The research was carried out in the spruce forests of Barania Góra (Silesian Beskids, Poland) affected by pandemic dying of trees. Twenty-seven samples were collected from the O layer in two plots: 17 in a cut down forest infested with insect pests (bark beetle) and ten in a 120-year-old healthy forest. The analyses covered basic parameters (pH(H2O), pH(KCl), w(org), C(tot), N(tot), CEC) and the concentrations of aluminium in the fractions leached with 0.1?M BaCl(2) (Al(exch)), 0.5?M CuCl(2) and 0.1?M Na(4)P(2)O(7) (Al(bio)) solutions. The total aluminium concentration in the soil was assayed digesting samples with hydrofluoric acid. The effect of pH and organic matter content on the amount of exchangeable (Al(exch)) and bioavailable (Al(exch)) aluminium in the soil was analysed. It has been found that the soils in both plots are strongly acidic and contain 550 to 1,700?mg?kg(-1) of exchangeable aluminium and 1,200 to 4,800?mg?kg(-1) of bioavailable aluminium. The lack of disease symptoms in the spruce trees in plot 2 can be explained by the higher content of organic matter in the soil. Unfortunately, one might expect that the high concentration of exchangeable aluminium will also cause the trees in the area to wither.  相似文献

This study aimed to evaluate the potential of soybean-promoted acidic nitrite reduction and to correlate this activity with the content of phenolics and with the bactericidal activity against Escherichia coli O157:H7. Extracts of embrionary axes and cotyledons enriched in phenolics increased ?NO formation at acidic pH at values that were 7.1 and 4.5 times higher, respectively, when compared to the reduction of the nonenriched extracts. Among the various phenolics accumulated in the soybean extracts, five stimulated nitrite reduction in the following decreasing order of potency: epicatechin gallate, chlorogenic acid, caffeic acid, galic acid and p-coumaric acid. Extracts of embrionary axes presented higher contents of epicatechin gallate and caffeic acid, compared to that of cotyledons, indicating a positive correlation between activity of the extracts and content of phenolics with regard to nitrite reducing activity. Soybean extracts enriched in phenolics interacted synergistically with acidified nitrite to prevent E. coli O157:H7 growth. The results suggest that soybean phenolics may interfere with the metabolism of ?NO in an acidic environment by accelerating the reduction of nitrite, with a potential antimicrobial effect in the stomach.  相似文献

A clear picture of the protein rearrangement in milk following UHT-treatment was drawn by a comparative analysis of the pH 4.6 soluble protein fraction (SPF) and the pH 4.6 insoluble protein fraction (IPF) recovered from raw and UHT-treated milk samples. The two protein fractions were analyzed by mono- or bidimensional gel electrophoresis under reducing and nonreducing conditions, and protein bands were identified by specific immunostaining. Results showed that bovine serum albumin, β-lactoglobulin, and, to a lesser extent, α-lactalbumin coprecipitated with caseins in UHT-treated milk samples at pH 4.6. These proteins were almost exclusively involved in high molecular weight aggregates held together by disulfide bonds. Partition of α-lactalbumin and bovine serum albumin in the protein fractions obtained upon acidification of milk at pH 4.6 was evaluated by competitive immunoassays. The ELISA-based results suggested the possibility of using pH 4.6 insoluble α-lactalbumin and bovine serum albumin, in addition to pH 4.6 insoluble β-lactoglobulin, as indicators of the intensity of the heat treatment applied to milk.  相似文献

The chemical composition of the essential oil of the Sardinian dwarf curry plant [Helichrysum italicum G. Don ssp. microphyllum (Willd) Nym] was studied. Genetic analysis suggested the presence of two chemotypes; morphological and chemical differences confirmed the presence of two chemotypes (A and B). The maximum yields were 0.18 and 0.04% (v/w) for flowering tops and stems, respectively. The concentrations of nerol and its esters (acetate and propionate), limonene, and linalool reach their highest values during the flowering stage both in flowers and in stems. Besides the essential oil, type B showed an interesting antifungal activity.  相似文献

