 μ m melatonin. In experiment 2, based on the results of experiment 1, to examine the synergistic effect of antioxidants, the oocytes were matured in culture medium (TCM199 + 10% SS), supplemented with both taurine at 1 m m and melatonin at 10 μ m concentration and the cleaved embryos were cultured in the same medium. Supplementation of taurine at 1 m m concentration in the culture medium resulted in a higher (p < 0.05) transferable embryo (TE) yield when compared with control (20.6% vs 14.1%). Supplementation of melatonin at 10 and 50 μ m concentration in the culture medium resulted in a higher (p < 0.05) meiotic maturation rate (90.3% and 88.8% respectively) and TE yield (28.4% and 27.2% respectively), than the other treatments. In experiment 2, the TE yield did not improve by supplementing the culture medium with both taurine and melatonin, when compared with melatonin alone. In conclusion, the results of this study demonstrated that, enriching the culture medium with taurine and melatonin, improves in vitro embryo production efficiency in buffaloes. In particular, a high TE yield was obtained by enriching the culture medium with 10 μ m melatonin.  相似文献

Starch granule composition and amylopectin structure affect starch digestibility, an important factor influencing wheat grain utilization for human food consumption. Six bread wheat cultivars with four belonging to the Canada Western Red Spring (CWRS) and two Canada Prairie Spring Red (CPSR) market classes were analyzed for the relationship between their grain constituents and in vitro enzymatic hydrolysis of starch. CPSR cultivars had higher starch and amylose concentrations compared with CWRS cultivars, which had a higher protein concentration. Starch granule size distribution did not differ among the genotypes, except AC Foremost, which had significantly (P < 0.05) higher volume percent of B‐type starch granules (≈15%) and lower volume percent of A‐type starch granules (≈9%) compared with other cultivars. Fluorophore‐assisted capillary electrophoresis revealed a lower content of R‐IV (DP 15–18, ≈6%) and a higher content of R‐VII (DP 37–45, ≈7%) chains in the CPSR cultivars compared with the CWRS cultivars. Starch in vitro enzymatic hydrolysis showed that compared with CWRS cultivars, the two CPSR cultivars had reduced amounts of readily digestible starch and higher amounts of slowly digestible starch and resistant starch. Consequently, the two CPSR cultivars also showed lower hydrolysis indexes in grain meal as well as extracted starch. CPSR cultivars, with higher starch and amylose concentrations, as well as a higher content of long chains of amylopectin, showed a reduced starch in vitro enzymatic hydrolysis rate.  相似文献
