水分状况对红壤水稻土中有机物料碳分解和分布的影响 总被引:2,自引:0,他引:2
以室内模拟的方法研究了好气、淹水和干湿交替3种水分条件下有机物料碳在红壤水稻土中分解和分布的差异。试验结果表明,干湿交替条件下有机物料的分解速率最高,好气条件下次之,淹水条件下较低。好气和淹水条件下添加物料促进土壤原有有机碳的矿化,产生正激发效应,而干湿交替条件显著抑制了土壤原有有机碳的矿化,呈负激发效应,随着培养时间的延长激发效应减弱。干湿交替条件下添加有机物料处理的土壤胡敏酸(Humic acid,HA)色调系数和E4/E6比值显著低于淹水和好气条件,淹水培养使土壤胡敏酸的结构简单化,干湿交替使胡敏酸芳构化和腐殖化程度增加,结构复杂化。有机物料碳在土壤腐殖质组分中的分配比例显示,干湿交替和好气条件促进了胡敏酸的形成,提高了土壤原有富里酸(Fulvic acid,FA)组分的转化和胡敏酸组分的分解;淹水条件下物料碳在富里酸组分中分布比例较高,且抑制了土壤原有胡敏酸组分的分解。有机物料碳在0.053 mm粒级团聚体中分布比例较大,干湿交替和淹水条件下更为明显。 相似文献

为探讨茭白冷藏期间衰老的分子机理,应用蛋白质组学技术,研究了茭白冷藏期间蛋白质表达谱的变化。结果显示,双向电泳胶上共检测到大约650个蛋白点,其中35个蛋白表达量存在2.0倍以上显著(p0.05)差异,经过串联飞行时间质谱分析,成功鉴定出29个蛋白,根据其功能可分为6类,即代谢(20.7%)、细胞结构(27.6%)、抗胁迫(20.7%)、衰老(6.9%)、蛋白质合成(13.8%)和功能未知蛋白(10.3%);其中:代谢相关蛋白3个上调表达、3个下调表达,细胞结构相关蛋白6个上调表达、2个下调表达,抗胁迫相关蛋白4个上调表达、2个下调表达,衰老相关蛋白2个上调表达,蛋白质合成相关蛋白4个及功能未知蛋白3个均下调表达。这些差异表达蛋白的功能分析表明,茭白采后衰老机理可能涉及物质代谢过程的调整、能量代谢途径的改变、活性氧清除能力的减弱以及细胞结构的解体。 相似文献

Correlation analysis of mineral element contents and quality traits in milled rice (Oryza stavia L.)

The relationships among potassium (K), calcium (Ca), sodium (Na), magnesium (Mg), iron (Fe), zinc (Zn), copper (Cu), and manganese (Mn) contents in milled rice (Oryza stavia L.) of 274 genotypes and the relationships between these mineral element contents and other rice quality traits including 3 cooking quality traits, 17 amino acid contents, and protein content were investigated. The results showed that there were significant correlations among most of mineral element contents. Mg, Fe, and Mn contents were significantly correlated with most of the other mineral element contents, while Cu content had significantly negative associations with the K and Mg contents of rice. The relationships between mineral element contents and cooking quality traits showed that gel consistency (GC) was significantly correlated with K, Cu, and Mn contents of rice. Amylose content (AC) was significantly associated with K, Na, Mg, Cu, and Mn contents. The alkali spreading value (ASV) had closely positive relationships with Ca, Mg, and Mn contents. In addition, 8 mineral element contents had obvious correlations with different amino acid contents. Mg, Ca, and Zn contents were significantly correlated with most of the 17 amino acid contents, but Na content did not correlate with amino acid contents except aspartic acid of rice. Furthermore, significant associations were found between protein content and Na, Mg, Zn, Cu, or Mn content. Six principal components were extracted to explain 84.50% of the total variances and contained the information provided by the original 29 variables according to the principal component analysis. 相似文献

A capillary electrophoresis-inductively coupled plasma mass spectrometric (CE-ICP-MS) method for the determination of cobalamin is described. Samples of cobalt-containing compounds were subjected to electrophoretic separation before injection into the microconcentric nebulizer (CEI-100) of ICP-MS. The Co-containing compounds studied include cyanocobalamin (CN-Cbl, vitamin B12), hydroxocobalamin (OH-Cbl), and Co(II). The species studied were well separated using a 70 cm lengthx75 microm i.d. fused silica capillary with the applied voltage set at +20 kV. A 25 mM Tris buffer (pH 9.0) containing 15 mM sodium dodecyl sulfate (SDS) was used as the electrophoretic buffer. The CE-ICP-MS detection limits were 0.3, 0.2, and 1.7 ng of Co mL(-1) for CN-Cbl, OH-Cbl, and Co(II), respectively. The concentrations of cobalamin compounds were determined in selected nutritive supplements and chlorella samples. A microwave-assisted extraction method was used for the extraction of these compounds. Over 92% of the total cobalt species was extracted using a 5% v/v HNO3 solution in a focused microwave field within a period of 10 min; spike recovery was in the range of 94-105% for various species. Although phytosterols in tobacco leaves have specific effects on tobacco quality, there is little research on the distribution of free and conjugated phytosterols in various tobacco leaves. In this paper, we explored the content and composition of phytosterols in representative cured tobacco leaves by gas chromatography of TMS-ether derivatives. We found that phytosterol contents in tobacco leaves ranged from 1.0 to 2.5 mg/g of dried leaf tissue, depending on different types of tobacco leaves. The majority of phytosterols (75-85%) were conjugated as ester and glycosides, with only about 15-25% existing in the free form. Furthermore, the genetic variability gives rise to the significant differences among different tobacco types with phytosterol levels: the contents of phytosterols in tobacco leaves decreased in the order of flue-cured tobacco, Oriental tobacco, Burley tobacco, cigar tobacco, and Maryland tobacco. The purification and characterization of a novel extracellular beta-glucosidase from Paecilomyces thermophila J18 was studied. The beta-glucosidase was purified to 105-fold apparent homogeneity with a recovery yield of 21.7% by DEAE 52 and Sephacryl S-200 chromatographies. Its molecular masses were 116 and 197 kDa when detected by SDS-PAGE and gel filtration, respectively. It was a homodimeric glycoprotein with a carbohydrate content of 82.3%. The purified enzyme exhibited an optimal activity at 75 degrees C and pH 6.2. It was stable up to 65 degrees C and in the pH range of 5.0-8.5. The enzyme exhibited a broad substrate specificity and significantly hydrolyzed p-nitrophenyl-beta- d-glucopyranoside ( pNPG), cellobiose, gentiobiose, sophorose, amygdalin, salicin, daidzin, and genistin. Moreover, it displayed substantial activity on beta-glucans such as laminarin and lichenan, indicating that the enzyme has some exoglucanase activity. The rate of glucose released by the purified enzyme from cellooligosaccharides with a degree of polymerization (DP) ranging between 2 and 5 decreased with increasing chain length. Glucose and glucono-delta-lactone inhibited the beta-glucosidase competitively with Ki values of 73 and 0.49 mM, respectively. The beta-glucosidase hydrolyzed pNPG, cellobiose, gentiobiose, sophorose, salicin, and amygdalin, exhibiting apparent Km values of 0.26, 0.65, 0.77, 1.06, 1.39, and 1.45 mM, respectively. Besides, the enzyme showed transglycosylation activity, producing oligosaccharides with higher DP than the substrates when cellooligosaccharides were hydrolyzed.
