Grazing behavior, diet selection and weight gain of heifers (with or without social models) were assessed in an unfamiliar site containing high plant diversity. The study was performed within a tropical forest ecosystem containing a mixture of grass forb/herb, shrub and tree species. Ten inexperienced crossbred Bos taurus × Bos indicus heifers (7 ± 1 months old) were randomly assigned to one of two groups: naive or naive + social model (sm) and two experienced heifers (16 months old) were assigned to the second group. Naive heifers were bottle-nursed from days 2 to 90 after birth and began grazing at 1 month of age in tropical grass monocultures, while experienced animals had been foraging in sites containing a high diversity of plant species for 3 months prior to the study. Each group grazed in separate paddocks for a 12-week period during the rainy season, and animals were observed using focal sampling from 7:00 to 19:30 h to assess diet composition based on bite counts. Weight gain was assessed every 14 days. Paddocks contained from 1481 to 1789 kg/ha of herbaceous dry matter, enough to support the heifers throughout the study without affecting diet selection. Shrub and tree cover ranged from 55.2 to 58.4% across treatments. Bites per minute were adjusted to a log-scaled quadratic-plateau model and the curves showed no differences between treatments (P = 0.756). However, diet composition differed between groups (P < 0.001), with naive + sm heifers ingesting a greater proportion of trees (P < 0.001) and shrubs (P = 0.02), while naive heifers ingested more forb/herbs (P = 0.02); no difference in grass consumption was observed (P = 0.92). Heifers in both treatments consumed the same plant species (50 from 26 families). Over time, utilization of several plant species increased or decreased (P < 0.05), eventually leveling off for the remainder of the study. Although differences in diet composition were observed, they did not affect overall weight gain (117 and 113 g/day in the naive and naive + sm groups, respectively; P = 0.913). However, initial post-weaning weight loss was avoided in the naive + sm group. Social learning facilitates a higher use of shrubs and trees in tropical pastures containing a high diversity of plant species with different growth habits. As many plant species have high potential as forage, more effort should be placed on developing mechanisms to increase their dietary inclusion by cattle.  相似文献

Background

The Magel2 gene is most highly expressed in the suprachiasmatic nucleus of the hypothalamus, where its expression cycles in a circadian pattern comparable to that of clock-controlled genes. Mice lacking the Magel2 gene have hypothalamic dysfunction, including circadian defects that include reduced and fragmented total activity, excessive activity during the subjective day, but they have a normal circadian period. Magel2 is a member of the MAGE family of proteins that have various roles in cellular function, but the specific function of Magel2 is unknown.

Methods

We used a variety of cell-based assays to determine whether Magel2 modifies the properties of core circadian rhythm proteins.

Results

Magel2 represses the activity of the Clock:Bmal1 heterodimer in a Per2-luciferase assay. Magel2 interacts with Bmal1 and with Per2 as measured by co-immunoprecipitation in co-transfected cells, and exhibits a subcellular distribution consistent with these interactions when visualized by immunofluorescence. As well, Magel2 induces the redistribution of the subcellular localization of Clock towards the cytoplasm, in contrast to the nucleus-directed effect of Bmal1 on Clock subcellular localization.

Conclusion

Consistent with the blunted circadian rhythm observed in Magel2-null mice, these data suggest that Magel2 normally promotes negative feedback regulation of the cellular circadian cycle, through interactions with key core circadian rhythm proteins.  相似文献

Five wildPhaseolus vulgaris beans were compared with five cultivatedPhaseolus vulgaris beans in proximate composition, total (true) protein, amino acid composition, and toxic and antinutritional factors. The wild beans contained more protein (25.5% vs. 21.7%), ash (5.15 vs. 4.15%), crude fiber (7.08% vs. 5.04%) compared to cultivated beans while the former contained less fat (0.56 vs. 0.89%) and carbohydrates (61.64 vs. 68.05%). Sulfur amino acids were found to be limiting in both groups of bean as expected; however, the cultivated beans had a higher content of the limiting amino acids. Therefore, the cultivated beans showed a better amino acid profile than the wild beans. Toxic factors were not found in either type of bean; the determinations included saponins, alkaloids, and cyanogenic glycosides. The antinutritional factors investigated were hemagglutinins (lectins) and trypsin inhibitors. The wild beans presented a higher content of trypsin inhibitors (28 TUI per mg) and lectins (9.6) than the cultivated beans did (21 TUI per mg and 7 respectively). From the chemical point of view, domestication seems to be positive; however, the better protein nutritive quality of the cultivated beans should be further confirmed by biological assays.  相似文献

The chemical composition, digestibility and toxin contents of two wild legumes: Styphnolobium burseroides and Acacia bilimekii, collected in a semi-arid zone of Mexico, were determined. Both legumes had a high fiber content. The seeds of Styphnolobium burseroides had a low protein content (14%), and the pod a high content of reducing sugars. However the seeds of Acacia bilimekii had a high protein concentration (35%). The seed proteins were low in sulphur amino acids and tryptophan in both legumes but were rich in lysine. Trypsin inhibitors and lectins were present in low concentrations; alkaloids and cyanogenic glucosides were not detected. The in vitro digestibility for monogastric animals was low but the same test with ruminal juice showed a high digestibility for both legumes. Based on their chemical composition and digestibility, these legumes could be a good alternative source in the feeding of ruminants.  相似文献

BACKGROUND: In the present study the principle of energy dispersive X‐ray microanalysis (EDX), i.e. the detection of elements based on their characteristic X‐rays, was used to localise and quantify organic and inorganic pesticides on enzymatically isolated fruit cuticles. Pesticides could be discriminated from the plant surface because of their distinctive elemental composition. RESULTS: Findings confirm the close relation between net intensity (NI) and area covered by the active ingredient (AI area). Using wide and narrow concentration ranges of glyphosate and glufosinate, respectively, results showed that quantification of AI requires the selection of appropriate regression equations while considering NI, peak‐to‐background (P/B) ratio, and AI area. The use of selected internal standards (ISs) such as Ca(NO3)2 improved the accuracy of the quantification slightly but led to the formation of particular, non‐typical microstructured deposits. The suitability of SEM‐EDX as a general technique to quantify pesticides was evaluated additionally on 14 agrochemicals applied at diluted or regular concentration. CONCLUSION: Among the pesticides tested, spatial localisation and quantification of AI amount could be done for inorganic copper and sulfur as well for the organic agrochemicals glyphosate, glufosinate, bromoxynil and mancozeb. The aim of this work was to study the effects of vermicomposting on selected properties of composts derived from animal and plant wastes. For this, two different worm beds were established and fed with two composts: C1, made from goat (Capra aegagrus hircus) manure and rosemary (Rosmarinus officinalis) pruning wastes, and C2, made from rabbit (Oryctolagus cuniculus) manure and grass clippings, to study the quality of the final vermicomposts. Physical and chemical properties were determined in the end products. Vermicomposting improved several properties of the composts, increasing their total water-holding capacity (in C1, from 333 to 451 mL/L, and in C2, from 371to 419 mL/L), reducing salinity (in C1, from 6.8 to 2.4 dS/m, and in C2, from 10.3 to 4.6 dS/m), and balancing pH in the final composts obtained, especially in C1 (from 8.57 to 8.02). The type of raw material used in the worm beds significantly influenced the final characteristics of the end products obtained and the development of the process, with more favorable results being obtained with the compost derived from rabbit manure and grass clippings (C2).  相似文献
