Detection of bovine Babesia spp. and Anaplasma marginale is based on the reading of Giemsa-stained blood or organ smears, which can have low sensitivity. Our aim was to improve the detection of bovine Babesia spp. and A. marginale by validating a multiplex PCR (mPCR). We used 466 samples of blood and/or organs of animals with signs and presumptive autopsy findings of babesiosis or anaplasmosis. The primers in our mPCR amplified the rap-1a gene region of Babesia bovis and B. bigemina, and the msp-5 region of A. marginale. We used a Bayesian model with a non-informative priori distribution for the prevalence estimate and informative priori distribution for estimation of sensitivity and specificity. The sensitivity and specificity for smear detection of Babesia spp. were 68.6% and 99.1%, and for A. marginale 85.6% and 98.8%, respectively. Sensitivity and specificity for mPCR detection for Babesia spp. were 94.2% and 97.1%, and for A. marginale 95.2% and 92.7%, respectively. Our mPCR had good accuracy in detecting Babesia spp. and A. marginale, and would be a reliable test for veterinarians to choose the correct treatment for each agent. 相似文献
The carotid artery of clinically normal goats was examined, using duplex ultrasonography and arteriography. The diameter of the carotid artery was measured by use of two-dimensional ultrasonography and Doppler ultrasonography, respectively, before and after xylazine administration. The diameter of the artery was also measured by use of an arteriography technique in halothane-anesthetized goats. There was no significant difference between the mean diameter of the carotid artery measured by ultrasonography in conscious nonsedated goats and that determined by arteriography in goats under halothane anesthesia. On the other hand, ultrasonography of xylazine-sedated goats revealed an increase of carotid artery diameter of 20 to 30%. Phymastichus coffea LaSalle (Hymenoptera:Eulophidae) is an adult endoparasitoid of the coffee berry borer, Hypothenemus hampei (Ferrari) (Coleoptera:Curculionidae:Scolytinae), which has been introduced in many coffee producing countries as a biological control agent. To determine the effectiveness of P. coffea against H. hampei and environmental safety for release in Hawaii, we investigated the host selection and parasitism response of adult females to 43 different species of Coleoptera, including 23 Scolytinae (six Hypothenemus species and 17 others), and four additional Curculionidae. Non-target testing included Hawaiian endemic, exotic and beneficial coleopteran species. Using a no-choice laboratory bioassay, we demonstrated that P. coffea was only able to parasitize the target host H. hampei and four other adventive species of Hypothenemus: H. obscurus, H. seriatus, H. birmanus and H. crudiae. Hypothenemus hampei had the highest parasitism rate and shortest parasitoid development time of the five parasitized Hypothenemus spp. Parasitism and parasitoid emergence decreased with decreasing phylogenetic relatedness of the Hypothenemus spp. to H. hampei, and the most distantly related species, H. eruditus, was not parasitized. These results suggest that the risk of harmful non-target impacts is low because there are no native species of Hypothenemus in Hawaii, and P. coffea could be safely introduced for classical biological control of H. hampei in Hawaii.
High elevation lakes are extreme ecosystems and serve as sentinels of various global changes.
An expedition to Volcán Llullaillaco in 1996 discovered an unstudied high-elevation lake (6,170 m a.s.l.) that probably was formed as a result of the past eruptive events or climatic processes such as glacial retreat in the lake basin.
This article describes an initial physical characterization of the lake and its microbial communities derived from two sampling expeditions in 2013 and 2016.
The microbial community in the lake, with an area between 1.2 and 1.4 ha and a depth of 6.8 m, was dominated by Proteobacteria, Actinobacteria and Haloarchaea. In addition, 26 bacterial isolates were identified within the genera Subtercola, Xylophilus, Rhodanobacter, Mesorhizobium and Pseudomonas.
Lago Llullaillaco is one of the highest recorded lakes in the world, and this study highlights the unique microbial diversity of this aquatic ecosystem and the importance of its preservation to understand the complex biological processes under polyextreme conditions.
Cardio-facio-cutaneous (CFC) syndrome is a sporadic developmental disorder involving characteristic craniofacial features, cardiac defects, ectodermal abnormalities, and developmental delay. We demonstrate that heterogeneous de novo missense mutations in three genes within the mitogen-activated protein kinase (MAPK) pathway cause CFC syndrome. The majority of cases (18 out of 23) are caused by mutations in BRAF, a gene frequently mutated in cancer. Of the 11 mutations identified, two result in amino acid substitutions that occur in tumors, but most are unique and suggest previously unknown mechanisms of B-Raf activation. Furthermore, three of five individuals without BRAF mutations had missense mutations in either MEK1 or MEK2, downstream effectors of B-Raf. Our findings highlight the involvement of the MAPK pathway in human development and will provide a molecular diagnosis of CFC syndrome. 相似文献
Although it is recognized that anthropogenic forest fragmentation affects habitat use by organisms across multiple spatial
scales, there is uncertainty about these effects. We used a hierarchical sampling design spanning three spatial scales of
habitat variability (landscape > patch > within-patch) and generalized mixed-effect models to assess the scale-dependent responses
of bird species to fragmentation in temperate forests of southern Chile. The abundances of nine of 20 bird species were affected
by interactions across spatial scales. These interactions resulted in a limited effect of within-patch habitat structure on
the abundance of birds in landscapes with low forest cover, suggesting that suitable local habitats, such as sites with dense
understory cover or large trees, are underutilized or remain unused in highly fragmented landscapes. Habitat specialists and
cavity-nesters, such as tree-trunk foragers and tapaculos, were most likely to exhibit interactions across spatial scales.
Because providing additional sites with dense understory vegetation or large habitat trees does not compensate the negative
effect of the loss of forest area on bird species, conservation strategies should ensure the retention of native forest patches
in the mixed-use landscapes. 相似文献
Anther culture is a convenient technique to obtain androgenic haploid and doubled haploid (DH) plants. In common eggplant
(Solanum melongena), this technique has been used to develop DH pure lines for producing uniform F1 hybrid seed of some commercial varieties.
However, a comprehensive study of the variation of this useful trait among different materials of common eggplant and related
species is still lacking. In this work, we studied the androgenic response of 12 accessions of common eggplant and related
materials from the primary (eggplant complex) and secondary genepools. We cultured anthers of all the accessions under the
same experimental conditions, and studied their competence to produce calli, embryos and plants, as well as the quality and
origin of the embryos produced. In our conditions, anthers of 11 out of the 12 accessions produced somatic calli, whereas
only 5 also produced microspore-derived embryos, with variable results in terms of embryo quality and of frequency of embryo
induction and plant germination. Embryos of responding accessions were initially haploid, and reached the DH status, verified
with SSR markers, after a defined period of culture. In addition to other aspects common to many androgenesis-responsive species,
our results allowed us to extract conclusions particular to common eggplant and relatives, including the difficulty for finding
sources of androgenic competence out of S. melongena, the reduced impact of calli in the production of non-DH individuals, and the need to avoid the occurrence of severe anatomical
and functional problems in the apex of most embryos, which seriously reduces their germinative success. 相似文献
