We have investigated the impact of shrimp farming waste on benthic invertebrates in a mangrove estuary in southern Thailand.
Stable carbon and nitrogen isotope analyses were used to assess the contribution of aquaculture feed as a nutrient source
for benthic invertebrates (fiddler crabs Uca annulipes, U. bengali, and U. forcipata) and to compare the organic origin of sediments in tributaries with and without a shrimp farm. The isotopic contributions
of shrimp feed to crabs varied between the tributaries: in the tributary with the farm, the crabs had higher contributions
from shrimp feed, whereas there was only a small contribution from aquaculture feed in crabs from the reference tributary.
In contrast, the contribution of shrimp feed to the sediment did not differ between the tributaries, the contributions to
sediments from aquaculture feed were, in general, quite low. Based on these results, we suggest that, in mangrove estuaries,
changes in the food sources of several surface feeder invertebrates would be an effective indicator of the possibility that
aquaculture waste was affecting the benthic ecosystem. 相似文献

A dihydropteroate synthase gene from the chromosomal DNA of the fish-pathogenic bacteria Lactococcus garvieae (formerly Enterococcus seriolicida ) was cloned. This gene was then chosen as the target for polymerase chain reaction (PCR). The designated PCR primer set only amplified a 709-bp DNA fragment from L. garvieae strains, and did not amplify the same molecular size fragment from related species of L. lactis, Enterococcus faecalis, E. faecium or β-haemolytic Streptococcus sp. The kidney tissue of yellowtail, Seriola quinqueradiata (Temminck and Schlegel), a species which is naturally infected with L. garvieae , and also kidney tissue samples of healthy yellowtail were stored in TNES-Urea. The DNA was extracted from tissue samples by a modification of the standard method and by a boiled-extraction method. In particular, template DNA was utilized within 30 min following extraction and purification by the boiled-extraction method. These species-specific PCR primers could amplify a L. garvieae target sequence from yellowtail which were naturally infected with L. garvieae . The total procedure for the diagnosis of L. garvieae infections in fish, from the point of DNA extraction to observation in an agarose gel following electrophoresis, can be performed in less than 4 h. 相似文献

A phospholipase gene of Photobacterium damselae ssp. piscicida (ppp) was cloned from a genomic library and its nucleotide sequence was determined. The open reading frame consisted of 1218 bp encoding a protein of 405 amino acids with a predicted molecular mass of 46 kDa. The PPP had identities (53-55%) with phospholipase and haemolysin of Vibrio spp., while it showed low identities (23-26%) with glycerophospholipid cholesterol acyltransferase of Aeromonas spp. A recombinant PPP (rPPP) with a His tag at the C-terminus expressed in Escherichia coli and purified showed phospholipase activity. The rPPP also showed lecithin-dependent haemolytic activity against mammalian erythrocytes and direct haemolytic activity against fish erythrocytes. The culture supernatant of wild-type P. damselae ssp. piscicida showed phospholipase activity, while that of a PPP gene knockout mutant did not. 相似文献

ABSTRACT: Restriction fragment length polymorphism analysis was performed on polymerase chain reaction-amplified DNA fragments containing the D-loop , ND2 , and CO I genes of fiddler crab Uca arcuata mitochondrial DNA. In total, 316 individuals from six populations in Japan and two populations in Taiwan were analyzed using five restriction endonucleases ( Afa I, Bcn I, Cfr 13I, Hae III and Hin fI), yielding 85 haplotypes. Samples were taken from Nakagusuku Bay, Okinawajima Island, which is the only known distribution of U. arcuata in the Ryukyu Archipelago. The Okinawajima Island population is isolated geographically from others and showed a marked low genetic variability ( h = 0.2539, π = 0.0005) and significant differentiation from other population samples in haplotype composition. We suggest that a substantial decrease in the genetic variability of the Okinawajima Island population was caused by genetic drift under the conditions of small population size and low gene flow from other populations. It is important to conserve the intertidal zone in Nakagusuku Bay for the maintenance of this endangered population. 相似文献

Various lines of evidence suggest that appetite‐related neuropeptides in the hypothalamus are regulated by adiposity signals such as leptin and insulin in mammals. In the present study, we examined age‐dependent changes in the weight of abdominal fat and hypothalamic mRNA levels of neuropeptide Y (NPY, an orexigenic neuropeptide) and proopiomelanocortin (POMC, a precursor of anorexigenic neuropeptides) in growing chickens at 7, 14, 21 and 28 days of age. Hypothalamic NPY mRNA levels were significantly (P < 0.05) decreased after 14 days of age, whereas hypothalamic POMC mRNA levels were significantly (P < 0.05) increased at 28 days of age. The percentage of abdominal fat was significantly increased after 14 days of age in chickens. We next examined the correlation of hypothalamic NPY and POMC mRNA levels and several parameters at 28 days of age. There were no significant correlations between hypothalamic mRNA levels of NPY or POMC and the percentage of abdominal fat. These findings suggest that the gene expressions of NPY and POMC do not depend on adiposity in chickens, at least in 28‐day‐old layer chickens. 相似文献
