The present study developed methods for quantifying foliar Zn uptake from suspension‐based Zn chemicals of low solubility, which were ZnO (particle size: 0.151.34 μm) and a newly synthesized Zn hydroxide nitrate crystal (Zn‐HNC) (50100 nm thickness and 0.21 μm lateral dimension). Recently matured leaves of citrus (Citrus aurantium L. cv. Valencia), capsicum (Capsicum annume L. cv. Giant Bell), and/or tomato (Solanum lycopersicum L. cv. Roma) were in vitro–treated with microdroplets (5 μL per droplet) of Zn‐HNC‐ and ZnO‐suspension solutions on the adaxial surface and incubated under controlled conditions for up to 72 h. Leaf‐washing protocols were compared, including: dilute ethanol (3%), dilute nitric acid (2%), and their combination. The methods for quantifying Zn uptake were: (1) whole‐leaf loading by which droplets of the Zn suspension solutions were loaded onto central regions of both left and right sides of leaf blades and (2) half‐leaf loading by which soluble‐Zn (ZnSO4) droplets were loaded onto only one side of leaf blades while the other was used as the background Zn control. Foliar‐surface characteristics of the plant species affected the effectiveness of the washing methods. The dilute nitric acid (2%; ± 3% ethanol) was required to remove residue particles of the ZnO and Zn‐HNC suspensions from foliar surfaces of capsicum and tomato (highly trichomatic), but the residue Zn chemicals on citrus leaves (nontrichomatic and highly waxy) were similarly and effectively removed by the three washing methods. For quantifying Zn uptake by the leaves, the half‐leaf loading method showed its advantages over the whole‐leaf loading method, because it did not stringently require similar background Zn concentrations in the control and treated leaves at the start and had little risks of secondary absorption of soluble Zn in the washing solution.  相似文献

The aquatic system of Hochiminh City comprises two main rivers: the Sai Gon and Nha Be rivers. Five canals discharge into these two rivers: NhieuLoc-ThiNghe, TauHu-BenNghe, TanHoa-LoGom, ThamLuong-BenCat and Doi-Te. The rivers and these canals collect effluent water from domestic and industrial sources. Most of these flows are not treated or at most are only primarily treated. A total of 33 sediment cores were taken from these rivers and canals. Chemical composition of these aquatic sediments has very high concentrations of several “urban” metals such as Cd, Cr, Cu and Zn. Most of the samples have exceeded the US EPA’s toxicity reference values for Cu, Zn and Cr (82, 82 and 70%, respectively). The highest concentrations of these metals appear to be associated with the uncontrolled and untreated industrial runoff to the discharge canals. These concentrations in fluvial sediment are relatively low, which indicates the dilution process of the contaminants. This finding indicates that the anthropogenic inputs play an important role in the elevation of heavy metals in the aquatic system and organic matter seems to exert a strong geochemical control on the amount of heavy metals. The Pearson correlation coefficients calculated for Cd, Cr, Cu and Zn, are 0.89; 0.72; 0.93 and 0.87, respectively.  相似文献

Adipose tissue gene expression in obese dogs after weight loss   总被引:1,自引:0,他引:1
Body weight (BW) mainly depends on a balance between fat storage (lipogenesis) and fat mobilization (lipolysis) in adipocytes. BW changes play a role in insulin resistance (IR), the inability of insulin target tissue to respond to physiological levels of insulin. This results in inhibition of lipogenesis and stimulation of lipolysis. Weight gain leads to IR whereas, weight loss improves insulin sensitivity (IS). The aim of this study was to evaluate the effect of weight loss and recovery of IS on the expression of genes involved in lipogenesis and lipolysis in weight losing dogs. Gene expression was studied in both subcutaneous and visceral adipose tissue. Obese dogs received a hypoenergetic low fat high protein diet (0.6 x NRC recommendation). Before and after weight loss, IS was assessed using the euglycaemic hyperinsulinaemic clamp. Gene expression of IRS-2, SREBP, intracellular insulin effectors, ACC, FAS, FABP, ADRP, PEPCK, lipogenesis key proteins, perilipin and HSL, lipolysis key proteins were quantified using real-time RT-PCR in subcutaneous and visceral fat. BW decreased from 15.2 +/- 0.5 to 11.4 +/- 0.4 kg (p < 0.05) over 78 +/- 8 days. When obese, dogs were insulin resistant. After weight loss, IS was improved. In the subcutaneous adipose tissue, the expression of only the IRS-2 was increased. In the visceral adipose tissue, the expression of the genes involved in the lipogenesis was decreased whereas one of the genes implied in the lipolysis did not change. The expression profile of genes involved in lipid metabolism, as measured after weight loss, is indicative for a lower lipogenesis after weight loss than in obese dogs. Our results also confirm dramatic differences in the lipid metabolism of visceral and subcutaneous fat. They should be completed by comparing gene expression during weight losing and normal weight steady state.  相似文献

