Foot-and-mouth disease (FMD) is a highly contagious aphthoviral infection of cloven-hoofed animals, inducing vesiculopustular stomatitis, pododermatitis, and thelitis. Vesicular fluid represents a major pathway of virus excretion, but bovine milk is another important source of virus shedding. We describe here the time course of FMD virus (FMDV) excretion in the milk and characterize associated lesions in the mammary gland. Three dairy cows were infected by nasopharyngeal instillation of FMDV and monitored over 12 d. Autopsy was performed at the end of the study, and specimens were collected for histopathology, IHC, and RT-qPCR. All 3 cows developed fever, drooling, vesiculopustular stomatitis, interdigital dermatitis, and thelitis. FMDV RNA was detectable in whole milk until the end of the trial, but only transiently in saliva, nasal secretions, and blood serum. Although histology confirmed vesiculopustular lesions in the oral and epidermal specimens, the mammary glands did not have unequivocal evidence of FMDV-induced inflammation. FMDV antigen was detectable in skin and oral mucosa, but not in the mammary gland, and FMDV RNA was detectable in 9 of 29 samples of squamous epithelia but only in 1 of 12 samples of mammary gland.  相似文献

Habitat fragmentation is considered a major cause of biodiversity loss, both on terrestrial and marine environments. Understanding the effects of habitat fragmentation on the structure and dynamics of natural communities is extremely important to support management actions for biodiversity conservation. However, the effects of habitat fragmentation on marine communities are still poorly understood. Here we evaluated whether habitat fragmentation affects the structure of epifaunal communities in the sublittoral zone, in the northern coast of São Paulo state, Brazil. Five experimental landscapes were constructed, each one forming a large continuous patch. After 4 weeks, each landscape was cut on three patches of different sizes. Epifaunal macroinvertebrate communities were sampled at the edge and interior of experimental landscapes before manipulation to evaluate edge effects. After four more weeks, communities from the three patch sizes were also sampled to evaluate patch size effects. We compared the diversity of communities at different levels of fragmentation by total abundance, rarefied taxon richness, Shannon–Wiener diversity index, Simpson’s dominance index, and abundance of dominant taxa. Higher taxon richness and gastropod abundance were recorded in the patch edges, but no significant differences were found among patch sizes. We found a significant effect of habitat fragmentation, with lower abundances of Gammaridea (the dominant taxon), Ophyuroidea, and Pycnogonida after the experimental fragmentation. Lower abundances of dominant taxa resulted in higher diversity and lower dominance in fragmented landscapes when compared to integral, pre-manipulation landscapes. Our results suggest that fragmentation of landscapes in the system studied can reduce dominance, and that even small patch sizes can be important for the conservation of macroinvertebrate diversity.  相似文献

Fusarium wilt, caused by Fusarium oxysporum, is a major disease of jojoba, causing serious economic losses. This study was aimed at characterizing the Fusarium populations associated with jojoba in Israel. Fifty Fusarium isolates used in this study included 23 isolates from the 1990s (“past”) and 27 recently isolated (“recent”). All the isolates were characterized by arbitrarily primed (ap)-PCR and 16 representatives were additionally delineated using multilocus (tef1, rpb1, rpb2) phylogeny and evaluated for their pathogenic potential. Consequently, 88% of the isolates were identified and characterized to the F. oxysporum species complex. The remaining 12% grouped within the F. fujikuroi, F. solani, and F. redolens species complexes. Variations in the infection rate (16.7%–100%), disease symptoms (0.08–1.25, on a scale of 0–3), and fungal colonization index (0.67–2.17, on a scale of 0–4) were observed within the tested isolates, with no significant differences between the past and recent isolates. The representative isolates were assigned to 11 groups based on ap-PCR. Pathogenicity tests showed that isolates from Groups II, IV, and V were the most aggressive, whereas isolates from Groups III, VIII, and IX were the least aggressive. Among the tested isolates, F. oxysporum sensu lato was the most aggressive, followed by F. proliferatum, while F. nygamai was the least aggressive. This study demonstrates the complexity and genetic diversity of Fusarium wilt on jojoba in Israel, indicating possible multiple introductions of infected germplasm into the country.  相似文献

Conspecific negative density dependence in tree species can have major consequences for structuring of communities, yet in temperate forests this phenomenon remains largely unexplored. We investigated density-dependent recruitment in the long-lived conifer yew (Taxus baccata L.), for which low levels of regeneration have been linked with reduced light availability beneath conspecific canopies and speculative autotoxic effects. We combined in situ assessment of yew regeneration in a temperate forest in southwest Ireland with ex situ pot experiments to assess whether light and/or beneath conspecific canopy conditions inhibit the germination and early growth of yew. In field experiments, recruitment was related to distance from adult conspecifics, with higher levels of regeneration found in areas not beneath yew canopies. However, reduced light availability beneath the dense yew canopy was not responsible for this pattern, with abundant seedlings found in areas of equivalent or even lower light availability. Pot experiments showed that seedling survival was highest in deep shade treatments. Experiments also demonstrated that the addition of yew needles negatively affected seedling growth, but not seed germination. Together, our results suggest that under natural forest conditions, the absence of regeneration beneath mature yew canopies, often attributed to low light availability, may at least be partly related to substances in yew canopy or leaf leachates.  相似文献

