ABSTRACT Transgenic tobacco plants (Tetr) expressing the mutant etr1-1 gene from Arabidopsis thaliana are insensitive to ethylene and develop symptoms of wilting and stem rot when grown in nonautoclaved soil. Several isolates of Fusarium, Thielaviopsis, and Pythium were recovered from stems of diseased Tetr plants. Inoculation with each of these isolates of 6-week-old plants growing in autoclaved soil caused disease in Tetr plants but not in nontransformed plants. Also, when 2-week-old seedlings were used, nontransformed tobacco appeared nonsusceptible to the Fusarium isolates, whereas Tetr seedlings did develop disease. Tetr seedlings were not susceptible to several nonhost Fusarium isolates. In contrast to results with Fusarium isolates, inoculation of 2-week-old seedlings with a Thielaviopsis isolate resulted in equal symptom development of nontransformed and Tetr tobacco. In order to explore the potential range of pathogens to which Tetr tobacco plants display enhanced susceptibility, the pathogenicity of several root and leaf pathogens was tested. Tetr plants were more susceptible to the necrotrophic fungi Botrytis cinerea and Cercospora nicotianae and the bacterium Erwinia carotovora, but only marginally more to the bacterium Ralstonia solanacearum. In contrast, the biotrophic fungus Oidium neolycopersici, the oomycete Peronospora tabacina, and Tobacco mosaic virus caused similar or less severe symptoms on Tetr plants than on nontransformed plants. Total peroxidase activity of Tetr plants was lower than that of nontransformed plants, suggesting a role for peroxidases in resistance against necrotrophic microorganisms. A comparable range of pathogens was examined on Arabidopsis and its ethylene-insensitive mutants etr1-1 and ein2-1. With the exception of one Fusarium isolate, ethylene insensitivity increased susceptibility of Arabidopsis plants to a similar spectrum of necrotizing pathogens as in tobacco. Thus, both ethylene-insensitive tobacco and Arabidopsis plants appear to be impaired in their resistance to necrotrophic pathogens.  相似文献

OBJECTIVE: To evaluate therapeutic efficacy of a high extralabel dose of ceftiofur for treatment of experimental salmonellosis in neonatal calves. ANIMALS: Forty-two 1- to 4-day-old Holstein bull calves. PROCEDURE: 36 calves were orally challenged with Salmonella enteritica serovar Typhimurium (6.5 x 10(8) colony-forming units). Six additional calves were retained as nonmedicated nonchallenged control calves. Four days following Salmonella challenge, surviving calves were randomly allocated to ceftiofur-treated (5 mg/kg, IM, q 24 h) or nonmedicated control groups. Calves assigned to the treated group were medicated daily for 5 days starting on day 4 after challenge. Calves were monitored for 18 days following Salmonella challenge. Outcome assessments included clinical parameters (attitude, appetite, fecal characteristics, and rectal temperature), mortality rate, and quantitative Salmonella culture of fecal samples, mesenteric lymph nodes, and cecal contents. RESULTS: Ceftiofur treatment was associated with a significant decrease in rectal temperature and diarrhea. Three of 15 medicated calves and 4 of 14 non-medicated calves died or were euthanatized between days 4 and 18. A significant decrease in fecal shedding of Salmonella organisms was observed in treated calves, compared with nonmedicated calves. Salmonella organisms were isolated from all 10 non-medicated calves at necropsy, whereas no Salmonella organisms were isolated from 5 of 12 medicated calves. CONCLUSIONS AND CLINICAL RELEVANCE: Treatment of salmonellosis in neonatal calves with a high extralabel dose of ceftiofur (5 mg/kg, IM, q 24 h) promotes animal welfare, reduces fecal shedding of Salmonella organisms, and may promote clearance of Salmonella infections when plasma ceftiofur concentrations are maintained above minimal inhibitory concentrations.  相似文献

The spider very high density lipoprotein (VHDL), which contains hemocyanin as the major apoprotein, transports most of the circulating lipids. In this work, the effect of the pesticide fenitrothion (FS) on the ability of VHDL-apoproteins to uptake different lipids was investigated. For this, VHDL was delipidated using Triton X-100 and recombined with different radiolabeled lipids in the presence or the absence of FS. The oligomeric structural integrity was maintained after delipidation as shown by non-denaturating PAGE. In the presence of the insecticide, palmitic acid uptake decreased by 28.2 and 62.4% after treating the apolipoprotein with 10 and 20 ppm FS, respectively. Decreases in the uptake of cholesterol, triolein, and phosphatidylcholine caused by FS were 29, 23, and 31% using 10 ppm, and 40, 44, and 29% using 20 ppm FS, respectively. Fluorescence measurements with the hydrophobic probes diphenylhexatriene (DPH) and diphenylhexatrienyl-propionic acid (DPH-PA) indicate that FS induces a red shift, decreases the intensity and increases the anisotropy of the emission of these probes in the VHDL. These results indicate that insecticide binding to the lipoprotein enhances the environment polarity and restricts the mobility of these probes at their binding site. These changes at the hydrophobic VHDL binding sites could lead to the decreased affinity for lipids and hydrophobic ligands. It is inferred that FS could alter the normal lipid exchange between this lipoprotein and tissues.  相似文献