Tomato rooting patterns, yield and fruit quality were evaluated in a field trial where three irrigation regimes [0.6 (DI), 0.9 (DII) and 1.2 ETc (DIII)] and three drip irrigation depths [surface (R0), subsurface at 20 cm depth (RI) and subsurface at 40 cm depth (RII)] were imposed following a split-plot experimental design, with four replications. The behaviour of the root system in response to the irrigation treatments was evaluated using minirhizotrons installed between two plants, near the plant row. Root-length intensity (L a)—length of the root per unit of minirhizotron surface area (cm cm−2)—was measured at four crop stages. For all sampling dates, none of the factors studied were found to influence L a or rooting depth significantly or the interaction between treatments. For all treatments most of the root system was concentrated in the top 40 cm of the soil profile, where the root-length density ranged from 0.5 cm cm−3 to 1.4 cm cm−3 . The response of tomato fruits to an increase in the water applied was similar in quantitative and qualitative terms for the different drip irrigation depths. Water applied by drip irrigation had the opposite effect on commercial yield (t ha−1) and soluble solids (°Brix) (r=−0.82, P<0.001), however, yield in terms of total soluble solids (t ha−1) was the same for the 0.9 and 1.2 ETc. The enzymatic transgalactosylation during lactulose hydrolysis was studied using the beta-galactosidase from Kluyveromyces lactis and an initial lactulose concentration of 250 g/L. During hydrolysis of lactulose, the formation of two novel trisaccharides was followed by high-performance anion-exchange chromatography with pulsed amperometric detection (HPAEC-PAD). A maximum trisaccharide yield of 14.05% was observed at 91.9% of lactulose hydrolysis. The two novel trisaccharides obtained by transglycosylation of lactulose were isolated and fully characterized by an extensive nuclear magnetic resonance (NMR) study. Complete structure elucidation and full proton and carbon assignment were carried out using 1D ( 1H, 13C, and 1D TOCSY) and 2D (gCOSY, TOCSY, ROESY, gHSQC, and gHMBC) NMR experiments. The trisaccharides were shown to be lactulose-based structures; the main one has a Gal unit linked to C-6 of the galactose moiety, and the other one has a Gal unit linked to C-1 of the fructose moiety. Transglycosylation of lactulose allows for the obtention of galacto-oligosaccharides with new glycosidic structures and would open new routes to the synthesis of prebiotics.  相似文献

The Sfh protein is encoded by self-transmissible plasmids involved in human typhoid and is closely related to the global regulator H-NS. We have found that Sfh provides a stealth function that allows the plasmids to be transmitted to new bacterial hosts with minimal effects on their fitness. Introducing the plasmid without the sfh gene imposes a mild H-NS(-) phenotype and a severe loss of fitness due to titration of the cellular pool of H-NS by the A+T-rich plasmid. This stealth strategy seems to be used widely to aid horizontal DNA transmission and has important implications for bacterial evolution.  相似文献

Despite recent reports in the literature that chitinases comprise approximately 50% of the soluble proteins present in mature Vitis vinifera L. (cv. Moscatel) grapes, with the other major proteins being thaumatin-like proteins, a careful inspection of the published data reveals inconsistencies as to which proteins actually accumulate in mature grapes. Mature Moscatel grapes were harvested in the same vineyard in two consecutive years, 1999 and 2000. The grapes exhibited widely distinct polypeptide patterns when analyzed by either FPLC cation exchange chromatography or two-dimensional electrophoresis: whrereas the 2000 grapes possessed a much higher protein content (1.96 versus 1.11 mg g(-)(1) of fresh weight), the 1999 grapes contained a greater heterogeneity of polypeptides. In addition, highly specific antibodies that recognize the pathogenesis-related proteins present in the grapes demonstrated that the 2000 harvest grapes had a wider variety of pathogenesis-related polypeptides. N-Terminal sequencing of the major polypeptides revealed differences in the relative abundance and amino acid sequence of thaumatin-like and osmotin-like polypeptides present in the 1999 and 2000 harvest Moscatel grapes and did not detect the presence of chitinase. As a whole, the data indicate that the expression and accumulation of the major proteins in grapes is essentially determined by the environmental and/or pathological conditions prevailing during grape development and maturation. The potential physiological and technological implications are discussed. The results of the present work suggest that it is not possible to base varietal differentiation of grapes on the profile of the pathogenesis-related polypeptides present in the mature berries.  相似文献