The control and/or removal of a dominant invasive species is expected to lead to increases in native species richness and diversity. Small pilot studies were performed on Santa Cruz Island (SCI), California, in the early 1990s to test the efficacy of different methods on the control of Foeniculum vulgare (fennel) and management’s effects on native species recovery. We chose a treatment that showed significant native species recovery, applied it at the landscape scale, and followed its effects on fennel infested plant communities. We tested the hypothesis that results from small-scale studies translate to the landscape level. We found that although the control of fennel translated from the small to landscape scale, decreasing from an average of 60% to less than 3% cover, native species recovery did not occur in the landscape study as it did in the pilot studies. Invasive fennel cover was replaced by non-native grass cover over time. Unexpectedly, fennel cover in untreated fennel plots decreased significantly (though not as drastically) from over 60% cover to just under 40% cover while native species richness in untreated areas increased significantly. The correlation between precipitation and changes in native and non-native species richness and abundance in this study imply that changes in species abundances were highly correlated with environmental fluctuations. The lack of a native seedbank and the accumulation of non-native grass litter likely prevented the recovery of native species in treated areas. Greater vertical complexity found in fennel communities, which increased visitation by frugivorous birds and likely increased native seed dispersal, may have been responsible for the increase in native species richness in the untreated areas. These results suggest that successful invasive species control and native species recovery experiments conducted at small scales may not translate to the landscape level, and active restoration should be an organic component of such large-scale projects.  相似文献

FeEDDHA (iron(3+) ethylenediamine-N,N'-bis(hydroxyphenylacetic acid) products are commonly applied to mend and prevent Fe deficiency chlorosis in soil-grown crops. Plants mainly take up Fe in the progressed vegetative and in the reproductive stages. This study examined which of the principal constituents of FeEDDHA products (the isomers racemic o,o-FeEDDHA, meso o,o-FeEDDHA, and o,p-FeEDDHA), most effectively meets the Fe requirements of soybean plants (Glycine max (L.) Merr.) grown on calcareous soil in the aforementioned growth stages. FeEDDHA isomers were applied once, separately or in mixtures, at t = 0, in the progressed vegetative stage or in the reproductive stage. o,p-FeEDDHA did not significantly contribute to Fe uptake in either growth stage. Both racemic and meso o,o-FeEDDHA were effective in supplying plants with Fe, approximately to the same extent. The moment of application had a significant effect on yield and FeEDDHA pore water concentrations at harvest, but not on Fe uptake. To optimize yield while minimizing FeEDDHA dosage, FeEDDHA is best applied to soybean plants prior to the onset of chorosis.  相似文献

Beyond its traditional function of food production, agricultural land offers public amenities such as the protection of natural resources and landscape scenery. This study investigates the preferences of non-farmers and farmers for nine landscape scenarios in the Swiss lowlands. The nine landscapes were the result of a photo editing process combining three land-use types (arable crops, grassland and a mixture of both) and three proportions of ecological compensation areas (0%, 10% and 30%). The landscape photographs were randomly arranged on one page of a paper-based questionnaire which was sent to a random sample of 4000 Swiss households (non-farmers) and 500 farmers. The respondents (1376 non-farmers and 276 farmers) rated each landscape by attractiveness. Both non-farmers and farmers preferred a mixed land-use type or one dominated by arable crops over one dominated by grassland. Non-farmers’ preference ratings were highly influenced by the proportion of ecological compensation areas (ECAs) in the rated landscape: Non-farmers rated a landscape with a mixed land-use type and 30% ECAs highest, whereas farmers rated a landscape dominated by arable crops and 10% ECAs highest. The results indicate that heterogeneous landscapes (mixed land use, high proportion of ECAs) influence scenic beauty positively. Thus, farming practices and agro-environment schemes such as ECAs can have an impact on the visual attractiveness of a landscape.  相似文献

This study presents concentrations of perfluorinated compounds in food and the dietary intake of perfluorooctane sulfonate (PFOS) and perfluorooctanoate (PFOA) in The Netherlands. The concentrations of perfluorinated compounds in food were analyzed in pooled samples of foodstuffs randomly purchased in several Dutch retail store chains with nation-wide coverage. The concentrations analyzed for PFOS and PFOA were used to assess the exposure to these compounds in The Netherlands. As concentrations in drinking water in The Netherlands were missing for these compounds, conservative default concentrations of 7 pg/g for PFOS and 9 pg/g for PFOA, as reported by European Food Safety Authority, were used in the exposure assessment. In food, 6 out of 14 analyzed perfluorinated compounds could be quantified in the majority of the food categories (perfluoroheptanoic acid (PFHpA), PFOA, perfluorononanoic acid (PFNA), perfluorodecanoic acid (PFDA), perfluoro-1-hexanesulfonate (PFHxS), and PFOS). The highest concentration of the sum of these six compounds was found in crustaceans (825 pg/g product, PFOS: 582 pg/g product) and in lean fish (481 pg/g product, PFOS: 308 pg/g product). Lower concentrations were found in beef, fatty fish, flour, butter, eggs, and cheese (concentrations between 20 and 100 pg/g product; PFOS, 29-82 pg/g product) and milk, pork, bakery products, chicken, vegetable, and industrial oils (concentration lower than 10 pg/g product; PFOS not detected). The median long-term intake for PFOS was 0.3 ng/kg bw/day and for PFOA 0.2 ng/kg bw/day. The corresponding high level intakes (99th percentile) were 0.6 and 0.5 ng/kg bw/day, respectively. These intakes were well below the tolerable daily intake values of both compounds (PFOS, 150 ng/kg bw/day; PFOA, 1500 ng/kg bw/day). The intake calculations quantified the contribution of drinking water to the PFOS and PFOA intake in The Netherlands. Important contributors of PFOA intake were vegetables/fruit and flour. Milk, beef, and lean fish were important contributors of PFOS intake.  相似文献