  • ? The aim of this study was to investigate the effectiveness of using cellulose content, measured by the diglyme-HCl method, as a selection trait in breeding programs for kraft pulp yield in Eucalyptus urophylla.
  • ? A total of 275 trees from sixty-two families were sampled from a thinned progeny trial of E. urophylla in northern Vietnam to evaluate cellulose content from breast-height increment cores. Among those, twenty unrelated trees were felled to evaluate cellulose content and pulp yield from breast-height disk samples.
  • ? The regression of pulp yield of disk samples on cellulose content was strong either from disks (R 2 = 0.83) or increment cores (R 2 = 0.69). There was no significant difference in cellulose content between the provenances. The narrow-sense within-provenance heritability of cellulose content was 0.50 and the coefficient of additive genetic variation was 3.9%. Genetic correlations between cellulose content and growth (0.28–0.45) or wood basic density (?0.02) were not significantly different from zero.
  • ? Breast-height increment core cellulose content measured by diglyme-HCl method is under strong genetic control and can be used to rank trees for pulp yield in E. urophylla plantations. Selection for increased cellulose content would have only minor effects on growth and wood basic density.

Dispersion is an important issue for clay leaching in soils. In paddy soils of the Red River Delta (RRD), flooding with fresh water and relatively high leaching rates can accelerate dispersion and the translocation of clay. For the clay fraction of the puddled horizon of a typical paddy soil of the RRD, the effect of various cations and anions as well as humic acid (HA) at different pH values on the surface charge (SC) were quantified and the dispersion properties were determined in test tubes and described by the C50 value. In the <2 µm fraction, dominated by illite, the proportion of 2:1 vs. 1:1 clay minerals is 5:1. The organic‐C content of the clay fraction is 2.2%. Surface charge was found to be highly pH‐dependent. At pH 8 values of –32 and at pH 1 of –8 mmolc kg–1 were obtained. Complete dispersion was observed at pH > 4, where SC is > –18 mmolc kg–1. The flocculation efficiency of Ca strongly depends on the pH. At pH 4, the C50 value is 0.33, 0.66 at pH 5, and 0.90 mmol L–1 at pH 6. At pH 6, close to realistic conditions of paddy soils, the effect of divalent cations on the SC and flocculation decreases in the order: Pb > Cu > Cd > FeII > Zn > Ca > MnII > Mg; FeII was found to have a slightly stronger effect on flocculation than Ca. An increase in concentrations of Ca, MnII, and Mg from 0 to 1 mmol L–1 resulted in a change in SC from –25 to approx. –15 mmolc kg–1. In comparison, the divalent heavy‐metal cations Pb, Cu, Cd, and Zn were found to neutralize the SC more effectively. At a Pb concentration of 1 mmol L–1, the SC is –2 mmolc kg–1. From pH 3 to 5, the dispersion of the clay fraction is facilitated rather by SO than by Cl, which can be explained by the higher affinity of SO to the positively charged sites. With an increase of the amount of HA added, the SC continuously shifts to more negative values, and higher concentrations of cations are needed for flocculation. At pH 3, where flocculation is usually observed, the presence of HA at a concentration of 40 mg L–1 resulted in a dispersion of the clay fraction. While high anion concentrations and the presence of HA counteract flocculation by making the SC more negative, FeII and Ca (C50 at pH 6 = 0.8 and 0.9 mmol L–1, respectively) are the main factors for the flocculation of the clay fraction. For FeII and Ca, the most common cations in soil solution, the C50 values were found to be relatively close together at pH 4, 5, and 6, respectively. Depending on the specific mineralogical composition of the clay fraction, SC is a suitable measure for the determination of dispersion properties and for the development of methods to keep clay particles in the soil in the flocculated state.  相似文献

