ABSTRACT Monoconidial isolates of Venturia inaequalis were collected in 1990 and 1991 from orchards in New York, Michigan, and Nova Scotia that had never or only sporadically been treated with fungicides acting as sterol demethylation inhibitors (DMIs). Sensitivities of isolates to two representative DMIs (fenarimol and myclobutanil) were determined by a sensitivity test based on the relative growth (RG) of mycelial colonies at one discriminatory dose. Mean isolate sensitivities were not significantly different (P > 0.2) for the majority of the populations tested, and all sensitivity data obtained from these sites were combined to provide a baseline distribution of isolate sensitivities for both fenarimol and myclobutanil. The baseline distributions were compared with isolate sensitivities determined for an experimental orchard in Nova Scotia with a documented case of DMI resistance and for a commercial orchard in Michigan with a long history of DMI use and first evidence of practical DMI resistance. For both DMIs tested and in both treated orchards, frequencies of isolates with RG values <80 had decreased or only slightly increased compared to the baseline population. In contrast, frequencies of isolates with RG values >80 had increased more than 20-fold over baseline levels. Thus, isolates with RG values >80 were rated DMI resistant. The validity of a qualitative isolate classification was tested in controlled infection studies. At doses of fenarimol and myclobutanil recommended for commercial control of apple scab, reproduction of a typical sensitive isolate on treated apple leaves was suppressed completely. Lesions caused by a resistant isolate continued to expand and produced abundant conidia. Statistical analysis of orchard sensitivities revealed that the analysis of isolate counts grouped into the categories DMI sensitive or resistant was most indicative in comparisons of orchard sensitivities aimed at detection of practical DMI resistance. A high degree of cross-resistance between fenarimol and myclobutanil indicated that sensitivity tests with one of the DMIs employed as the diagnostic tool in this study can serve as a test for other DMIs. 相似文献

ABSTRACT Several wheat lines and cultivars of wheat (Triticum aestivum) originating from the southwestern region of China were found to be highly resistant to stripe rust by inoculation with the prevalent races (CYR30, CYR31, and CYR32) and newly emerged races (H46-4, SY11-4 and SY11-14) of the pathogen. An inheritance study of the resistance to stripe rust was carried out by crossing resistant AIM6 with susceptible BeiZ76. Results indicated that the resistance to stripe rust was controlled by a single dominant gene. The 112 F(2) plants chosen from the cross BeiZ76/ AIM6 were analyzed with 218 pairs of microsatellite primers to determine the map location of the resistance gene. A simple sequence repeat marker on chromosome arm 2BS, Xgwm410, showed polymorphism and co-segregation between stripe rust resistant and susceptible plants. From the pedigree, inheritance, molecular marker, and resistance response, it is concluded that the stripe rust resistance gene in wheat cv. Chuan-nong19 (CN19) and wheat lines AIM5 and AIM6 is a novel gene, designated YrCN19. The microsatellite primer Xgwm410 is a diagnostic marker of the resistance gene YrCN19, which has potential for application in the marker-assisted breeding of wheat. 相似文献

Soil adjacent to new brick veneer work is likely to have a higher pH owing to the mixture of cement with the soil. In the Gainesville, FL, area, soil samples taken from such locations had a range of pH values from 9.0 to 10.1; similar soils used in bioassays had a pH of 5.6 before the addition of cement. Addition of 15 mg of Portland cement to 33 g of soil increased the pH to 6, and addition of 291 mg of Portland cement increased the pH to 9. The pH of soil amended with cement was stable for the first 5 months. After 10 months, soil pH values decreased from alkaline to near neutral in all cases. Eastern subterranean termite workers, Reticulitermes flavipes (Kollar), were exposed to the treated soil at pH 6-9 for 24 h, and percentage mortality was recorded at 5 days, 5 months and 10 months. Termite mortality significantly decreased at higher soil pHs for bifenthrin, chlorpyrifos, fipronil and imidacloprid treatments at 5 months and similarly for bifenthrin, permethrin, chlorpyrifos, fipronil and imidacloprid treatments at 10 months. There was an inverse linear relationship between soil pH and mortality. Our previous study had demonstrated cytotoxicity of EtOAc fraction from aerial part of Cimicifuga foetida on various cancer cell lines. In the present study, we investigated the cytotoxicity of five cycloartane triterpenoids isolated from the EtOAc fraction. Compounds (1,2,3) showed moderate cytotoxic activity on R-HepG2 cells and drug resistant sub line R-HepG2 but exhibited less toxicity against primary cultured normal mouse hepatocytes. Refinement of an orthotopic lung cancer model in the nude rat 总被引:5,自引:0,他引:5
Over 85% of people with lung cancer eventually succumb to this disease, largely because current chemotherapies are ineffective. The testing and validation of promising new approaches generally rely on achieving responses with cell lines in vitro or in tumor xenografts in nude mice. However, quite often the results seen with these models are not recapitulated in the clinic, thus necessitating the need for better animal models of lung cancer for preclinical testing of new therapies. One promising model is that of orthotopic lung cancer, where xenografts of human lung cancer are established in lungs of immunodeficient rodents. The problems associated with this model include poor rates of engraftment, limited tumor multiplicity, and a heightened risk for surgical trauma. The purpose of our study was to develop an efficient approach to engraftment of orthotopic tumors throughout the lungs of the Rowett nude rat. Initially, we augmented immunosuppression in the rats with whole-body X-irradiation and then used orotracheal cannulas to intratracheally instill human cancer cells from the Calu-6 cell line. This protocol produced a low rate of engraftment and low tumor multiplicity. The hypothesis that slight disruption of the pulmonary epithelium or the surfactant layer would allow better tumor engraftment was tested by coadministration of either pancreatic elastase or ethylenediaminetetraacetic acid (EDTA) along with the cell instillations. Lung tumor engraftment was evaluated 8 weeks after instillation. The inclusion of elastase or EDTA with the Calu-6 cells resulted in an 80-100% engraftment rate, respectively. Coadministration of EDTA resulted in significantly larger and greater numbers of tumors/lung than those in elastase-treated animals. Temporal studies demonstrated that small nodules were scattered throughout the lung parenchyma 5 weeks after instilling Calu-6 cells and EDTA. These nodules grew to coalesce and form large masses that effaced >75% of the parenchyma at 9 weeks postinstillation. The refinements made through our studies have led to the development of an orthotopic lung cancer model that should facilitate the evaluation of novel therapies designed to treat or impede lung cancer development. 相似文献

The significant decrease in fertility observed in dairy cattle during the last few decades and increasing requirements by the farmers have made a regular control of reproduction indispensable to urgently identify and solve potential problems affecting reproductive efficiency. Traditionally, the main diagnostic methods used for reproductive control in cattle included rectal palpation, inspection of vaginal discharge and vaginoscopy. Since the 1990 s, the use of ultrasound (US) has become a common diagnostic method as a result of the new advances made in the development of US scans: smaller size, high level of autonomy, high image quality and accessible prices. Ultrasound improves accuracy in the diagnoses of stages of the oestrous cycle, ovarian and uterine pathologies, and pregnancy diagnosis. In addition, it facilitates the diagnosis of alterations during pregnancy (embryo mortality, foetal malformations, etc.) and helps determining foetal sex from day 55 of pregnancy. 相似文献