A probabilistic spatial model was created based on empirical data to examine the influence of different fire regimes on stand structure of lodgepole pine (Pinus contorta var. latifolia) forests across a >500,000-ha landscape in Yellowstone National Park, Wyoming, USA. We asked how variation in the frequency of large fire events affects (1) the mean and annual variability of age and tree density (defined by postfire sapling density and subsequent stand density) of lodgepole pine stands and (2) the spatial pattern of stand age and density across the landscape. The model incorporates spatial and temporal variation in fire and serotiny in predicting postfire sapling densities of lodgepole pine. Empirical self-thinning and in-filling curves alter initital postfire sapling densities over decades to centuries. In response to a six-fold increase in the probability of large fires (0.003 to 0.018 year−1), mean stand age declined from 291 to 121 years. Mean stand density did not increase appreciably at high elevations (1,029 to 1,249 stems ha−1) where serotiny was low and postfire sapling density was relatively low (1,252 to 2,203 stems ha−1). At low elevations, where prefire serotiny and postfire lodgepole pine density are high, mean stand densities increased from 2,807 to 7,664 stems ha−1. Spatially, the patterns of stand age became more simplified across the landscape, yet patterns of stand density became more complex. In response to more frequent stand replacing fires, very high annual variability in postfire sapling density is expected, with higher means and greater variation in stand density across lodgepole pine landscapes, especially in the few decades following large fires.  相似文献

OBJECTIVE: To provide an epidemiologic investigation of the seroprevalence of antibodies to Toxoplasma gondii in populations of cats and wild rodents in Rhode Island and to address the possible epidemiologic role of wild rodents in the spread of toxoplasmosis. ANIMALS: 200 cats and 756 small wild rodents. PROCEDURE: Serum samples were obtained from 84 cats in animal shelters and 116 cats in veterinary hospitals. Serum samples were also obtained from 756 small wild rodents from multiple sites in Rhode Island. Sera from rodents and cats were assayed for antibodies to Tgondii by use of the modified agglutination test RESULTS: Overall, 42% (84/200) of cats had serum antibodies to Tgondii. Seroprevalence was not significantly different between stray (50%; 42/84) versus client-owned (36%; 42/116) cats, between male (43%; 40/94) versus female (42%; 39/93) cats, or between indoor (26%; 7/27) versus outdoor (39%; 35/89) cats. Seroprevalence rate of trapped rodents was 0.8% (6/756). Six rodents captured in Washington County accounted for of the seropositive rodents. Four of 6 of the seropositive rodents were trapped at a single site in Washington County (an abandoned barn). Five stray cats, known to have resided at the same site in Washington County as 4 of the seropositive rodents, were also found to be seropositive for antibodies to T gondii. CONCLUSIONS AND CLINICAL RELEVANCE: Seroprevalence rate in rodents was not correlated with the seroprevalence rate in cats. Stray cats, especially those known to be feral, may be more likely to perpetuate the cat-mouse cycle of T gondii than client-owned cats.  相似文献

Postharvest biological treatment entails a range of different approaches, including strengthening of the commodity’s natural defense mechanisms and application of antagonistic microorganisms and natural antimicrobial substances. Postharvest biological treatment has highlighted the potential of antagonistic microorganisms (fungi, bacteria and yeasts) against a limited number of pathogens, and only on specific hosts. Further studies are therefore required to identify antagonists with a broad spectrum of activity. The resistance of fruits and vegetables to postharvest diseases is closely linked to the ripening process, and drops markedly with the onset of tissue senescence: It is now possible to protect the product by inducing disease resistance. Plants produce a large number of secondary metabolites with antimicrobial effect on the main postharvest pathogens. Detailed studies have been conducted on aromatic compounds, essential oils and volatile substances. Combination of the above complementary techniques could well lead to effective control of postharvest fungal diseases.  相似文献

BACKGROUND: Serum protein analysis in both humans and experimental animal species has so far been carried out by labor-intensive techniques, such as agarose gel electrophoresis (AGE). OBJECTIVE: The objective of this study was to evaluate capillary electrophoresis (CE) as an alternative technique to AGE for the analysis of serum proteins from healthy animals. METHODS: Blood samples were collected into tubes without anticoagulant from 6 fasted healthy male mice, rats, dogs, marmosets, and humans. Serum proteins were separated by CE using a technique standardized for the analysis of human proteins, and the results (efficiency, resolution, and precision) were compared with those obtained through AGE. RESULTS: Compared with AGE, CE resulted in narrower peaks and more peaks. The efficiency of protein separation by CE was significantly higher for all species, and resolution (R) was significantly higher in samples from dogs. Using rat serum, intraday reproducibility was lower for all protein fractions, and interday reproducibility was lower for most peaks, compared with AGE. CONCLUSIONS: We conclude that CE is a viable alternative to AGE for the determination of protein electrophoresis in a routine veterinary clinical pathology laboratory. The minimal sample requirement (2 microL), complete automation, and quantitative results make CE an especially valuable technique for protein analysis in experimental animal models.  相似文献

Fish stocks vary in abundance. The causes behind the fluctuations may be difficult to determine, especially ones caused by natural fluctuations, but long‐term data series may provide indications of the mechanisms. Assessments show that the recruitment to the Norwegian spring‐spawning herring (Clupea harengus, Clupeidae) has remained low since 2004, a year which produced the last really rich year‐class. Long time‐series of estimated recruitment and mean winter temperature in the ocean showed a significant positive correlation for the period 1921–2004. Here, we show that this positive correlation did not continue from 2005 onwards as the winter temperature increased to high levels while herring recruitment decreased and has remained low. The density of zooplankton in the drift route of the herring larvae dropped significantly after 2004, and their centre of gravity shifted northwards. There may currently be heavy predation on the larvae by Atlanic mackerel (Scomber scombrus, Scombridae), and top‐down regulation is suggested to hamper successful recruitment. Our analysis indicates that the presence of food and overlap with high food concentrations are likely important regulators of survival in herring larvae. The findings may be important for future management and planning of fisheries of this stock because recruitment failure may continue if temperature remains high and food abundance remains low.  相似文献