Silver nanoparticles (AgNPs) have significantly increased in production and use for anti-microbial propose. This agent, after used, is released into sewerage system resulting in possibility to inactivate non-targeted microorganisms in wastewater treatment plants. In this study, the inhibitory effect of AgNPs on ammonia oxidation was investigated using respirometric assay. The initial concentrations of AgNPs and ammonia ranged 0.25?C10.00 and 14?C280?mg/L, respectively. Half saturation constant (K s) for ammonia oxidation was found to be 15.9?mg?N/L. Under the presence of AgNPs, the maximum oxygen uptake rate and K s declined. The effect of AgNPs was proved to follow an uncompetitive-like inhibition kinetic type with the inhibition coefficients (K i) of 5.5?mg/L. Increasing AgNPs from 0.25 to 10.00?mg/L inhibited 4 to 50?% of ammonia-oxidizing activities at the initial ammonia concentrations from 14 to 280?mg/L. Based on transmission electron microscopic observation, AgNPs could damage the microbial cells. All findings indicated that AgNPs substantially reduced ammonia-oxidizing microorganisms and their activities. Thus, special attention should be made to manage discharge of AgNPs into the environment.  相似文献

Background, Aim and Scope   Coastal and river plains are the surfaces of depositional systems, to which sediment input is a parameter of key-importance. Their habitation and economic development usually requires protection with dikes, quays, etc., which are effective in retaining floods but have the side effect of retarding sedimentation in their hinterlands. The flood-protected Dutch lowlands (so-called dike-ring areas) have been sediment-starved for up to about a millennium. In addition to this, peat decomposition and soil compaction, brought about by land drainage, have caused significant land subsidence. Sediment deficiency, defined as the combined effect of sediment-starvation and drainage-induced volume losses, has already been substantial in this area, and it is expected to become urgent in view of the forecasted effects of climate change (sea-level rise, intensified precipitation and run-off). We therefore explore this deficiency, compare it with natural (Holocene) and current human sediment inputs, and discuss it in terms of long-term land-use options. Materials and Methods: We use available 3D geological models to define natural sediment inputs to our study area. Recent progress in large-scale modelling of peat oxidation and compaction enables us to address volume loss associated with these processes. Human sediment inputs are based on published minerals statistics. All results are given as first-order approximations. Results: The current sediment deficit in the diked lowlands of the Netherlands is estimated at 136 ± 67 million m3/a. About 85% of this volume is the hypothetical amount of sediment required to keep up with sea-level rise, and 15% is the effect of land drainage (peat decomposition and compaction). The average Holocene sediment input to our study area (based on a total of 145 km3) is ~14 million m3/a, and the maximum (millennium-averaged) input ~26 million m3/a. Historical sediment deficiency has resulted in an unused sediment accommodation space of about 13.3 km3. Net human input of sediment material currently amounts to ~23 million m3/a. Discussion: As sedimentary processes in the Dutch lowlands have been retarded, the depositional system's natural resilience to sea-level rise is low, and all that is left to cope is human countermeasure. Preserving some sort of status quo with water management solutions may reach its limits in the foreseeable future. The most viable long-term solutions therefore seem a combination of allowing for more water in open country (anything from flood-buffer zones to open water) and raising lands that are to be built up (enabling their lasting protection). As to the latter, doubling or tripling the use of filling sand in a planned and sustained effort may resolve up to one half of the Dutch sediment deficiency problems in about a century. Conclusions: Conclusions, Recommendations and Perspectives. We conclude that sediment deficiency – past, present and future – challenges the sustainable habitation of the Dutch lowlands. In order to explore possible solutions, we recommend the development of long-term scenarios for the changing lowland physiography, that include the effects of Global Change, compensation measures, costs and benefits, and the implications for long-term land-use options.
