Differences in aroma components and total volatiles between a single unpasteurized Marsh grapefruit juice and its 65 Brix concentrate reconstituted to 10 Brix were examined using GC-olfactometry (GC-O) and GC-FID. Total volatiles (FID) in the reconstituted concentrate were reduced to less than 5% of initial values, but 57% of total aroma (GC-O) remained. Forty-one aroma-active compounds were observed in unpasteurized single strength juice, whereas 27 components were found in the unflavored reconstituted concentrate. Aroma-active compounds were classified into grapefruit/sulfury, sweet/fruity, fresh/citrusy, green/fatty/metallic, and cooked/meaty groups. Five of six components in the sweet/fruity and 14 of 18 green/fatty/metallic components survived thermal concentration. However, only 4-mercapto-4-methyl-2-pentanone in the grapefruit/sulfury group, and linalool and nootkatone from the fresh/citrusy group, were found in the reconstituted concentrate. Methional was the only aroma compound in the cooked/meaty category found in both juice types. beta-Damascenone and 1-p-menthen-8-thiol were found only in the reconstituted concentrate. 4-Mercapto-4-methyl-2-pentanol was found for the first time in grapefruit juice.  相似文献

Fumonisins are polyketide-derived mycotoxins produced by Fusarium verticillioides, a fungal pathogen of corn plants. Although a gene cluster for the biosynthesis of fumonisins has been cloned, the biosynthetic pathway is still not clear. We have used three gene-disrupted mutants, designated DeltaFUM1, DeltaFUM6, and DeltaFUM8, to study the early steps of the pathway. Fumonisins were not produced in single-strain cultures of the DeltaFUM1, DeltaFUM6, and DeltaFUM8 mutants. However, fumonisins were produced by DeltaFUM1 or DeltaFUM8 mutants when they were cocultured with the DeltaFUM6 mutant. No fumonisins were produced when the DeltaFUM1 and DeltaFUM8 mutants were cocultured. These results suggest that the DeltaFUM6 mutant produces a fumonisin intermediate that can be further metabolized by fumonisin biosynthetic enzymes in the DeltaFUM1 and DeltaFUM8 mutants. To isolate the potential intermediates produced by DeltaFUM6, we followed a time course of cocultures of the DeltaFUM1 and DeltaFUM6 and the DeltaFUM8 and DeltaFUM6 mutants. Liquid chromatographic-mass spectrometric data suggested that metabolites having the general carbon skeleton of fumonisins with 1-4 hydroxyl groups were accumulated over a 7-day period. These results indicate that fumonisin biosynthesis starts with Fum1p-catalyzed carbon-chain assembly followed by the Fum8p-catalyzed alanine condensation. The resulting product then can be further oxidized by Fum6p and other enzymes.  相似文献

The third variable region (V3) of the HIV-1 gp120 envelope glycoprotein is immunodominant and contains features essential for coreceptor binding. We determined the structure of V3 in the context of an HIV-1 gp120 core complexed to the CD4 receptor and to the X5 antibody at 3.5 angstrom resolution. Binding of gp120 to cell-surface CD4 would position V3 so that its coreceptor-binding tip protrudes 30 angstroms from the core toward the target cell membrane. The extended nature and antibody accessibility of V3 explain its immunodominance. Together, the results provide a structural rationale for the role of V3 in HIV entry and neutralization.  相似文献

To complement the sequencing of the three kinetoplastid genomes reported in this issue, we have undertaken a whole-organism, proteomic analysis of the four life-cycle stages of Trypanosoma cruzi. Peptides mapping to 2784 proteins in 1168 protein groups from the annotated T. cruzi genome were identified across the four life-cycle stages. Protein products were identified from >1000 genes annotated as "hypothetical" in the sequenced genome, including members of a newly defined gene family annotated as mucin-associated surface proteins. The four parasite stages appear to use distinct energy sources, including histidine for stages present in the insect vectors and fatty acids by intracellular amastigotes.  相似文献
